Abstract

This study was conducted to investigate if silymarin can prevent the adverse effect of oxidative stress induced by carbon tetrachloride(CCl4) on some characters of male rat reproductive system. Twenty male albino rats were divided into 4 groups, 5 for each included, control group, CCl4 group, silymarin group and silymarin plus CCl4 group. Groups with silymarin received dose (150mg/kg) and group with CCl4 received (3ml/kg) at the end of experiment. The period of the study was 35 days. CCl4 can induce male reproductive toxicity through damage testis structure, weights, reduction in epididymis, sperm count, morphology, viability and motility. Silymarin plays its antioxidant role with scavenging free radical resulted by CCl4 and showed significant increasing in body and testis weight, normal sperms counting and vital sperms in compared to CCl4 group

انجزت هذه الدراسة لمعرفة كفاءة السلمارين في منع التاثير الضار للاجهاد التاكسدي الذي يحثه رباعي كلوريد الكربون CCl4 على بعض خصائص الجهاز التكاثري لذكور الجرذان البيض. عشرون ذكراً من الجرذان البيض قسمت الى اربع مجموعات، خمس حيوانات لكل منها، تضمنت المجموعات المختبرية، مجموعة السيطرة، المجموعة المعاملة بـCCl4 3m/Kg))، مجموعة السلمارين (150mg/Kg) ومجموعة السلمارين مع CCl4. كانت مدة الدراسة 35 يوما. اظهر CCl4 تاثيرا سميا على الجهاز التكاثري من خلال تضرر نسيج الخصية وانخفاض وزن البربخ ،عد النطف،الشكل الطبيعي للنطف، حيوية وحركة النطف. اظهر السلمارين فعاليته المضادة للاكسدة لازالة الجذور الحرة الناتجة عن تاثير CCl4 ولوحظ ارتفاعا معنويا في وزن الجسم والخصى، النطف الطبيعية، عد النطف وحيوية النطف بالمقارنة مع المجموعة المعاملة بـCCl4

Abstract

The semen was collected from five adult Iraqi black local bucks by theartificial vagina and using estrous induced doe the semen was diluted with egg yolk10% extender and frozen by liquid nitrogen for three months, macroscopic andmicroscopic evaluation were done after collection, dilution and thawing, threemedia(TCM-199, DMEM and SOF)were used to see the their effect on activation ofspermatozoa. The results showed that TCM-199 media presented higher significancein sperm motility, progressive motility compared with DMEM and SOF media.There was a great decrease in the concentration of spermatozoa after activation usingthese media which didn’t differ significantly among them and they were highersignificantly compared with control in normal morphology.

4.EFFECTS OF CIPROFLOXACIN ON MALE FERTILITY PARAMETERS AND SPERM DNA INTEGRITY IN RATS

Abstract

Background: The administration of antibiotics is of great value in the therapy of infections of male genital tract, which may have an effect on fertility. Antibiotics are generally prescribed for a variety of infections. A number of patients requiring assisted conception occasionally show evidence of reproductive tract infections. While fluoroquinolones being excessively prescribed in the treatment of male genital tract infections, too little information concerning their outcome on fertility are present.Objective: To estimate ciprofloxacin effects on sperm function parameters in addition to inspect whether ciprofloxacin can affect the integrity of sperm DNA.Methods:In the present study, 48 male adult rats were enrolled. The animals were randomly allocated into six groups; four ciprofloxacin treated groups, which were treated with either (40 mg/kg/day) or (80 mg/kg/day) of ciprofloxacin and 2 control groups. For each dose, the treatment maintained for either 14 days or 28 days. At the end of each duration of treatment, certain epididymal sperm function parameters: sperm morphological normality, sperm concentration and sperm motility were analyzed together with sperm DNA integrity analysis.Results: A significant reduction in sperm concentration, motility and percentage of morphologically normal sperm (in a dose dependent manner) was observed when ciprofloxacin administered for 28 days. The level of DNA fragmentation was significantly elevated with a significant reduction in sperm chromatin quality in ciprofloxacin treated groups whereas serum testosterone level was not significantly affected.Conclusion: Ciprofloxacin can adversely influence fertility parameters in male rat.Keywords: Fertility, Ciprofloxacin, Sperm DNACitation: Abd AH, Al-Dujaily SS, Al-Saray DA. Abstract

