Abstract

Some agar alternatives were used for solidifying the half strength MSmedium using corn starch, potato starch, ice cream thickener (Thalabia) and amixture of potato and gelatin. Those agents were tested for themicropropagation of three European grape varieties Vitis vinifera L. namely,Ollivate blane (Fransi Aswad), Halawani and Kamali. Results showed that usingcorn starch or potato starch instead of agar inconsistent in solidifying theculture media. Meanwhile, mixing potato starch with gelatin or ice creamthickner individually proved promising solidifying agents. The addition of 1 mg/1BA to the culture media increased the number and length of shootssignificantly. No significant differences appeared between such gelling agentused. The three grape varieties produced roots characterized with its length andleaser branching when cultures on auxin free medium compared with thosegrown on auxin containing media. It is concluded that the three varieties ofgrape can be micropropagated on some cheap agar alternatives instead of thecostly agar.____________________________________________________________________

الخلاصةبنصف قوة الاملاح MS استخدمت بعض المواد كبدائل لمادة الآكار في تصليب الوسط الزرعيومنها نشاء الذرة الصفراء ونشاء البطاطا ومادة الثعلبية (مثخنة مصلبة للمثلجات) بتركيز 2.4 % وخليطVitis vinifera نشاء البطاطا مع الجيلاتين بتركيز ( 4 و 8) % في اكثار ثلاثة أصناف من العنب الاوربيوهي فرنسي اسود، حلواني وكمالي خارج الجسم الحي بهدف تقليل كلف الانتاج. L.أظهرت النتائج ان استعمال نشاء الذرة او البطاطا بدلاً عن مادة الآكار كان غير فعال في تصليبالأوساط الزرعية في حين وجد ان استخدام خليط نشاء البطاطا مع الجيلاتين وكذلك مادة الثعلبية قد اعطتالى الوسط الغذائي قد اظهر BA نتائج جيدة في تصليب الوسط الغذائي، ووجد ان إضافة 1 ملغم/لترمعنوية في زيادة عدد الافرع واطوالها ولم تظهر فروقات معنوية بين هذه المصلبات ومادة الآكار في نمو. BA وتطور الأجزاء النباتية المزروعة عند إضافةلم تؤثر المادة المصلبة للوسط الغذائي في نشوء الجذور كما لم تظهر اختلافات معنوية بينالأصناف الثلاثة في اعداد الجذور. وان أصناف العنب المدروسة قد انتجت جذوراً ولكنها تميزت بطولهاوقلة تفرعاتها مقارنة بالوسط الغذائي المجهز بالاوكسينات.نستنتج من ذلك بأمكانية اكثار بعض أصناف العنب على أوساط غذائية مصلبة ببدائل الآكارمنخفضة الكلفة بدلاً عن مادة الآكار غالية الثمن.

Abstract

Background: HROM agar Orientation is a chromogenic medium used for the detection and differentiation of Gram’s negative and Gram’s positive pathogenic microorganisms in urine samples.
Evaluation of CHROM agar Orientation for identification of urinary pathogens and susceptibility determinations in comparison to the ordinary media used.
Patients and Methods: A total of 375 midstream urine sample collected from patients with urinary tract infection (UTIs). CHROM agar Orientation, blood agar and macConkey agar media were used for direct inoculation.
Results: CHROM agar Orientation succeeded in detecting all the urine pathogens that were detected by the reference media, and antimicrobial Susceptibility tests were performed directly from primary isolates in all cases without the need for subcultures.Conclusion: HROM agar Orientation medium excellent detection of urinary pathogens and antimicrobial Susceptibility tests without the need for subcultures. Therefore, can replace the standard primary plating media used in routine diagnosis of urinary tract infection.

Synthesis of Fibrous Hydroxyapatite through Sol-Gel Route

Abstract

The sol-gel route using an agar gel with calcium nitrate and phosphate solution as starting materials for producing hydroxyapatite (HAP). The product formed were needle like, zigzag and straight fibres. The fibrous products on sintering transformed into stoichiometric HAP with a biological Ca/P ratio of 1.67. The influences of pH, temperature, nature of base and phosphate solution on the growth of fibrous HAP were studied. The pH of the solution was found to greatly influence the growth rate and morphology of the resultant product. The optimum gel temperature was found to be 60oC and sintering temperature of 900oC for 1 hour. The crystalline, thermal, functional and morphological characteristics of the fibrous HAP were investigated.

