Abstract

Implantation of the blastocysts into the maternal uterus is a crucial step in mammalian reproduction, which is controlled by a number of complex molecules like hormones, cytokines, and growth factors and their cross talk. A network of these molecules plays a crucial role in preparing receptive endometrial and blastocysts. This study aimed to found out the role of Leukemia inhibitory factor (LIF) , gene expression, concentration and genetic polymorphisms in the endometrial that may interfere with implantation process of polycystic ovary syndrome(PCOS) and non-PCOS women. A convenient blood (6ml)sample of 80 infertile women undergoing in vitro fertilization (IVF) program were intentionally divided according to the cause of infertility into 40 healthy women their husbands complaining from male infertility factors, and 40 infertile women with polycystic ovary syndrome. Leukemia inhibitory factor( LIFs) were measured on ovulatory and luteal phase of cycle (CD14-CD16,17) at the day of ovarian pickup and embryo transfer by using quantitive polymerase chain reaction(qPCR ), Elisa technique ,polymerase chain reaction(PCR) and sequencing to determine any genetics polymorphisms in LIFgene study. Results of the present study indicate that no mutation was detected in LIF gene in the healthy and PCOS women. The LIF gene expression in addition to levels of serum leukemia inhibitory factors are more valuable in predicting the pregnancy out come in infertile PCOS women than in non-PCOS women .This result can be utilized to be used as predictors of implantation window for successful implantation and pregnancy.

Abstract

The aim of this search was to study the levels of follistatin and a number of sex hormones of polycystic ovary syndrome patients (PCOS).The study included 70 blood samples of women infected with PCOS and 20 samples from healthy women as control. The study had been perfigureed in Baghdad / Kamal al-Samarrai Hospital. The patient had been diagnosed with ultrasound waves (sonar device) by specialized physicians in the hospital. The research requirements for the period from September 2016 to March 2017. The results showed high significant difference (p <0.01) in the concentration of follistatin in women with PCOS compared with healthy women. On the other hand, there was a significant decrease (p <0.01) in the concentration of follicle stimulating hormone (FSH) in women with PCOS compared to control. The results showed high significant increase (P<0.01) in the concentrations of prolactin hormone and testosterone hormone in patients with PCOS compared to control.

الهدف من هذه الدراسة هو بحث مستوى الفوليستاتين وعدد من الهرمونات الجنسية في متلازمة تكيس المبايض لدى مجموعة من النساء 70)) امرأة مصابة بتكيس المبايض مع عدد من النساء السليمات(20) امرأة كسيطرة. وكان موقع الدراسة في مستشفى كمال السامرائي/ بغداد . وقد تم تشخيص المرضى بجهازفوق الامواج الصوتية (السونار) من قبل الطبيب المختص. واستمرت مدة الدراسة من شهر ايلول 2016 إلى شهر اذار 2017 أظهرت النتائج وجود ارتفاع عالي المعنوية (P<0.01) في تركيز الفوليستاتين لدى النساء المصابات بالمرض مقارنة مع النساء السليمات من ناحية أخرى، كان هناك انخفاض عالي المعنوية P<0.01) ) في تركيز الهرمون المحفز للجريبات (FSH) في النساء المصابات بمتلازمة تكيس المبايض مقارنة مع السيطرة. أظهرت النتائج وجود ارتفاع عالي المعنوية (P<0.01) في تركيز هرمون البرولاكتين و هرمون التستوستيرون في المريضات اللاتي يعانين من متلازمة تكيس المبايض مقارنة بمجموعة السيطرة.

Abstract

ABSTRACT: Background Polycystic Ovary Syndrome - PCOS- is the most common endocrine disease that affects women of reproductive age. The Aim is to assess the levels of thyroid antibodies in women with PCOS in Iraq, and to learning the link between PCOS and thyroid hormones. Patients & Methods: Two study groups have been investigated; the first group consists of one hundred and five infertile women with PCOS. The second group is healthy control which consists of forty six healthy fertile women. Hormonal analysis was performed by using a ready kit in accordance with the manufacturer's instruction (Biomerieux SA Etoile-France) and minividas full automated system (Triiodothyronine) .Thyroxine is performed by a one-step enzyme immunoassay competition method with a final fluorescent detection (ELIFA) (Enzymelinked immunoflourecent assay) and minividas full automated system (Biomerieux SA Etoile- France). Thyroid stimulation hormones , anti -thyroid peroxidase autoantibodies ,anti-TPO and anti -thyroglobulin antibodies Tg is performed by means of immunoassay sandwich with a final fluorescent detection (ELIFA) method with the use of a ready kit in accordance to the manufacturer's instruction (Biomerieux) and MiniVidas Full automated system (Biomerieux). The Results: It was found a highly significant difference (p< 0.01) between the thyroid stimulation hormone means (7.03± 13.45) μIU/ml of the infertile women with a mean of (3.51±4.43) μIU/ml of the control group, while the mean of Triiodothyronine and Thyroxine was found a slightly significant difference (0.05>p>0.1),). A positive result was found for thyroid peroxidase antibody (TPO-Abs)in infertile women (19.95 1± 39.53) IU/ml was higher than that of the control (4.82±4.7 ) IU/ml; anti-Tg autoantibodies in infertile women were (31.31±65.86) IU/ml compared to control (11.75±27.30) IU/ml with a significant difference(P=0.001).

