Spectrophotometric determination of progesterone and dopamine in breast cancer serum

Abstract

Dopamine (DA) is one of the most important catecholamine neurotransmitters in the human central nervous system in the brain and plays a key role in the functioning of the renal, hormonal, and cardiovascular systems. Abnormal release of DA will contribute to some diseases such as Alzheimer’s and Parkinson's disease. Therefore, the sensitive determination of DA becomes increasingly significant in the field of clinical disease diagnosis and the research of physiological functions. Progesterone is an essential for the development and cyclical regulation of hormone responsive tissues including the breast and reproductive tract. In the breast, progesterone acts in concert with estrogen to promote proliferative and pro-survival gene programs. Progesterone has actions in breast cancer.This paper shows a spectrophotometric method for determination dopamine and progesterone concentration in serum samples, fifty six veins blood samples collected from healthy control and breast cancer patient groups. The analytical data obtained by using UV-Spectrophotometer (max = 250, 266 nm), linearity (0.5 - 2.0, 0.25 - 1.5 ng/ml) for dopamine, progesterone respectively. The molar absorptivity (), correlation coefficient (R2) and limit of detection (LOD) for dopamine, progesterone ( =2.5156  106, 4.1935 106 L.mol-1 cm-1), (R2= 0.9979, 0.99957) and (0.64, 0.57 ng/ml) respectively. The results show significant differences between the concentrations of dopamine and progesterone in control and breast cancer patient groups (p0.05).It is fast, sensitive, selective and reliable quantification spectrophotometric method used in complex biological samples.

الدوبامين هو احد اهم النواقل العصبية في الجهاز العصبي المركزي في دماغ الأنسان و يلعب دور اساس في وظيفة الجهاز ( الكلوي ، الهرموني و الوعائي القلبي). ان الإفراز غير الطبيعي للهرمون يؤدي الى امراض مثل الزهايمر ومرض باركنسون، لهذا إزدادت اهمية تقدير الدوبامين في مجال تشخيص الأمراض السريرية وبحوث الوظائف الفسيولوجية.البروجستيرون له دور اساسي في التطوير والتنظيم الدوري لإستجابة الأنسجة كالثدي والقنوات اللبنية للهرمون ، ففي الثدي يعمل مع الأستروجين لتطوير التكاثر والمحافظة على بقاء برامج الجين كما ان للبروجستيرون دور في سرطان الثدي. يوضح هذا البحث الطريقة الطيفية لتقدير الدوبامين والبروجستيرون في نماذج مصل الدم حيث جمعت 56 عينة دم وريدي لكل من مرضى سرطان الثدي واخرى للأصحاء. اوضحت النتائج التحليلية التي توفرت بأستخدام جهاز المطياف الضوئي عند الطول الموجي الأعظم (250،266) نانوميتر وخطية ضمن المدى ( 0.5-2.0 و 0.25-1.5) للدوبامين والبروجستيرون على التوالي. معامل الإمتصاص المولاري ، معامل الإرتباط وحد الكشف، ( 0.9979 ، 0.9995) و ( 0.64، 0.57) ( لتر مول-1 سم -1 4.1935  106, 2.5156 106) للدوبامين والبروجستيرون على التوالي.اوضحت النتائج وجود اختلافات معنوية بين تركيز الدوبامين والبروجستيرون في مجموعة مرضى سرطان الثدي ومجموعة السيطرة وهي طريقة طيفية سريعة وحساسة ويمكن الوثوق بها واستخدامها عند التقدير الكمي لنماذج بيولوجية معقدة.