back ground: Agglutination of spermatozoa means that motile spermatozoa stick to eachother in the ejaculate. It can be due to infection or antibodies that react with sperm. Spermagglutination has a negative impact on fertility due to impeding of sperm function, inhibitionof fertilization and implantation. Zinc is an essential trace element for male sex function. It is acofactor in more than 300 enzymes influencing various organ functions. Zinc found in seminal fluid,increase sperm count and motility and blood testosterone levels. Zinc had direct effects on theproduction, maturation and function of leucocytes and can modulate antibody production.Objective: The objective of this study to assess the effect of zinc sulfate treatment on spermagglutination among subfertile patients with asthenospermia.Design: Prospective study.Setting: Infertility clinic and assisted reproduction unit at the Institute of Embryo Research andInfertility Treatment, Al-Nahrain University.Patients: Fifty - three subfertile male patients.Materials and Methods: Semen analysis was performed before zinc treatment using direct slideexamination for fifty - three infertile patients. A daily dose of 440 mg of zinc sulfatesupplementation was orally administered for a period of two weeks, three weeks, four weeks, andsix weeks. After each period semen samples were obtained by masturbation after a recommendedperiod of 3-5 days of sexual abstinence, and evaluated for determinations of semen volume, pH,concentration, motility, normal morphology, total progressive motile sperm count, round cells, andsperm agglutination, according to WHO recommendation.Results: subfertile men demonstrated a significant (P< 0.05) decrease in round cells /HPF after thirdweek of zinc treatment. Sperm agglutination was significantly (P< 0.01) decreased after second,third, fourth week, and (P< 0.02) at six weeks of zinc treatment compared to sperm agglutinationbefore treatment. Also the study showed significant (P< 0.05) improvement in progressive spermmotility [grade –A after three week and grade –B after three and four week of zinc treatment],significant (P< 0.01) increase of total progressive motile sperm count /ejaculate, and ofmorphologically normal sperm after third week of oral zinc sulfate supplementation.Conclusions: It was concluded from the results of the work that the oral zinc sulfatesupplementation is effective in decreasing of abnormal sperm agglutination and round cells, withsubsequent improvement of progressive sperm motility.

Abstract

Background : The exact pathogenic mechanism by which varicoceles cause impaired testicular function remains poorly understood . One of the possible mechanisms is the generation of seminal oxidative stress. In addition, the impact of bilateral varicocele on spermatogenesis is still poorly studied . Aims : To study the changes in seminal antioxidant defenses in relation to unilateral and bilateral varicocele, and to correlate them with changes in sperm concentration and total motility. Patients and methods : A prospective, randomized, controlled study , whereby 60 semen samples were collected and analyzed. Twenty samples were from healthy subjects , having no varicocele ( control group ), 20 samples from patients with unilateral varicocele and 20 samples from patients with bilateral varicocele . For each semen sample, measurement of the seminal concentration of the antioxidants ( Albumin, Glutathione peroxidase and Superoxide dismutase ) was done and the seminal concentration of the oxidative stress marker, Malondialdehyde. In addition sperm concentration and total motility were also estimated. Comparison of the results in the 3 groups was done . Results : Measurement of semen parameters revealed a drop in the mean value of the 3 antioxidants and an increase in the mean value of Malondialdehyde, which was statistically significant in the 2 varicocele groups in comparison with control, and in bilateral varicocele in comparison with unilateral varicocele . Another drop was in the mean sperm concentration and mean percentage of sperm total motility, which was statistically significant in the 2 varicocele groups in comparison with control, but was statistically not significant between the 2 varicocele groups . Conclusions : Bilateral varicocele causes more significant drop in seminal antioxidant defenses than unilateral varicocele which seems to add a more negative impact on spermatogenesis .

