Protective effect of Silymarin against kidney injury induced by carbon tetrachloride in male rats

Abstract

The herbal drugs have a protective effect for kidney function against chemical toxicity. 24 male rats divided into 4 groups and treated as following, control group administrated orally with 1ml/kg. B.W physiological solution (0.9%), One dose Carbon Tetrachloride (CCl4) 3 ml/kg. B.W, Silymarin 150 mg/kg. B.W and Silymarin150 mg/kg. B.W with CCl4 3 ml/kg. B.W for 30 days. Oxidative stress resulted by CCl4 caused increasing in Creatinine, Urea, total protein, Albumin, malondialdehyde (MDA) levels decreasing in Glutathione (GSH) and superoxide dismutase (SOD) levels in serum and congestion, degeneration and desquamation in kidney tissue. We concluded that Silymarin showed protective effect via increasing GSH, decreasing creatinine, Urea, total protein and MDA levels in serum and protect kidney tissue in rats.

تمتلك العقاقير الطبية تأثيراً وقائيا لوظائف الكلى ضد سمية المواد الكيميائية. تم تقسيم 24 من ذكور الجرذان إلى أربع مجموعات وعوملت كالاتي، مجموعة السيطرة تم معاملتها فمويا بالمحلول الملحي الفسلجي (0,9%)، مجموعة CCl4 أخذت جرعة مفردة من مادة رباعي كلوريد الكربون CCl4(3 مل/كغم وزن جسم)، مجموعة السلمارين (150 ملغم/كغم وزن جسم) ومجموعة السلمارين مع CCl4 ولمدة ثلاثين يوما. الجهد التاكسدي الناجم عن CCl4 تسبب بزيادة في مستوى الكرياتنين، اليوريا، البروتين الكلي، الألبومين، المالوندايالديهايد وانخفاضا في مستوى الكلوتاثيون والسوبر اوكسيد دسيميوتيز في مصل الدم واحتقان دموي وتحطم وتوسف في نسيج الكلى بينما اظهر السلمارين تاثيرا وقائيا من خلال زيادة مستوى الكلوتاثيون والالبومين وخفض الكرياتنين واليوريا والبروتين الكلي والمالوندايالديهايد في مصل الدم وحماية نسيج الكلى في الجرذان.

Abstract

This study aims to evaluate the effect of aqueous extract stem of dried of Senna Alexandrıa using male mıce once daily at varying doses for 2 and weeks on hematological and histological changer in liver and kidney. In the acute toxicity aqueous extracted of dried stem of Senna Alexandrıa were administrated orally to mice up to 20-25 gr of body weight, in sub-acute study mice was daily administrated orally with same extraction at dose 500,1000,2000 mg/kg for 14 days . Hematological findings have shown in male group increase of RBC, HGB, HCT in increase of PIT in group treated with 500mg/ kg of body weight.Body weight was increase after application of extract at the dose of 1000 and 2000 mg/ kg . The histology in liver includes congestion of central vein with inflammatory cell in their lamen, vascular dilation ,fatty infiltrations cytoplasmic vacillation necrotic hepatocyte . Whereas in the kidney the changer includes shrunken glomeruli increasing of renal space, swelling and cytoplasmic vaculation of epithelial cells lining the renal tubule. Abstract

Background:The incidence of cancer is increasing worldwide. Xanthium strumarium may possess anticancer activity, the plant extract for in vitro anticancer activity against a panel of three human cell lines (Breast MCF7, Renal TK10 and Melanoma UACC62), exhibit anticancer activity against these three human cell lines screened by National Cancer Institute.Objective:To investigate the safety of chloroform extract of Xanthium strumarium leaves, which showed potent cytotoxic activity against tumor cells.Methods:The leaves were dried and grounded into fine powder, and extracted with chloroform, that showed potent cytotoxic activity has been tested against two animal species mice and rats for testing its safety. Toxicity was evaluated in Swiss albino mice by feeding with serial doses of extract between 1.0 to 20.0 gm/kg orally and observed continuously for the first 4 hr and hourly for the next 24 hr, then every 6 hr for 48 hr (72 hr, acute toxicity). Rats were also fed with extract single dose of 5 gm/kg, the toxicity was carried out by assessing the effects on biochemical parameters, body weight and relative organ weights for both male and female rats.Results:LD50 of chloroform extract was 3.07 gm/kg. The biochemical finding showed no significant differences compared to control. No significant weight changes occur throughout the study.Conclusion:Chloroform extract has low LD50 (3.07 gm/kg), but the acute toxicity study showed no mortality or signs of toxicity, with non-significant changes in body weights, relative organ weights, and biochemical tests among treated groups compared to their controls.Keywords:Xanthium strumarium, chloroform extract, cytotoxic herb, acute toxicityCitation:Alsabah AS, Abd AH, Al-Shammari AM.
