PURIFICATION AND IDENTIFICATION OF MANNITOL PRODUCING FROM THE ISOLATION OF LOCALLY ISOLATED BACTERIA LEUCONOSTOC MESENTEROIDES

Abstract

Study included the use of locally isolated bacteria Leuconostoc mesenteroides in the production of mannitol from production media contains 10g Trypton, 10g yeast extract, 10g dipotassium phosphate, 75g glucose, 150g fructose, 10ml solution of vitamins and minerals, dissolved in the liter of distilled water. The conditions of the primary grow was pH 6.5 incubation at 25 ْC for 48hrs and the inoculum was 1%. The production was done by batch fermentation. The amount of output mannitol 11g/100ml from the media of production. The produced mannitol was identified by paper chromotagraphy which revealed the Rf value of standard mannitol. The produced mannitol from standard medium amounting to 0.55, highly similar the infra red spectra analysis (IR) for standard mannitol and the produced one were all similar to that of the Aldrich library of Infra red spectra of mannitol .
