تأثير الاكسدة الضوئية وبعض المثبطات في فعالية انزيم البروتيز المنقى من بذور الماش Phaseolus aureus Roxb.

Abstract

The aim of study included the effect of photoxidation and some of chloride and chemical parameter on enzyme protease activity that purified from Phaseolus aureus Roxb. Seeds. The results of the effect of photooxidation on protease enzyme activity showed that there was decline in the activity when exposed to the light in the presence of mehylene-blue in the absence of the substrate; the remaining activity was 25%. While the enzyme remaining 75% of the activity when exposed to the light in the present of substrate.The effects of some chemicals on protease activity proved that the enzyme retained its original activity when it was incubated at 5, 10 mM of NaCl. The results showed that using ZnCl2, CoCl2 at concentrations of 5, 10 mM the remaining activities were 54%, 35% at 5 concentration respectively and 66%, 40% at 10 concentration respectively.The incubation of the enzyme with chelating agent EDTA 5, 10 mM has an inhibitory effect on enzyme activity indicating that the enzyme is metalloenzyme. The results showed no effect of PMSF on protease activity indicating that the enzyme is not a serine protease and this results showed the activity was decline in use of 2-ME.