أن العمل المعروض في هذا البحث يتضمن طريقة جديدة لتصنيع مادة الهيدروكسي ابتايت (هاب) الشعيرية ذات الاستخدام البايولوجي. محلول نترات الكالسيوم ]المخلوط بنسبة محسوبة لمادة (agar) الهلامية[ وفوسفات الامونيوم تمثلان المواد الرئيسية في التحضير، والتي ينتج عنها مادة (هاب) وبالاشكال (الابري، المتعرج والمستقيم) مع الاخذ بنظر الاعتبار المحافظة على البنية الاساسية لمادة الهاب وبالنسبة المعتمدة بايولوجيا (1.67). لاجل الحصول على تلك المادة وبالمواصفات البايولوجية الدقيقة، يجب السيطرة على المتغيرات الحاكمة وبمنتهى الدقة (الدالة الحامضية، درجة حرارة التفاعل، طبيعة المادة القاعدية وتركيز محلول الفوسفات). الفحوصات الطورية والحرارية والصور المجهرية بينت بان المادة المحضرة مطابقة للمواصفات القياسية.

Medicine, University of Basrah,basrah,Iraq(Received 19 June 2012,Accepted 28 June 2012)Keywords; milk, Mannitol egg-yolk agar, chloramphenicol.ABSTRACTTwo hundred twenty seven samples of milk, soft cheese, curls cheese and yogurt were taken to isolate B. cereus. The percentage of these bacteria was 32.7 %, 16.6 %, 18 % and 26% in milk, soft cheese, curls cheese and yogurt, respectively. Mannitol egg-yolk agar (MYP) supplemented with polymyxin B sulfate is used for isolation. Identification of the isolates was done by biochemical tests. Screening of isolates by polymerase chain reaction ( PCR ) revealed the presence of diarrheal toxin cyt K gene in 87.09 % of isolates. Susceptibility of isolates to 7 antimicrobial agents revealed neomycin, chloramphenicol and gentamycin 100% for each, streptomycin 96% and erythromycin 93.5 % to be the most effective antimicrobial, while the highest resistance was noted against penicillin 100 %.

Abstract

The present study had thrown the light on the in vitro antimicrobial potential of the ethanolic extract of three local medicinal plants; Coriandrum sativum (Coriander), Vitis vinifera (Grape seeds), and Zingiber officinale (Ginger) against the growth of pathogenic Staphylococcus aureus isolated from milk of some local cows infected with clinical mastitis. The antibacterial activity was carried out by using agar well diffusion technique in Mueller-Hinton agar. Four concentrations could be prepared from each plant extract, these concentrations were 50, 100, 200, and 400 mg/ml.The results were obtained by measured the zone of inhibition around the well that could be exhibited by each plant concentration that followed incubation of bacterial plates and expressed as mean±Standard error (SE). Ethanolic extract of Coriandrum sativum was possessed the strongest antibacterial effect among the tested plants, the results were: 29.44±1.17, 29.22±0.32, 27.77±0.99, and 26.11±1.27 mm at a concentration of 50, 100, 200, and 400 mg/ml respectively, Followed by Vitis vinifera extract which showed moderate values recorded as 20.88±0.77, 20.11±0.58, 18.22±0.36, and 20.88±0.35 mm at a concentration of 50, 100, 200, and 400 mg/ml respectively. The least antibacterial activity was exhibited by the extract of Zingiber officinale that produced the following inhibition zones; 15.11±0.80, 15.77±1.12, 17.66±0.33, and 17.55±0.44 mm at a concentration of 50, 100, 200, and 400 mg/ml respectively. On the other hand, S.aureus was variably susceptible to five of the used standard antibiotics; Lomefloxacin, Erythromycin, Amoxicillin, Ciprofloxacin, and Rifampin. Means of their inhibitory zones were; 29.44±0.41,23.22±0.46, 21.77±0.36, 19.88±0.42, and 11.11±0.26 mm respectively. Whereas Cefprozil showed no effect against the growth of the tested organism.

