Effect of hawthorn extract on blood pressure and lipid profile in patients with stage I hypertension: A placebo-controlled, double-blind randomized trial

Abstract

Aims: To investigate the possible effects of hawthorn extract on blood pressure & blood lipid profile in patients with stage I hypertension. Patients and Methods: This 12 week randomized, double -blind, placebo controlled study was conducted on newly diagnosed patients with stage I hypertension (n=60). Patients were selected from outpatient clinic in Al-Yarmuk Teaching Hospital, Baghdad and were divided into two groups each comprising of 30 patients, they were given hawthorn extract capsule 450 mg (Willmar Schwabe Pharmaceuticals) twice daily and identical placebo capsule respectively. Systolic and diastolic blood pressure was recorded and fasting lipid profile was done initially and at the end of 4th, 8th and 12th week of treatment. Both of hawthorn and placebo treated group were given diet and exercise plan. Results: After 12 weeks the hawthorn treated group had a significant reduction in both of systolic and diastolic blood pressure (p<0.01), serum total cholesterol (p<0.001) and low density lipoprotein (p<0.001), while the placebo treated group had a non-significant decrease in their blood pressure (p>0.05), serum total cholesterol and low density lipoprotein (p>0.05). High density lipoprotein was significantly increased in patients treated with hawthorn (3.01 mg/dl, 8.13% P= <0.05) compared with placebo group (1.06 mg/dl, 2.8 % P= > 0.05) but there was no significant difference in triglyceride detected between two groups. Conclusions: The study suggests that hawthorn does have an effective antihypertensive ability, as well as lipid lowering effects.

Abstract

Background: Garcinia cambogia is a herbal remedy extracted from pumpkin like fruit used for treatment of various health conditions. Hydroxycitrate, the principle ingredient of Garcinia cambogia, has been shown to be a powerful lipogenic inhibitor. Blood viscosity regarded as an important factor for many diseases. Many parameters are affecting the blood viscosity such as packed cell volume, fibrinogen and serum cholesterol. Aim of the study: This study was conducted to determine the effects of Garcinia cambogia on blood viscosity profile. Methods: This study was conducted in the Department of Pharmacology, College of Medicine, AL-Mustansirya University from September to December 2009. Twenty healthy male volunteers aged between 20 and 22 years enrolled in this study. Those volunteers randomly assigned equally, in a double-blind manner into two group; group (A) take Garcinia cambogia oral tablets and group (B) take placebo tablets ,the duration of therapy was two weeks. Blood viscosity, packed cell volume, plasma fibrinogen, and serum cholesterol were performed at day one and at the end of 1st and 2nd week of treatment.Results: This study showed that after one week treatment with Garcinia cambogia there was significant reduction in blood viscosity and plasma fibrinogen. Moreover two weeks treatment with Garcinia cambogia produced significant reduction in blood viscosity, plasma fibrinogen and serum cholesterol (P<0.05). An exception was the packed cell volume which is not altered (P>0.05).Conclusion: It can be concluded that Garcinia cambogia favorably affect blood viscosity. Though further clinical studies are recommended, the use of Garcinia cambogia in treatment and prevention of diseases that related to high blood viscosity is promising.

أجريت هذه الدراسة لغرض تقيم تأثير الغارسينيا كامبوغيا على لزوجة الدم التحق عشرون متطوعا من الذكورالاصحاء أعمارهم بين 20 و 22 سنة في هذه الدراسة. هؤلاء المتطوعين وزعوا عشوائيا بالتساوي إلى مجموعتين، المجموعة (أ) أخذت أقراص غارسينيا كامبوغيا ، ومجموعة (ب) أخذت أقراص وهمي، وكانت مدة العلاج أسبوعين. تم قياس اللزوجة في الدم ، ومعبأة حجم الخلية والفبرنيوجين ، والكوليسترول في الدم وأجريت في اليوم الأول ، وفي نهاية الأسبوع الأول والثاني من العلاج. وأظهرت النتائج بعد أسبوع واحد من غارسينيا كامبوغيا كان هناك انخفاض معنوي في اللزوجة في الدم و الفيبرينوجين. أما بعد أسبوعين فأن الغارسينيا كامبوغيا تسبب في انخفاض كبير في اللزوجة في الدم ، والكوليسترول الفيبرينوجين. استثناء هو حجم الخلية معبأة الذي لم يتغير معنويا. ويمكن أن نخلص إلى أن الغارسينيا كامبوغيا تؤثر بشكل إيجابي على لزوجة الدم. وإن كان من المستحسن إجراء دراسات سريريه أخرى، استخدام الغارسينيا كامبوغيا في العلاج والوقاية من الأمراض التي تتعلق اللزوجة الدم المرتفع واعدة.

Abstract

ABSTRACT:BACKGROUND:Alpha1-adrenoceptor blocking agents showed several effects beyond their action on the vascular smooth muscles. They improve the lipid profile and inhibit the aggregation of blood platelets.OBJECTIVE:To investigate the clot-lysis effect of selective α1-adrenoceptor antagonists and its relation to peroxynitrite level in vitro experimental model.MATERIALS AND METHODS:Venous blood samples obtained from ten healthy subjects. To each pre-weighed clot, 100 μL of either distilled water as a negative control, prazosin (10 μg), terazosin (20 μg) and alfuzosin (25 μg) were added. Peroxynitrite level was measured in sera and sangious fluid that formed after clot-lysis.RESULTS:Prazosin, terazosin and alfuzosin,in order, significantly reduced the clot weight up to 3.7%. Peroxynitrite level in sangious fluids was higher in treated groups than that of negative control or sera levels.CONCLUSION:α1-adrenoceptor antagonists induced clot-lysis effect. This effect is associated with generation peroxynitrite

Abstract

background: Ergogenic denote rising in the capacity of the body for eliminating exhaustion and enhancing physical performance, with broad variety of drugs are apparent to be ergogenic in their effects. Yohimbine is an alkaloid recognized as an antagonist to the noradrenergic α2-receptors which normalize noradrenaline release in the central nervous system and their activation decreases the discharge of noradrenaline in the synaptic terminal.Objectives: This study was designed to evaluate the ergogenic effects of yohimbine in normal healthy volunteers.Patients and Methods: The present study was conducted in Department of Pharmacology, College of Medicine, Al-Mustansiriya University, Baghdad-Iraq, in collaboration with Department of Pharmacology, College of Medicine, University of Kerbala, Kerbala-Iraq, during April of 2013. The subjects of this study were medical students. 20 male volunteers were conformist to join up this single blind random experimental study. The exercise program consists of cycling on an ergometer. Pulse was measured using ear pulse sensor and maximum pulse rate achieved in this study and the entire exercise parameters were recorded as baseline data, then each subject take yohimbine tablets 5 mg (orally) and after 2 hours, each subject run on cycling ergometer to fatigue state, then, the display functions of bicycle were scanned. Besides, maximal oxygen uptake (VO2 max) was measured to estimate oxygen consumptions.Results & discussion: Yohimbine significantly improved all ergogenic parameters (efforts, calories, distance, speed and exercise time) from baseline after single oral dose of 5mg, where p values were ˂ 0.05. Moreover; yohimbine significantly accelerate the oxygen consumption in both oxygen consumptions and velocity at maximal oxygen uptake regarding maximum heart rate and resting heart rate with p values ˂ 0.05.Conclusions: Yohimbine accelerates and improves cycling performances via positive ergogenic effects .So, yohimbine can be used as ergogenic agent for ameliorating the physical fatigue.
