Inhibitory effect of gundelia extract on urinary α-amylase activity of type-I diabetes mellitus

Abstract

Background and objective: α-amylase is an enzyme that degrades starch into maltose and glucose by hydrolyzing α-1,4-glucan bonds.It is known that the enzyme is found to be elevated in patients suffering from diabetes mellitus.A potent extracts of locally distributed wild plant,the Gundelia was found to inhibit elevated activity of α-amylase with dose responses.Methods: Samples of 50 diabetes mellitus type-I has been investigated for urinary α-amylase activity.An inhibitory dose of Gundelia extract were used for the inhibition of the elevated enzyme activity.The data obtained were compared with that of healthy controlled samples﴾30﴿.The adopted protocol was a colorimetric determination﴾Anonymous,1980﴿ .Results: α-amylase activity was found to be elevated﴾281.70±10.03 IU/24hr) in patient’s serum compared with that of controlled samples﴾43.38±3.33 IU/24hr﴿.The activity of the enzyme was found to be inhibited﴾160.11±250.4 IU/hr﴿ in patients serum using dose response of 15mg/ml of Gundelia extract. The study were comprehensive to determined the physical parameters of the enzyme activity and a values of Vmax and Km were obtained.Conclusion: It was known that Gundelia is used in prevention and treatment of liver diseases. The plant has a role in the body as an antioxidant factor.It has a hypolipemic effect, therefore, a use of such plant extract could have a hypoglycemic activities on patients with DM-I.

Abstract

Acute mayocardial infarction (MI) remains a leading cause of morbidity and mortality world wide. MI occurs when myocardial ischaemia exceeds a critical threshold and overwhelms mayocardial cellular repair mechanisms designed to normal operating functions and homostatus. Activity of xanthine oxidase (XO), MDA and vit-C concentrations were determined in sera of 100 patients with MI. Their mean±SD age was (57.05±1.25) year. The same parameters were determined in sera of 100 apparently healthy matched subjects as a control group. The mean ±SE of XO, MDA, and vit-C in patients with MI was 0.031±0.016 unit/ mg protein, 1.28±0.053 µ mol/L, 0.520±0.137 mg/dl respectively while the mean ± SE of control group was 0.012±0.006 unit/ mg protein, 0.819±0.019 µ mol, and 0.648±0.302 mg/dl respectively. There were significant differences in the mean value of XO and MDA in patient with MI compared to control group (P<0.001). However the mean value of vit-C showed no significant differences in both groups.

Abstract

Background: Liver enzymes are commonly found in liver cells and when the liver is damaged, liver cells release their enzymes into the blood stream. Vitamin C is a low-weight molecular antioxidant and is effective in the aqueous phase in protecting different parts of cells against free radicals. An enzyme inhibitor is a molecule, which binds to enzymes and affect their activity.Objective: This study was carried out to investigate the effect of new vitamin C derivative on serum liver enzymes.Research design and methods: Eight samples were used. Liver enzymes activity measured with and without vitamin C derivative. Results: The result showed a reduced activity of aspartate aminotransferase (AST), and alanine transferase (ALT) after addition of vitamin C derivative.Conclusion: It is concluded that the decrease in ALT, and AST activities were due to noncompetitive inhibition of enzymes by vitamin C derivative.

أنزيمات الكبد موجودة عادة في خلايا الكبد وعند تلف الكبد يتم الإفراج عن الانزيمات الخاصة في مجرى الدم. فيتامين C هو منخفض الوزن الجزيئي للأكسدة وفعال في المرحلة المائية في حماية أجزاء مختلفة من الخلايا ضد الجذور الحرة. مثبط الإنزيم هو جزيء، الذي يرتبط بالانزيمات ويؤثر على نشاطهم. أجريت هذه الدراسة لمعرفة تأثيرمشتق فيتامين C الجديد على انزيمات الكبد في الدم. استخدمت ثمانية عينات. النشاط قياس انزيمات الكبد بدون ومع فيتامين C المشتق. أظهرت النتيجة انخفاض نشاط ASTو ALT بعد إضافة فيتامين C المشتق. ويستنتج من ذلك أن الانخفاض في ALT، AST نتيجة التثبيط غير التنافسي على الانزيمات الذي اظهره مشتق فيتامين C.

