Abstract

Background: To determine the DNA content in subfertile patients and to correlate it
with seminal sperm concentration.
Design: Prospective observational study.
Setting: College of Medicine, Dept. of Physiological chemistry and Institute for
embryo Research and infertility treatment-University of Baghdad.The study was conducted through years 2004-2005.
Methods: A random sample of 61 subfertile male patients undergoing semen evaluation and aged from 20-45 years were studied. Semen samples were assessed for seminal sperm concentration microscopically and were classified into 3 different groups according to count (million/ml).Then sperm D N A content (µg/ml) was estimated using a microchemical spectrophotometric method.
Results: The three groups were statistically of significant difference( P <0.01 ).The D N A concentration per spermatozoan in Gr.III ( 1 20 million/ml) was higher than the other two groups It was also noticed that there was a statistically significant (P< 0.05 ) correlation in Gr.III b etween D N A content and sperm concentration . No significant (P >0.05 )correlation was observed in the other two groups (Gr. I & Gr. II ).
Conclusion: Significant positive correlation was obtained between sperm concentration and DNA content. in the oligozoospermic subfertile patient , but no significant correlation was found in the normozoopermic patients . D N A content per spermatozoan from oligozoospermic patients was higher than that in normozoospermic patients.

Abstract

The objective of the present study is to measure some biochemical and hormonal contents of oviductal flushing medium (OFM)used for in vitro fertilization and embryonic development of mice as a model for mammals.Oviductal flushing medium of superovulated and spontaneously ovulated mice was analyzed for certain biochemical and hormonal contents. Amino acids( taurine, alanine, aspartate, glutamine and glycine) were measured using HPLC. Glucose and fructose were estimated using glucose oxidase method and fructose indol-reagent colorimetric method ,respectively. Magnesium , calcium ,sodium and potassium ions were determined using flame photometry and atomic absorption spectrophotometry. Luteinizing hormone (LH) and follicular stimulating hormone(FSH) were measured by enzyme linked fluorescent assay technique.The results of this investigation show that there is no significant (P>0.05) difference in the concentration of taurine ,glutamine and alanine in the OFM of SUO and SO mice. The OFM contents of aspartate and glycine in SUO is significantly(P<0.05) higher than that of SO group .A significant (P<0.05) decrease was observed in the concentration of glucose (µmol/L) in OFM of SUO compared to SO females.The concentration of Ca++ in the OFM of SUO (66.5µg/ml) was significantly (P<0.05) higher than that of SO mice(60.2 µg/ml). There was a significant(P<0.05) decrease in the level of LH in the OFM of SUO compared to SO mice .It is concluded from the present study that the OFM of superovulated mice contains higher amounts of different amino acids ,electrolytes and hormones that lead to offer a proper environment for in vitro fertilization and embryonic development of early cleavage stages in mice .

هدفت الدراسة الحالية لتحديد كمية بعض محتويات الكيمياء الحيوية والهرمونية للسائل الدافق لقناة البيض والمستخدم في الإخصاب ونمو الأجنة في الزجاج للفئران والتي استخدمت نموذجاً للبائن.حلل الوسط الدافق لقناة البيض من فئران محفزة واخرى غير محفزة بالهرمونات تحليلا كيميائيا وهرمونيا فقد تم حساب كمية بعض الأحماض الامينية ( taurine, alanine, aspartate, glutamine and glycine) باستخدام جهاز HPLC .تم تقدير كمية سكر الكلوكوز والفركتوز باستخدام طريقة glucose oxidase وطريقة fructose indol-reagent colormietric على التوالي.حددت مستويات أيونات المنغنيز والكالسيوم والصوديوم والبوتاسيوم باستخدام طريقتي flame photometry and atomic absorption spectrophotometry . تم حساب هرموني المحفز الجريبي((FSH واللوتيني (LH) بأستخدام تقنيةEnzyme linked fluorescent assay. أظهرت النتائج الحالية عدم وجود فرق معنوي(P>0.05) في تركيز التورين والكلوتامين والألنين في محتوى السائل الدافق من قناة البيض بين مجموعة الفئران المحفزة بالهرمونات وغير المحفزة. محتوى السائل الدافق من الأسبرتيت والكلايسين في قناة بيض الفئران المحفزة بالهرمونات أعلى معنويا (P<0.05) من الفئران غير المحفزة . وجد انخفاض معنوي(P<0.05) في تركيز الكلوكوز في السائل الدافق لقناة بيض الفئران المحفزة بالهرمونات بما يقابلها في الفئران غير المحفزة. تركيز الكالسيوم في الفئران المحفزة بالهرمونات(66,5 ملغم/مللتر) أعلى معنويا P<0.05)) مـن تركيزه في الفئران غير المحفزة بالهرمونات(60,2 ملغم/مللتر) . كمـا لوحظ وجود انخفاض معنوي( P<0.05) في معدل الهرمون اللوتيني الموجود في السائل الدافق لفئران محفزة بالهرمونات بالمقارنة مع فئران غير محفزة.لذا نستنتج من هذه الدراسة أن السائل الدافق من قناة بيض الفئران المحفزة بالهرمونات يحتوي على كميات أكبر من مختلف الأحماض الأمينية والأيونات والهرمونات مما أدى الى توفير بيئة ملائمة للأخصاب في الزجاج والتطور الجنيني لمراحل الأنقسام المبكرة في الفئران.

