Salivary alkaline phosphatase and acid phosphatase levels in gingivitis diagnosis and treatment

Abstract

Background and objective: Gingivitis is the presence of clinical signs of inflammation in gingiva and is associated with teeth showing no attachment loss. Human saliva contains informative components that can be used as diagnostic markers for human diseases. This study is directed to evaluate salivary phosphatase enzymes levels in diagnosis and follow up the treatment of gingivitis.Methods: Saliva samples were collected from 100 healthy persons, and 50 patients before treatment (scaling and polishing) and 20 days after the treatment. Their age ranged between 20-30 years. Alkaline phosphatase and acid phosphatase activities, some kinetics and thermodynamic parameters were measured.Results: Salivary alkaline phosphatase and acid phosphatase activities elevated in gingivitis, while their levels returned to the control values after treatment. The Michaelis constant (Km) values for salivary alkaline phosphatase and acid phosphatase decreased in gingivitis, while the maximum velocity (Vmax) values increased. These values returned back to normal values after treatment. The changes in the thermodynamic parameters (∆H*, ∆G* and ∆S*) values of transition state for salivary alkaline phosphatase were non-significant, while their changes were significant for salivary acid phosphatase, except for ∆G* value.Conclusion: Salivary alkaline phosphatase and acid phosphatase levels can be relevant for diagnosis and the follow-up of gingivitis treatment.

Abstract

Background and objective: Cigarette smoke is a leading preventable cause of many diseases and death, because it contains a complex mixture of chemical compounds. Few studies about the relation between cigarette smoking and glycoconjugate were found in literature, while no study was observed regarding passive smokers. Thus this relation is obscure till now. Therefore this work was designed to study the effect of cigarette smoking on serum glycoconjugates levels.Methods: One hundred twenty one completely healthy males from different areas of Duhok governorate were included in this study. Their age was ranged between 20 and 50 years. They were classified according to the number and duration of cigarette smoking into: heavy smokers (31), moderate smokers (26), passive smokers (30) and non-smokers (control). Fasting blood was withdrawn from all groups and used for determination of serum fucose and its related parameters.Results: The results showed that there was a significant decrease in serum total protein in heavy smoker groups when compared to non-smokers. The levels of serum total fucose and protein bound fucose significantly increased in smoker groups, while the results indicated that there was a significant increase in serum lipid associated fucose levels in heavy and passive smoker groups comparing to non-smoker group. A significant decrease in serum protein bound hexoses was observed in both heavy and moderate smokers. The result also showed that serum total calcium concentration, Ca/TP and Mg/TP ratios significantly decreased in all smoker groups.Regarding to serum Manganese (Mn) and Mn/TP ratio, their values increased significantly in case of passive smokers only.Conclusion: The results revealed that cigarette smoking had significant effects on glycoconjugates status (expressed in serum fucose and its related parameters).

Abstract

Background and objective: Periodontitis is one of the most widespread oral diseases in Kurdistan. Saliva can be used as a noninvasive diagnostic fluid to measure chemical biomarkers released during oral disease, so this work is directed to focus on the study of salivary fucose (a glycoprotein component) and its related parameters in periodontitis, to assess the possibility of using them as indicators for the disease and its progress.Methods: The present work included 79 individuals. They were grouped into healthy (32), advanced periodontitis (20) and moderate periodontitis(27) subjects. Their age was ranged between (20-60) years. Unstimulated whole saliva samples were collected from all the groups. The samples were used for the estimation of salivary; total fucose (TF), protein bound fucose (PBF), protein bound hexose (PBH), lipid associated fucose (LAF), total protein (TP), total calcium (TCa), zinc (Zn), and salivary magnesium (Mg).Results: Salivary TF, LAF, FF, PBF, TP and TCa were significantly increased in both advanced and moderate periodontic groups comparing to healthy individuals, while there were no significant differences in salivary PBH, Mg and Mg/Ca levels in periodontic groups when compared to normal. Salivary TF/TP, PBF/TP, PBH/TP and FF/TP ratios and salivary zinc were significantly reduced in periodontic males.Conclusion: On the basis of the above results, it can be concluded that these biochemical parameters, may be used as indicators for the extent of periodontal tissue damage, thus they can be used in the identification of periodontitis progresses and treatment follow up.
