Study of Signal Transduction Hypothetical Proteins of Helicobacter pylori

Abstract: Hypothetical proteins of Helicobacter pylori strain 26695 were screened using MBGD database. There were about 714 (41.58 %) hypothetical proteins, 82.2 % were virulent proteins. Hypothetical proteins were categorized according to Gene Ontology grouping using ProtFunc server. Proteins in signal transduction group were selected for studying their characters. Post-translation modifications (PTMs) such as phosphorylation gave unclear results, Glycosylation such as N-linked Glycosylation was more frequent than O-linked Glycosylation among proteins. Secondary and tertiary structures were determined and pbd homologous (assigned with X-ray and NMR techniques) were assigned. Most proteins showed high interaction with cellular proteins and components.

Abstract: In vitro effect of salinity in response of growth of four Potato varieties(Ambo, Arizona, Lusa and Rivera) were studied .Result showed difference in the response varieties for tissue culture, Where cultivar Lusa surpassed in response to the growth amounted to 100% of the shoot tips developing planted on the medium, Followed by product Arizona 90%while the response of Rivera was50%.Three concentrations of salinity levels used NaCl(8,10,12 dS/m) were used in addition to the treatment control (6 dS/m)to study the growth. The result showed that there was no significant differences among the varieties, but it was appeared significant differences among salt concentrations .The highest length of the branches was (4.90 cm) at 8 dS/m .The average number of branches at 12 dS/m significantly different from the others and gave the lowest rate (1.125 branch/plant).The number of nods at 10 dS/m gave the highest rate reached 4.125 nod/plant . According to the interaction between varieties and salt concentrations, Arizona gave the highest rate of stem length (5.85cm) and branches number (2.4 branch /plant) at 6 dS/m and Rivera variety gave the highest rate of number of nods which reached 5 nod/plant at 10 dS/m NaCl.

Abstract: Two types of pesticides were selected as organic matter for their negative influence on human health and aquatic life. Two bacteria Bacillus badius and Bacillus subtilis were used to examine their ability to degradation the insecticide Diazinon and herbicide Glyphosate with initial concentrations 50, 60, 70, 80, 90 and 100 ppm in water, of laboratory conditions of 25 Co and pH 7.2-7.6 in batch culture system for 24 hr treatment period. Bacillus badius showed high capability to reduce Diazinon concentrations by 66.6% , 62.3% ,43.7%, 34.1% , 23.8% and Glyphosate concentrations by 75.4% , 61.8% , 47.8%, 37.5% and 30.5%.While, Bacillus subtilis showed lower ability to reduce Glyphosate concentrations by 62.2%, 52.3% , 42.4%, 32%, 23% and Diazinon concentrations by 43.2%, 39.3% , 25.1%, 24.6% and 21.8%. This study proved that Bacillus badius and Bacillus subtilis have no ability to survive in pesticides concentration of 100 ppm.

Abstract: The study was conducted on 90 blood samples divided into 35 samples Hypothyroidism 35 samples hyperthyroidism (group of patients ) and 20samples from healthy people ( control group). DNA of two groups was extracted and exon 10 was amplified by using PCR. Mutations were diagnosed and analyzed by using Sequencing. The present study found seven novel mutations in exon 10, divided into five mutations among hypothyroidism patients compared to control (who did not appear to have any type of mutations diagnosed). All mutations diagnosed were substitution included transition or transversion depending on the nature of the mutant base, three missense mutation in three hypothyroidism patients with a proportion of 20% each. They are as follows c.1330 T>C, Y444H; c.1424 T>C, L475P and c.1435 T>C, S479P and two silent mutation in two hypothyroidism patients (c.1338G>A, L446L and c.2300 G>A Untranscrib).While the result showed two mutation in hyperthyroidism, one was missense mutation (c.1832 C>A, P610Q) and the other silent mutation (c.2103 C>A, R701R). The current study pointed out the occurrence of the disease linked with mutations in specific areas of linking stimulating hormone and the interaction between the protein receptor and α-subunit for G-protein.

Abstract: Interleukin-18 also known as interferon-gamma inducing factor, Interleukin 18 is a cytokine that plays an important role in the T helper cell 1 response, by its ability to induce IFN- c production in T cells and natural killer cells. A functional variant of IL-18 gene has been reported as associated with type 1 diabetes (T1D). In the present study were analyzed two promoter single nucleotide polymorphisms (SNPs), at -607 (rs#1946518) and -137 (rs#187238) position, in 60 patient with type 1diabetes and 60 healthy individuals, both from Egyptian population. Allele and genotype frequencies of IL18 SNPs were compared in patients and controls .The genotype CC in -137 show significance according to the patient and their control (P<0.004) while the SNP – 607 don’t showed any significance between patient and control (P=0.641), respectively. Our finding suggested an association between IL18 promoter SNP -137 and susceptibility toT1DM in Egyptian population.