Abstract

Defective Sperm parameters was suggested to play an important role in determining fertility and was proven to be closely related to fertilization and pregnancy rates in the natural fertilization process as well as in ART.To evaluate the influence of defective sperm parameters including concentration, morphology and motility on intracytoplasmic sperm injection outcome represented by fertilization rate, cleavage rate, embryo quality and pregnancy rate.This a cohort study included 60 infertile couple attended the clinics of infertility treatment center Al-Sadder teaching hospital and underwent intracytoplasmic sperm injection. They are classified into three groups according to their spermiogram and ICSI outcome evaluated for each group and compared with each other.The main results of this study showed no significant difference in fertilization rate,cleavage rate ,embryo quality and pregnancy rate among studied groups i.e., no significant difference in ICSI outcome between normozoospermic group and those with defective spermiogram. Intracytoplasmic sperm injection outcomes represented by fertilization rate, cleavage rate embryo quality and pregnancy rate were identical with no significant differences between couples with defective sperm parameters and those with normal spermiogram.

Abstract

This study was aimed to investigate the effect of sperm parameters on the response to in vitro sperm activation (ISA) and to compare sperm parameters according to the age and type of infertility.One hundred twenty-eight subjects employed in this study, with age range (18-49) years and duration of infertility (1-28) years. From each subjects semen sample was obtained and seminal fluid analysis (SFA) was done. Simple medium for assisted reproductive techniques; (SMART) was used for in vitro sperm activation by direct swim-up technique and centrifugation Swim-Up technique. Sperm concentration, sperm motility (%), sperm grade activity (%), normal sperm morphology (%) and sperm agglutination (%) were assessed. Crude data were statistically analyzed. From the results of present study, it was appeared that most sperm parameters were significantly (P<0.05) enhanced post-activation in vitro as compared to pre-activation using SMART medium by direct swim-up technique and centrifugation Swim-Up technique. Sperm concentration for post-activation was significantly reduced (P<0.05) compared to pre-activation groups and the percentages of progressive sperm activity with grade A, total progressive sperm motility (grades A and B) and normal sperm morphology were significantly increased (P<0.05). Based on the results of this study, it can be concluded that sperm motility and concentration were considered most important parameters that have the importance of activation and sperm morphology one of the sperm parameters that have role of the influence on the sperm qualities after activation.

صممت هذه الدراسة لتقييم مدى تأثير متغيرات النطف على الاستجابة للتنشيط خارج الجسم ولمقارنة متغيرات النطف مع العمر ونوع العقم. شارك في هذه الدراسة مئة وثمانية وعشرون شخص، وكان متوسط أعمارهم (18-49) وفترة العقم (1-28) سنة. أخذت عينة السائل المنوي من كل مريض واجري لها فحص السائل المنوي. استعمل الوسط الزرعي البسيط للتقنيات المساعدة على الإنجاب لتنشيط النطف خارج الجسم باستخدام تقنية سباحة النطف للأعلى مباشرة وتقنية الطرد المركزي. تم تقيم تركيز النطف والنسبة المئوية للنطف المتحركة والنسبة المئوية لنشاط النطف والنسبة المئوية لشكل النطف الطبيعي والنسبة المئوية لتكتل النطف.كما تم تحليل النتائج الأولية إحصائيا. أظهرت نتائج الدراسة الحالية تحسن معنوي (P<0.05) في اغلب متغيرات النطف بعد التنشيط خارج الجسم عند المقارنة مع قبل التنشيط باستخدام وسط سمارت وتقنية سباحة النطف للأعلى مباشرة وتقنية الطرد المركزي. تركيز النطف بعد التنشيط انخفض بشكل معنوي( P<0.05) عند المقارنة مع قبل التنشيط والنسب المئوية لكل من الحركة التقدمية (A) للنطف والحركة التقدمية الكلية((A+B للنطف وشكل النطف الطبيعي أظهرت زيادة معنوية(P<0.0).لذلك نستنتج اعتمادا على معطيات هذه الدراسة بأن حركة النطف وتركيزها من أهم الصفات التي لها أهمية بالتنشيط خارج الجسم وشكل وشكل النطف الطبيعي واحد من الصفات التي لها دور بالتأثير على صفات النطف بعد التنشيط خارج الجسم.

