Inducing of callus production and anticancer vinblastine and vincristine from Catharanthus roseus in vitro using Trichoderma harzianum and Fusarium oxysporum fungi in different techniques .

Abstract

Experiment was carried out in 2014/2015 in Labs. Of agriculture collage to study the effect of Trichoderma harzianum and Fusarium oxysporum on callus inducing and production of anticancer vinblastine and vincristine in vitro from leaves of Catharanthus roseus. Callus was induced using different combinations of plant growth regulators 1+1 ,2+2, 3+3 mg/L 2,4-D and BA ,the fungi were added to the tissue culture medium /MS by two ways first direct on callus till covering and second way as extract with MS medium in percent 10% volume/volume . Results showed that the best combination was 1+1 mg/L 2,4-D and BA with significant differents compared with all treatments. Fungi gave increase in quantities of bioactive compounds and the high value was for F. oxysporum as direct inoculation to callus 35.750 ug/g vincristine and 40.620 ug/g vinblastine followed by fungi extract treatment which gave 32.413 ug/g vincristine and 34.512 ug/g for bioactive compound vinblastine with significant different compared with control treatment which gave 1.870 ug/g vincristine and 2.120 ug/g vinblastine ( Plant leaves extract) .
