Abstract

Polycystic Ovarian syndrome (PCOS) is being the most frequent cause of anovulatory infertility, Adiponectin is the most abundantadipocytokine and may play a role in the regulation of insulin sensitivity and IR in PCOS and count for 0.01% or 3–30 μg/ml of total plasma proteinthe adiponectin gene contains 3 exons spans 16 kb on chromosome3q27.The aim of the present study was to evaluate the genetic influence of the adiponectin gene polymorphisms in the development of PCOS among women of Babylon Province/ Iraq. sixty three women were studied ,and were classified into two groups of : first group consists of 32 women infected with polycystic ovaries syndrome, the second consists of 31 healthy women to detect the presence of T45G polymorphism within the gene. From all subjects a whole-blood sample was taken and was used for isolation of peripheral blood leukocytes. The adiponectin T45G polymorphism, located in exon 2, was genotyped by amplification of genomic DNA.The present study included study of the relationship of this gene with PCOS women in reproductive age, A statistically significant difference was observed in the frequency of TT,TG and GG genotypes between women with PCOS and controls .

Abstract

Background: Staphylococcus aureus is a major pathogen that associated with serious community acquired and nosocomial diseases. In view of the present problem the study aimed to isolation of S. aureus within osteomyelitis samples and amplified the mecA gene of some antibiotic resistant isolates.Methodology: S. aureus were isolated from osteomyelitis and all isolates screened for antibiotic susceptibility test to detection of mecA gene.Results: 29 swabs were taken from both sex patients related to Al- Sammawa Teaching hospital at period from September 2013 to march 2014. Only 14(48.27%) patients had S. aureus and 19 isolates yielded from all samples. These isolates showed pronounced resistance for a group of active antibiotics. Five isolates have full (100%) resistance to oxacillin, penicillin and cefoxitin so considered as Methicillin Resistant Staphylococcus aureus (MRSA). Genetic identification showed that five S. aureus isolates have mec A gene in their genome.Conclusion: This study highlights the presence osteomyelitis infection in both sex with little incidence. S. aureus one of the important pathogens that have resistance to many antibiotics due to mecA gene.

المقدمة: تعد المكورات العنقودية الذهبية Staphylococcus aureus من اهم الممرضات المرتبطة بالأمراضالمكتسبة واصابات المستشفيات. من خلال ذلك تهدف الدراسة الحالية للتحري عن بكتريا المكورات العنقوديةالذهبية في اصابات العظام ومن ثم تضخم جين mecA لعدد من العزلات المقاومة للمضادات الحياتية.طرق العمل: عزل بكتريا المكورات العنقودية الذهبية من اصابات العظام ومن ثم فحص جميع العينات لعدد منالمضادات الحياتية لأجل التحري عن جين mecA .النتائج: تسعة وعشرون عينة تم الحصول عليها من كلا الجنسين المراجعين لمستشفى السماوة التعليمي. فقط%42.11 ( هم من المصابين ببكتريا المكورات الذهبية (74 Staphylococcus. aureus و 71 عزلة تمالحصول عليها من تلك العينات. اظهرت العزلات مقاومة واضحة لعدد من المضادات الحياتية. من بينها 5عزلات مقاومة بشكل كامل) 711 %( لمضادات الاوكساسلين والبنسلين و السيفوكسيتين وعلى اساس ذلكاعتبرت المكورات العنقودية المقاومة للمثيسلين ) MRSA (.بينت نتائج التشخيص الجيني ان خمس عزلاتتمتلك الجين المشفر للمقاومة mec A في مادتها الوراثية.التوصيات: بينت الدراسة الحالية حدوث اصابات العظام في كلا الجنسين وبنسب قليلة. تعد بكتريا المكوراتالعنقودية الذهبية S. aureus من اهم الممرضات التي تمتلك مقاومة لعدد من المضادات بسبب امتلاكها جينmecA

Abstract

To examine the recovered strains phenotypically by conventional methods and genotypically by using synthetic oligonucleotides of 21 and 24 bases in polymerase chain reaction to amplify a sequence of nuc gene , which encoded thermostable nuclease of Staphylococcus aureus. A DNA fragment of approximately 270 bp was amplified from isolated DNA, PCR products was detected by agarose gel electrophoresis. Our results showed that 100% of Staphylococcus aureus were positive to nuc gene .The PCR for amplification of nuc gene has potential for rapid diagnosis of S. aureus infections by direct testing of clinical specimens .

