Construction of New Ethambutol Selective Electrodes for Determination Ethambutol in Pharmaceutical Drugs

Abstract

Ethambutol selective electrodes were constructed based on ethambutol-phosphotungstate (EMB-PT) complex with different plasticizers; di-butyl phosphate (DBP), tri-butyl phosphate (TBP), di-butyl phthalate (DBPH), o-nitro phenyl octyl ether (NPOE) and di-octyl phthalate (DOPH). The electrodes based on DBP, TBP and DOPH plasticizers gave slopes near to Narnistain slope which are 27.5, 25.2 and 34.5 mV/decade with detection limit of 1.9x10-5 M and concentration range 10-1 to 10-4 M and pH range 3.0 – 8.0. The electrodes based on DBPH and NPOE showed non-Nernistain slopes, 21.5mV/decade for both plasticizers. Interfering of some cations was investigated and shows no interfering with electrodes response. Potentiometric methods were used for measuring ethambutol in pharmaceutical drugs (tablets) and the electrode based on DOPH was used for determination. The recovery obtained from measuring was in good agreements with that given in British Pharmacopeias.

في هذه الدراسة حضرت اقطاب انتفائية لمادة الايثمبوتول الدوائية معتمدة على معقد الدواء مع dodeca-Tungstophoric acid مع مجموعة من المواد الملدن di-butyl phosphate (DBP), Tri-butyl phosphate (TBP), di-butyl phthalate (DBPH),(NPOE) o -phenyl octyl ether وdi octyl phthalate (DOPH) - .اظهرت الاقطاب مع الملدنات DBP, TBP, DOPH انحدر27.5,21.5,34.5 mV/decade على التوالي وبحد تحسسس 1.9x10-5 مولاري وبمدى تركيز خطي 10-1-10-4 مولاري ومدى دالة حامضية 3-8.اما الاقطاب للملدنات DBPH, NPOE اظهرت استجابة غير نرنستية بانحدار 21.5 mV/decade .اظهرت الدراسة عدم وجود تداخل لعدد من الايونات الموجبة الاحادية والثنائية والثلاثية .تم تطبيق هذا القطب على المستضرات الصيدلانية باستحدام قطب المستندعلى DOPH كملدن وقد اعطت نتائج جيدة

Abstract

Derivative spectrophotometric techniques (first, second, third and fourth derivative) were
developed for the determination of four â-lactam antibiotic binary mixtures; in combinations
containing these compounds. The simultaneous determination of these compounds was
accomplished by derivative (1D, 2D, 3D and 4D) spectrophotometric technique and applying zerocrossing
technique amoxiclline trihydrate with cephalexin monohydrate (mix I) using 1D spectrum
zero crossing at valley 238.2 nm, and 2D spectrum at valley 263.8 nm, respectively, amoxiclline
trihydrate with cloxacillin sodium (mix II) using 2D at valley 281 and 4D at valley 230.6,
respectively, cephalexin monhydrate with cloxacillin sodium (mix III) using 1D spectrum zero
crossing at 272 nm and 2D spectrum at 253.2 nm, respectively and ampicillin trihydrate with
cloxacillin sodium (mix IV) using 1D at 282nm and 2D at 271.6 nm, respectively. From this
studying it was noticed that in all the applications of the previous methods the correlation
coefficient of calibration curves not less than 0.999 and the relative standard deviation not exceed to
0.214. The four described procedures were successfully applied to the determination of these
compounds in synthetic mixtures and in preparations with high percentage of recovery, accuracy
and precision .The procedures do not require any separation step.

Abstract

Amoxicillin trihydrate selective electrodes were prepared based on amoxicillin-phosphotungstate (Amox-PT) as an active sensor with different plasticizers, di-butyl phosphate (DBP), di-butyl phthalate (DBPH), di-octyl phthalate (DOPH), tri-butyl phosphate (TBP) and o-nitro phenyl octyl ether (NPOE) in PVC matrix membranes. The study was carried out to investigate the electrode parameters, effect of pH, selectivity and UV-derivative spectrophotometer technique. Internal filling solution of 10-3 M + 10-3 M NaCl was used to fill the electrodes. The best electrode was based on DBP plasticizer which gave a slope 58.7 mV/decade and detection limit of 2 x 10-6 M displayed good stability and reproducibility. The electrode was used to determine the amoxicillin in oral suspension and capsules. The results were compared with UV-derivative spectrophotometer technique and the recovery for the drugs obtained by second derivative are quite comparable with the recovery obtained by amoxicillin electrodes.

حضرت أقطاب ﺇنتقائية للأموكسيسلين بتحضير المادة الفعالة (أموكسيسلين- فوسفوتنكستيت) Amox-PT مع الملدنات المختلفة وهي داي بيوتيل فوسفيت (DBP) , داي بيوتيل فثاليت (DBPH) ,داي اوكتيل فثاليت (DOPH) ,ثراي بيوتيل فوسفيت (TBP) و اورثو نيترو فنيل ﺇيثر (NPOE) في غشاء اصله من بولي فاينيل كلورايد .(PVC) الدراسة شملت خواص ألأقطاب كتأثير الدالة الحامضية pH, المحلول الداخلي للأقطاب, الأنتقائية . ﺇن أفضل قطب المحضر باستعمــال المـلدن DBP والمحـلول الداخلـي (10-3M Amox+10-3M NaCl) حيث ان الميل 58.7 mV/decade وبحدود تحسس 2x10-6M وله ثبوتية وتكرارية جيدة. ﺇن هذا القطب أستخدم لتقدير الأموكسيسلين في المستحضرات الدوائية الكبسول والمعلق الفموي. النتائج تطابقت مع نتائج المشتقة الطيفية الثانية.

