Influence of succharides on secondary metabolites production from leaves induced callus of Salvia officinalis in vitro

Abstract

An experiment on the effect of sucrose , glucose, fructose, sorbitol and mannitol on leave induced callus of Salvia officinalis was conducted from April/2008 to October /2009. Succharides concentrations added to MS medium were 20 ,40 ,60 ,80 g/l. Succharides concentration were added to MS medium separately. Callus induction was achieved by culturing true leaves of the plant on MS medium supplemented with 1.0 mg/l of 2,4-D and 0.2 mg/l of BA. Result could be summarized as follows:Adding 20 g/l of sucrose to the medium was more effect on Thujone and Borneol production, the values were 137.2 and 151.2 µg/g . dry weight of callus respectively. While 40g/l was superior on 1-8Cineol production the value was 109.9 µg/g. The highest level of sucrose (80g/l) increased Camphor production. Lowest level of glucose (20g/l) enhanced Thujone , Camphor , Borneol and 1-8Cineol amount. Highest values of Thujone and 1-8Cineol were achieved from the explant cultured on MS medium supplemented with 20g/l of fructose ; they were 25.4 and 14.9 µg/g . However , 60 g/l was the best on Camphor and Borneol production. Adding 40g/l of sorbitol was more effective on Thujone , Camphor and 1-8Cineol production , while 80 g/l enhanced Borneol production ; the value was 189.3 µg/g . callus dry weight. Sorbitol at 40g/l increased Thujone , Borneol and 1-8Cineol levels on callus. Mannitol at 20g/l increased Camphor level ; the value was 132.0 µg/g.
