Salivary Physicochemical Characteristics in Relation to Oral Health Status Among Institutionalized Autistic Adolescents in Baghdad/Iraq

Abstract

Background: Autism spectrum disorder (ASD) is characterized by impairments in social interaction and communication, restricted patterns of behavior, and unusual sensory sensitivities. Saliva may provide an easily accessible sample for analysis. Some salivary constituents levels altered in adolescents with ASD including antioxidants . This study aimed to investigate salivary physicochemical characteristic in relation to oral health status among adolescents with ASD.Materials and methods: Two groups were included in this study: 40 institutionalized autistic adolescents and 40 apparently healthy school adolescents control group with age range (12-15 years old, only males) selected randomly from Baghdad. Each group subdivided into two groups according to the severity of dental caries: caries free group (20 child, DMFT=0) and high caries group (20 child, DMFT≥6). Decayed, missing and filled surfaces (DMFS), plaque (PlI), Gingival (GI) and calculus (CI) indices were used to measure oral health status for both groups. Copper (Cu), zinc (Zn) and thiocyanate (SCN) in saliva measured by atomic absorption spectrophotometer. Salivary alpha amylase (sAA) and glutathione (GSH) assessed by enzyme-linked immunosorbent assay (ELISA). Salivary pH and flow rate were measured directly. The data of current study was analyzed using SPSS version 21.Results: A higher value of salivary pH, flow rate, sAA, SCN, Cu and Zn were found among study group than control group with significant difference, also higher in caries free subgroup than high caries subgroup. While GSH was significantly higher in control group than study group. Moderate negative correlations between sAA, Cu, Zn and PlI, CI, GI with highly significant and salivary pH correlate moderately with PlI and CI with highly significant.Conclusion: There was alteration in salivary constituents levels which related to oral health status in adolescents with ASD and can act as adjunctive diagnostic aid for diagnosing autism.

Salivary and Serum Oxidant/Antioxidant Level in Behçet’s Disease Patients

Abstract

Background Behçet’s disease (BD) is a multi-systemic inflammatory disorder characterized by recurrent oral and genital ulceration. Free oxygen radicals may have a role in the pathogenesis of BD and can relate to the existence of the disease.Objectives This study had been designed to assess salivary and serum lipid per-oxidation as malondialdehyde (MDA) and status of antioxidant as total glutathione (GSH) in patients with BD in comparison with healthy subjects to detect the possible involvement of oxidative stress in BD. Materials and methods This is a case–control study, salivary and serum samples were taken from ffty BD patients with an age range 20 to 56 years with mean age 35.3 ± 7.6 years and ffty healthy control subjects with age range 16 to 58 years and mean age 34.8± 9.1 years. All were analyzed for MDA and GSH spectrophotometrically. Data was analyzed using descriptive statistics, t-test, P (ANOVA) test, Chi-square (2X) test, Pearson correlation and Receiver Operating Characteristic statistical analyses. Results The study showed that salivary and serum MDA level was higher in BD patients than in healthy subjects (P<0.05 and P<0.001 respectively). The levels of saliva and serum total GSH were signifcantly lower in BD patients than in healthy subjects. (P <0.001). Conclusion The results of this study revealed that BD patients were subjected to oxidative stress damage and saliva could be used as pain-free alternatively to serum in determining oxidative stress in Behçet’s disease.

Growth inhibition and apoptosis induction in two types of women cancer cell lines using laser ablated silver nanoparticles in polyvinylpyrrolidone solution

