In vivo histological evaluation of the effect of the topical application of estrogen hormone on wounds healing in ovariectomized rabbits

Abstract

Background: Wound healing, as a normal biological process in the human body, is achieved through four preciselyand highly programmed phases: hemostasis, inflammation, proliferation, and remodeling. Growth factors released inthe traumatized area promote cell migration into the wound area (chemotaxis), stimulate the growth of epithelialcells and fibroblasts (mitogenesis), initiate the formulation of new blood vessels (angiogenesis), and stimulate matrixformation and remodeling of the affected region. One of factors that effects on wound healing is a sex hormonesand one of these hormones is an estrogen hormone. A wide range of cutaneous cell types (eg, fibroblast,endothelial, epithelial, and inflammatory) expressed estrogen receptors, indicating potential estrogenresponsiveness.Materials and methods: Thirty two female New Zealand rabbits were used in this study. All animals wereovariectomized, and incisional wounds were done on the right (experimental for estrogen hormone application) andleft (control) sides of face for each animal, the control side was left to heal normally. Histological assessmentregarding the count of inflammatory cells was performed for healing intervals (3, 7, 10, 14 days).Results: Topical estrogen hormone application revealed enhancement of wound healing by reducing wound sizeand stimulating matrix deposition in comparison to control.Conclusion: Topical estrogen cream application results in significant progress of cutaneous wound healing, leavingno scar or crust formation and can minimize the probable wound complications

Abstract

Background: Herbal medicine can be called one of the branches of medicine in various forms. Turmericcurcuminhas proved its efficiencies a coloring, flavoring agent and has been traditionally used in medicine, exhibitingremarkable anti-inflammatory and antioxidant properties. The varied biological properties of curcumin and lack oftoxicity even when administered at higher doses makes it attractive to explore its use in various disorders like diseasesof skin. It is good potential agent for wound healing.Materials and methods: Sixty four new Zealand rabbits were used in this study ,they were divided into fourgroups,each group was subdivided as follows: Experimental groups(8 rabbits) right facial side of animals for essentialoil application and left facial side animals for curcumin powder application, 8 animals as control group(right facialsides) .Histological assessment regarding the count of inflammatory cells was performedon all studied samples for thehealing intervals (3, 7, 10, and14 days).Results: Histological findings of the study showed that re-epithelialization, wound contraction were accelerated aftertopical application of curcumin material especially the essential oil at wound site.Conclusion: Topical application of curcumin essential oil was significantly effective in skin wound healing ascompared to curcumin powder

Effect of Punica granatum rinds ethanolic extract on healing of fungated wounds in rabbits

Abstract

Pomegranate (Punica granatum) is an ancient fruit used in various systems of medicine for a variety of ailments. The study has been designed to investigate the role of ethanolic extract of rinds of Punica granatum as antimicrobial on 6 types of bacteria includes; Pseudomonas aeruginosa, Salmonella typhi, Staph aureus, E.coli, Streptococcus spp. and Klebsiella pneumonia and antifungal activity on 4 types of fungi includes; Candida albicans, Aspergillus niger, Aspergillus fumigates and Aspergillus valvas. Also the study build to assessment the ability of rinds ethanolic extract of Punica granatum to reduced the oxidative intention as a result of fungal infection and to accelerate the wound healing in normal and wound model infected with fungi using local rabbits as experimental animals. To achieve this goal 30 adults rabbits were used and divided into three groups, one as control group and the others as treated groups. Two wounds models via incision were make and infected with fungi. The first infected group was treated with ethanolic extract of Punica granatum and the second group treated with 0.5 g ointment of ethanolic extract of Punica granatum. The results of antimicrobial assessment exhibited significant antibacterial and antifungal activity against almost all tested bacteria and fungi, and the used of rinds ethanolic extract of Punica granatum for each treated groups (300 mg/kg and 500 mg/kg) also showed significant (P<0.05) improvement in percentage of wounds contraction and healing. In addition the oxidative stress was assessed by estimating the serum levels of antioxidant enzymes: malondialdehyde (MDA), catalase (CAT), glutathione (GSH), and SOD. Also the results shows that the treatment with ethanolic extract of rind of Punica granatum markedly prevented oxidative stress by increasing of catalase (CAT), glutathione (GSH) and decreasing the level of MDA and SOD. In conclusion the study revealed that use of rinds ethanolic extract of Punica granatum as a lotion and ointment improve the wounds healing properties.