Abstract

IGF-1 is a protein produced by the liver in response to growth hormone stimulus.One important key in effectively preventing and treating osteo arthritis, is establishing a healthy balance of IGF-1.Leptin is a hormone produced by adipose tissue, acting as a sensor of fat mass in part of a negative feedback loop that maintains a set point for body fat stores.Leptin plays an important role in the progression of Osteoarthritis (OA), prompting some to classify OA as a metabolic disease. it, has been found in synovial fluid from patients with OA , and are thought to have local effects on joint tissues . Atherogenic index of plasma (AIP) is newly marker of atherogenectiy provide prediction to accelerated development of atherosclerosis in diabetes mellitus patients.Objective This study was designed to find out the variation of some parameters in OA with & without T2DM. Patients and methods This study included ( 88) subjects aged between (40-60) years (all females , newlydiagnoses and obese) who were attending the Al- Kadhimiya Teaching Hospital . Baghdad andAl-Mustansiriya University . The enrolled patients were divided to four groups: OA(24) , T2DM (20), OAwithT2DM (24) and control (20) . Venous blood samples from women were taken for laboratory investigation which included : Fasting plasma glucose , lipid profile (TC ,TG , HDLc and LDLc) , IGF-1 , Leptin were measured and Atherogenic index of plasma calculated as molar ratio of log (TG/ HDL-C). Results The current study shows a significantincreasedatherogenic index , Leptin value , in three patients groups diabetes , osteoarthritis and OAwith DM when compared with control. But IGF-1 level was significantly decreased in the three patients groups T2DM, OA , T2DM with OA when compared with control .ConclusionOsteoarthritis has a direct effect on dislipidemia ,Leptin& AIP value in Diabetes mellitus type2

هرمون النمو الشبيه بالانسولين هو بروتين ينتج من الكبد كرد فعل لهرمون النمو وهو المفتاح المؤثر في منع وعلاج سوفان المفاصل. وهناك توازن بين هرمون النمو الشبيه بالانسولين واللبتين الذي ينتج من النسيج الدهني ويعمل كمؤشر لكتلة الدهون , واللبتين له دور مهم في تطور مرض سوفان المفاصل . وقد تضمنت هذه الدراسة (88) عينه من النساءكلهم في باية التشخيص تتراوح اعماره مبين(40-60) سنه, تم جمع العينات من مستشفى الكاظمية التعليمي والمركز الوطني لعلاج وبحوث السكري / مستشفى اليرموك وقد قسمت هذه العينات الى اربعة مجاميع وهي مجموعة مرضى السكري20عينة) , ( مجموعة مرضى سوفان المفاصل 24 عينه) ,( المجموعة التي تحمل كلا المرضين24 عينه ) و مجموعة السيطرة (20) عينه وقد تم اجراء الفحوصات المختبرية التالية : قياس مستوى الكلوكوز بالدم و قياس مستوى الدهون وهرمون النمو الشبيه بالانسولين ومستوى اللبتين وايضا حساب قيم مؤش ر معصد البلازما. في هذه الدراسة لوحظ زيادة معنوية في قيم مؤشر معصد البلازما واللبتين في كافة المجاميع اعلاه بالمقارنة مع مجموعة السيطرة. لكن لوحظ انخفاض معنوي في هرمون النمو الشبيه بالانسولين في نفس المجاميع اعلاه بالمقارنة مع مجموعة السيطرة . ومن الدراسة نستنتج ان لمرضى سوفان المفاصل تاثير مباشر على زيادة اختلال الدهون ومستوى هرمون اللبتين وقيم مؤشر معصد البلازما في مرضى السكري.
