الخلاصة
درس تأثير تراكيز مختلفة من منظمات النمو في إستحداث الكالس من البذور الناضجة لخمسة أصناف من
الرز العراقي هي عنبر المناذرة وعنبر بغداد وعنبر 33 وبسمتي العراق والعباسية، وكان التركيز 3 ملغم/ لتر
قد أعطى أفضل MS و 300 ملغم/ لتر كازئين متحلل مائياً المضاف إلى وسط BA 2,4- و 0.2 ملغم/ لتر D
بالتراكيز 0.25 و 0.5 و 0.75 و 1 و 1.25 و 1.5 NaCl إستجابة. كما درس تأثير ملح كلوريد الصوديوم
% في نمو الكالس وإخلاف النباتات منه. بينت النتائج وجود إختلافات بين التراكيب الوراثية في إستجابتها
لتكوين الكالس من البذور الناضجة وكان صنف العباسية أقل الأصناف إستجابة ولم تختلف باقي الأصناف
معنوياً فيما بينها فضلاً عن كونه الأكثر تأثراً بالملح بالنسبة إلى وزن الكالس الطري إذ إنخفض معدل الوزن
فيه بعد إضافة أقل تركيز للملح وهو 0.25 % يليه صنفا المناذرة وعنبر 33 الّلذان تأثر الوزن فيهما عند
MS التركيز 0.5 %. لم يتم الحصول على نبيتات من كالس الرز ضمن التراكيز المستعملة وتفوق وسط
في إعطاء الجذور فقط والتي تباينت نسبها بين الأصناف K و NAA المحتوي على 2 ملغم/ لتر لكل من
كذلك إذ تفوق صنف البسمتي بإستجابته التي بلغت 100 % وكانت أقل إستجابة للصنفين عنبر 33 والعباسية
.% إذ بلغت 28.5

Abstract

Study included the use of locally isolated bacteria Leuconostoc mesenteroides in the production of mannitol from production media contains 10g Trypton, 10g yeast extract, 10g dipotassium phosphate, 75g glucose, 150g fructose, 10ml solution of vitamins and minerals, dissolved in the liter of distilled water. The conditions of the primary grow was pH 6.5 incubation at 25 ْC for 48hrs and the inoculum was 1%. The production was done by batch fermentation. The amount of output mannitol 11g/100ml from the media of production. The produced mannitol was identified by paper chromotagraphy which revealed the Rf value of standard mannitol. The produced mannitol from standard medium amounting to 0.55, highly similar the infra red spectra analysis (IR) for standard mannitol and the produced one were all similar to that of the Aldrich library of Infra red spectra of mannitol .

Abstract

Twenty seven bacterial local isolates of Bacillus sp. were obtained from soil samples. The isolates were tested for agarase production on solid media; fourteen isolates were able to develop clear zone around the bacterial growth after floating the plates with iodine reagent (Lugol´s solution), with clear zone diameter between (1.08-1.11cm). There were further treated in submerged culture which led to selection of Bacillus sp. H12, since it was the most efficient that gave higher specific activity of agarase 11.73 unit/mg protein. The optimum conditions for agarase production using submerged cultures were 0.3% agarose as carbon source, 0.5% beef extract and peptone as nitrogen sources, 0.5% sodium chloride, at initial pH 8 and inoculum size 1×108cell/ml at 37˚C for 24 hrs using shaker incubator at 150 rpm.

Abstract

This study was conducted to detect the toxic and mutagenic and antimutagenic effects of the alcoholic plant extract Salvia officinalis. The optimal concentration of alcoholic extract was 150micrograms/ml, and compared its effectiveness against Methotrexate (MTX) as a mutagenic chemical, and ultraviolet radiation (UV) as a mutagen and using physical test system (in vitro) individually for each nested transactions of the extract with MTX and UV before, within, and after treatment using bacterial system (G-system) depending on plant's survival frection (Sx) to study the effects of mutation and induced resistance to the antibiotic streptomycin and rifampicin as genetic markers. The results showed the effect of interference between the optimal concentration of the extract and mutagen on the survival factor (Sx), increasing of the values of the survival factor of the system isolates to normal values compared to positive control (MTX or UV only), and also showed the results of the overlap between the optimal concentration of the extract and treatment with MTX in induction resistance mutations for streptomycin and rifampicin. MTX had no effect in induction resistance to two anti-biotics before, within, and after treatment with MTX for the three isolates and thus the alcoholic extract was suppress or repair mutations and provided 100% protection of bacterial cells. While the results of the effect of interference between the optimal concentration of the extract and UV treatment showed no any effect in induction resistance to rifampicin for all treatment and all the three isolates. The same as streptomycin, this indicate that treatment of bacterial isolates with plant alcoholic extract with and after exposure to altraridet radiation, while the percentage of mutations repair and protection before UV exposure was 93.6 - 98.3% .

