Morphological, Histological and Ultrustructural Study of the Tongue in House Geecko (Hemidactylus flaviviridis) Lizard

The aim of this study to investigate the tongue morphology and histology in house gecko Hemidactylus flaviviridis using light and scanning electron –microscopy (SEM ) technique.The morphology result revealed the presence of three parts : apex , body and lingual root .The light microscopy result showed that the tongue is covered with mucous membrane composed of non keratinized stratified squamous epithelium based on lamina propria which contain bundles of striated muscles its fibers in different directions including connective tissue , blood vessels and nerves. The lingual mucosa of the dorsal surface is covered with different pattern of lingual papillae which are widely distributed all over the dorsal surface except the apex. Noticed on the dorsal surface different types of papillae were like fungiform, long and short folliolate, conical and cuboidal papillae. SEM examination showed that the tongue in Hemidactylus vlaviviridus appeared elongated with triangular shape and bifurcated free tip, also it is divided into three parts: apex, body and root; many types of papillae are distributed on the dorsal surface of the body and root of tongue. Fungiform papillae are widely distributed all over the dorsal surface of the tongue body .In the root of tongue, there are different types of papillae like filiform and foliate papillae , a lot of goblet cells have been showed in this region .

Potential Healing Effect of Topical Stem Cell Transplantation and Methandrostenoloneon in Induced Cutaneous Wounds in Dogs

This study was done to explore the influence of stem cells on cutaneous wounds in comparison with treatment with methandrostenoloneon drug. Cutaneous wound was induced on the back of each dog using disposable dermal biopsy punches . Twenty five dogs were divided into five groups: 1stgroup (T1) was control. 2nd group (T2) was induced without any treatment. 3rd group (T3) was induced wound and treated with phosphate buffer saline, 4rth group (T4) was induced and treated with 5mg/kg/B.W. of Methandrostenolone, 5th group (T5) was induced and treated with stem cells 1x106cell/250 µL of phosphate buffer saline. After 7 and 15 days post treatment level of Hb and RBCs count in T2 and T3 group were decline as compared with control group. The WBCs, Alkaline phosphatase , acid phosphataseas in T2 and T3 showed enhance as contrast with T1. After 7 days, Hb and RBCs in T4 and T5 showed important increase as compared with T2. Moreover the WBCs, alkaline phosphatase, acid phosphatase and total protein levels significantly elevated in the T4 and T5 as compared with T1. After 15 days of treatment the Hb, RBCs, levels of Alkaline phosphatase, acid phosphataseas and total protein levels in T4 showed significant increase as compared with T1, The Hb, RBCs, levels of Alkaline phosphatase, acid phosphataseas and total protein in T5 showed no significant changes as compared with control group (T1). The histopathological section in (T2) explained less collagen and more inflammatory cells, the section of wound tissue in (T4) appeared incomplete healing wound tissue and presence little inflammatory cells, the histopathological section of wound treated in (T5) showed return the wound to normal tissue. From this result it can be concluded that the MSCs have ability to access healing of the wound in comparison with methandrostenolone without any side effect.

Biosynthesis and Characterization of Silver Nanoparticles Using Olive Leaves Extract and Sorbitol

This study developed a rapid, ecofriendly and convenient green method for the synthesis of stable silver nanoparticles (AgNPs) with average diameter of 40 ± 5.0 nm and like spherical in shape, using the aqueous solution of Olive tree (Olea europaea) leaves extract as reducing and capping agent along with D-sorbitol. The reaction is carried out at 10−3M of silver nitrate, and the effect of temperature on the synthesis of AgNPs was investigated by stirring at room temperature (25OC) and at 60OC. The AgNPs synthesized were confirmed by their change of color to (dark brown-grey). The characterization studied was done using UV-Visible spectroscopy, Scanning Electron Microscopy (SEM), Atomic Force Microscopy (AFM) and Fourier Transmission Infrared Spectroscopy (FTIR)”.

Effect of G1705A SNP in Growth Hormone Gene on the Productive and Physiological Performance in Broiler Chicken

In this study two commercial broiler hybrids were used (Cobb500 and Hubbard-F15) to detect the G1705A SNP in the third intron of chicken GH gene and investigate its effect on the productive and physiological performance in broiler chicken. PCR- RFLP method was used to identify this SNP, three genotypes were found when using of Eco RV restriction enzyme wild genotype GG, heterozygous GA and homozygous AA. Highly significant difference (p<0.01) was found between the distribution of the different genotypes, the genotype GG had the highest percentage followed by GA then AA and allele G had the superiority over allele A in both broiler breed, no significant effect of the various genotypes on the productive traits in both broiler breeds of study, significant effect (p<0.05) of the various genotypes were found on the serum total protein and triglycerides concentrations of Cobb500 at 14 days of age and the genotypes GG and GA were gave highest mean followed by the genotype AA.

