Inhibitory Effect Of Pomegranate (Punica Granatum L.) Juice Against Some Gram Positive And Negative Bacteria

Abstract

This study aimed to see the inhibitory activity of Iraqi and Syrian Pomegranate juice against Two gram positive (Staphylococcus aureus, Staphylococcus epidermidis)bacterial isolates and three gram negative(E. coli,Pseudomonas aeruginosa,Proteus spp., and to compare with the inhibitory activity of some antibiotics. Several granatum juice concentration of 100%,75%, 50%, 25%, 10%and 5% which were diluted with distilled water and with honey bee after filtration through bacterial filter(Millipore).Result indicated that Iraqi Pomegranate juice diluted with distilled water showed bigger inhibition zone on all isolates than Syrian pomegranate juice and the largest effect was on Staphylococcus aureus isolate, while pomegranate juice of both Iraqi and Syrian diluted with honey bee had inhibitory effect in all concentration, on the other hand all isolates were sensitive to Amikacin, Ciprofloxacin, Gentamycin, Nalidixic acid and Tobramycin and these results were close to the results of Pomegranate juice used in this study

هدفت هذه الدراسة معرفة الدور التثبيطي لعصير الرمان العراقي و السوري المضاد للبكتريا ضد اربع عزلات ومقارنتها مع تأثير بعض المضادات الحياتية.تمت الدراسة بأستخدام تخافيف من عصير الرمان بعد تعقيمه بالمرشحات البكتيرية مع الماء المقطر وكذلك مع العسل وهي 75%,50%,25%,10%و5%.وقد اشارت النتائج ان عصير الرمان العراقي المخفف بالماء المقطر اظهر مناطق تثبيط اكبر لكل العزلات البكتيرية من عصير الرمان السوري وكان التأثير الاكبر على عزلة Staphylococcus aureus ,وقد ازداد التأثير التثبيطي لعصير الرمان العراقي و السوري بعد تخفيفه بالعسل حيث اظهرت النتائج مناطق تثبيط في كل التخافيف ومن ناحية اخرى كل العزلات البكتيرية كانت حساسة لمضادات الاميكاسين و السبروفلاكساسين وجنتميسين وحامض النالديكسك و التوبرميسين

Abstract

ABSRACT:BACKGROUND:Phytotherapy is an alternative for antibiotics therapy in these days since resistance to many antibiotics used in treatment are increasing along with high cost of some antibiotics and side effect on patients. Punica granatum and Vaccinium macrocarpon have antimicrobial properties. this paper suggest that P. granatum and Vaccinium macrocarpon may be more effective than some antibiotics used in the treatment of some Gram negative urinary tract pathogens.OBJECTIVE:Punica granatum juice, cranberry juice and antibiotics were used to investigate the antimicrobial activity by using well diffusion method.MATERIALS & METHODS:A total of four different urinary tract causing pathogens collected from patients with urinary tract infection (UTI).RESULT:The result indicate that extract obtained from Punica granatum pericarp exhibited antimicrobial activity against all organisms almost similar to the effect of cranberry which is not grown in Iraq and that Punica granatum and Vaccinium macrocarpon are more effective than some antibiotics used in the treatment of urinary tract infection.CONCLUSION:The antibacterial activity of crude extract of P. granatum against urinary tract causing organisms is reported for the first time and it showed similarity with the effect of cranberry juice that are not found or imported to the local market. Further phytochemical elucidations are required to determine the nature of compound(s) responsible for the antibacterial effects. This study is generally considered an effective approach in the discovery of new antibacterial agents from P. granatum.

Abstract

To overcome the development of drug resistance, it is necessary to find a new class of antimicrobial compounds possessing different mechanism or chemical properties from those that are used commonly for the treatment of the diseases which are caused by Candida albicans. This study was designed to investigate the antifungal activity of Diacetyl against Candida albicans isolates with a total of forty eight Candida albicans isolates which was isolated from sputum, vagina and urine sample of different patients were tested against several concentrations of Diacetyl compound (30,60,120,180)µg/ml and for different incubation time to examine the inhibitory activity of the compound in vitro. All urine isolates showed 100% complete inhibition to(30,60,120,180) µg/ml concentrations after 24,48,72 hrs incubation at 37C°,while only three vaginal isolates 30% and eleven sputum isolates 42.3% showed resistance after 48 hrs incubation to 30 and 60 µg/ml concentrations. It seems that Diacetyl compound has a potent antifungal activity against Candida albicans in vitro.

للتغلب على مقاومة المضادات من الضروري ايجاد اصناف جديدة من المركبات الضد ميكروبية ذات ميكانيكية وتركيب كيميائي مختلف عن المواد الضد ميكروبية المستخدمة حاليا لعلاج الاصابات الناتجة عن الاصابة ب Candida albicans .اجريت التجربة على ثمان واربعون عزلة من Candida albicans المعزولة من القشع,المسحات المهبلية و الادرار من مختلف المرضى وتم استخدام تراكيز مختلفة من Diacetyl 30,60,120,180 مايكروغرام/مل وباوقات حضن زمنية مختلفة 24,48,72 ساعة في درجة حرارة 37° مئوية.كل عينات الادرار لم تظهر اي نمو في كل التراكيز المستخدمة بعد 24,48,72 ساعة بينما اظهرت ثلاث عزلات فقط من Candida albicans المعزولة من المهبل 30% واحدى عشر عزلة من العزلات المعزولة من القشع 42.3% مقاومة بعد 48 ساعة من الحضن لتركيز 30 و 60 مايكروغرام/مل من Candida albicans.التجربة تشير الى ان مركب Diacetyl له فعالية ضد ميكروبية ضد بعض عزلات Candida albicans.
