Abstract

ABSTRACT:BACKGROUND:Chronic myeloid leukaemia can be accurately modeled in laboratory mice by the retroviral transfer of a BCR-ABL gene into murine hematopoietic stem and progenitor cells, followed by transplantation of these cells into irradiated recipient mice.OBJECTIVE:The use of Retroviral vector for transfer of BCR-ABL gene into murine bone marrow cells (BMC) and measurement of efficiency of transfection by flowcytometry.METHODS:Murine bone marrow cells obtained from mice were cultured in a medium containing the supernatant of BCR-ABL (p210) transfected platinum E cells which is rich in retroviral vector carrying the BCR-ABL (p210) gene.The vector express green fluorescent protein (GFP) as well so that the efficiency of transfection of murine BMC with the target gene was able to be measured using flowcytometry.RESULTS:The use of the retrovirus packaging cell line enhanced the transduction of BMC with the retroviral vector and efficiency of transfection was 72% as measured by the flowcytometry.CONCLUSION:Transfer of BCR-ABL gene into murine BMC by retroviral vector that carry GFP marker which allowed the estimation of transfection efficiency by the flowcytomery

Abstract

ABSTRACT:BACKGROUND:The Th-1 cytokines were well implicated in adversely affecting pregnancy and the mechanism by which they cause pregnancy loss was suggested to be mainly vascular by activation of mediators of inflammation like certain adhesion molecules.OBJECTIVE:To study the pathological effect of IFN-γ and VCAM-1 in recurrent pregnancy loss.PATIENTS AND METHODS:The study included three groups of women; Group A: patients had recurrent abortion (n=24), Group B: patients had spontaneous abortion for the first time (n=10), Group C: women with elective pregnancy termination (n=6). Curate samples obtained from these women were subjected for in situ hybridization technique to determine the in situ expression of IFN-γ, and immunohistochemistry analysis to detect the expression of VCAM-1.RESULTS:The in situ expression of IFN-γ was significantly higher in women with RSA as compared with normal pregnant and first abortion groups (p=0.000 and 0.002) respectively, and the expression of VCAM-1 was also significantly higher (p=0.005) in women with RSA as compared with those who had abortion for the first time, with a positive correlation between the expression of IFN-γ and VCAM-1 (r=0.418; p<0.05) in women with RSA.CONCLUSION:The data of this study strengthen the possibility that type-1 immune response may have the upper hand in the pathology of RSA on multidirectional bases including up regulation of the surface expression of VCAM-1

Abstract

Background: The possible immunological bases of recurrent spontaneous abortion (RSA) are still largely unknown, aberrant type 1 cytokine production; interferon-γ (IFNγ), and a defective type 2 cytokine; Interleukin-10 (IL-10) has been suggested to be related to the incidence of unexplained RSA.Objective: To study the relation between the in situ expression of IFNγ and IL-10 in women with recurrent spontaneous abortion.Materials and Methods: The study included three groups of women; Group A: patients had recurrent abortion (n=24), Group B: patients had spontaneous abortion for the first time (n=10), Group C: women with elective pregnancy termination (n=6). Curate samples obtained from these women were subjected for in situ hybridization technique to detect and determine the in situ expression of IFN-γ and IL-10.Results: The in situ expression of IFN-γ was significantly higher in women with RSA as compared with normal pregnant and first abortion groups (p=0.000 and 0.002 respectively), while IL-10 expression was significantly lower in women with RSA as compared with first abortion group (p=0.005), and the ratio of IFN-γ/IL-10 was 1.97 in women with recurrent abortion, while that of normal pregnant and first abortion groups were 0.67 and 0.73 respectively.Conclusion: The data of this study strengthened the possibility that type-1 immune response may have the upper hand in the pathology of RSA in association with reduction in the type-2 immune response.Key words: RSA, IFN-γ, IL-10

