Frequency of HLA Antigens in a Sample of Iraqi Brucellosis Patients

Abstract

Fifty one patients with serologically confirmed brucellosis and 70 healthy controls were phenotyped for HLA-A, -B, -DR and -DQ antigens by using standard microlympho-cytotoxicity method, and lymphocytes defined by their CD markers (CD3, CD4, CD8 and CD19). The results revealed a significant (Pc = 0.001) increased frequency of HLA-DR8 (41.18 vs. 10.0%) in the patients . A significant increased percentage of CD8+ lymphocytes was also increased in the patients (25.15 vs. 22.0%; P = 0.006), while CD3+ lymphocytes were significantly decreased (75.1 vs. 79.4%; P = 0.02).

نمط مظهريا احدى وخمسون مريضا من مرضى داء البروسيلا المشخصين مصليا وسبعين من الأصحاء لمستضدات خلايا الدم البيض البشرية A و B و DR و DQ باستخدام الطريقة القياسيةMicrolymphocytotoxicity test ، وفضلا عن توصيف الخلايا اللمفية في ضوء بعض الواسمات (CD3 و CD4 و CD8 و CD19). أوضحت النتائج زيادة معنوية(الاحتمالية المصححة = 0.001) بتكرار المستضد HLA-DR8 (41.18 مقابل 10.0%) في المرضى. كما ارتفعت في المرضى ايضا وبفرق معنوي النسبة المئوية للخلايا اللمفية الحاملة للواسم CD8 (25.15 مقابل 22.0%؛ الاحتمالية = 0.006)، في حين انخفضت معنويا الخلايا الحاملة للواسم CD3 (75.1 مقابل 79.4%؛ الاحتمالية = 0.02).

Abnormal Lymphocyte Subsets in Children with Type 1 Diabetes Mellitus

Abstract

This study was designed to investigate the cellular changes of peripheral blood lymphocytes (PBL) in children with type 1 diabetes mellitus (T1DM) by determining the surface antigens (CD markers). Sixty T1DM patients who were newly diagnosed as type1diabetetics (duration of their illness is less than five months) were selected from the National Diabetes Center at Al-Mustansiriya University during the period May 2004 to October 2005. All the patients were treated with daily replacement doses of insulin. Their age ranged from (3-17) years. Fifty apparently healthy control subjects, matched with age (4-17) years and sex underwent the PBL phenotyping. Phenotyping of lymphocyte surface antigens was done by direct Immunoflurocent (IF) technique using mouse antihuman CD3, CD4, CD8, CD19 and CD56.T1DM patients showed a remarkable lowering in CD3+, CD8+, and CD56+ cells but the decrease in CD4+ cells percentage was not significant in patients in comparison to healthy controls. In contrast, a significant elevation of CD19+ cell percentage and CD4+: CD8+ ratio were observed in the patients.

Abstract

SUMMARY:BACKGROUND:In Type 1 Diabetes Mellitus (T1DM), numerous changes in the cellular as well humoral immune response have been identified. However, it is not known whether both the CD4+ and CD8+ subpopulation or only one of these or CD19+contains increased numbers of activated cells.OBJECTIVE:The aim was to study the activated lymphocyte subpopulation by use of monoclonal antibodies to T-cell and B-cell antigens which is known to be expressed on activated cells.METHODS:A total of 60 T1DM patients who had newly onset of the disease (diagnosed was from one week up to five months) were included in the present study, all the patients were treated with daily replacement doses of insulin. Fifty apparently healthy control subjects underwent the PBL phenotyping. Phenotyping of surface antigens was done by direct Immunoflurocent (IFT) technique using mouse antihuman CD3, CD4, CD8, CD45RA, CD19, and activated markers CD45RO, DR-antigen and CD38.RESULTS:T1DM patients showed a remarkable lowering in CD3+, CD8+, and CD45RA+ cells (p<0.0001), but the decrease in CD4+ cells percentage was not significant. In contrast, a significant elevation of activation markers includes (CD45RO+, HLA-DR+ and CD38+ cells) were observed in patients in addition to a significant increase of CD19+ cell percentage and CD4+: CD8+ ratio in the patients.CONCLUSION:This study provides evidence that abnormalities of T-cells regulation are detectable in patients with T1DM.

Abstract

Background: Visceral leishmaniasis is an endemic protozoan disease in Iraq Recovery from this disease confers a solid and permanent immunity.Objective: Determining the type of lymphocyte phenotype by using; monoclonal antibodies against (CD 4, CD8).Patients and methods: Indirect immunofluorescence technique analysis was performed to detect the percentage of CD4, CD8 and CD4/CD8 ratio positive lymphocytes.Results: Our results showed that the patients groups have decrease in both the percentage of CD4 cells &CD4/CD8 ratio and increase in the percentage of CD8. Follow up of patients after treatment showed a significant increase in the percentage of CD4, CD4/CD8 ratio but they were still lower than their normal range.Conclusion: CD4/CD8 ratio is reversed in VL infection then increase after treatment but it was less than its normal range.Key words: CD-Markers profile, leishmania donovani, amastigote form.

