Estimation of total IgE, blood eosinophils and phagocytic activity in human scabies

Abstract

Background: Scabies is a contagious disease. It can be easily misdiagnosed with many other dermatological conditions.
Objective: This study was designed to evaluate some laboratory methods, (e.g. IgE,NBT) which might be useful in confirming the diagnosis of scabies, and to follow up the response of the patients after treatment.
Patients and method: This study was conducted on 103 patients from Al- Zenjelli Custody (jail) in Mosul City (Iraq) over a period of 6 months (Oct. 2001 to April 2002). Blood samples from 103 scabietic patients and 40 control subjects were analysed by using various tests including total white blood cell count, eosinophil count and nitroblue tetrazolium (NBT) positive neutrophils count. Plasma total IgE levels were measured for only 70 patients and 20 control subjects by using Enzyme Linked Immunosorbent Assay (ELISA).
Results: Plasma total IgE level showed a significant raise in patients with scabies in comparison to control group. (p< 0.05)The NBT test revealed a significant raise in both treated and non-treated subjects in comparison to control group. (p<0.05) Also, the untreated patients showed a higher NBT positive neutrophils than the treated ones. Eosinophil counts were higher in patients with scabies than the control subjects in the first few postinfection days. (p<0.001) Total white blood cell counts revealed no significant variation between patients and control groups.
Conclusion: The results indicate that nitroblue tetrazolium (NBT) test was the best in comparison to other tests. It was used to study the role of phagocytosis in scabies. Its increase reflected an efficient innate immunity. This test might explain to us some immunopathological aspects of this disease and its symptoms which might help in diagnosis and follow up of patients with scabies.

Abstract

This study was conducted in Basrah at period from March through July 2000.The protein extract from Bovine dander was prepared with extraction followed bypurification and fractionation by gel filtration into tow peaks. Elisa inhibition withserum samples from 77 patients who were sensitive to bovine dander, revealedsimilar lgE,lgG binding patterns for each peak. Also, there is a cross —reaction .between these two peaks.PeakII exhibited the highest IgE binding inhibition andshowed a specific allergenic activity about two fold higher than that of peakI.Theallergenecity ofpurified allergen was not lost during purification process.

Abstract

Abstract Sixty male patients with asthma were included in this study, their age ranged between 18-24 years (mean 20 ±1.3). Forty-five were in attack state and fifteen were in rest (between the attacks). All cases were admitted to Al-Rasheed military hospital during November 2001 and April 2002.Twenty age matched apparently healthy en were included as a negative control. Eight males with signs and symptoms of asthma (mostly chronic bronchitis patients) were regarded as a positive control. Ten ml of blood sample were withdrawn from each patient as well as from controls.IgE level was measured by enzyme linked immunosorbant assay (ELISA) in which a significant elevation were noticed in both groups attack and rest groups in comparison with control groups.Key wards: IGE, Asthmatic, immune response.

الملخص:شملت هذه الدراسة ستون مريضا ذكرا مصابا بالربو القصبي وبعمر يتراوح بين 18- 24 سنة (معدل20±1.3).خمسة وأربعون منهم في حالة نوبة ربوية حادة وخمسة عشر في فتره مابين النوبات سجل جميع المرضى دخول إلى مستشفى الرشيد العسكري للفترة مابين تشرين الثاني 2001 إلى نيسان 2002. شملت مجموعة السيطرة عشرون رجلا سالما واعتبرت مجموعة السيطرة السالبة, شملت مجموعة السيطرة الموجبة ثمانية مرضى يعانون من أعراض الربو الناتجة من التهاب القصبات المزمنة. تم سحب 10 مل من الدم من كل المجاميع الذين هم قيد الدراسة لفصل المصل.استعملت طريقة (ELISA) لتحديد مستوي الضد . IgE وقد أظهرت النتائج ارتفاعا معنويا في كلتا المجموعتين عند مقارنتها مع مجاميع السيطرة.

Abstract


Background: Asthma is an allergic disease characterized by airway obstruction as a result of cellular accumulation due to the liberation of certain mediators. Among those mediators are the cytokines such as IL-5.
Patients & Methods: Interleukin-5 concentration has been estimated in 94 sera samples of Asthmatic patients in comparison with 41 non-asthmatic bronchitis as patient controls in addition to 30 apparent healthy control group using ELISA method.
Results & Conclusions: There is highly significant elevation of IL-5 in the asthmatic cases in comparison with healthy controls (P< 0.001). We conclude that this cytokine may play the major role in asthmatic attack & it may be a good marker for the disease.