Abstract

Silver nanoparticles (AgNPs) coated with polyvinylpyrrolidone (PVP) was prepared by pulsed laser ablation method inPVP solution, 1000 pluse was used with laser energy of 800 mJ/pulse. Formation of AgNPs-PVP was confirmed by UVvisiblespectrophotometer detecting surface plasmon resonance chang in the abilation solution. Concentration of AgNPs, sizedistribution, and surface charge of the ablated AgNPs in the produced solution was determined using atomic absorbtion spectroscopyand zeta potential analysis respectivly. Nanoparticles shape was determined using transmission electron microscopeimaging. Ability of the senthezied AgNPs to inhibit the growth of cancer cells was compared to that of normal fibroblast cellsusing two cancer cell lines (HeLa and SKOV-3) in different concentrations (0.78, 1.56, 3.125, 6.25, 12.5 and 25μg/ml). Theapoptosis events were also detected in both types of cancer cell lines using mitochondrial membrane permeability, nuclearmorphology, and genomic DNA fragmentation. The contribution of AgNPs to the levels of glutathione s-transferase was alsodetermined. According to the results, AgNPs size was 28.43nm with spherical shape and peak UV-Vis absorbance rangedbetween 402-410nm. Concentration of AgNPs in ablated solution was 50μg/ml and used as stock concentration in cell linesexperiments. Ablated silver nanoparticles was capable of inhibit the growth of both HeLa and SKOV-3 cancer cell lines indose and time dependent manner, whereas it was less inhibitory against normal fibroblast cells. All three apoptosis detectionmethod conducted indicated positive characteristics for apoptosis, while GSH levels did not varied significantly in both HeLaand SKOV-3 cells.

بطريقة فيزيائية من خلال الازالة بالليزر النبضي وبطاقة ) 800 ( ملي جول/نبضة. تم توصيف جسيمات الفضة )PVP( صنعة جسيمات فضة نانوية مغلفة بمادةالمصنعة باستخدام مطياف الاشعة الفوق البنفسجية وقياس جهد الزيتا وتم قياس تركيز الجسيمات داخل المحلول المحضر بواسطة طيف الامتصاص الذري. كماتم توصيف شكل وحجم جسيمات الفضة المصنعة باستخدام المجهر الالكتروني. جرى مقارنة تاثير جسيمات الفضة النانوية على نوعين من الخطوط السرطانية)سرطان عنق الرحم وسرطان المبيض( مع خط الخلايا الطبيعي )خلايا جنين الفار(. اظهرت النتائج بأن متوسط حجم جسيمات الفضة النانوية المصنعة كانت402-410 (, وان التركيز الكلي لجسيمات الفضة المصنعة كانت nm( وذات شكل كروي تقريبا, كما اظهرت رنين بلازمون سطحي يتراوح بين )nm 28,43(وبفترات حضن ) 24 و 48 )μg/ml عوملت الخلايا بعدة تراكيز متدرجة لجسيمات الفضة النانوية ) 0,78 و 1,56 و 3,125 و 6,25 و 12 و 5 و 25 .)μg/m 50(لها تاثير تثبيطي عالى على كل من خلايا سرطان الرحم والمبيض اعتمادا )PVP( و 72 ساعة(. بينت فحوصات السمية, ان جسيمات الفضة المصنعة المغلفة بمادةخلال 24 ساعة من التعريض )IC على التركيز والوقت, بينما اظهرت مستوى تثبيطي واطئ على خلايا الفأر الطبيعية. تم تحديد التركيز المثبط النصفي ) 50على التوالي, بينما لم يكن هناك مستوى تثبيطي نصفي على خلايا الفأر الطبيعية. )μg/ml و 12,5 μg/ml في كل من خلايا سرطان الرحم والمبيض ) 3,75على كل من خلايا سرطان الرحم والمبيض. وقد اظهرت النتائج بان جسيمات الفضة النانوية المصنعة قد حثت )IC اختبرت سمية التركيز المثبط النصفي ) 50كثيرا مقارنة GSH بينما لم يتغير مستوى ,DNA على حدوث الموت المبرمج في المايتوكوندريا وفي انوية الخلايا وكذلك حدوث تقطع في مادة الخلايا الوراثيةفي علاج انواع مختلفة من السرطانات. )PVP( بخلايا السيطرة )الغير معاملة(. تشير هذه النتائج الى امكانية استخدام جسيمات الفضة المصنعة والمغلفة بمادة