Serum Magnesium Concentration in Patients with Leukemia and Lymphoma

Abstract

Background: Leukemias and lymphomas are malignant disorders that occur in the blood forming organs and lymphoid tissue respectively. They are classified to types and several subtypes such as acute or chronic, lymphocytic or myelocytic and T-cell or B-cell lymphocytic for leukemias and histologically into Hodgkin’s and Non-Hodgkin’s for lymphomas. Literatures do not contain many research work on magnesium in patients with these disorders, although this mineral is essential for many metabolic, enzymic, regulatory and immune reactions in the human body. Therefore, the present study was aimed to evaluate the level of magnesium in the sera of patients with different types of leukemia and lymphoma. Patients and Methods: Fifty five patients with leukemia and lymphoma and twenty five healthy controls were studied. The patients were attendants of Baghdad Teaching Hospital and Pediatric Teaching Hospital from September 2006 to January 2007. The diagnosis was confirmed by examination of both peripheral blood, lymph node biopsy and/or bone marrow examination. Patients were categorized according to French-American-British Criteria. Thirty five patients with ALL, one with CLL, six with AML, three with CML, three with HL and seven with NHL. Venous blood was collected from each patient or control person and sera were obtained by centrifugation. S[Mg] was measured by Atomic Absorption Flame Spectrophotometry.Results:The Mean ± SD of S[Mg] in mg/dl of all types of leukemic and lymphomatous patients was lower than the controls. There was a high statistically significant difference (P<0.01) in patients with ALL and significant difference (P<0.05) in patients with AML and NHL and non-significant difference (P>0.05) in patients with CML and HL, compared with the control. Total patients group showed high significant difference (P<0.01) compared with control group. The mean distribution of S[Mg] among total patients indicated decreased (58.2%), normal (38.2%) and increased (3.6%) levels.Conclusion:The present study disclosed the existence of normal to decreased level of S[Mg] in patients with leukemia and lymphoma suggesting an influence of many variable factors. Although the decreased S[Mg] was statistically significant, it was still within lower normal range.

Abstract

ABSTRACT:BACKGROUND:Fibromyalgia syndrome (FMS) is a common rheumatologic syndrome with multiple manifestations and associated with many diseases, it characterized by chronic wide spread muscular pain and tenderness. Further, circulatory levels of pro-inflammatory cytokines like IL-8 and others may be altered in FMS patients, possibly associated with their symptoms.The objective of the study is to measure IL-8 concentration and to evaluate its role in the pathogenesis of primary FMS.OBJECTIVE:Fifty patients with primary FMS were included in the study (37 females and 13 males) the age range of (17-65) years (Mean ± SD) (40.13 ± 12.0) years, and thirty healthy individuals volunteers (21 females and 9 males), whose age and sex matching with FMS patients, age ranging (18 - 63) years, (Mean ± SD) (36.1 ±10.0) years.METHODS:IL-8 concentration was measured in sera of patients and controls by ELISA kit. Anthropometric measurements like body mass index (BMI) and waist circumference (WCr) were taken ,besides other features like sleep disturbance , emotional distress , and fatigue were reported.RESULTS:IL-8 concentration was higher in FMS patients than controls(40.24±22.0) pg/ml vs. (19.16±7.8) pg/ml. This elevation was highly significant statistically (p=0.000).Other measurements in patients group like BMI, WCr were(28.39±5.0) Kg/m2 and (100.34±13.21) cm respectively .These values were highly significant when compared to their control group (p=0.002)and (p=0.008) respectively .Clinical features like sleep disturbance, emotional distress ,and fatigue showed highly significant difference between the two groups . No significant differences were reported with respect to age and sex.CONCLUSION:The result of the current study suggest that interleukin-8, (IL-8) might have a role in the pathogenesis of FMS.