تم فحص العترات المعزولة من الجروح من مستشفى الرمادي العام التعليمي مظهريا باستخدام الطرق التقليدية وجينيا باستخدام تفاعل الكوثرة PCR للكشف عن جين nuc الخاص ببكتريا المكورات العنقودية الذهبية والمسؤول عن انتاج انزيم thermostable nuclease باستخدام قاعدات البوادئ المصنعة بحجم 21 و24 قاعدة على التوالي , ولقد ضخم الجين ذو الحجم 270 زوج قاعدي المستخلص من الجينوم البكتيري بتفاعل الكوثرة وان نواتج هذا التفاعل رحلت على هلام الاكاروز بعملية الترحيل الكهربائي . ومن خلال نتائج هذه الدراسة تم عزل المكورات العنقودية الذهبية بنسبة 57,5% (96/167) بالاعتماد على الطرق التقليدية وكانت جميع هذه العزلات 100% موجبة لــــ جين nuc . ان استخدام تفاعل البلمرة لتضخيم جين nuc يعتبر مؤشر قوي للتشخيص السريع لالتهابات المكورات العنقودية الذهبية بطريقة مباشرة في العينات السريرية وحتى دون الحاجة الى استخدام الطريق التقليدية المعتاد عليها في مختبرات الوحدات الصحية.

Abstract

Background: Candida albicans is the principal fungal infectious agent in human infection. Adhesion is thought to be an essential step for colonization and establishment of Candida infections.Objectives: Identification and comparison of ALS1 virulence gene of adhesion family among different isolates of Candida albicans by PCR. Patients and methods: One hundred eight samples were collected from different group of Iraqi patients. All samples were culture on Sabouraud′s agar, CHROMagar for identification while API Candida kit confirmatory test and extracted DNA was done for just Candida albicans isolates, detected the ALS1 gene, extracted RNA for synthesis of cDNA and detected of gene and compare between isolates. Result: C. albians isolated from each of vaginal and oral swabs expressed the stronger gene that mean these isolates were the most virulent once. The two isolates from each of urine sample and nail clip which expressed only one strong fluorescence represented by ALS1 gene. C. albicans isolated from cutaneous scrap sample showed the only one moderate fluorescence represented by ALS1 gene. Conclusion: C. albians isolated from each of vaginal and oral swabs expressed the stronger gene that’s mean these isolates were the most virulent once.Key words: Candida albicans, ALS1 gene, PCR virulence gene.

المقدمة:تعتبر مبيضات البيضاء عامل فطري معدي اساسي في عدوى الانسان. يعتبر الالتصاق الخطوة الاساسية لتمركز و لتكوين الاصابات المبيضيةالاهداف:التعرف ومقارنه الجين بين العزلات المختلفة العائدة للمبيضات البيضاء بطريقة تفاعل البلمرة التسلسلي.المرضى وطرق العمل: تم جمع 108 عينه من مختلف عزل المبيضيات البيضاء من مجاميع مختلفة لمرضى العراقيين . تم زرعهم على السابرود اكار والكروم اكار وتم اجراء أكار وApi Candiaللتاكد . تم استخلاص الدنا والتحري عن الجينات وتم استخلاص الرنا للعزلات الثمانية الموجبة لصنع نسخة للدنا منه والمقارنة بينها .النتيجة : كانت شدة التألق المبيضيات البيضاء المعزولة من كل من المسحات الفموية والمهبلية قوي لكلا الجينين بينما شدة التألق للعزلات المأخوذة من البول والأظافر فكان قوي فقط للجين ALS1 اما شدة التالق للعزلات المأخوذة من القشطات الجلدية كانت متوسطة التألقالاستنتاج : كانت العزلات للمبيضات البيضاء المعزولة من المسحات المهبلية والفمويه الأكثر تألق للجين هذا يدل على انها أكثر مرضيةمفتاح الكلمات: المبيضات البيضاء , جين ALS1

Abstract

In this study, (100) clinical samples were obtained from nasal passage from healty individuals in Hilla city who work in restaurants in Hilla province, during a period of three months (from November 2013 to January 2014) . A total of (100) samples only twenty – six of S.aureus were isolates from nasal passage . Molecular detection of Enterotoxin A and B was also studied by using specific markers . It was seen that Enterotoxin A was positive in (9) isolates where Enterotoxin B was detected in ( 5 ) isolates . On the other hand, molecular detection of bap gene by PCR technique showed that (4) S.aureus isolates (50 %) gave positive results for this marker only in the plasmid but non was seen in the chromosomal DNA .

Abstract

Abstract: The aim of this study was to determine the genotypic characteristics of S. aureus isolates by using coagulase gene typing and polymerase chian reaction-restriction fragment length polymorphism (PCR-RFLP) of coagulase gene. One hundred and seventy two wound swab specimens were collected from patients in AL-Kindy Teaching Hospital, Al-Wasity Hospital, Baghdad Al-Taalemey Hospital and Al Shaheed Gazi Hospital in Baghdad during the period from September 2013 to January 2014. All isolates were diagnosed depending on microscopical and biochemical tests. It was found that 47(27.33%) isolates were identified as S.aureus. The amplification of the coa gene of S.aureus strains generated 5 different genotypes: I, II, III, IV, V based on different size of polymorphism raning from 500-900bp. The majority (44.7%) of the strains were of coa gene type II. PCR-RFLP of coa gene exhibited 11patterns which were obtained with AluI digests of PCR products. The number of fragments varied from one to three with sizes of the fragments varied from 80 to 700 bp. Type IX was the most common and accounted for 27.7% of the isolates. PCR-RFLP analysis, showed discriminatory power, reproducibility, easy interpretation and can be considered as a promising alternative for the epidemiological typing of S.aureus isolates.

