16- DETECTION OF BRUCELLA SPECIES IN APPARENTLY HEALTHY COWS AND GOATS RAW MILK BY PCR

Abstract

Brucellosis an intracellular pathogen capable of infecting animals and humans.The aim of this study was to identify Brucella spp in apparently healthy cow and goatraw milk samples by a polymerase chain reaction (PCR)-based method. A total of 75cow milk samples(55 direct and 20 indirect) and 50 goat milk samples wereexamined by PCR. To establish a PCR protocol for diagnosis of brucellosis, DNAwas extracted from the milk samples by using a commercial kit. PCR amplificationwas done for detection of Brucella DNA using BCSP31 target gene and IS711 locus.The PCR assay showed that an amplicon of 223 bp was obtained in 28% (21/75) and8% (4/50) samples of cow and goat tested milk using primers (B4/B5) derived fromaBCSP31gene encoding the 31-kDa Brucella abortus antigen. In another PCR, anamplicon of 498 bp was obtained in 100% (21/21) of the brucella genus BCSP31-PCRpositive cow milk samples using Brucella abortus-specific primers derived from alocus adjacent to the 3’-end of IS711, and also an amplicon of 731 bp was produced in100% (8/8) of the BCSP31-PCR positive goat milk samples using Brucellamelitensis-specific primers.PCR positive results for brucella genus and brucellaabortus species were observed in all (100%) cow,s milk samples(n=11) with clinicalhistory of abortion.As a single PCR product, no B. abortus was detected in goat milksamples alsoB. Melitensis was not detected in cow milk samples

Abstract

The aims of the study were to determined incidence ratio of buffalos brucellosis in Salahaldeen governorate and evaluating the efficiency of serological tests used in current study. For this purpose 135 buffalos (adult males, pregnant females and aborted females) were tested.From each animal two blood samples were collected, one for PCR test and other for serological tests (Rose Bengal test, tube agglutination test, mercaptoethanol test and ELISA test).The incidence ratio of buffalo brucellosis in Salahaldeen governorate were 14.8% in PCR test, while in serological tests were 21.4%, 13.3%, 16.2% and 7.4% for rose Bengal test, tube agglutination test, mercaptoethanol test and ELISA test respectively. Highest positive and negative agreement recorded between rose Bengal test and mercaptoethanol test were 34.9% & 63.7% . The current study recorded highest sensitivity recorded in rose Bengal test (75%), highest Specificity in ELISA (97.1%). Highest postive predictive values s and negative predictive values s recorded by tube agglutination test were: 77.7% and 96.2%

هدفت الدراسة تحديد انتشاء الاصابة بداء البروسيلات في الجاموس في محافظة صلاح الدين وتحديد كفاءة الاختبارات المصلية المستخدمة في التشخيص، ولهذا الغرض اخضع للدراسة 135 (اناث مجهضة ، حوامل، اناث وذكور بالغة). من كل حيوان اخذ نموذجين من الدم : الاول لاجراء اختبار تفاعل البوليميرات المتسلسل والثاني فصل المصل منه واجري عليه اربع الاختبارات المصلية (اختبار وردية البنكال، اختبار التلازن الانبوبي، اختبار المركابتوايثانول واختبار الاليزا) .بلغت النتائج الايجابية الكلية اعتمادا على اختبار تفاعل البوليميراز المتسلسل 14.8% اما الاختبارات المصلية فقد اعطت نتائج ايجابية بلغت 21.4% و 13.3% و 16.2% و7.4% لاختبار وردية البنكال, واختبار التلازن الانبوبي واختبار المركابتوايثانول واختبار الاليزا على التوالي. سجلت اعلى نسبة توافق بين اختبار وردية البنكال واختبار المركابتوايثانول اذ بلغت ، 34.9% و 63.7% للتوافق الايجابي والسلبي على التوالي. اعطى اختبار وردية البنكال اعلى حساسية بلغت 75% واعطى اختبار الاليزا اعلى نوعيةً بلغت 97.1% وقد كان اختبار التلازن في الانابيب الاعلى على مستوى التنبؤ الايجابي والسلبي اذا بلغا 77.7% و 96.2% على التوالي.