Abstract

ABSTRACT:BACKGROUND:Nodal Osteoarthritis is a subset of OA characterized by polyarticularinterphalangeal and thumb base OA ,Heberden’sand Bouchard’s nodes formation.C-reactive protein (CRP) is one of the first acute phase response proteins to be elevated in an inflammatory processes.OBJECTIVE:The aim of this study is to investigate whether the inflammatory processes have roles in the development of nodal osteoarthritis (NOA) in Iraqi femalepatients using serum highly sensitive C-reactive protein as aninflammatory marker.SUBJECTS AND METHODS:The study included eighty four females, divided into two main groups,sixty were patients with (NOA), and twenty-four represented the control group. Other subdivisions were applied according to body mass index and menopause status. Measurement of serum (hsCRP), wasapplied to all subjects included in this study, by ELISA sandwich method.RESULTS:Serum highly sensitive C-reactive protein (hsCRP) level was significantly higher (18.08±8.15) (P value < 0.05) in patients with NOA in comparison to control group (2.23±1.73) .CONCLUSION:Highly significant elevated level of serum (hsCRP) observed in the NOA patients compared to control suggest that the inflammatory processes have roles in the development of NOA.

Abstract

Summary:Background: To determine the DNA content in subfertile patients and correlate it with seminal sperm parameters, (count, motility and morphology).Design: Prospective observational study.Setting: College of Medicine, Department of physiological chemistry and Institute for Embryo research and Infertility treatment, University of Baghdad.Methods: A random sample of 58 subfertile male patients undergoing semen evaluation and their age ranged from 20-45 years were studied. Semen samples were assessed for seminal sperms (count , motility and morphology) by direct light microscopy. Sperm DNA content was estimated using a microchemical spectrophotometric method .The data were categorized into four groups according to normal and abnormal sperms count ( million / ml ),percent motility and percent morphology. The normal limits of those sperm function parameters were according to WHO criteria. The four groups were named as Normozoospermic (NZS) (control) , Normoasthenozoospermic (NAZS) ,Normoasthenoteratozoospermic (NATZS) and oligoasthenoteratozoospermic (OATZS) groups.Results: The Mean ± SD of the four groups were of significant difference (P> 0.05 ) with respect to sperm count, sperm percent motility and morphology with exceptions of certain groups. Moreover, a non significant difference (P< 0.05) was found with respect to DNA content (µg /ml) and (µg/sperm) except for Gr. IV which showed a significant difference when compared to others.The correlation coefficients (r values) between sperm count and sperms motility, morphology and DNA content (µg/ml) were non significant in the four groups. Noticeably, the DNA content (µg/sperm) was statistically of significant (P>0.05) negative correlation with sperm count in all groups.Conclusion: All groups were of significant difference (P<0.05) among their Mean ±SD values of their count, motility and morphology. There were few exceptions. No significant differences (P>0.05) were found with respect to DNA content (µg/ml or µg/sperm) except for oligoasthenoteratozoospermic group compared to others. No significant correlation was found between sperm count and each of sperms motility , morphology and DNA content (µg/ml) in the four groups .However, the DNA content (µg/sperm) had a statistically significant (P<0.05) negative correlation with sperm count in all groups.Keywords:DNA, sperm concentration(count million/ml ), Normozoospermic(NZS),Normoasthenoszoopermic(NAZS),Normoasthenoteratozoospermic(NATZS), Oligoasthenoteratozoospermic (OATZS ) .