Abstract

An accurate and sensitive spectrophotometric method has been developed for the determination of cefotaxime (CEF) in pure and pharmaceutical samples. The suggested method depended on the coupling reaction between diazotized cefotaxime and 3,5-dimethyl phenol (3,5-DMPH) in basic medium to form light orange, water soluble dye, that is stable and has a maximum absorbance at 497nm. The calibration graph was liner over the concentration range (1-70) μg.mL-1 with LOD of 0.750 μg.mL-1 and LOQ of. 2.740 μg. mL-1, sandal sensitivity of 0.0526 μg. cm-2 . molar absorptivity 11328 Lmol-1 cm-1 . The stoichiometry composition was found by Jobs and mole ratio methods to the formation dye (1:1). The suggested method has been applied successfully to the determination of CEF in pure and pharmaceutical synthetic sample.

Abstract

A simple, rapid and sensitive spectrophotometric method for the determination of baclofen in pharmaceutical preparations were developed by combining a spectrophotometric detector with Flow injection analysis. This method based on the oxidation of iron (II) to iron (III) by baclofen and formation of complex between iron (III) and potassium hexacyano ferrate (II) in acidic solution. The absorbance from prussian blue dye is recorded at 700 nm. Chemical and physical parameters of this system were investigated. The linearity of baclofen is ranged from (0.05-25) mmol.L-1 with correlation coefficient r = 0.9972. The limit of detection (S/N=3) was equaled 0.01 mmol.L-1 the L.O.Q was 1.05 mmol.L-1 and a relative standard deviation for 6 replicate determinations of baclofen in 10 mmol.L-1 solution was 1.3 % using 100 µL sample volume. The proposed method was successfully applied to the determination of baclofen in pharmaceutical formulation with recovery 99%. Using paired t-test it was shown that there was no significant difference between the proposed method and official method on that basis the proposed method can be accepted as an alternative analytical method. Abstract

Simple Spectral method was developed to estimate the methyl and propyl paraben through charge- transfer (CT) complexes. The method involves the interaction of parabens compounds with o-chloranil as π-acceptor. Under the optimized conditions, the complexes were found to be absorbed at 415 and 385 nm with in the linearity range of 5-40 and 5-55 μg ml-1 respectively. The molar absorptivity values were in the range 3803.75 and 2342.6 L.mol-1.cm-1 respectively. And the lower limit of detection were in the rang 0.073 and 0.086 µg/ml-1 for methyl and propyl respectively. The stoichiometry of the paraben compounds o-chloranil complexes was found to be 1:1 method. The procedure was characterized by its simplicity with accuracy and precision. The proposed method was applied successful in some detergents (shampoo and lotion) containing paraben compounds.

Abstract

Three simple, inexpensive and nontoxic spectrophotometric methods have been used for determination folic acid in pure and market formulation tablets. Linearity was founded in the range 6- 20 mg.l-1 for the all three methods, the detection limits was found to be 0.261 for zero order method, 0.006 for AUC method and 3.800 for first derivative method. The RSD% was found to be less than 0.856 indicating a good accuracy and precision of three proposed methods. The results of zero order, AUC and first derivative methods were statistically compared with those obtained by the official standard method using the F-test and t-test and found to be a good agreement.

Abstract

A newly developed FIA-merging zones spectrophotometric system, the method is rapid, accurate and sensitive for metformin hydrochloride determination through the oxidation of 1- naphthol by sodium hypochlorite and coupling with metformin.HCl in the presence of sodium hydroxide to form a blue soluble ion pair and this product was determined using homemade CFIA-Merging zones techniques , at 580 nm. Data treatment shows that linear range is (0.5-35) μg/ ml. The optimization conditions for various chemical and physical conditions of [MTF- NaOCl-α-naphthol-NaOH] system were investigated. The LOD was 0.01μg / ml and LOQ 0.1μg/ml from the lowest concentration of the calibration graph with r2% 99.18 and RSD% did not exceed 3%, sample through put 48 sample/h while percentage recovery (Rec.%) were from 97-100% , indicate no interferences of the tablet excipients. The method was applied successfully, for estimation of metformin in environmental samples of water and pharmaceutical drugs. Statistical analysis of results was compared, shows the method can be regarded as an alternative analytical way for determining of metformin. hydrochloride in bulk and dosage forms samples