Abstract

This study was conducted in the Institute of Genetic Engineering and Biotechnology for Postgraduate Studies, University of Baghdad from August 2007 to March 2008, it is an attempt to determine ethnic distribution of IVSInt.6 mutation associated with β-thalassaemia in Iraqi population. Ninety EDTA blood samples of clinically thalassaemic patients including Arab, Turkman and Kurd were collected from two thalassaemic center in Iraq. Also blood sample from 30 apparently healthy individuals were collected as a control group. The DNA samples were subjected for molecular detection of IVSInt.6 β-thalassaemia mutation by Amplification Refractory Mutation System (ARMS) based on Polymerase Chain Reaction (PCR). It was found that the ethnic specificity of the studied mutation were reported for the first time within Iraqi population. The ethological distribution of the positive diagnosed mutation showed that the studied mutation was detected only in 10(33.33%) Turkman patients, while it was not detected in Arab and Kurds.

Abstract

Here we describe the lipofecation of the human larynx carcinoma cell line (Hep-2) using TransFastTm reagent as a liposome after its mixing with pUC18 plasmid. Optimization of the charge ratio for liposome/DNA was carried out. The DNA was extracted and PCR analysis for the presence of the plasmid is carried out using two specific primers for pUC18. Result indicates that lipofecation with TransFastTm reagent is successful for Hep-2, the optimum charge ratio of transfection reagent to DNA was 1:1, and the optimum DNA concentration was 0.75-1ng.

Abstract

Swabs were taken from 20 patients from root canal (Endodontic Department, Al-Elwiya Specialized Centre for Oral and Dental Surgery), Streptococcus oralis, Streptococcus anginosus, Enterococcus durans, Enterobacter cloacae and Staphylococcus epidermides were identified according to their cultural properties, microscopic examination and biochemical Api and miniapi tests. Antibacterial action was evaluated on these bacterial isolates by using six different concentrations of Salvadora persica(Siwak), Nigella sativa (black seed), aluminum potassium sulphate (alum) and mixture of them, the concentrations are 3.125, 6.25, 12.5, 25, 50 and 100%. At 100% concentration of alum, Salvadora persica, Nigella sativa and the mixture of them, the maximum inhibition zone diameters were 42mm on Enterococcus durans, 27mm on Enterobacter cloacae, 26mm on Enterobacter cloacae and 25mm on Staphylococcus epidermides, respectively.

Abstract

A new formulation of Sn-2,3meso dimercapto succinic acid“Sn-DMSA" kit to be labeled with penta valent technisum-99m. The DMSA kit was prepared by simple and fast procedure. It was prepared by reaction of kit that is contained (1.2mg 2,3 meso dimercapto succinic acid, 0.4mg stannous chloride dihydrate, 50mg of Inositol and 1.0mg of ascorbic acid as anti oxidant ) with Na99mTcO4 at pH equal to 8.0 which is the optimal condition of the formation of 99mTc (v) DMSA complex the radiochemical yield and the stability of kit in liquid and lyophilized forms were determined by using gel filtration column scanning "GCS". The radiochemical yield of complex formation was greater than 99% after 15min. from addition of Na 99mTcO4. The complex in liquid form was stable during six hours while the lyophilized form for six months. Biodistribution of 99mTc(v)DMSA in New Zealand white rabbits demonstrated greater uptake in bone marrow and liver 50% and 40% while lower uptake in liver 6.2% during one an hour after injection respectively, however a negligible amount of radioactivity was detected in the non target organs. The study suggests that this kit was an excellent agent for skeleton and bones scintigraphy which was used in diagnosis and scanning of metastases.

Abstract

The objective of this project was to establish the molecular genetics of β-thalassemi in the Iraq. Genomic DNA was isolated from the blood of 75 patients who were diagnosed clinically and hematologically as β-thalassemia major and from 20 blood samples of apparently healthy individuals as control, all these samples was collected from Baghdad thalassemic center at Ibn-Al-Baladi pediatrics hospital. Polymerase chain reaction (PCR) was used for amplification of 4 regions in β-globin gene. The data show that IVS-1 nt 110 mutations are the most common in the population. Codon 39 mutation was also found with relatively high incidence. One of the subjects had a compound mutation which included IVS-1 nt 110 and CD8 mutations. Eighteen of the β-thalassemia major patients showed none of the four common mutations.
