Isolation and Identifying of Bacillus subtilis bacteria and efficiency test in phosphate soluble in broth culture

Abstract

AbstractThis study was conducted to isolate and identify Bacillus subtilis from different rhizosphere soil in Almuthna governorate ( Tomato ,wheat, Corn, Barley, Gumbo, Berseem) and test its efficiency to solubilize phosphate in broth media.Twenty five rhizosphere soil samples were collected subjected to serial dilution and plated on Nutrient agar medium.Microscopical and biochemical characterties were used to diagnosis these bacterial isolates.Results showed that ten isolates were belong in its characterties to B. subtilis ,The results showed also that there ten isolates differ in its ability to solubilize tricalcium phosphate and droping the P it of the medium.Spectro photpmeter was used to quantify the amount of solubilize phosphate in the broth medium at 430 nm . Results showed that isolates which have local number B11 & B6 was superior in its efficiency to solubilize phosphate (.3403,31.9 mg.L-1 ) respectively where as isolates number B22 & B17 gave the lowest amount of solubilized phosphate (20.63 , 23.09 mgP.L-1) respectively .In addatio medium pH drop to 6.5 in medium inoculated with B11 isolate compared with 7.0 in control while reach to 6.54 with B6 isolate with strong relationship with the increasing of solubilized phosphate.