Abstract

A simple, accurate and sensitive spectrophotometric way is used to determine Bisacodyl in pure and pharmaceutical preparations. The proposed method depends on using 2,4-Dinitrophenylhydrazine as chromogenic reagent . The method was based on the oxidative coupling reaction of Bisacodyl with 2,4-Dinitrophenylhydrazine with Sodium periodate in the presence of sodium hydroxide as alkaline media to form red water soluble dye product , that has a maximum absorption at λmax 522nm . Beer ,s law is obeyed in the concentration of (2.00–20.00) μg.ml -1 .The molar absorptivity is (6505) L.mol-1.cm-1,a sandall sensitivity of(0.0555) μg.cm-2), correlation coefficient of (0.9970) , Limitof detection (LOD) (0.0312 μg.ml-1), limit of Quantitation (LOQ) (0. 3125 μg.ml-1) and the relative standard deviation of RSD% (1.6). The method gave a successful determination for Bisacodyl in pharmaceutical preparations and the value of recovery % was better than (100.16%) .

يتضمن البحث تطوير طريقة طيفية بسيطة ومضبوطة وحساسة لتقدير العقار بسكودايل في صيغته النقية وفي مستحضراته الصيدلانية.الطريقة المقترحة تعتمد على تفاعل ازدواج البسكودايل مع الكاشف اللوني داي نايتروفنيل هيدرازين وبوجود بيرايودات الصوديوم وهيدروكسيد الصوديوم كوسط قاعدي حيث يتكون مركب لونه احمر يعطي اعظم امتصاص عند الطول الموجي 522 نانوميتر.النتائج اظهرت ان مدى الخطية بين 2.00-20.00 مايكروغرام/مل ومعامل امتصاص مولاري مقداره((6505) لتر.مول -1 سم -1ودلالة ساندل مقدارها(0.0555) مايكروغرام.سم -1 ومعامل ارتباط (0.9970) وحد كشف نوعي للطريقة (0.0312) مايكروغرام/مل ،وحد كشف كمي (0.3125) ومعدل انحراف قياسي نسبي (1.6).الطريقة طبقت بنجاح لتقدير البسكودايل في المستحضرات الدوائية وبنسبة استرجاعية افضل من (100.16%).

Abstract

The research includes, spectrophotometric determination for Metronidazole drug (MND) in pharmaceutical form (tablet), using multi-wavelengths technique, the results obeyed Beer’s law in the concentration ranges of between (0.5-25 µg.ml-1), recovery percentage Rec% (99.99- 100.05), RSD% (0.0011-0.0019), and detection limit (0.2042-2.9772 µg.ml-1) This method been successfully applied to the determination of MND in pharmaceutical form tablet. The results also showed there is no interference from additives.

تضمن البحث تقدير عقار الميترونيدازول طيفيا في شكله الصيدلاني (حبوب) باستخدام تقنية الاطوال الموجية المتعددة، وبينت النتائج مطاوعة لقانون بير-لامبرت بمدى خطية تراوح بين (0.5-25 مكغم.مل-1)، كما وبلغت الاستعادية المئوية (99.99-100.05)، وقيمة الانحراف النسبي المعياري بين(0.0011-0.0019)، وتراوحت قيمة حد الكشف بين (0.2024-2.9772 مكغم.مل-1)، وتم تطبيق الطريقة بنجاح على المستحضر الصيدلاني، كما واشارت النتائج أيضا الى عدم وجود تداخل من قبل المضافات الدوائية.