Abstract

The aim of this study was to investigate the antibacterial activity of three different concentrations of a newly produced CHX powder prepared locally in Mosul College of Dentistry in the Department of Dental Basic Sciences( approved by Nineveh Drug Industry (NDI,licence no.2256 in26/9/2002), and to compare it with the same concentrations of commercially available CHX solution against three bacterial isolates commonly isolated from infected root canals, using the agar diffusion test. Astandard agar diffusion test was utilized to investigate the antibacterial activity of different concentrations of experimental chlorhexidine gluconate solution(NDI no.2256)and acommercially available chlorhexidine solution.The diameter of the microbial inhibition zones were measured in millimeter around each paper disk containing the experimental solutions. Data were analyzed using Duncan’s Multiple Range Test (a = 0.05). The results revealed that all tested microorganisms were affected by different concentrations of CHX powder(NDI no.2256) except for the lowest concentration (0.1%). Similar results were observed for CHX solution. The new prepared CHX powder(NDI n.2256)can be used aroot canal irrigant.

Abstract

Atotal of 100/270 (37%) fermented Mannitol salt agar (MSA) isolates were obtained: 40/180 (22.2%) were from buffaloes milk with subclinical mastitis and 60/90 (66.7%) were from hands of milk workers. All suspected Staphylococcus aureus isolates which tested microscopically and biochemically were 15/100 (15%) of suspected isolates, 5/40 (12.5) from milk and 10/60 (16.7) from hands swabs of milk workers were diagnosed as S. aureus.

Abstract

Background: Antibacterial action of root canal filling is an important factor for successful root canal treatment, so theaim of the study was to identify and to compare the antimicrobial effect of new sealer (GuttaFlow) to commonlyused endodontic sealers (AH Plus, Apexit and EndoFill) against four endodontic microbes.Materials and methods: Twenty patients aged (30-40) years with infected root canals were selected. Four types ofmicroorganisms were isolated from root canals (E faecalis, Staphylococcus aureus, E coli and Candida albicans) andcultured on Mueller Hinton agar Petri-dishes. After identification and isolation of bacterial species, agar diffusionmethod was used to assess the antibacterial action of four contemporary endodontic sealers used in root canalobturation (AH Plus, Apexit, EndoFill and GuttafFlow). Four wells measuring (5mm depth and 4mm diameter) werecreated in each Petri dish and sealer was applied into them incubated overnight at 37 C° for bacterial species and48 hr. at 37 C° for Candida albicans prior to determination of results. Zones of inhibition (no growth of bacteria) wereexamined around the wells containing sealer & diameters of the zones were measured in mm. The mean of inhibitionzones for each group was measured and statistically analyzed among groups using ANOVA and between groupsusing LSD tests.Results: There was a highly significant difference (P<0.001) among all the tested groups. EndoFill showed themaximum antibacterial action against tested microorganisms. GuttaFlow showed moderate to weak antimicrobialeffect, Apexit had weak effect, while AH Plus had no antibacterial action.Conclusion: All the tested materials except AH Plus had antibacterial efficacy against E faecalis, Staphylococcusaureus, E coli and Candida albicans. EndoFill had favorable results among tested sealers and E faecalis was the mostresistant bacteria, but none of the materials totally inhibited microbial growth. Thus, endodontic treatment must beperformed under aseptic conditions

Abstract

The study aimed to evaluate the distribution of CTX-M genes, which coding for ,Extended Spectrum β- lactamases (ESBLs) which hydrolyzed the extended spectrum β- lactams . Total of 800 urine samples taken from patients suffering from urinary tract infections were collected during the period from February to September from Hospitals in Al-Najaf province. All samples were cultured on MacConkey agar , were the results revealed that 250 isolates were lactose fermentative versus to 50 isolates were lactose non fermentative. The isolates succumbed to conventional tests (IMVC), Indol, Methyl red , Voges- Proskauer and Citrate utilization as well as motility test. Lactose fermentative and non motile isolates were candidate to be confirmed by Vitek2 system. Initial screening tests by Disk Diffusion test and by MacConkey supplemented with 2 mg / liter of Ceftazidime had been conducted, the resultsof disk diffusion test revealed different resistance phenotypes as well as multi drug resistance patterns Ceftriaxone were found to be the most resistant antibiotics in our isolated bacteria with resistance of 100%. The next most resistant antibiotics were Ceftazidime (98.11%), followed by aztreonam (90.57%); and cefotaxime (90.27%). While the result of screening with Ceftazidime reported that 31/53(58.49%) of isolates were initially ESBLs producers. The results of genotyping tests showed that out of 53 isolates which had been screened for the prevalence of bla CTX-M genes, 47(88.6%) were positive. Nucleotide sequence of bla CTX-M of amplicons (19, 20, 22, 23, 24) showed high identity (99%) with CTX-M-15 of NCBI-BLAST. As concerning with protein sequence, the results showed high identity (98%) with reference protein of gene bank. The results of other protein sequences revealed great degree of identity reach to (100%) of identity.