اللشمانيا الحشوية هي مرض وبائي مستوطن في العراق وعند الشفاء منه يحصل المريض على المناعة الدائمة. أظهرت الدراسة المناعية لهؤلاء المرضى بان هنالك نقصان معنوي في نسب الأنماط المظهرية (CD4) وكذلك (CD4/CD8) بينما ظهرت زيادة معنوية في نسبة CD8 مقارنة مع مجموعة السيطرة.المواد وطريقة العمل:استعملت طريقة الومضان غير المباشر (IFAT) وقد لوحظ إن النسبة المئوية لعدد T cell CD8 في مجموعة المرضى المصابين باللشمانيا الحشوية قد اتخذت التفاعل العكسي وأبدت ارتفاعاً معنوياً حاداً عند المقارنة مع المجموعة الضابطة. كما أظهرت فرقاً معنوياً حاداً في هبوط معدل النسب CD4 Tcell و CD4/CD8 .النتائج: أظهرت النتائج إن هناك انخفاضا ملحوظا في النسبة المئوية للمعلومات الخاصة بالخلايا اللمفية المساعدة والنسبة بين الخلايا التائية المساعدة والخلايا السمية لمرضى اللشمانيا الحشوية . كما أظهرت ارتفاعاً بالخلايا اللمفية السمية (P < 0.01) . عند متابعة معلومات هذه الخلايا بعد العلاج نلاحظ إن هناك ارتفاعاً في النسبة بين الخلايا التائية المساعدة والخلايا اللمفاوية السمية مع بقاء هذه النسبة اقل من معدلاتها الطبيعة (P < 0.01). الاستنتاجات: إن نسبة الخلايا اللمفية المساعدة إلى الخلايا اللمفية السمية معكوسة في مرض اللشمانيا الحشوية ولكن هذه النسبة ترتفع بعد العلاج ولكن تبقى اقل من المعدل الطبيعي للنسبة

Abstract

Immunological assessment of our patients was carried out and the results showed a significant reduction in the percentage of CD3, CD56 and a significant increase in the percentage of CD19 in the peripheral blood lymphocyte of VL in comparison with control group.Patients and methods: Indirect immunofluorescence technique analysis was performed to detect the percentage of CD3, CD19and CD56 positive lymphocytes.Results: Our results in the patients groups showed decrease in the percentage of CD3, CD56 and increase in the percentage of CD19. Follow up of patients after treatment showed a significant increase in the percentage of CD3, CD56, but they were still lower than their normal range . Conclusion:- CD3,CD56, were decreased in VL infection then increased after treatment but it was less than its normal range, while CD19 was increased in VL when compared with control group.Key words: CD-Markers profile, leishmania donovani, amastigote form

Abstract

ABSTRACT:BACKGROUND:Among most basic and clinical investigations for detection and genotyping HPV, only In Situ Hybridization (ISH) is effective in studing the relation of HPV genome to that histopathological entity. Tumor Infiltrating Lymphocytes (TILs) represent the local cellular immune response against both cervical cancer and the associated HPV.OBJECTIVE:This study is aimed to investigate ISH in detection and genotyping of HPV and immunohistochemical (IHC) study in phenotyping of TILs in Iraqi women with different grades of cervical neoplasia.MATERIALS AND METHODS:This retrospective research included a total number of 129 cervical tissue blocks , 64 with invasive and 27 with pre-invasive cervical neoplasia,12 with condylomata acuminata,11 with chronic cervicitis and 15 with apparently- healthy cervices .Molecular detection and genotyping of HPV DNA were performed by using ISH whereas immunophenotyping was done by IHC using monoclonal antibodies for that specific CD markers representing some of TILs.RESULTS:The overall prevalence of HPV DNA in total group of invasive cervical neoplasia was 28.4% , 12 out of 48(25%) with squamous cell carcinoma(SCC) and 8 out of 16(50%) with adenocarcinoma (AC). HPV 16 constituted (58.3%) of HPV- positive invasive cervical SCC , where as HPV 31/33 and lastly HPV 18 constituted (25%) and (16.7%) , respectively .In AC group, HPV 16, 18, 31/33 occupied 50% , 25% , 12.5% of this group, respectively .The prevalence of HPV DNA in pre-invasive cervical neoplasia group was 22.2%. While no case with HPV 18 was detected , HPV 16 and HPV 31/33 genotypes occupied 40% and 60% , respectively .Although CD3+,CD4+and CD8+ lymphocytes showed high mean values , significant differences present only between HPV DNA- positive tissue blocks and control group (p<0.05) . The CD20+and CD56+ lymphocytes showed low mean values.Compared to control group,however,statistical analysis showed significant differences (p<0.05) regarding CD20+ and non-significant regarding CD56+ lymphocytes .CONCLUSION:Using ISH proved more powerful and sensitive tool in revealing precised effects of HPV in cervical neoplasia compared to histopathological examination .High mean values of CD4+ and CD8+ versus low mean values of CD20+ and CD56+ lymphocytes could reflect an important and specific role of these cellular responses against HPV viruses during initiation and progression of HPV-associated cervical cancers.