Abstract

A sensitive, rapid and economical spectrophotometric processes for estimationof two drugs; methyldopa (MTD) and Fluvastatin Sodium (FLV), by “Sodium 1,2-naphthoquine- 4-sulfonate (NQS)” as reagent in an alkaline interemediate. Thesemethods are digested on the forming of “complexes” among these drugs and thechromogenic reagent (NQS). Orangish-brown colored complex formatted at (pH12) and λmax. 462 nm for (MTD), and orange colored at (pH 11.7) and λmax. 466 nmfor (FLV). Beer’s Law is obeyed in a concentrations range of (4-40 μg/mL), (16-64 μg/mL), with molar absorptivity (14.101×104 L/mol.cm), (22.7×104 L/mol.cm),and correlation coefficient 0.9995, 0.9987, successively, The detection limit andquantification limit were (1.03×10-1 μg/mL, 3.12×10-1 μg/mL ), and (1.36×10-1μg/mL, 4.12×10-1 μg/mL ), successively. The suggested methods were prosperityimplement to the estimation of “these drugs” in pure forms and in theirpharmaceutical formulations.

استخدمت طرائق حساسة وسريعة واقتصادية لتقدير عقاقير مختلفة وهي: مثيل دوبا، وفلوفاستاتين صوديوم، باستخدام كاشف صوديوم 2،1- نفثوكينون-4-سلفونيت في الوسط القاعدي، هذه الطرائق تعتمد على تكوين معقدات بين تلك العقاقير والكاشف الكروموجيني (NQS). يتكون معقد برتقالي-بني اللون عند (pH 12) وله أعلى امتصاص عند 462 نانومتر للمثيل دوبـا، ومعقــد برتقالي اللون وعند (pH 11.7) وله أعلى امتصــاص عند 466 نانومتر للفلوفاستاتيـــن صوديــوم، طُبق قانون بير في مدى من التراكيز (4-40 µg/mL)، (16-64 µg/mL)، وبامتصاصية مولارية (14.101×104 L/mol.cm)، (22.7×104 L/mol.cm)، ومعامل ارتباط 0.9995، 0.9987، على التوالي، وكان حد الكشف وحد الكمية (1.03×10-1 µg/mL, 3.12×10-1 µg/mL)، (1.36×10-1 µg/mL, 4.12×10-1 µg/mL) على التوالي. طُبقت الطرائق المقترحة بنجاح في تقدير هذه العقاقير بشكلها النقي وفي مستحضراتها الصيدلانية على شكل (أقراص).

Spectrophotometric Determination of Cu(II) by Complex with Ethyl Cyano(2-Methyl Carboxylate Phenyl Azo Acetate) (ECA)

Abstract

A new simple and sensitive spectrophotometric method for the determination of trace amount of Cu(II) in the ethanol solution have been developed. The method is based on the complexation of Cu(II) with ethyl cyano(2-methyl carboxylate phenyl azo acetate) (ECA) in basic medium of sodium hydroxide givining maximum absorbance at (λmax = 521 nm). Beer's law is obeyed over the concentration range (5-50) (μg / ml) with molar absorptivity of (3.1773 × 102 L mol-1 cm-1) and correlation coefficient (0.9989). The optimum conditions for the determination of Cu(II)-complex and have been studied and applied to determine Cu(II) in synthetic water sample using simple and standard addition methods.

تم تطوير طريقة طيفية جديدة بسيطة وحساسة لتقدير كميات قليلة من النحاس الثنائي في المحلول الكحولي. تعتمد الطريقة على تكوين معقد بين النحاس الثنائي و اثيل سيانو (2- مثيل كاربوكسليت فنيل ازو خلات) في وسط قاعدي من هيدروكسيد الصوديوم الذي يعطي اعظم امتصاص عند الطول الموجي 521 نانوميترا". وجد انه يطيع قانون بير ظمن التراكيز (5-50) مايكروغرام / مللتر وقيمة الامتصاصية المولارية (3.1773 × 102) لتر مول-1 سم-1 ومعامل الارتباط (0.9989). تم دراسة الظروف المثلى لتقدير المعقد وطبقت الطريقة لتقدير النحاس في نموذج مصنع من الماء وباستعمال الطريقة الاعتيادية وطريقة اضافات القياس.
