Evaluation of the effect of local exogenous application of osteopontin on wound healing in rats

Abstract

Background: Wound healing is a complex dynamical interaction between various cell types, the extracellular matrix, cytokines, and growth factors. osteoponetin is a substance that acts as an anti-inflammatory.Aims of study: The study was designed to identify the role of local exogenous applications of osteopontin on wound healing (in cheek skin).Materials and methods: Thirty adult male albino rats weighting an average of (250-300gm) used in this study, incisional wounds were made in the skin of the cheek of rat and they were divided into the following groups:A-Control group: 15 rats treated with 1μ l of normal salineB-Experimental groups: 15 rats treated with topical application of 1μl osteopontin.The scarification of animals were done for the healing intervals (1, 5 and 10 days) Histological analysis and assessment of number of inflammatory cell, Thickness and contraction of incision were performed for both experimental and control groups for all healing periods.Results: Histological analysis revealed that osteopontin accelerate wound healing of cheek skin and there was highly significant difference among studied groups in periods 1,5,10 days. Regarding mean values of epithelial thickness, inflammatory cell count and contraction of wound area have reported a highest value different in 1, 5 and 10 days durations.Conclusion: It can be concluded that application of osteopontin shown efficacy in the healing of skin wounds induced in rats

Abstract

Background and objective: Rhus coriaria, a well-known spice grown in the Kurdistan region of Iraq, commonly known in the Middle East Region by (Sumac), is reported to enclose many medical benefits. This study assessed the effects of topical application of Rhus coriaria extract on the rate of wound closure, the Hydroxyproline (HXP) and Nitric Oxide (NO) levels were measured using the healed skin tissue homogenate the elemental composition of the plant was screened.Methods: Adult male Sprague Dawley rats were topically treated with 0.2 mL of the vehicle (gum accacia), Intrasite gel (positive control), 100 and 200 mg/kg of Rhus coriaria extract.Results: Wounds dressed with the extract and Intrasite gel healed significantly earlier than those with the vehicle. The high content of HXP and NO proves that the effect of Rhus coriaria extract on an excision wound model was significantly higher than that of the vehicle itself. The highest mineral contents were in Potassium, Calcium, Phosphorus, Magnesium and the extract was rich in energy content.Conclusion: The current study concluded that Rhus coriaria showed high potential in wound healing activity.

Effects of autologous platelet-rich plasma on skin healing in dogs

Abstract

This study aimed to evaluate the benefits of using platelet– rich plasma (PRP) on wound healing. The study performed on ten adult male dogs, aged 2-3 years and their weight were 18-25 kg. Under the effect of deep sedation and local subcutaneous anesthesia at the site of wound, four of (2x2) cm of full-thickness square cutaneous wounds were induced, two on each side of the lateral thoracic region of each animal. A pair of left wounds was treated by injection with 5 mL of autonomous PRP (treatment group), 2 mm lateral to the wound edges and in the wound center., immediately after wounding while, right wounds, were injected with a single dose of 5ml of sterile saline (control group) by using the same procedure as in treatment group. The clinical and histopathological evaluation of each group for five weeks, histological biopsies were taken in the days (3, 7, 14, 21, 28 and 35) of the experiment. The clinical evaluation of PRP treated wounds showed that the wound healing process (% contraction, % re-epithelization and % total wound healing) were significantly more (P<0.05) than the control wounds during the five weeks study. The histopathological results revealed that PRP treated wounds have enhanced cellularity, increased vascularity, with increased amount of granulation tissue accompanied by increase numbers of skin appendages suggesting enhanced cutaneous regeneration, than those in untreated wounds. Depending on the clinical and histopathological findings, this study confirms that local implantation of PRP leads to acceleration and improvement cutaneous wound healing.

هدف الدراسة الى تقييم فوائد استخدام الصفائح الدموية الغنية بالبلازما على التئام الجروح. أجريت هذه الدراسة على 10 كلاب محلية وكان اغلبها من الذكور, عمرها من 2-3 سنوات وكانت تزن من 18-25 كغ. تحت تأثير التهدئة العميقة والتخدير الارتشاحي الموضعي, استحداث أربعة من الجروح الجلدية مربعة الشكل بأبعاد 2×2 سم في كل حيوان, اثنان على كل جانب من جوانب الوجه الوحشي لمنطقة الصدر. تم علاج زوج من الجروح اليسرى عن طريق الحقن بجرعة 5 مل من PRP الذاتية (مجموعة العلاج) ، 2ملم وحشيا لحافات الجرح وفي مركز الجرح, مباشرة بعد استحداث الجرح, بينما مجموعة السيطرة فقد شملت على الجروح اليمنى والتي حقنت بجرعة 5 مل من المحلول الملحي, بنفس طريقة الحقن التي اتبعت في مجموعة المعالجة. تم تقييم ومقارنة شفاء الجروح سريريا )عيانيا) ونسجيا في مجموعتي السيطرة والمعالجة مدة خمسة اسابيع حيث تم اخذ الخزع النسجية في الايام (3, 7, 14, 21, 28, 35) من التجربة. أظهر التقييم السريري للجروح المعالجة بالـ PRP أن عملية التئام الجروح (نسبة التقلص ٪, اعادة بناء الظهارة ٪, والشفاء الكلي للجروح ٪) كانت أكثر بكثير (P <0.05) من جروح السيطرة خلال دراسة الخمسة أسابيع. كشفت النتائج النسيجية أن الجروح المعالجة بالـ PRP عززت الخلوية، زيادة الأوعية الدموية، مع زيادة كمية النسيج الحبيبي مصحوبة بزيادة أعداد الملحقات الجلدية مما يوحي بتجديد جلدي محسّن، أكثر من الجروح غير المعالجة. اعتمادا على النتائج السريرية والتشريح المرضية، تؤكد هذه الدراسة أن الغرس الموضعي للـ PRP يؤدي إلى تسريع وتحسين شفاء الجروح الجلدية.