Abstract

ABSTRACT:BACKGROUND: Sperm motility is essential in fertilization because non active or mal active sperms never reach the ovum, but the progress in the assisted reproductive techniques makes it easy for doctors to select viable sperm to be used in intracytoplasmic sperm injection thus the differentiation between viable and non-viable sperms is very important especially in sever asthenospermic samples.OBJECTIVE:To evaluate low level Helium Neon laser irradiation of the whole seminal sample as a method to activate mal or non-active sperms and to study and follow up the late effect of laser irradiation on the seminal samples.MATERIALS AND METHODS: 100 samples were included in this study from patients attending seminal fluid examination lab. After digital seminal fluid examination, samples were divided in 2 parts, the first was incubated for 120min at 37C and the other was irradiated with 623.8 wave length Helium Neon laser and an output power of 2mW at 1cm distance for 10min and then examined digitally after irradiation then incubated with the first part to 120min also to be examined again with the first part.RESULTS: It was found that there is a significant increase in the progressive motile sperm percentage in the laser irradiated samples from about 33.6% to 41.6% (p<0.005) and also decrease in the percentage of the immotile sperms from about 44.1% to 34.4% (p<0.001). While after 120min incubation, active sperms percentage in the irradiated sample was about 43% which is higher than the non-treated part (31.6%; p<0.01) that showed also higher percentage of immotile sperms (52.8%) against (39.9%) in the treated part.CONCLUSION: Helium Neon laser irradiation to the whole seminal sample increase the sperms activity and decrease the percentage of immotile sperms which mean that it stimulates viable but immotile sperm to move. And also, sperms in laser irradiated samples preserved their increased activity for more than 2hours after laser irradiation.

Abstract

Objective: This study aimed to determine the relationship between the addition of Glutathione to sperm freezing medium and cryosurvival rate of sperm motility and viability in asthenozoospermic patients.Methodology: fifty nine semen samples of asthenozoospermic patients were obtained from the male partner of infertile couples attending the fertility center of al-sadder medical teaching city in Al-Najaf during the period 1st October 2016 until 1st March 2017.Each semen sample was divided into four equal parts, volume of each part is 0.5 ml: first part (before activation), second part (after activation), third part (cryopreservation with sperm freezing medium for one month), and fourth part (cryopreservation with sperm freezing medium plus glutathione for one month ) .Sperm concentration , sperm progressive motility, sperm viability and sperm normal morphology were counted in first two parts, cryosurvival rate of sperm motility, viability was counted in third and fourth parts .Statistical analysis of the study performed by using Independent t-test and paired test to assess the differences between two groups .Results: After sperm activation by using the swim up technique ,the study results showed significant increase (P < 0.01) of progressive sperm motility, normal sperm morphology and sperm viability, while there were a significant decrease (P < 0.01) of sperm concentration. cryosurvival rate of sperm motility and viability were significant decrease (p < 0.01) after cryopreservation and thawing processes while the results were revealed significant improvement in cryosurvival rate of sperm motility, and viability ( p < 0.01 ) after the addition of glutathione to the sperm freezing medium.Conclusion: it can conclude from this study that addition of glutathione to sperm freeze medium improves. (p < 0.01) cryosurvival rate of sperm motility and viability during cryopreservation and thawing processes .Recommendations: the study recommended to addition 50 microliter of glutathione to sperm freezing media in concentration 1 mM .Keyword: Glutathione, sperm freezing medium, cryosurvival rate, sperm motility, viability, asthenozoospermic .