Cytotoxic Effect of Pyomelanin Pigment Produced from Local Pseudomonas aeruginosa Isolates on Different Cell Lines Using MTT Assay

This study was designed to determine the most significant (IC50) values of purified pyomelanin pigment which was produced by local isolates of Pseudomonas aeruginosa on four cell lines using3-(dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT assay). 143 samples from burns, urine and sputum were collected from patients hospitalized into eleven hospitals which are located in Baghdad, Al-Anbar and Karbala provinces during the period of July 2013 to April 2014. All isolates were identified as Pseudomonas aeruginosa according to morphological, cultural, biochemical characteristics, VITEK-2 and 16S rRNA . Eight pyomelanogenic isolates (5.6%) were produced pyomelanin pigment on pyomelanin production medium. The results were showed that the isolate of sputum from cystic fibrosis (CF) was greatest in production of pyomelanin. The cytotoxicity of extracted and purified pyomelanin was assessed on Lung A 549, Skin A375, Macrophage RAW264.7 and VERO cell lines. The results showed that pyomelanin has cytotoxic effect on all tested cell lines but the most significant was on Skin A375 cell line with dose dependent effect.

Determination the Activity of Some Physical and Chemical Factors on the Polyphenol Oxidase in Hot Pepper and Cabbage (Brassica oleracea Var)

The activity of polyphenol oxidase (PPO) in hot pepper pericarp and cabbage was evaluated using spectrophotometric method. The enzyme was extracted from the pepper pericarp with suitable volume of 0.1 M phosphate buffer solution pH (7. 0) .PPO activity was determined using catechol as a substrate. The effects of the enzyme concentration, substrate concentration, pH and temperature were investigated. The highest activity of ppo at (2.5 mg /ml) from hot pepper and the maximum activity of ppo from cabbage was at (1.5 mg /ml) .The highest activity of enzyme catechol concentration was at 100 mM for PPO from hot pepper and cabbage respectively. The optimum pH was 6.0 and 7.0 for PPO from hot pepper and cabbage. The optimum temperatures were 40°C, 30 °C for hot pepper and cabbage respectively. These optimum conditions were used to determine the enzyme activity in hot pepper and cabbage.

Plasma IL-10 Concentration and Its Role in the Pathogenesis of Acute Myeloid Leukemia: a Prospective Study

Many suggestions support that cytokines have pathogenesis role of acute myeloid leukemia (AML) patients and that may be related with disease progression and patient survival. But their prognostic significance in this disease is unknown. The study aimed to estimate the plasma level of IL-10 before and after treatment in patients with AML. Plasma concentrations of IL-10 were estimated in 26 newly diagnosed patients with AML and follow up 19 of them after treatments, 7 patients were died after induction, matched 16 individuals' healthy donors. IL-10 levels were estimated using the enzyme‑linked immunosorbent assay (ELISA) technique. Patients were divided into two groups: 26 before treatment and 19 after treatment. The results showed that plasma concentrations of IL-10 were significantly higher in AML patients compared to control group. Moreover, plasma concentrations of IL-10 were significantly associated with non-responding (NR) AML patient after treatment compare to complete remission (CR). While it observed non-significantly before treatment. The study demonstrated that AML NR patients have increased plasma concentrations of IL‑10 after induction, suggesting that this cytokine is involved in the pathophysiological process of the disease and may be associated with poor prognosis of clinical outcomes.

