Al-Nasiri PY - 2012 VL - 11 IS - 2 SP - 481 EP - 493 JO - Iraqi Journal of Biotechnology المجلة العراقية للتقانات الحياتية SN - 18154794 25207245 AB - Four local isolates of Klebsiella pneumoniae subsp. pneumoniae from different sources were tested towards twenty six antibiotic disks. The results showed that all selected isolates were sensitive to nalidixic acid, amikacin, and ciprofloxacin and the isolate MK6 showed further sensitivity to tetracycline, kanamycin, and streptomycin; but all isolates showed resistance toward the rest of antibiotics used. The minimal inhibitory concentration (MIC) test of four selected Klebsiella pneumoniae isolates (MK1, MK2, MK6 and MK20) to nine antibiotics showed the highest MIC values for the selected isolates were for amoxicillin and chloramphenicol (≥ 16384 µg/ml), while the lowest MIC was for streptomycin as it ranged from (8 – 16 µg/ml). Agarose gel electrophoresis was performed to detect plasmid profile for the four selected isolates. The results showed that all isolates exhibited two bands of small plasmid DNA with molecular size ranged from less than 4kb for the smaller one to more than 12kb for the larger one as compared with DNA of molecular size marker )lambda phage restricted with EcoRІ and HindIII). Besides two bands of large plasmid DNA were obtained from the isolates MK20, MK1, and MK6, but the isolate MK2 didn’t show any band of large plasmid DNA. This may be due to a technical error or to the low DNA concentration during extraction process In order to determine whether the plasmids of selected isolates play a role in antibiotic sensitivity, plasmid curing experiment was performed by using physical agent (elevated temperature). The results indicated that high temperature (47°C) was efficient to cure all the plasmids, and the isolates MK20, MK1, and MK2 became sensitive to streptomycin, kanamycin, and tetracycline.

أختبرت الحساسية الدوائية لأربع عزلات من بكترياKlebsiella pneumoniae subsp. pneumoniae تجاه 26 مضاد حيوي. بينت النتائج بأن جميع العزلات المنتخبة كانت حساسة لحامض النالدكسك والاميكاسين والسبروفلوكساسين هذا وقد أظهرت العزلة MK6 حساسية إضافية للمضادات تتراسايكلين وكانامايسن وستربتومايسين، غير إن جميع العزلات كانت مقاومة لبقية المضادات الحيوية المستخدمة. كما أجري أختبار التركيز المثبط ألأدنى (MIC) لعزلات Klebsiella pneumoniae المنتخبة MK20,MK1,MK2, MK6 تجاه تسعة مضادات حيوية وبعدد خلايا حوالي 105 خلية/مليلتر إذ لوحظ بأن أعلى قيمة لهذا التركيز كان لمضادي ألأموكسسلين و الكلورامفينكول إذ سجل كل منهما ≥16384) مايكروغرام/مليلتر)، في حين اقل تركيز مثبط أدنى كان لمضاد الستربتومايسين إذ سجـل (8-16 مايكروغرام/مليلتر). تم التحري عن دنا المحتوى البلازميدي للعزلات المنتخبة بإستعمال طريقة Miniprep. مع إستعمال دنا العاثي lambda مقطع بكل من إنزيمي التقييد ІEcoR و HindIII كدليل حجمي. وقد دلت النتائج على إحتواء جميع العزلات على دنا حـزمتين بلازميديتين صغيرتين إذ قدرت الصغيرة منها بأقل من (4 كيلو قاعدة) والكبيرة بأكثر من (12 كيلو قاعدة)، فضلاًعن إحتواء العزلات MK20,MK1,MK6 على دنا حزمتين بلازميديتين كبيرتين، بينما لم ُتظهِـِر العزلة MK2 أي حزمة بلازميدية كبيرة، وقد يعود ذلك أما الى حصول خطأ تجريبي أو إنخفاض تركيز الدنا اثناء عملية الإستخلاص. ولغرض معرفة مدى الدور الذي تلعبه البلازميدات للعزلات المنتخبة تجاه الحساسية للمضادات الحيوية، تم إجراء تجربة تحييد البلازميدات من العزلات المنتخبة بإستعمال للعوامل الفيزياوية (الحرارة المرتفعة). أظهرت النتائج بأن إستعمال درجـة (47 ºم) كان له تأثير كبير في إزالة البلازميدات من جميع العزلات بألأضافة الى أن العزلات MK20,MK1,MK2 قد أصبحت حساسة للمضادات الستربتومايسين، و الكانامايسين، والتتراسايكلين.