Abstract

Phenytoin selective electrodes were constructed based on penytoin-phosphotungstate (Ph-PT) complex with different plasticizers; di-butyl phosphate (DBP), tri-butyl phosphate (TBP), di-butyl phthalate (DBPH),and o-nitro phenyl octyl ether (NPOE) phthalate. The electrodes based on DBPH, ONPOE plasticizers gave Narnistain slope which are, 56.4 and 55.3mV/decade with detection limit of 1.9x10-5 M , 1.8x10-5 and concentration range 10-1 to 10-4 M and pH range 3.0 – 8.0. The electrodes based on TBP and DBP showed non-Nernistain slopes, 40.2,40.5 mV/decade for both plasticizers. Interfering of some cations was investigated and shows no interfering with electrodes response. Potentiometric methods were used for measuring phenytion in pharmaceutical drugs (tablets) and the electrode based on DBPH was used for determination. The recovery obtained from measuring was in good agreements with that given in British Pharmacopeias.

في هذه الدراسة حضرت اقطاب انتقائية للفنيتون معتمدة على معقد الدواء مع dodeca-Tungstophpsphoric acid ومجموعة من المواد الملدنة Di-butyl phthalate(DBPH),Di-bytyl phosphate (DBP),Tri-butyl phosphate (TBP),O-nitro phenyl octyl ether (ONOPE), اظهرت النتائج ان اقطاب الفنيتون مع الملدنات (DBPH (و ONPOE اعطت انحدرارا= 56.4 , 55.3 mV/decadeعلى التوالي ومدى تركيز خطي 1x10-1 -1x10-4وحد تححسس 10-4 1.9x .اما اقطاب المعتمدة على الملدنات و TBP و DBPاعطت انحدارا لانرنستي 40.2 , mV/decade40.5على التوالي .اظهرت الدراسة عدم وجود تداخل للعدد من الايونات الموجبة الاحادية والثنائية والثلاثية . لدراسة الخواص العملية في محاليل قياسية محضرة مختبريا وكذلك في نماذج دوائية تم استعمال الاقطاب كاقطاب كاشفة في عملية التسحيح النرنستي واستعملت طريقة الاضافات القياسية لنماذج دوائية للفنيتون.

Abstract

Chloride selective electrodcs based on PVC matrix membrane consist of various alkyl quaternary ammonium salts such as tetra heptylammonium chloride (THACI), and tetraoctylammonium chloride (TOACI) as a sensors with various mediators were prepared and studied.The study includes the electrodes parameters determination (slope, linear response range, detection limit, pH effect , life time and selectivity). Chloride electrode based on (TOACI) as sensor with di-n-octyl phthalate (DOPH) as mediator gives excellent response with a slop equal 60,1 mV.'decade which is the near to the Nernstian slope. Concentration was ranged from 10-2 M to 10-4 M with detection limit around 10-5 M. This electrode was used for chloride determination ion in samples using different potentiometric methods.

تأثير تداخل بعض الايونات على قياس ايونات السيانيد والكبريتيد في الماء باستخدام الطرق الطيفية والبولاغرافية

Abstract

Spectrophotometric and polarographic techniques were used for cyanide and sulfide ions determination. Chloramine-T and barbituric acid were used as reagents for complex formation and 0.5M H3BO3+ 0.1M KOH and 0.02 M ascorbic acid + 0.018 M disodium EDTA + 100 mL of 1M NaOH supporting electrolytes for polarography. The Interference effect of the interfering ions Cl -, Ca2+, Mg2+, Fe3+, and Cu2+ was studied. For eliminating the effect of interference , distillation method was used for distillation of cyanide solutions. The concentration of cyanide ion in several samples of drinking water, natural water and waste water were determined by both techniques. The linearity of cyanide determination by spectrophotometric was ranged from (0.05 - 0.5 ppm) at a wavelength (578 nm). The linearity for cyanide ion determination by polarography was ranged from (0.02 - 0.2 ppm).

تم تعيين تركيزي ايوني السيانيد و الكبريتيد باستخدام الطرق الطيفية والبولاروكرافية و قد تم تثبيت مدى التحسس بالطريقة الطيفية التي تتراوح في المدى ( ppm 0.5 - 0.05 ) بتكوين معقد مع Chloramine – T و Barbituric acid عند طول موجي(578 nm) . كانت درجة التحسس بالطريقة البولاروكرافية تتراوح من ( ppm 0.2 – 0.02 ) . وكما درست التداخلات الحاصلة نتيجة تواجد الايونات التالية ( Cl - , Fe2+ , Mg2+ , Ca2+ , S 2- , Cu2+) قبل و بعد عملية التقطير لمحلول السيانيد و قد وجد ان الايونات الموجبة لها تاثير مباشر على تعيين السيانيد اما ايون الكبريتيد فله تداخل عالي جدا لا يمكن ازالته بعد التقطير و يمكن ازالة تاثيره باضافة كاربونات الكادميوم او تحويل السيانيد الى حامض الفورميك .تم تعيين السيانيد في نماذج من ماء الحنفية و المياه الطبيعية ومياه الفضلات.
