This study was conducted to investigate the role of seed extracts of Loranthus europeus Linn. and Glycine max (L.) Merrill and leaf extract of Cassia senna L. and their mixtures in the inhibition of genotoxic effect of 1,2-dimethylhydrazine in mice depended on recording some cytogenetic markers including mitotic index, chromosomal aberration assay and micronucleus test in the bone marrow of the mice. These plants extract showed anti-mutagenic activity against the mutagen DMH by increasing the mitotic index and decreasing chromosomal aberration and micronucleus in the bone marrow.

The aim of study was to record the effect of radiation and pollution on Iraqi sheep fertility, semen physical characteristics and lymphocyte cell chromosomes changes. Blood was collected from sixty sheep, of them 30 grazing in Al-Tuwaitha area, for lymphocytes cultivation, cytogenetics to do analysis and chromosomal examination by using BudR for DNA molecule. The results indicated that there was chromosomal changes in the animals were exposed to the pollution represented in reduce in Blasto index 22.75±0.5, mitotic index 0.954±0.07 and cell cycle progression, while there was a high significant in sister chromatid exchange(SCE) 7.81±0.30. Semen were collected from ten rams divided into two equal groups, control group were obtained from Veterinary Medicine and Al-Shuala, the second group were obtained from Al-Tuwaitha in Baghdad. The results showed significant differences at P<0.05 level in dead and abnormal sperms percentages between two groups. Also, there was a decrease in fertility and number of birth in 24 ewes which was naturally inseminated from rams grazing in Al-Tuwaitha area, comparing with 24 ewes inseminated from normal rams. The study concluded chromosomal changes in chromatied exchanges with decrease in lymphocyte division. The testes functions was effected when abnormal chromosomes sperms appeared in semen which indicated the genetics changes in sperm chromosomes.

This study includes a physio-chemical and spectral characterization for a biological active compounds in the ANAB Al-Theab plant (Solanum nigrum L.). Three different extracts from dry leaves and fruits (a,b and c) were used to realize this purpose by using different techniques (pre–TLC, m. p., UV–Visible and IR). The obtained results indicated that a major content of those extracts was total crude alkaloid. Whereas, when using (prep–TLC) technique, the value of the Rf of the dispersed spots of extracts was 0.83–0.93 which give positive test with Dragendroff reagent, and the measurements of the melting point (m. p.) of the extracts were 200, 210 and 279°C that closely related to the alkaloid compound ranges. But when Uv–Visible spectroscopy was applied, the value of λ –max was 1.410–2175 measured by spectrophotometer in a wavelength 227.6– 232.0nm. It is known that the absorption of the alkaloid compounds was within the 200–245nm wavelengths. Finally, the data obtained from th IR spectroscopy of the samples, were determined between 600–1360cm which includes the absorption of OH, CH, CO and CN groups, considering as the finger print. These results confirm being the Solanum nigrum plants as indicator for the alkaloids like substances.

In this study, the geographical and demographical distribution of eighty clinically thalassaemic patients and fifty six apparently healthy individuals as a control group were studied. The geographical distribution of the samples revealed that the collected samples were from fifteen governorates which represent various regions in Iraq. The distribution of the samples according to some demographical parameters including: gender, age, and parent relationship was studied to reveal the effects of these parameters on the diversity of β-thalassaemia in both patient and control groups. Also, after molecular diagnosis of causative β-thalassaemia mutations using Amplification Refractory Mutation System (ARMS), the geographical distribution of positive diagnosed patients was studied in order to learn about the distribution of β-thalassaemia mutations, the results revealed the heterogeneity of diagnosed mutations within the Iraqi regions.

EXTRACTION AND PURIFICATION OF RESVERATROL FROM GRAPE SKIN FRUIT

This study was conducted with the aim to extract and purify a polyphenolic compound “Resveratrol” from the skin of black grapes Vitis vinifera cultivated in Iraq. The partial purified resveratrol is obtained after chromatography on silica gel G60 column. The preperative thin layer chromatography elution yields pure crystals identified as resveratrol (mixture of the two isomers cis and trans ) in relation to resveratrol standard trans- resveratrol (35 mg resveratrol crystals/0.5kg fresh grape skin obtained as a result of these processes). Chemical investigations and tests for identification and qualification of the extracted partial and purified crystals involved: general tests for polyphenoles, aromatic unsaturated compounds, spectrophotometric scanning for λmax screening, High Performance Liquid Chromatography (HPLC) analysis, Fourier Transform Infra-Red (FTIR) assay and melting point estimation in relation to resveratrol standard for the all applied tests.