Abstract: The aim of this study is to investigate the association between polymorphisms Exon1 of Dopamine Receptor D3 (DRD3) receptor and Exon 2 of 5-hydroxytryptamine type 2a (HTR2A) genes and schizophrenia patients from Baghdad Iraq. To achieve this goal, blood samples were collected from 50 patients with schizophrenia (25 samples of male and 25 samples of female) and 25 samples of healthy individual, DNA was isolated and the DRD3 receptor and HTR2A genes were amplified by using specific primers for exon1 and exon 2 of this genes, then sequencing of nucleic acid of genes was performed by machine is AB13730XL, (Applied Biosystem, Macro gen company, USA). The DNA sequencing results of flank sense of DRD3 receptor and HTR2A genes from healthy individual was found to be compatible 100% with wild type of Homo sapiens from the Gene Bank, the DNA sequencing results of flank sense of DRD3 receptor from 50 cases schizophrenia patients was found to be compatible 99% and score 398 and expect 0.0 with the wild type sequences of gene bank, and the differences may be attributed to one transition mutations (G/A), at position 1804 of exon 1. It is a missense mutation that leads to changes in amino acid from Serine (S) to Glysine (G). Our results showed that the incidence of G/A mutation was highly significant (X2=100, P<0.01). In total, 8% of Shizophreina patients had two transition mutation +1804 G/A and +1830 G/A single nucleotide polymorphism, and compatibility 99%, score 397 and expect 0.0 with the wild type sequences of gene bank. The +1830 G/A SNP was silent mutation which result change of codon from GCG to GCA (Alanine to Alanine). However, the DNA sequencing results of flank sense HTR2A genes from 50 cases schizophrenia patients was found to be compatible 99% and score 259 and expect 5e-132 with the wild type sequences of gene bank, there was no significant correlation between shizopherina and incidence of transition mutation G/A (Serine to Serine) at nucleotide 156 of exon 2 HTR2A gene (X2=0.055, P>0.05). In conclusion, our case study suggests that the 1804 A/G SNP of the DRD3 gene is strongly associated with genetic susceptibility to schizophrenia in the Baghdad / Iraqi population.

Mycobacterium bovis tuberculins: Preparation and antigenic properties

Abstract: Three types of tuberculin were prepared from one isolate of M. bovis which was isolated from clinically tuberculosis cow. The isolate was purified and identified after it had been subjected to all biochemical tests and laboratory animal inoculation. These tuberculins include: The purified protein derivative, new tuberculin and lysozymal extract. The preparations were subjected to different in Vivo and in Vitro tests of potency and specificity .The results showed that the lysozymal extract had the highest potency in the diagnosis of the diseases in the experimentally infected animals, while the other tuberculin ( purified protein derivative and new tuberculin) were equal in their potency as that of the imported one .

Abstract: Psoriasis is a common, chronic, disfiguring, inflammatory, hyperproliferation, and abnormal differentiation of keratinocyte, in which both genetic and environmental influences have a critical role. The study was designed to detection the levels of IL-23 and IL-17A among psoriatic patients compared with healthy group, also studied the correlation between them and some parameter such as:-age of onset and severity of psoriatic patients. Fifty psoriatic patients were selected randomly from both sexes with ages from (10-70) years. Patients were diagnosed clinically by dermatologist. Sera samples of both groups were collected from all individuals for the estimation levels of IL-23 and IL-17A by ELISA technique. All mean values sera levels (IL-23 96.74 pg/mL and 6.56 pg/mL for IL-17A ) of patients were significantly higher than those of controls (18.74 and 3.01 pg/mL for IL-23and IL-17A respectively). There was a high significant in levels of patients sera of IL-23 and IL-17A, in compared with healthy control group according to these normal values. Furthermore, there was a significant correlation between the levels of IL-23, and age of disease onset among psoriatic patients. In contrast there was no significant association between the severity & interleukins (Il-23 and Il-17A). Emerging data in humans reveals a critical contribution of IL-23 and IL-17A in the pathogenesis of psoriasis. The IL-23 and IL-17A pathways were atherapeutic targets for biologic agents and systemic therapies in psoriasis treatment

Abstract: Viola odorata L. (Violaceae ) Cultivated in Iraq, it is an important folklore medicine for the treatment of various ailments as antioxidant and antibacteria. In the previous study two phytosterol (Stigmasterol and β-sitosterol) were detected and identificated from the methanol extracts in roots, leaves, flowers and seeds by using chemical test,TLC and HPLC techniques.The results showed all the plant parts contain high amount of sterols the quantitative data revealed that the concentrations of Stigmasterol in Viola odorata were in roots(4.5mg/100g) ,in leaves(2mg/100g),in flowers(7.3mg/100g) and in seeds(2.4mg/100g). also the content of β-sitosterol in roots(9.2mg/100g),in leaves(3.2mg/100g),in flowers(5.3mg/100g) and in seeds (3mg/100g). Results showed high amount of Stigmasterol in flower while high amount of β-sitosterol in root.

Abstract: Biosorption can be an effective technique for the treatment of heavy metals bearing waste water resulting from human and industrial activities. several bioadsorbents have the ability to remove the heavy metals and thereby making water contaminant free. In the present study the biosorption of heavy metals using the black tea leaves waste and parameters affecting the biosorption of Cr ion; such as contact time, pH, temperature, dosage and particles size have been investigated. The present study showed that the contact time taken for maximum sorption of Cr ion was 120 min. The maximum percent removal of Cr ions on adsorbent was observed at pH 6 and significantly decreased by reducing the pH values and slightly decreased at higher pH values. The results indicated that the increasing of temperature from 35°C to 50°C induced that the adsorption capacity of Cr ions with highest adsorption capacity at 45°C and the concentration of 2 g was sufficient for maximum biosorption. It was obvious that the adsorption capacity of metal ion on tea waste increased by decreasing the particle size and the best particles size was 200 μm. The experiment results showed that maximum removal of Chromium ion by tea waste is 89.5 % at optimum condition.