Abstract

Abstract: Fifty-eight Clinical isolates of Klebsiella pneumoniae were collected from patients at Al-Kindey teaching hospital, Al-Kadhymia teaching hospital, Ibn-Albalady hospital and Al-Imam-Ali hospital in Baghdad from Urinary tract infection (UTI) patient. These isolates were diagnosed using different morphological and biochemical test followed by the complementary API 20E, isolates all screened for antibiotic resistance by rapid detection of β-lactamases production method and their minimum inhibitory concentrations(MICs) were conducted by polymerase chain reaction assay. Detection of blaCTX –M gene was performed and highly resistant isolates (n=17) were selected to determine aminoglycoside resistance mediated by methylation 16S rRNA that combined with the detection of blaCTX –M gene responsible for Extended Spectrum β-lactamase (ESβLs) production. Seven 16S rRNA methylase genes were amplified, the ArmA, RmtA, RmtB, RmtC, RmtD, RmtF and npmA beside amplifying blaCTX –M gene. This study explained the physically link between blaCTX-M gene (ESβLs) production among Klebsiella pneumoniae in correlation with 16S rRNA methylation.

Abstract

Background: Multidrug resistant methicillin-resistant Staphylococcus aureus (MRSA) is a major cause of nosocomial and community acquired infections. The glycopeptides vancomycin has been proposed as the drug of choice for treating such infections; this lead to the emergence of vancomycin intermediate sensitive S. aureus (VISA) and vancomycin resistant S.aureus (VRSA).Objectives: To identify the vancomycin resistance both phenotypically and genotypically among MRSA isolates from different hospitals and to determine the sensitivity of these isolates to different antimicrobial agents.Methods: A total of 204 S. aureus isolates were obtained randomly from various clinical specimens including (wound swab, burn swab, ear swab, urine, sputum, blood and other body fluids) from different inpatient and outpatient who were attending different hospitals in Baghdad. The susceptibility pattern of the S. aureus isolates to different antibiotics was determined by disk diffusion method and vancomycin minimum inhibitory concentration (MIC) for MRSA isolates were determined using broth dilution method following clinical laboratory standard institution (CLSI) guidelines. Van A gene was amplified by PCR using standard primers.Results: All VRSA isolates were MRSA. Twelve VRSA isolates were positive for van A gene, while the remaining ten isolates were negative. All VRSA had a vancomycin MIC of 16μg/ml or more. In the present study, VRSA showed resistance to a wide range of antimicrobial agents (ampicillin, cephalothin, cefoxitin, erythromycin, gentamicin, oxacillin, penicillin, rifampin, tetracycline and trimethoprim).Conclusions: There were high incidences of resistance to the commonly used antibiotics among VRSA isolates compared to VISA and VSSA. Further molecular studies such as PCR technique to identify genes rather than van A (e.g. van HAX analogue) might be suitable to predict VRSA lacking the van A gene.

Abstract

Background: The study of human leukocytes (HLA) alleles, and haplotype frequencies within populations provide an important source of information for anthropological investigation, organ and hematopoietic stem cell transplantation as well as disease association, certain diseases showed association with specific alleles specially those of known or suspected hereditary origin or immunological basis, whether simple renal cyst is congenital or acquired is still unclear and need to be investigated.Objectives: To study the genetic aspect of simple renal cysts by detecting the gene frequency and the haplotype of HLA class I of patients with simple renal cysts, and to find the presence of these cysts in other family members.Method: Thirty patients with simple renal cysts who were attending the outpatient clinic of urosurgery in the medical city were tested for HLA class I antigen using the microlymphocytotoxicity technique, in the period from February to June 2004 compared to 50 unrelated apparently healthy individuals. Gene frequency were calculated using square root formula (g=1-√1-f), full history were taken including the family history.Results: Certain gene frequencies were higher in the patients group than in the controls, yet not reached to a statistical significant level. No haplotype association with simple renal cysts was detected in this study; family history was detected in two patients which were proved by ultrasound examination.Conclusion: Increasing the sample size may contribute to best results regarding gene frequency, haplotype and family study.Key words: Gene frequency, Haplotype, Human Leukocyte Antigens.

Abstract

A total of 50 P. aeruginosa isolates are leading reasons of hospital acquired infections (burns) were collected from AL-Kindy Hospital. This study focused on the quick synthesis of silver nanoparticles by this bacteria as assay for periplasmic nitrate reductase enzyme. The silver nanoparticles were described by using Atomic Force Microscopy (AFM) and found the average size was 93.55nm. From the screening profile of 50 isolates enlisted in this study the percent of napA gene positive encoding periplasmic nitrate reductase was 100%. Results revealed that nitrogen bases sequencing for PCR product of one isolate correspond reaching up to 88% as compared with nitrogen base sequence of the napA gene of P. aeruginosa strain of National Center for Biotechnology Information (NCBI).
