Estimation of lipopolysaccharide concentration in the content of ruminal fluid and feces of dairy cows that suffering from subacute ruminal acidosis

Abstract

Subacute ruminal acidosis'(SARA) is a common health and productions problem in dairy cattle and described as repeated incidences of drop pH. of the rumen. The aim of the study was to estimate the lipopolysaccharide concentration in the content of ruminal fluid and feces of dairy cows as a diagnostic tool for subacute ruminal acidosis. Four hundred and twenty- seven cows examined in this study in Al-Qadisiyah Province, from these cows, Ninety lactating cows, were have affected with subacute ruminal acidosis. Moreover, fifty healthy lactating cows were considered as a control. Both diseased and control cows were exposed to additional tests of ruminal fluid and fecal samples. The difference observed in the lipopolysaccharide (LPS) concentration in the ruminal fluid and fecal samples 97.772 and 108.823 Eu/ml respectively, of SARA group which were significantly (P≤0.05) higher than the control group. The ruminal and fecal LPS can be used as diagnostic tools to identify the SARA.

Abstract

The objective of this study was to find the relationship of acidity (pH) among following constituents: ruminal fluid, blood, milk, and feces, and evaluating it as an indicator and diagnostic tool for this disease in crossbreed dairy cows in Al-Diwaniyah province. A randomized selection of crossbreed dairy cows in Al-Diwaniyah province in two seasons Winter and Autumn (Sep 2013 – Feb 2014), about 427 total heads, 90 as SARA group and 50 as control. Our results of mean values of pH in the subacute ruminal acidosis (SARA) group in the ruminal fluid, milk, blood, and fecal samples were (5.22), (6.70), (7.0), and (5.56) respectively, which were different and lowest from the control group. The data obtained were processed statistically and compared together to detect the presence of significance revealed that pH of ruminal fluids and pH of fecal samples took the significant differences between groups (p≤0.05). While there were no significant difference in the milk and blood pH between the control and SARA sets (p≥0.05). Moreover, the study revealed that pH levels were in both rumenocentesis and stomach tubation sets did not have any significant difference (P ≥ 0.05). According to the results, which were confident, can be considering the pH is an important diagnostic tool to identify the SARA in dairy cows.

أن الهدف من هذه الدراسة هو لإيجاد العلاقة للدالة الحامضية (pH) في كل من عينات سائل الكرش والحليب والدم والبراز، وتقيم هذه الدالة كمؤشر ووسيلة تشخيصية مهمة لمرض حموضة الكرش تحت الحادة في الابقار الحلوب المضربة في محافظة الديوانية. تم اختيار حيوانات الدراسة بشكل عشوائي في محافظة الديوانية وخلال فصلي الخريف والشتاء (من أيلول 2013 ولغاية شباط 2014) وكان المجموع الكلي للأبقار427 رأسا تضمنت 50 رأسا كمجموعة سيطرة و90 رأسا هي المجموعة التي أصيبت بالحموضة تحت الحادة. أظهرت النتائج ان قيم المتوسط الحسابي للدالة الحامضية في مجموعة الدراسة في سائل الكرش والحليب والدم والبراز هي كالاتي: 5.56, 7, 6.70, 5.22 على التوالي والتي كانت مختلفة واقل مما هو عليه في مجموعة السيطرة. تم تحليل البيانات احصائيا لتكشف ان هنالك فرقا معنويا في الدالة الحامضية لسائل الكرش والبراز مقارنة مع مجموعة السيطرة (p≤0.05) في حين ان عينات الحليب والدم لم تبدي أي فرقا معنويا (p ≥0.05). أضف الى ذلك ، ان هذه الدراسة لم تجد اي فرقا معنويا في الدالة الحامضية بين طريقة جمع سائل الكرش من خلال الانبوب المعدي او من خلال عملية بزل الكرش (p ≥0.05). إذا طبقا لهذه النتائج المطمئنة والدراسات السابقة يمكن ان نعتبر قياس الدالة الحامضية وخصوصا في الكرش وسيلة تشخيصية مهمة للتعرف على حموضة الكرش تحت الحادة في الابقار الحلوب.