Abstract

ABSTRACT:BACKGROUND: Osteoarthritis is a degenerative joint disease, occurring primarily in older persons. Although many factors initiate this disease, progress to overt OA needs many biological substances like IL-6 and IL-8 that reduce cartilage repair ability or like reactive species that induce cartilage destruction through direct degradation of matrix components. OBJECTIVE: To shed the light on the expression of serum IL-6 and IL-8 levels, and production of serum malondialdehyde level (marker of oxidative stress), then estimation of the correlation among serum IL-6, IL-8 and MDA levels in patients with knee osteoarthritis.PATIENTS AND METHODS: This study was conducted in AL-Kadhemiya Teaching Hospital during the period from April 2011 to December 2011. The study included two groups: knee osteoarthritis( KOA) group of patients and the control group. Each group was further sub-divided into: obese & non-obese according to their BMIs. Sixty osteoarthritic patients aged 40-70 years were chosen depending on medical history, clinical examination, and radiographic observations. 10 ml of blood needed for assessment of the above makers. Thirty healthy control (age and sex-matched) were enrolled in this study. RESULTS: Mean serum IL-6 and IL-8 levels were significantly higher in KOA patients compared to control group for both (P<0.001), with the highest levels seen in the obese KOA group. Mean serum MDA level was significantly higher in patients with KOA compared with that in control (P<0.001). Significant negative correlation was observed between serum IL-6 and IL-8 levels. CONCLUSION: IL-6, IL-8 & MDA have important role in the pathogenesis of KOA. Receptors of IL-6 and IL-8 on chondrocytes could be considered as target for future therapy of any arthritic inflammation and also to consider MDA reduction in patients with knee osteoarthritis, as part of treatment of osteoarthritis.

Association of Erythropoietin, Adiponectin and Leptin levels with Anemia in uremic diabetic patients (Under hemodialysis):

Abstract

Background: End stage renal disease in patients with diabetes mellitus has been called a medical catastrophe of worldwide dimensions .It was recognized that anemia is a complication of diabetes, particularly in patients with diabetic kidney diseases. The purpose of the present study is to determine whether erythropoietin, adiponectin, and leptin levels correlate with anemia in uremic diabetic patients (under hemodialysis).
Patients and Methods: The studied groups were comprised of (30) diabetic patients(9 patients with type1 and 21 type2 diabetes) with renal failure (group2) who had been under hemodialysis compared with (30) healthy controls (group1). Blood film was obtained in addition to blood tests include fasting serum glucose (FSG), glycosylated hemoglobin (A1c), insulin, urea, creatinine, uric acid, estimated glomerular filtration rate (eGFR), serum erythropoietin (EPO), Adiponectin (ADPN),and leptin.Results: It was found that uremic diabetic patients in group2(G2) had higher FSG, A1c, insulin and higher levels of urea, creatinine, uric acid and lower levels of eGFR when compared to healthy controls (P <0.05). Anemia was confirmed in the diabetic patients by Hemoglobin (Hb) and hematocrit (Hct) that were lower than healthy control (group1) (G1) (P<0.05).EPO, ADPN levels were increased in uremic diabetic patients as compared to control subjects (P <0.05). While, leptin levels were higher in type2 diabetes when compared to type1 diabetes and control subjects (group1) (P <0.05). There were positive significant correlations between (FSG and A1c), (Hb and Hct), (EPO and leptin), (EPO and Hct), (ADPN and urea). In addition there were significant negative correlations between (FSG and EPO), (A1c and leptin), (ADPN and leptin).Creatinine was negatively associated with Hb and Hct. ADPN was negatively associated with Hb in uremic diabetics.Conclusions: Anemia is a common accompaniment to diabetes, particularly in those with renal failure or under hemodialysis .Adipocytokines like leptin and adiponectin may involved with anemia in these patients. Leptin in the present study was positively associated with EPO levels (the hormone that stimulates red blood cells production in the bone marrow). ADPN was negatively associated with Hb. These elevated levels of ADPN may antagonize EPO and leptin to increase the hemopoiesis rate and Hb levels in uremic diabetic patients in addition to other factors involved with diabetes.