أجريت الدراسة لمعرفة الفعالية التثبیطیة للمستخلص الایثانولي لقشور نبات الرمان granatum) (Punica اتجاه 6 أنواع بكتيرية متمثلة Pseudomonas pycoaneous و Salmonella typhiو Staph.aureusوE. coli و Streptococcus spp. و Klebsiella pneumoniaفضلا عن خمائر مثل Candida albicansوفطريات مثل Aspergillus niger و Aspergillus fumigatusو Aspergillus valvas وكذلك تهدف الدراسة إلى تقييم فعالية المستخلص الايثانولي لقشور الرمان في تقلل الإجهاد التأكسدي الناتج من الاصابة الفطرية وشفاء وإعادة بناء الجروح المستحدثة والمصابة بالفطريات في الأرانب المحلية. استخدم في هذه الدراسة 30 أرنبا بالغا تم تقسيمها إلى ثلاث مجاميع احدها مجموعة سيطرة والأخيرتان مجاميع معالجة وتم احداث الجروح واصابتها بالفطريات وتم علاج إحدى المجموعتين بسائل المستخلص الايثانولي والمجموعة الثانية بمرهم المستخلص الايثانولي. وأظهرت النتائج وجود تأثير تثبيطي معنوي بالنسبة للأحياء المجهرية كافة , وان استخدام المستخلص الايثانولي لنبات Punica granatum لكل المجموعات المعالجة (300 ملغ/كغ و 500 ملغ/كغ) قد أظهرت وجود تحسن معنوي(P<0.05) في النسبة المئوية لسرعة تقلص الجروح المعالجة وشفاءها. وتم خلال فترة المعاملة قياس مستوى الإنزيمات المضادة للأكسدة MDA,TBARS,CAT,GSH,SOD وأظهرت الدراسة إمكانية المستخلص الايثانولي في تقليل الاجهاد التأكسدي من خلال زيادة CAT و GSH ونقصان MDAو SOD. وأخيراً كشفت نتائج هذه الدراسة أن استخدام المستخلص الايثانولي لنبات Punica granatum كمحلول او مرهم يعمل على تحسين خصائص شفاء الجروح.

Abstract

The present study reports the potential effect of Punica granatum L. peels aqueous extract on the in vivo wound healing. A 10 % extract based-ointment was formulated and evaluated for its wound healing in rabbit comparing with topical use of honey and penicillin-streptomycin antibiotic. The ointment was applied in vivo on the paravertebral area of induced excisional wounds once a day for 10 consecutive days. The ointment significantly enhanced the wound contraction and the period of epithelialization as assessed by daily measurement of wound diameter and the histopathological characteristics. Such investigation was encouraged by the efficiency of the pomegranate peel extract as antimicrobial. Indeed, the extract exhibited significant antibacterial activity against isolated bacteria from induced contaminated and infected wounds (Staphylococcus aureus). The formulated ointment might well find use as skin repair agent without hazard to health based on these results and on the fact that it has been well established that the extracts of pomegranate used in conditions similar to those applied by traditional medicine, showed no toxic effects.

سجلت الدراسة الحالية تأثير فعال للمستخلص المائي لقشور الرمان على شفاء الجروح. حيث تم تحضير مرهم بتركيز 10% من المستخلص المذكور وقيم تأثير هذا المرهم على شفاء الجروح في الأرانب مقارنتا مع الاستعمال الموقعي للعسل ومع المضاد الحيوي البنسلين- ستريبتومايسين واسع الطيف. استعمل هذا المرهم بوضعه على الجروح الخزعية المستحدثة في المنطقة الظهرية وذلك بالاستعمال اليومي ولمدة 10 أيام متتالية. حيث عجل وبشكل ملحوظ عملية تقلص الجروح ومرحلة اعادة تكوين الخلايا الطلائية وكان ذلك التقييم من خلال القياس اليومي لقطر الجرح والوصف النسجي المرضي. أن فعالية مستخلص قشور الرمان كمضاد للجراثيم هي التي شجعت على أجراء هذه الدراسة. وبالفعل فأن المستخلص قد أظهر تأثير فعال ضد الجراثيم (Staphylococcus aureus) المعزولة من الجروح الملوثة والخمجة المستحدثة. ان من الممكن استخدام هذا المرهم لاصلاح الجلد وبدون اي مخاطر صحية وذلك استنادا لنتائج البحوث السابقة التي اكدت على ان مستخلص الرمان قد استخدم في حالات مماثلة في الطب التقليدي وبدون اي تأثير سمي.
