Observations on the ultrastructure of a rat mammary gland treated with harmal and borage

Abstract

Background : It had been indentified by histological, histochemical and morphometrical studies that peganum harmala is a mammogenic herb and borage officinalis is a lactogenic one . To complete our investigation about these two herbs , we performed electron microscopical study . Materials and methods : Rats were grouped according to their physiological status into three groups . Each group was subdivided in to three subgroups : one control and two experimental . The two experimental group were treated daily; the 1st one with an aqueous extract of peganum harmala seeds and the 2nd with an aqueous extract of borage officinalis flowers . After two weeks of treatment , mammary glands were employed for electron microscopical study . Results : In virgin rats , the epithelial and myoepithelial cells were partially differentiated when harmal was given and completely differentiated when borage was given . In pregnant rats , harmal and borage optimize mammary parenchymal growth and induce lactation when these herbs were given. In lactating rats ,these herbs exhibited a picture similar to control lactating group but the budding of lipid droplets and the swelling of secretary vesicles were markedly increased . Conclusion: Both harmal and borage stimulate the release of prolactin and induce galactogenesis during pregnancy and promote it during lactation . Abstract

ABSTRACT: BACK GROUND : Herbal remedies are used in induction of lactation . In view of that ,Borage Officinalis was employed to ensure an abundant milk supply or rectify milk insufficiency .However ,this remedy has not been scientifically tested . METHODS: The animals were treated with the aqueous extract of Borage Officinalis flowers at a daily concentration of 100 mg / kg body weight /ml for each rat through Oro-gastric tube for 14 days . Animals were subdivided into subgroups according to their physiological status . Mammary glands of these animals were processed for histological , histochemical and immunohistochemical studies . RESULTS: The results of all parameters indicated that the aqueous extract of Borage Officinalis flowers induced lactogenesis in the mammmary glands of virgin and pregnant rats and promoted lactation when had given to lactating rats . CONCLUSION : Borage Officinalis is a lactogenic herb

Abstract

The experiment was applied on /45/ healthy male of golden Syrian hamsters, were divided into three groups as follows: Control group(C), High Fat-High Cholesterol group(HF-HC) and Borage Oil group (BO). All groups had laboratorial diet freely during/15/ days(first period), after that each of HF-HC and BO groups took diet consisting of 80% laboratorial diet+ 20% fats(13.5% sheep fat+6.5% coconut oil) for /4/ weeks(second period). The treatment of BO group changed after second period and hamsters had laboratorial diet and dosed borage oil 2g/kg of the body weight daily for /4/ weeks(third period). The results showed a significant increase(p=0.0000) of the total cholesterol(TC), low density lipoprotein cholesterol (LDL-C) and triglycerides(TG) concentrations approximately(163%, 224%, 232%) respectively in each of HF-HC and BO groups in the second period in comparison with first period. Our results showed, in third period, a dramatic significant decrease(P=0.0000) in the TC, LDL-C and TG concentrations approximately (56%, 69%, 40%) respectively in BO group in comparison with the same group of second period

Abstract

This study was designed to show the inhibitory effect of different concentrations of alcoholic extract of Borage officinalis on the Monoamine oxidase (MAO) and Acetylcholinesterase (AChE) enzymes in human serum. The results obtained from the study exhibited that alcoholic extract of Borage officinalis caused inhibition to enzymes activity with all concentrations of the extract. The results also showed that when the concentration of the extract was (0.001 mg/ml), the percentage of inhibition was (4.3% with MAO and 15.2% with AChE) and this percentage increases until reaching up to (74.7% with MAO and 84.18% with AChE) when the concentration of the extract was (0.1 mg/ml). From the kinetic parameters, studies found that alcoholic extract of Borage officinalis is acting as competitive inhibitor with MAO enzyme and as uncompetitive inhibitor with AChE enzyme.

في هذا البحث تم تحضير المستخلص الكحولي لنبات Borage officinalis (لسان الثور) ودراسة تأثير تراكيز مختلفة من هذا المستخلص على الفعالية الانزيمية للانزيمات MAO و AChE في مصل الدم البشري. وقد اظهرت النتائج ان كافة تراكيز المستخلص الكحولي لنبات لسان الثور لها تأثير تثبيطي على الانزيمات وانه كلما زاد تركيز المستخلص زاد التأثير التثبيطي على الانزيم. كما اظهرت النتائج انه عندما يكون تركيز المستخلص الكحولي للنبات (0.001mg/ml) فأن النسبة المئوية للتثبيط للانزيمن MAO و AChEكانت (4.3%)، 15.2% على التوالي، كما وجد ان بزيادة تركيز المستخلص الكحولي للنبات تزداد النسبة المئوية للتثبيط لكلا الانزيمين قد تصل الى (74.70%، 84.18%) على التوالي عندما يكون تركيز المستخلص الكحولي (mg/ml 0.1). و وجد من خلال دراسة الخواص الحركية للمستخلص مع الانزيمات ان المستخلص الكحولي لنبات لسان الثور يسبب تثبيطا تنافسيا مع انزيم MAO وتثبيطا غير تنافسي مع أنزيم AChE.
