Omar مصطفى محمد عمر}, journal={Iraqi Journal of Agricultural Sciences مجلة العلوم الزراعية العراقية}, volume={48}, number={6}, pages={1493-1504}, year={2017}, abstract={Twenty-five bacteria isolates (Bacillus sp.) were isolated from different local sources (soil of college of Agriculture/university of Baghdad and from Amiriya); the obtained isolates were screened for their capabilities for producing alkaline proteases. It has been noticed that isolates designated (2,5,6,8,11,12 at 37°C) and (14,17 and 18 at 55°C) were showed high capability for producing alkaline protease. All isolates subjected to secondary screening. The results revealed that isolate (12) coded MH12 was the highest producers with enzyme activity of (81.00U/ml). The Identification tests of this isolate were carried out by studying morphological, microscopic characteristic and using of Vitek2 compact system. These tests were conformed by identification of 16S rRNA gene using PCR and its nitrogen base sequencing and the results revealed that the isolate was belongs to Bacillus cereus.

أمكن الحصول على 25 عزلة من البكتريا المنتجة لأنزيم Alkaline Protease من أصل 50 عزلة والتي تعود إلى جنس Bacillus من نماذج من ترب مختلفة أخذت من حقول كلية الزراعة/جامعة بغداد ومن منطقة العامرية في بغداد وأخضعت هذه العزلات لعملية الغربلة الأولية إذ تميزت العزلة 2 و5 و6 و8 و11 و12 المعزولة بدرجة 37 مْ والعزلات 14 و17 و18 المعزولة بدرجة 55 مْ بإنتاجيتها العالية للأنزيم بدلالة أقطار الهالة المتكونه حول المستعمرات البكتيرية لهذه العزلات، ثم أخضعت تلك العزلات لغربلة ثانوية فتبين أن العزلة 12 والتي رمز لها MH12 هي أكفأ تلك العزلات إذ بلغت الفعالية الأنزيمية 81.00 وحدة/مل. تم تشخيص العزلة من خلال دراسة الصفات المظهرية المزرعية والمجهرية والتشخيص بجهاز Vitek2 compact system كما تم تأكيد التشخيض على المستوى الجزيئي لتلك العزلة بالتحري عن جين16S rRNA بإستخدام تقنية PCR ودراسة تتابعات القواعد النتروجينية. وأظهرت النتائج أن العزلة تعود إلى بكتريا Bacillus cereus.} }