Abstract

ABSTRACT: Background Endometrial hyperplasia is relatively a common gynecological condition that affect women of all age groups. The aim was to find out the effect of some factors and diseases associated with endometrial hyperplasia in Salahuddin province during January 2003-December 2013. Material & Methods: Random samples included 316 women attended Tikrit general hospital for curettage, hysterectomy, dilates and endometrial biopsy were collected. Results: There are many factors and diseases which are associated with endometrial hyperplasia as weight, medication, abortion, hypertension, diabetes and polycystic ovarian syndrome (PCOS). Hypertension, diabetes and PCOS affect significantly more women with endometrial hyperplasia. Conclusion:In conclusion, E.H is a multifactorial case and not depending on one factor. There are several factors affecting the E.H. Among these are the weight, age, as well as some diseases. Recommendation:To carry out more studies related to E.H. in the province of Salahuddin. The effect of some medication as hormonal replacement may clear out more the condition with E.H.

Abstract

Follistatin has been identifiedas a nominee gene for polycystic ovary syndrome (PCOS) from relationship studies.Acting to control the developmentof ovarian follicles and as an antagonist to aromatase action, changes in Follistatinrole or expression may affect main features of PCOS alteration.The aim of the recent study was to study the association of FST gene to define if genomicdifferencesarerelated with phenotypic structures of PCOS patients in a case–control suggestion study.Methods:This study involved 30 women with PCOS and 20 healthy women, whowerematched by their ageand anthropometric and biochemical profiles.The study was designed to examine theseparation of deoxyribonucleic acid (DNA), and genotype analysis was done for all the study participants using PCR-SNP.Results:Polymorphisms (SNPs) in the exon 6 FST gene region were determined for PCOS patients and controls, it was found that PCOS patientshad17 SNP in different samples, and after performing amino acid sequences alignment for these SNPs, it was observed that the last two SNPs of sample C12 constituted a non-synonym mutation, and the functional conformation of the produced FST gene weremodified with regard to exon 6 in the genotype C12. When this exonic SNP appears in the three-dimensional structure of the resulting proteins of C12 genotypes, the positively charged amino acid, Asp, is converted into another positively charged amino acid, His, in position 331 of the whole amino acid sequence of FST protein.The conclusionis that this, in turn doesn’t change the final phenotypic manifestation of the protein, and there was no alteration in the phenotypes of the PCOS patients.

يعرف الفولستاتينعلى انه من اكثر الجينات ارتباطا بمرض تكيس المبايض اذ انه يعمل على السيطرة على نمو الحويصلات المبيض وكذلك السيطرة على فعالية انزيم الاروماتيز وان أي تغيير في هذا الجين يؤدي الى زيادة امراضية النساء المصابات بتكيس المبايض وان الهدف من الدراسة الحالية هو دراسة العلاقة بين الجين المذكور ومرض تكيس المبايض مقارنة مع النساء الطبيعيات.تضمنت هذ الدراسة 30 امراة مصابة بتكيس المبايض و20 امراة غير مصابة و تمت المقارنة بينهن من الناحية المظهرية وقياس المعايير الهرمونية وكذلك دراسة التغيرات الجينية.لقد وجد من نتائج الدراسة ان النساء المصابات بتكيس المبايض يعانين من الزيادة في الوزن وكذلك ظهور شعر في مناطق غير مرغوب بها وزيادة في هرمون اللوتيني ونقصان في الهرمون المولد للحويصلات اما من الناحية الجينية فقد لوحظ وجود 17 تغير جيني عند النساء المصابات بتكيس المبايض وبعد مطابقة هذه التغيرات الجينية مع التغيرات المحتملة في البروتين وجد ان فقط 2 من التغيرات الجينية التي تسمى الطفرة الوراثية الصامتة التي تؤدي الى التغير من بروتين موجب الشحنة الى بروتين موجب الشحنة ايضا الا انه هذا التغير لا يؤثر على التغيرات المظهرية او امراضية تكيس المبايض .