الدراسة كانت تهدف لتقييم انتشار جينات انزيمات السيفوتاكسيم المقاومة للمضادات الحياتية بيتا لاكتام واسعة الطيف ودراسة تتابع النيوكليوتيدات والبروتين لبعض منها.جمعت 800 عينة ادرار للفترة من شباط الى ايلول من العام 2012 لمرضى مصابين بالتهابات المجاري البولية من عدة مستشفيات في محافظة النجف الاشرف. جميع العينات تمزراعتها على وسط الماكونكي الصلب , 300 منه نمت على وسط الماكونكي و اذ كانت 250 منها مخمرة للاكتوز بينما كانت 50 منها غير مخمرة للاكتوز. العزلات خضعت لفحوصات الاندول و احمر المثيل وفحص فوكس بروسكاور واسهلاك السترات بالاضافه الى فحص الحركة .رشحت العزلات الخمرة للاكتوز وغير المتحركة لفحص الفايتك 2. اجريت فحوصات انتشار الاقراص واستخدام الماكونكي الحاوي على 2 ملغم / لتر من السيفتازيديم . نتائج فحص انتشار الاقراص بين انماط مقاومة مختلفة بالاضافه الى انماط تعدد المقاومة , كانت 100% من العزلات مقاومة للمضاد الحياتي سيفترياكزون , بينما كانت المقاومة 98.11% , 90,57% و 90.27% للسيفتازيديم والازترونام والسيفوتاكسيم على التوالي. بينت نتائج الماكونكي الحاوي على السفتازيدبم 2 ملغم / لتر ان 31 / 53 (58.49%) من العزلات ابتداءاً منتجة لانزيمات المقاومة للبيتا لاكتام الواسعة الطيف.بينت نتائج الفحوصات الوراثية لدراسة انتشار جينات انزيمات المقاومة لمضادات البيتا لاكتام واسعة الطيف ان 47(88.6%) من العزلات كانت موجبة . دراسة تتابع نيوكليونتيدات لخمسه من العزلات ( 19 , 20, 22, 23, 24 ) بينت درجة عالية من التشابه وصلت الى (99%) مع CTX-M-15 للNCBI - BLAST . فيما يتعلق بدراسة تتابع البروتينات فان الدراسة بينت ان التشابه كان ( 98%) مع البروتينات القياسية لبنك الجينات كما ان بعض البروتينات اظهرت (100%) من التشابه مع البروتينات القياسية.

Abstract

Specimens have been collected from one hundred and seventeen patients residing in local hospitals, 33 with burns and 84 wound injuries,. Three different methods ,Cefoxitin disk diffusion, EPI-M Screening Kit and Crome agar (MeReSa agar)with selective supplement were used to detect methicillin-resistant Staphylococcus aureus (MRSA) . A comparison was made between these 3 methods according to the results. It was found that the results of the Cefoxitin disk diffusion test were compatible with the results of culturing on Crome agar, while those obtained from the EPI-M Screening kit were not accurate and some of them gave false negative results.

تم جمع العينات من 117 شخص راقدين في المستشفيات المحلية, 33 مصاب بحروق و 84 شخص مصاب بجروح. استخدمت ثلاثة طرقCefoxitin disk diffusion, EPI-M Screening KitوCrome agar (MeReSa agar) مع الملحق الخاص بالوسط, للتحري عن بكتريا المكورات العنقودية المقاومة للمثسلين (MRSA) وقورنت هذه الطرق بالاعتماد على نتائج استخدام كل طريقة. لقد وجد ان نتائج طريقة Cefoxitin disk diffusion و الزرع على وسط Crome agarكانتا متطابقتين, بينما النتائج الحاصلة من استخدام EPI-M Screening kitلم تكن دقيقة و اعطت لقسم منها نتائج سلبية خاطئة.