الهدف: تهدف الدراسة إلى تحديدالعلاقة بين إضافة الكلوتاثايون الى وسط تجميد النطف ومعدل الحركة والعيوشية النطفية بعد النجاة من التجميد في مرضى وهن النطف.المنهجية: تم الحصول على تسعة و خمسين عينة من السائل المنوي من مرضى وهن النطف من الشريك الذكر للأزواج العقيمين الذين يراجعون مركز الخصوبة في مدينة الصدر الطبية التعليمية في النجف وأمتدت الدراسة من المدة(1تشرين الاول 2016 حتى 1 اذار 2017). تم تقسيم كل عينة من السائل المنوي إلى أربعة أجزاء متساوية , حجم كل واحدة 0,5 مل: الجزء الأول (قبل التنشيط)، الجزء الثاني (بعد التنشيط)، الجزءالثالث (جمد بإستخدام وسط تجميد النطف لمدة شهر واحد)، والجزء الرابع (جمد بإستخدام وسط تجميد النطف مضافا اليه الكلوتاثايون لمدة شهر واحد). تركيز النطف، حركة النطف التقدمية ، و المظهر الطبيعي للنطف و العيوشية تم حسابها في كل جزء,ومعدل الحركة والعيوشية بعد النجاة من التجميد، تم حسابها في الجزء الثالث والرابع.تم إجراء التحليل الإحصائي للدراسة بإستخدام إختبارt المستقل وإختبار t المقارن لتقييم الفروق بين المجموعتين.النتائج: بعد تنشيط النطف بإستخدام تقنية السباحة ، أظهرت نتائج الدراسة زيادة معنوية (p < 0.01) في حركة النطف التقدمية، و المظهر الطبيعي للنطف وعيوشية النطف, في حين كان هناك إنخفاض معنوي(p < 0.01) في تركيز النطف.أما معدل الحركة والعيوشية بعد النجاة من التجميد إنخفضت بشكل معنوي (p < 0.01) بعد عمليات التجميد والإذابة , بينما بينت النتائج تحسنا معنويا (p < 0.01) في معدل الحركة والعيوشية النطفية بعد النجاةمنالتجميد بعد إضافة الكلوتاثايون الى وسط تجميد النطف .الإستنتاج: يمكن أن نستنتج من هذه الدراسة أن إضافة الكلوتاثايون إلى وسط تجميد النطف حسن(p < 0.01) معدل الحركة والعيوشية النطفية بعد النجاة من التجميد خلال عمليات التجميد والإذابة .التوصيات : أوصت الدراسة بإضافة 50 مايكرو مول من الكلوتاثايون إلى وسط تجميد النطف بتركيز 1 ميلي مولار.

Abstract

BackgroundSemen samples are prepared for assisted reproduction by selecting a population of highly motile, morphologically normal sperm and removing the seminal plasma, leukocytes and bacteria. Culture media provide the spermatozoa with needs that maintain optimal function of spermatozoa to give rise excellent results during semen preparation.ObjectiveThe aim of this study was to investigate the effect of alcoholic extraction of turmeric (AET) on sperm parameters during in vitro sperm activation (ISA) for asthenozoospermic (AZ) and oligoasthenozoospermic (OAZ) patients.MethodsSeventy four infertile patients were included, and classified into two groups according to their sperm parameters. Semen sample was divided into 3 aliquots. One mL of Earl's medium either alone (control group) or supplied with one concentration of AET (5 µg/mL or 10 µg/mL) was over layered the pellet, and the three tubes were incubated at 37 oC for 30 min in air incubator. Sperm concentration, motility, grades activity, progressive motility, normal morphology and agglutination were assessed pre- and post-activation in vitro.ResultsResults revealed an enhancement of most sperm parameters for control and both treated groups post-activation as compared to pre-activation. Post-ISA, sperm parameters for both treated groups were better than the control group. However, best results for improvement of sperm parameters were assessed within treated group (5 µg/mL of AET).ConclusionsThe lower concentration of alcoholic turmeric extraction enhanced human sperm parameters during ISA without any harmful effects on sperm physiology. The results are also useful as a guide for further standardization of turmeric extracts used for pharmaceutical purposes in the techniques of assisted reproduction.Key words Male infertility, Sperm activation, Turmeric.
