Abstract

Diabetes mellitus is a group of metabolic diseasesdue to defect in insulin secretion or action or both.Hyperglycemia in diabetes creates free radicals .These free radicals produce oxidative stress and thus debilitate the endogenous antioxidant defense system.If the amount of insulin available is insufficient, cells response badly to the effects of insulin or if the insulin itself is abnormal. The net effect is persistently elevating levels of blood glucose, low protein synthesis.The current study evaluate the biochemical changes in diabetes mellitus patients in using different medications and investigate the glutathione S-transeferase M1 gene deletion in different treatments. The present study was conducted on (75) diabetic patients, (25) of them were treated with insulin, (25) were using insulin and metformin and the last (25) were on metformin and glibinclimide .The study also included (25) apparently healthy subjects were taken as control group. The blood sampleswere collected from Merjan Teaching Hospital from November 2013 to April 2014.The total serum protein was measured by colorimetric method described by Gornall.LSD test showed that there was significant difference between insulin & metformin group and metformin &glibinclimide in total serum protein (p ≤0.05). Metformin &glibinclimide group was less mean difference with the control group. In the gene level of the study, genotyping of glutathione S-transferase mu 1gene by PCR were defined as GSTM1 and GSTM0 or deletion association to the present and absences of the guanine nucleotide in the gene sequence. There was statistically significant difference in the genotyping distribution and the frequency of GSTM0 among study groups were 44% for insulin and metformin group, 68% for metformin and glibinclimide, 44% insulin and 28% for control healthy group.there was significant decrease in serum total protein in diabetic patients. Genetic polymorphism of glutathione S-transferase mu 1gene may be considered as risk factor for both types of diabetes mellitus.

Abstract

This study aimed to investigate the effect of administration of depakene (valproic acid)(VPA) (15 mg/Kg of body weight, orally) on serum protein levels, including immune proteins, because its functions and the importance of these proteins in the body and they are overlapping, and that was by measured protein levels before and after treatment as concentrations of total protein (Tp), albumin and globulin in normal male rats and in rats that treated with VPA drug along the period of experiment (30) days. The results showed a significant decrease (P<0.05) in concentrations of total protein and albumin in comparison with normal control group, whereas did not observe significant differences (P<0.05) in globulin concentration compared with normal control group, despite of, there was a little decrease in globulin concentration in rats group that administrated the drug under study but non-significant. It could be concluded of the present study that the VPA drug have an greater side effects on concentration and function of proteins in serum, and subsequently, may extending these effects to comprise its effect on the immunity, osmotic pressure, blood volume, nutrients exchange between the blood and tissues, glomerular infiltration rate, transport many of materials and hormones, and several of physiological and biochemical functions which correlated with the serum proteins.

استهدفت هذه الدراسة التحري ومعرفة تأثير إعطاء الديباكين (Valproic acid)(VPA) بجرعة (7.14 ملغم/كغم من وزن الجسم) عن طريق التغذية الانبوبية على مستويات البروتين في مصل الدم بضمنها البروتينات المناعية، وذلك لما يوجد من وظائف واهمية لتلك البروتينات في الجسم وبصورة متداخلة، وذلك تم من خلال قياس مستويات البروتينات قبل وبعد المعاملة كتركيز البروتين الكلي (Total protein)، الالبومين (Albumin) والكلوبيولين (Globulin) في مصل دم ذكور الجرذان البيض السليمة والمعاملة بعقار (VPA) وطيلة فترة التجربة البالغة (30) يوماً. النتائج اظهرت وجود انخفاض معنوي (P<0.05) في تركيزي البروتين الكلي والالبومين مقارنة مع مجموعة السيطرة السليمة. في حين لم يلاحظ فرق معنوي (P˃0.05) في تركيز الكلوبيولين مقارنة مع مجموعة السيطرة السليمة، بالرغم ان لوحظ انخفاض قليل في تركيز الكلوبين في مجموعة الجرذان التي اعطيت العقار قيد الدراسة لكن غير معنوي. وقد استنتج من هذه الدراسة ان لعقار (VPA) تاثيرات جانبية كبيرة على تركيز ووظيفة البروتينات في مصل الدم وبالتالي قد تمتد هذه التاثيرات ليشمل تاثيرها على المناعة والضغط الازموزي وحجم الدم وتبادل المواد الغذائية بين الدم والانسجة ومعدل الترشيح الكلوي، نقل العديد من المواد والهرمونات في الدم والعديد من الوظائف الفسلجية والكيموحيوية المرتبطة بالبروتينات في مصل الدم.

Total protein and some minerals in late stage of gestation and early lactation in dairy cows

Abstract

The study was conducted on 50 dairy crossbred pregnant cows from January until May 2014, for evaluating the total protein and some minerals parameters. Blood samples were collected from the jugular vein three times; at one week pre parturition, at parturition, and at a week after parturition, for measurement of serum total protein, calcium, and phosphorus, magnesium and iron concentrations. The results were indicated a significant (P˂0.05) decreases of total protein, calcium, phosphorus, magnesium and iron concentrations in calving period compared to pre calving and post calving periods. In conclusion, there was a significant decrease in minerals concentration (calcium, magnesium, phosphorus, iron) during pregnancy and lactation periods.

اجريت الدراسة على 50 بقرة من الابقار الحلوبة المضربة الحوامل وللفترة من شهر كانون الثاني حتى شهر ايار2014 وذلك لقياس البروتين الكلي وبعض المعادن في الابقار. جمعت نماذج الدم من الوريد الوداجي للأبقار في ثلاث فترات: سبعة ايام قبل الولادة , واثناء الولادة ، وسبعة أيام بعد الولادة وذلك لقياس البروتين الكلي في مصل الدم وقياس تراكيز الكالسيوم والفسفور والمغنسيوم والحديد. اظهرت النتائج انخفاض معنوي بتركيز البروتين الكلي والكاليسيوم والفسفور والمغنسيوم والحديد في فترة الولادة مقارنة بفترات قبل الولادة وبعدها. نستنتج مما سبق ان تركيز المعادن والبروتين الكلي ينخفض خلال فترة الحمل والولادة.