Role of Oxidative Stress and Some Antioxidant Enzymes in Male Infertility

More than half of all infertile male suffer from oxidative stress. Seminal plasma have antioxidant mechanisms to suppression the reactive oxygen species. The aims of the project was to evaluate the levels of protein peroxidase, lipid peroxidase and antioxidant enzymes such as glutathione peroxidase, superoxide dismutase in blood sample of infertile male and control group. Samples were collected from 50 infertile men and 25 apparently healthy men to measure the levels of oxidative stress biomarkers: protein peroxidase (PPO), lipid peroxidase (LPO) and antioxidant enzymes: glutathione peroxidase (GPX), superoxide dismutase (SOD). Results show highly significant differences in patients with male factor infertility in PPO and LPO (76.48+9.51; 58.22+5.33) compared with control group (54.72+8.63; 39.05+4.74) respectively. While GPX and SOD enzymes levels show significant differences in patients with infertility (8.01+1.04; 12.26+2.17) compared with control group (11.23+1.74; 18.26+3.24) respectively. In conclusion oxidative stress was associated with infertility and this may help for treatment of male factor infertility by using proper antioxidants such as vitamin C, vitamin A, vitamin E and selenium.

Epstein Barr Virus and P53 Gene Expression Correlation with Gastric Adenocarcinoma Patients in Baghdad City

Gastric carcinoma is the most prevalent cancer related deaths worldwide today . The current study was carried out to shed the light on Epstein Barr virus, Tumor suppressor gene p53 associated gastric cancer of some Iraqi patients. According to insitu hybridization technique , this study was determined EBV and p53 gene correlates with some aspects of gastric cancer patients like age, gender , histological grade and stage of the tumor . Fourty six biopsies were obtained from patients with adenocarcinoma who had undergone gastrectomy. The tissue sections were collected during the period between September 2010 until June 2013. Epstein Barr virus was detected insitu hybridization in 18 patients (39%) out of 46. Whereas, the positive results of relation between p53 oncogene and gastric adenocarcinoma patients were detected in 30 (65.2%) out of 46. Out of 46 patients , this study included 32 male and 14 female with mean age 54 ranged between 30-72 years. The histological types included 18 well, 4 moderate and 24 poor differentiated respectively . Most of cases 32 (69.5%) falling in stage I-II and the remaining 14 were in the stage III and IV. The positive results revealed that EBV correlate in highly significant association with each of age, gender, grade and stage of the tumor , also expression of tumor suppressor gene p53 was correlation with all patients aspects in highly significant association.

Levan Production using Pseudomonas Brassicacearum Isolated from Rhizosphere Soil of Cowpea Farm in Iraq

The present investigation, was designed in order to isolate bacteria from rhizosphere soil of Cowpea farm in Iraq which is capable of levan production. The selected levan producing bacterium was identified as Pseudomonas brassicacearum based on the phenotypic identification by Bergey’s Manual of Systematic Bacteriology and confirmed with the 16S rRNA gene sequencing. Acid hydrolyzed levan sample was subjected to TLC technique which revealed that, levan is composed of only one sugar which is fructose. Rf values of levan produced by P. brassicacearum was 0.42 which is identical or close to Rf values of fructose of 0.42. The effect of some factors on levan production by P. brassicacearum was investigated. The results showed that the best carbon source for levan production was sucrose 7.77 g/l, and casein was the best nitrogen source for levan production which gave 8.56 g/l, followed by ammonium sulfate and corn steep liquor which they gave 8.52, 8.09 g/l respectively. The highest levan yield was at the sucrose concentration of 300 g/l which gave 7.95 g/l. At initial pH of 7.8, P. brassicacearum gave their highest levan production that was 7.77 g/l. Levan production by P. brassicacearum continued increasing until it reached its maximum production at 40ºC which was 8.24 g/l. The optimal incubation period for levan production, was estimated at 8.70 g/l after 72 h of incubation.

Evaluation the Activity of Rosemary (Rosmarinus officinalis L.) Essential Oil Against Some Cyanobacteria

Freshwater resources are threatened by harmful algal blooms (HABs ) internationally. The HABs are sometimes direct result of eutrification status caused by pollution entering water bodies, such as partially treated nutrient-rich effluents discharged in the rivers, in addition to fertilizers and animal wastes. The anti-cyanobacterial activity of Rosmarinus officinalis essential oil on Microcystis aeruginosa and Chroococcus minor was evaluated. The essential oil was extracted by steam distillation method, the yield of extraction was 1 % (v/w),and the concentrations of this oil 0.05, 0.1 and 0.15% were prepared. Results revealed very strong antagonistic effects of the essential oil on M. aeruginosa and C. minor growth rates by measuring the amount of chlorophyll- a of each alga. Generally growth rates of both algae severely decreased as the time and the concentrations of the essential oil increased comparing with the growth rate of control (algae without added essential oil) which exponentially increased along with time. M. aeruginosa was more sensitive to the allelopathic effect of the oil.