خلفية الدراسة: لا تزال الأسس المناعية الممكنة للإجهاض التلقائي المتكرر غير معروفة بدرجة كبيرة لحد الآن. وقد أُعتبِرت الزيادة في السايتوكينات من النوع الأول (Type 1 cytokines) مثل (IFN-γ), والإنخفاض في السايتوكينات من النوع الثاني (Type 2 cytokines) مثل (IL-10) من الأسباب المحتملة لعدد من حالات الاجهاض المتكرر.هدف الدراسة: دراسة العلاقة بين التعبير الموضعي للانترفيرون كاما والانترلوكين العاشر في حالات الاجهاض التلقائي المتكرر.المرضى وطريقة الدراسة: تضمنت هذه الدراسة ثلاثة مجموعات من النساء، المجموعة A : حالات اجهاض متكرر(n=24)، المجموعة B : حالات اجهاض تلقائي للمرة الأولى (n=10)، المجموعة C : حالات انهاء حمل ارادي (n=6). اجريت تقنية التهجين الموضعي للكشف عن وتحديد كل من IL-10 , IFN-γ في عينات الجرف الرحمي لهذه الحالات.النتائج: اظهرت النتائج زيادة معنوية كبيرة في التعبير الموضعي IFN-γ في حالات الاجهاض المتكرر مقارنة مع حالات الاجهاض لأول مرة و الحمل الطبيعي, مع نقصان معنوي كبير في التعبير الموضعي IL-10 في حالات الاجهاض المتكرر مقارنة مع الحالات الأخرى. أما النسبة بين IFN-γ - IL-10 فقد كانت 1.97 في حالات الاجهاض المتكرر, بينما كانت 0.67 و 0.73 في حالات الحمل الطبيعي و الاجهاض لأول مرة على التوالي. الاستنتاج: لقد قوّت نتائج هذه الدراسة احتمالية الدور المهم للنوع الأول من الاستجابة المناعية في مرضية الاجهاض التلقائي المتكرر مترابطة مع انخفاض واضح في النوع الثاني من الاستجابة المناعية.مفاتيح الكلمات: الاجهاض المتكرر, IFN-γ, IL-10

Abstract

Background: To elucidate the possible role of human cytomegalovirus in pregnancy loss through induction of certain pro-inflammatory adhesion molecules.Methods: Paraffin embedded sections of curate samples were obtained from 34 women had spontaneous abortion, and 5 women had elective termination of pregnancy (as control), and then subjected for immunohistochemistry analysis to detect human cytomegalovirus (HCMV) early protein and VCAM-1 molecule.Results: Nine out of 34 women with spontaneous abortion were positive for HCMV early protein, with asignificantly higher expression of VCAM-1 in HCMV positive cases as compared with HCMV negative and the control groups (p = 0.05, 0.001 respectively).Conclusion: HCMV infection may play an important role in the pathology of pregnancy loss on multidirectional bases include inducing the surface expression of pro-inflammatory adhesion molecules like VCAM-1.Key wards: human cytomegalovirus, VCAM-1, Abortion

Abstract

background: human epidermal growth factor receptor-2 (her2/neu) is related to growth factor receptors with alkaline kinase activity and it is regarded as important prognostic and therapeutic factor that can depended on in breast cancer therapy. HER2/neu expression by immunohistochemistry (IHC) is submitted to a great in terob server inconsistency. Subsequently additional confirmatory tests for assessment of gene alterations and amplification status are needed for patients with early or metastatic breast cancer. In situ hybridization techniques and specifically Chromogenic in situ hybridization (CISH) was arise as a practical, cost-effective, and alternative to fluorescent in situ hybridization in testing for gene alterationAims of the study: was to determinate her2/neu gene amplification in (27) breast cancer cases with equivocal her2/neu (2+) detected by IHC by using dual-color chromogenic in situ hybridization (CISH) method and to compare the results with clinic-pathological parameters of those patients like age, stage, grade, ER, PR and ki-67.Methods: twenty seven breast-cancer cases with equivocal HER2-IHC(2+) test results were prospectively collected and reanalyzedusing dual-color CISH (Zyto- Vision) detection kit for further identification of her2/neu gene amplification.Results: Cases with equivocal staining for her2/neu were about 27 cases; these cases were involved in chromogenic in situ hybridization technique to determine the status of her2/neu gene. After CISH study 12(44.4%)cases show her2/neu gene amplification (her2/neu/CEN17 ratio > 2.2).In15 (55.5%) cases out of 27 cases, no amplification of the HER2 gene was shown (her2/neu/CEN17 ratio <1.8).Conclusions: A significant proportion of patients with equivocal IHC(2+) test results show amplification of the HER2 gene by ISH.CISH method has the advantage of diagnosis of gene amplification and revealing tissue histopathology under light microscope.

Abstract

Background: Breast cancer is the most common malignancy affecting females worldwide. The association of Epstein-Barr virus (EBV) with this cancer is a long-standing interest to this field.Aim: to investigate the presence of EBV in breast tumor tissue in relation to age.Patients and Methods: Paraffin-embedded tissue blocks from 45 female patients with breast tumors (ranged in age from 28 to 85 years) were retrieved. The cases were grouped into two categories: group (A): included 30 cases with breast carcinoma and group (B): included 15 cases with benign breast diseases as a control group .The expression of EBV protein was examined immunohistochemically. Results: Twelve (40%) of the 30 breast cancer cases (group A) were reported as positive for EBV expression and significantly higher in patients less than 40 years age (P= 0.026). Conclusion: the results of the current study might refer to the significance of EBV expression in breast tissue of young patients with breast cancer.
