The effect of autologous bone marrow-derived stem cells with estimation of molecular events on tooth socket healing in diabetic rabbits (Immunohistochemical study)

Abstract

Background: Healing of a tooth extraction socket is a complex process involving tissue repair and regeneration. Itinvolves chemotaxis of appropriate cells into the wound, Transformation of undifferentiated mesenchymal cells toosteoprogenitor cells, proliferation and differentiation of committed bone forming cells, extracellular matrix synthesis,mineralization of osteoid, maturation and remodeling of bone. These cellular events are precisely controlled andregulated by specific signaling molecules. Some of these like transforming growth factor beta (TGF-β), vascularendothelial growth factor (VEGF), bone morphogenetic proteins (BMP) and insulin like growth factors (IGF) are wellconserved proteins involved in the initial response to injury and repair in soft and hard tissue.Materials and Methods: 48 rabbits weighting an average of (2.5 - 3 kg) were used in this experimental study, and dividedinto 3 groups as follows; group A ( contains 16 healthy rabbits regarded as control group ) , Group B ( contains 16diabetic rabbits not received treatment ), group C ( contains 16 controlled diabetic rabbits received insulin as atreatment ), the lower incisor for each rabbits was extracted, after 2, 10, 20 and 30 days of healing periods afterscarification , the socket was analysed by immunohistochemical (IHC) estimation of growth factors : TGFbeta-3 , VEGF ,IGF-1R , BMP-4 .Results: IHC findings revealed high positive expression of TGFbeta-3, BMP-4 on fibroblasts, osteoproginetor cells,osteoblasts and osteocytes,high positive expression of VEGF on endothelial cells and high positive expression of IGF-1Ron endothelial cells and moderate expression on osteoblasts.Conclusions: The inhibition of proliferation and migration of osteoblasts, or differentiation from progenitor cells, isimplicated in the delay of teeth sockets healing. For this fact the results of the present study concluded that in thediabetic healing bone (rabbits of group B), the onset of cell proliferation and osteoblast differentiation were delayedand subsequently prolonged healing process when compared with the other groups (rabbits of group A,Cand D).

Abstract

Back ground: Type 1 diabetes is the result of an autoimmune attack against the insulin-producing beta cells of thepancreas. Current treatment for patients with type 1 diabetes typically involves a rigorous and invasive regimen oftesting blood glucose levels many times a day along with injections of recombinant insulin. Many recent researcheshave shown that stem cell therapy can be the best choice for treatment of this disease. The aims of this researchwere investigating regeneration of pancreatic beta cells of type 1 diabetic rabbits after stem cell transplantation.Materials and Methods: 32 rabbits weighting an average of (2.5 - 3 kg) were used in this experimental study, anddivided into 2 groups as follows; group A ( contains 16 controlled diabetic rabbits received insulin as a treatment )and group B ( contains 16 diabetic rabbits received autologous mesenchymal stem cells as a treatment).Theinduction of diabetes was achieved by a single dose of intravenous injection of the Alloxan, which was administeredto the rabbits via the marginal ear vein, mesenchymal stem cells were differentiated into insulin – producing cells andreimplanted into the rabbits of group B with daily monitoring of blood glucose level and body weight.Results: The insulin – producing cells regulated the hyperglycemia resulted from diabetic rabbits , 7 to 9 days afterreimplantation the blood glucose level were decreased from about( 400 mg/dl into 180 mg/dl).Conclusions: Islet-like functional cells can be differentiated from bone-marrow mesenchymal stem cells (MSCs),which may be a new procedure for clinical diabetes stem -cell therapy, these cells controlled blood glucose level indiabetic rabbits as the effect of insulin. MSCs play an important role in diabetes therapy by islet differentiation andtransplantation.

Abstract

The results of the IHC showed there was positive expression for bcl2 in the control group with high score (++) high and this expression will decrease with the increase of the dose of the PC extract in the treated group .And the results of the ultrastructural examination by using TEM showed there was increase in the N/C ratio, well developed ER, increase number of variably sized mitochondria in the control group while the treated group revealed there was many vacuoles in the cytoplasm , plasma membrane blebbing and formation the apoptotic bodies. From our study we concluded that there was clear effect on the ultrastructural after using PC as anti- tumor.