Abstract

Background: Ovine pulmonary adenocarcinoma (OPA), formerly known as sheep pulmonary adenomatosis is a contagious tumor of the respiratory epithelium.Aim: The aim of this study to investigate about ovine pulmonary adenocarcinoma (OPA) in Iraqi Awassi sheep.Material and methods: One hundred and twenty blood samples from Awassi sheep clinically had respiratory problematic were collected as well as sixty tissues samples. included twenty of each of (lung, lung fluid, mediastinal lymph nodes ) were collected from lungs with abnormal pathological lesions , all samples tested by RT-PCR by using specific primers and probe were designed in this study by using conserved region in envelope protein gene (NCBI-Gen Bank Code: KT279065.1) . Results: Ten sheep (5.55%) were found to be positive for JSRV by RT-PCR with higher infection rate in lung fluids 20% , mediastinal lymph nodes 10%, lung tissues 5% , and the less infection rate in blood . 2-5 years aged sheep were more susceptible for infection than other age ,but gender of examined sheep not affected on APO infection .Conclusion: The first detection of Ovine pulmonary adenocarcinoma (OPA) in Iraq in Awassi sheep in field and abattoir.

Abstract

This study aimed to highlight on hernial types that affect animals especially in cattle, the ways of diagnosis and treatment by using different techniques, which way is more suitable either the highlight on advantage and disadvantage of these techniques. There are many types of a hernia may happen in cattle's but the most frequently noted in cattle was a ventral and umbilical hernia. The researches confirmed that an umbilical hernia usually accrued in calves due to failure to close of the umbilical cord, while a ventral hernia is usually present in cattle due to trauma, heavyweight. A hernia was easy to diagnoses by case history, inspection and palpation. There are several techniques to treat a hernia, surgical or non-surgical depending on the size and the type of a hernia. In case of surgical techniques, there are a two-type open and closed surgical operation. The recent searches commend that in case of a simple and small hernia in size they treat by suturing the edges of a ring by absorbable suture (Hernioraphy), but in case of a large hernia must be used mesh to support the muscle in technique named (Hernioplasty). Mesh may cause irritation in its site, cause post-operative pain, and delay the time of healing so the recent searches to make a solution to this problems mesh must be covert by special material to make a barrier between it and muscle and lubricated a site hernia.

Phylogenic analysis of the sheep Orf virus in Iraq

Abstract

Orf disease is a highly contagious, zoonotic viral skin disease affects sheep, goats and some other domesticated and wild ruminants, therefor this study is designed to evaluate predominant genotypes circulating Orf virus of sheep in Al-Qadisiyah Province in Iraq as the first time by using the sequencing and phylogenic analysis of isolates Orf virus strains. The study was carried out on (94) skin samples of affected sheep animals diagnosed previously by RT-PCR technique in different ages and sexes between September /2013– March/ 2014. PCR assay was performed for amplification of 408bp GM_CSF/IL-2 inhibition factor (GIF) gene Orf virus in positive samples of real-time PCR assay. Eight purified DNA products isolated from different areas of Al-Qadisiyah Province (one sample for each area) were analyzed by using sequencing method to obtain the nucleotide sets of GIF gene. DNA sequencing technique was performed for Phylogenic relationship analysis study of local Orf virus positive samples isolates with NCBI-GenBank Global Orf virus isolates. The results of endpoint polymerase chain reaction for detection of specific GIF gene (408 bp) of Orf virus showed positive results for all collected examined samples. Sequencing analysis of our isolates in the present study recorded and published under accession numbers were (KJ508895.1, KJ634611.1, KJ634842.1, KJ648451.1, KJ653445.1, KJ653446.1, KJ659373.1 and KJ697772.1). The results of phylogeny tree construction showed that five of our published isolates of this study (KJ508895.1, KJ648451.1, KJ653445.1, KJ653446.1, and KJ697772.1) are located in the same first tree branch, which shared highest and closed relationship with AF192803.1, DQ184476.1 Orf virus strain NZ2 isolated from New Zealand. The KJ659373.1 our published Iraqi Orf virus isolates of this study showed high homology and closed relationship with AY605973.1 Orf virus isolated from reindeer in Norway. Phylogenic analysis of KJ634611.1 and KJ634842.1 our published Iraqi Orf isolates of the present study showed high homology with AY605977.1 Orf virus isolated from goat in Norway. Sequencing study and phylogenic analysis have been considered useful in understanding Orf virus scenario in Iraqi endemic area and which important in application of control measures and selective efficient vaccines to use in vaccination programs in Iraq.