The activity of the local diatom Nitzschia palea extract against bacteria was studied. The diatom was extracted using aceton. High activity of crude extract was recorded against gram positive strains. Whereas no activity against gram negative was recorded except Pseudomonas aeruginosa standard and pathogenic strains.

Liquid-side mass-transfer coefficients (KLa) were measured in air-suction type fermentors using physical absorption of oxygen. A fermentor of 0. 5 m i.d. was used with a working capacity of 60 liters of liquid. Tap water was used as the liquid phase, and air was used as the gas phase. The bioreactor mixing system consists of shrouded-disk/curved-blade turbine with six evacuated bending blades. The effect of liquid submergence (S) was investigated. Further, the effects of the ratio of the impeller diameter (D) to the tank diameter (T), and the clearance of the impeller from the tank bottom(C) were also studied. The agitation speed (N) was varied in the range of 50-800 rpm. It was found that the value of KLa increased as the impeller diameter increased, while it was decreased continuously with increasing the clearance. The effect of impeller submergence on the value of mass transfer coefficient (KLa) is not much pronounced. Suitable correlation was developed for estimating mass transfer coefficient (KLa) in this type of bioreactors.

THE THROMBOCYTES AS AN IMMUNE INDICATOR

This study investigated the effect of vaccine as an immune enhancing on chicken thrombocyte number. Also the relation between thrombocytes and other blood cells like erythrocytes and leukocytes were studied in regard to age. A total of 100 broiler chicks of one day old were used in this study. They were divided equally into two groups. One group was kept without vaccination and the other one was vaccinated twice against Newcastle disease at 7 and 20 days of age. During the whole experiment which is lasted for eight weeks, blood samples were collected daily for the first two weeks and weekly for the other six weeks. Thrombocytes, erythrocytes and leukocytes number were calculated for both groups. The results show a significant increase in thrombocyte number as well as in leukocytic number post vaccination. Beside that a high correlation coefficient is reported between thrombocyte and leukocyte in both vaccinated and control groups (99.8% and 97.5 %, respectively). The three cell types increase gradually from the first day of age and as the animal grows up. They seem to stabilize at the fourth week of age. On conclusion, these results investigate the role of thrombocytes in immunity. Certainly, the over all results reflect that we could depend on thrombocytes as an indicator of stress as much as the H/L ratio. It could be an indicator of the physiological and immunological state of the animal. Further studies are needed to investigate the role of thrombocytes in some immune-supressive diseases.

In this study, MerE gene related to Mer operon (Mercury resistance) from the original Tn501 transposon was amplified using PCR technique. Plasmid pUB3466 represent the cloning vector pBR322 containing the transposon Tn501 was used as a template. The PCR process was fully successful and only one band about 1.2 kbp was appeared in the gel electrophoresis. The amplified gene was cloned using pGEM-T easy vector, then transformed to a competent strain E. coli DH5α. Both of the cloning and transformation process succeeded and transformed cells were selected on selective media containing Ampicillin final concentration 100 µg/ml. Result of plasmid content analysis revealed that the transformed cell harboured one plasmid band represent the vector with MerE gene. E. coli DH5α with and without the cloned vector were used in swimming motility assay using semisolid LB media in the present and absent of HgCl2 final concentration 1µg/ml in order to study MerE function. The result showed that there was diminishment in swimming movement and the growth zone clearly decreased when HgCl2 was added. This result revealed that MerE gene may play role in transport mechanism also modified the behavior of the microorganism leading to altered motility.

This study investigated thirty patients with chronic hepatitis B who were admitted to Hepatology and Gastroenterology Teaching Hospital in Baghdad, aged from 20-65 years during the period from beginning of February 2008 to the end of February 2009. The patients were suffering from different clinical symptoms with previous risk factors for transmission of HBV infection, also blood samples were collected from thirty healthy HBs Ag carrier was discovered accidentally through attending blood bank for donation of blood, aged from 18-52 years and served as a control group. A cross sectional study was conducted in the period between beginning of February 2008 to the end of February 2009.The establishment of the molecular techniques for identified HBV DNA in chronic patients and carrier group gives an obvious image especially when uses the Nested PCR. The current results confirm that a nested PCR protocol is more sensitive method to detect HBV DNA than PCR direct protocol. The positively of HBV DNA in carrier group and chronic group reached to 46.7% and 30.0%, respectively by employing direct protocol. Whereas, HBV DNA is 100% in both groups by Nested protocol with significant difference (p=0.001).
