Evaluation of Antibacterial Efficacy Against Enterococcus Faecalis of Newly Prepared Endodontic Irrigant Solution (an In vitro study)

Abstract

To evaluate the efficacy of a newly prepared endodontic irrigant solution, against E. faecalis.Materials and Methods: Sixty human extracted single rooted teeth samples were prepared by usingprotaper NiTi rotary system. The apical foramen of each sample was sealed by acrylic resin on theapical 3mm. Each root was embedded in silicon impression material block and autoclaved. A ten μl(4X10P5P) of inoculated broth media (nutrient broth) with E. faecalis was injected gently inside thecanals of group A,B&C was incubated for 24hours at 37CPoP. Group D was left without inoculation toserve as a control negative. The samples of group A,B and C were disinfected as fallow: Group A wasdisinfected with MCP solution(mixture of 20% castor detergent and 4% papain). Group B wasdisinfected with freshly diluted 2.5% NaOCl solution. Group C not treated with any solution controlpositive. The disinfected solutions were left inside the canal for 5 min then each canal was dried withsterilized paper point, then bacterial samples were taken from the samples of 4 groups using size 40sterilized file which was inserted to full canal length and rotated 360 degrees in clock wise direction fordentin engagement. The file was put inside the sterilized plastic tube containing 1ml of normal salinethen 200 μl was inoculated and spread on Enterococcus agar media and incubated at 37CPoP for 18hours,the numbers of bacterial colonies were counted. Results: Both solutions (MCP and NaOCl) producecomplete eradication of E Faecalis within 5 minutes. Conclusion: Castor detergent 20% and papainenzyme 4% (MCP) has ability to completely eradicate Enterococcus Faecalis bacteria from theinfected root canal in vitro in 5 min. It's antibacterial action is similar to the action of 2.5 % NaOCl

Abstract

Bacterial water pollution is a genuine general wellbeing concern since it causes various maladies. Antimicrobial nanofibers can be integrated by incorporating nanobiocides, for example, silver nanoparticles into nanofibers. Nylon 6 was dissolved in formic acid at a concentration of (25 wt. %) and tough antibacterial (AgNO3/Nylon) nanofibers were produced utilizing electrospinning system. Polymer solution was tested before accomplishing electrospinning process to acquire its surface tension, electric conductivity and viscosity, where every one of those parameters increased relatively with increasing concentration of (AgNO3) additions. SEM and EDX spectra were utilized to focus on the morphology, surface elemental membrane, fibers and porosize diameters. The resulted nanofiber membrane has an average fiber diameter of 139 nm for pure nylon 6 and 247 nm for (1.2 wt. % AgNO3/Nylon). The resultant polymer membrane was then tested for their ability to destroy microorganisms in water; antimicrobial tests showed that the prepared nanofibers have a high bactericidal effect against Escherichia Coli Bacteria with inhibition zone (10 mm) and antibacterial activity (99%). Likewise, these results highlight the potential utilization of these nanofibrous mats as antimicrobial agents.

التلوث الجرثومي للمياه مشكلة خطيرة تهدد صحة العامة لأنه يسبب العديد من الأمراض. ألياف النانو المضادة للميكروبات يمكن تصنيعها عن طريق دمج مبيدات نانوية مثل الفضة النانوية في ألياف النانو. النايلون 6 تم اذابته في حمض الفورميك بتركيز (25٪) ، الاليلاف النانوية مضادة للجراثيم نايلون / نترات القضة اعدت باستخدام تقنية البرم الكهربائي. تم اختبار محلول البوليمر قبل عملية البرم الكهربائي للحصول على خواص محلول ( توتر سطحه، التوصيل الكهربائي واللزوجة) وجميع تلك العوامل تزيد بشكل متناسب مع زيادة كمية الإضافة نترات الفضة. واستخدمت فحص المجهر الالكتروني الماسح و فحص مطياف الاشعة المتتشتتة لدراسة مورفولوجية، سطح التركيب العنصري،قطر الألياف و حجم المسامية للغشاء. حيث كان معدل قطرالالياف النانوية للغشاء النتائج )139 نانومتر( للنايلون النقي و )247 نانومتر( ، في (1.2 %) نترات الفضة . ثم تم اختبار الاغشية البوليمرية الناتجة فحص لقدرتها على تدمير البكتيريا في الماء؛ وأشارت الاختبارات المضادة للميكروبات أن ألياف النانو لها تأثير عال مضاد للجراثيم ضد البكتيريا القولونية و كانت منطقة التثبيط (10 ملم) الفعالية المضاد للبكتيريا (99٪). وعلى وفق ذلك، تبين هذه النتائج إمكانية استخدام اغشية الالياف النانوية بوصفها عوامل مضادة للميكروبات