وأظهرت نتائج IHC كان هناك تعبير إيجابي لbcl2 في السيطرة على المجموعة مع درجة عالية (++) مرتفعة وهذا التعبير ستنخفض مع زيادة جرعة من مستخلص الكمبيوتر في المجموعة المعالجة. ونتائج التركيبية وأظهر الفحص باستخدام تيم كان هناك زيادة في ن نسبة / C، ER متطورة، وزيادة عدد الميتوكوندريا الحجم بنسب مختلفة في المجموعة الضابطة في حين كشفت المجموعة التي تلقت العلاج هناك العديد من الفجوات في السيتوبلازم، blebbing غشاء البلازما وتشكيل الهيئات أفكارك . من دراستنا استنتجنا أن هناك تأثير واضح على التركيبية بعد استخدام الكمبيوتر كما المضادة للورم.

Production of polysaccharide from Local Isolates of Lactobacillus and Using as Antioxidant

Abstract

Lactobacillus bacteria was Isolated from different food samples included banana, tomato, apple, mandarin orange, cheese, buffalo milk, local yogurt and imported yogurt and from infant feaces. A Total of 65 isolates were obtained out of 64 food samples, and 11 samples of newborn feaces belong to Lactobacillus spp included 20 isolates of Lactobacillus acidophilus, 23 isolates of Lactobacillus plantarum, 6 isolates of Lactobacillus casei, 12 isolates of Lactobacillus bulgaricus, 3 isolates of Lactobacillus johnsonii and one isolates of Lactobacillus helveticus. Detectine of polysaccharides for all obtained bacteria was done . Results showed that the bast amount of produced of polysaccharide was from Lactobacillus plantarum while isolated from imported yoghurt (yR5) 0.850 g.L-1 . which, Lactobacillus plantarum isolated from Tomato (T4) had 0.014 g.L-1 .Results of antioxidant actiivity showed that the highest activity was belong to Lactobacillus bulgaricus isolated from local yoghurt (yL6) was 67.51%. which the lower activity was belong to Lactobacillus acidophilus isolated from local chees (cha2) was 29.1%. compared with the industrial antioxidant (BHT) and α- tocopherol. with activites of antioxidant was (82.70 and 73.96) % respectively.

عزلت بكتريا Lactobacillus من عينات غذائية مختلفة وشملت (الموز والطماطة والتفاح واللالنكي (اليوسفي) والجبن (الأبيض الطري) وحليب الجاموس و اليوكرت المحلي واليوكرت المستورد) وكذلك من براز الأطفال حديثي الولادة, وتم الحصول على 65 عزلة من أصل 64 عينه غذائية و11 عينة من براز الأطفال عائدة الى جنس .Lactobacillus spp وشملت 20 عزلة من بكتريا Lactobacillus acidophilus و23 عزلة من بكتريا Lactobacillus plantarum و 6 عزلات من بكتريا Lactobacillus casei و 12 عزلة من Lactobacillus bulgaricus و3 عزلات من بكتريا Lactobacillus johnsonii وعزلة واحدة من بكترياLactobacillus helveticus. وتم الكشف عن البولي سكرايد وتقدير كميته لجميع أنواع بكتريا حامض اللاكتيك المعزولة. وكانت أفضل كمية أنتاج للبولي سكرايد من العزلة Lactobasillus plantarium المعزولة من اليوكرت المستورد (yR5)، أذ بلغت كمية الأنتاج 0.850 غم. لتر-1، بينما العزلة Lactobacillus plantarum المعزولة من الطماطةT4) ) فقد بلغت كمية الأنتاج 0.014 غم. لتر-1. أما بالنسبة لفعالية البولي سكرايد المضادة للأكسدة كانت أعلى فعالية من العزلة Lactobacillus bulgaricus المعزولة من اليوكرت المحلي (yL6) أذ بلغت 67.51 % بينما كانت أقل فعالية من العزلة Lactobacillus acidophilus المعزولة من الجبن المحلي Cha2 والتي بلغت %1.29. وبالمقارنة مع مضادات الأكسدة الصناعي BHT ومضاد الأكسدة الطبيعي α-tocopherol, أظهرت النتائج إن الفعالية المضادة للأكسدة باستعمال المضاد الصناعي BHT بلغت 82.70 % في حين فعالية مضادة الأكسدة الطبيعية α-tocopherol كانت 73.96%.
