Abstract

This study is to evaluate plasma levels of several cytokines in preeclamptic pregnancies compared to those of healthy pregnancies.Ninety pregnant women with preeclampsia (37 mild & 53 severe) and thirty healthy pregnant women were enrolled in the study. Blood samples were taken and plasma levels of IL-8, IL-10, and IFN-γ were measured by enzyme-linked immunosorbent assay (ELISA).Preeclamptic women and their severe cases but not mild cases had significantly (P<0.05) increased levels of plasma IL-8, and IFN-γ as compared with healthy pregnancies. By contrast, plasma levels of IL-10 was significantly (P<0.05) increased in healthy pregnant women as compared to all groups of preeclampsia. Preeclampsia is associated with an imbalance between pro-inflammatory cytokines (IL-8, IFN-γ) and anti-inflammatory cytokines (IL-10), and these support our suggestion of altered immune response in preeclampsia.

تضمنت هذه الدراسة تقييم مستويات كل من الحركيات الخلوية γ IL-8, IL-10, IFN- في بلازما النساء العراقيات المصابات بمقدمة الارتعاج, شملت هذه الدراسة 120 امراة حامل قسمت الى اربعة مجاميع:المجموعة الاولى : مجموعة النساء ذوات مقدمة الارتعاج المعتدل (37) المجموعة الثانية : مجموعة النساء ذوات مقدمة الارتعاج الشديد (53) المجموعة الثالثة : مجموعة النساء ذوات مقدمة الارتعاج الكلي (90)المجموعة الرابعة : مجموعة النساءالسليمات كسيطرة (30) اظهرت نتائج الدراسة الحالية ارتفاع معنوي (P<0.05) في مستوى الحركيات الخلوية IL-8,IFN-γ في البلازما في المجموعة الثانية والثالثة مقارنة بالمجموعة الاولى و الرابعة وكذلك ارتفعت معنويا (P<0.05) مستوى IL-10 في بلازما النساء الحوامل السليمات (مجموعة السيطرة) مقارنة بمجاميع المرضى الثلاث .نستنتج من خلال الدراسة الحالية اختلاف في مستوى الحركيات الخلوية في بلازما النساء المصابات بمقدمة الارتعاج الشديد مما يؤكد الاختلاف في الاستجابة المناعية لهؤلاء النساء.

Abstract

Summary: Background: The role of cytokines in cancer immunity and carcinogenesis in general has been well established, which play an important role in the pathogenesis of many solid cancers.This study aimed to estimate serum levels of IL-2 and IL-4, and to shed light on the correlation of these interleukins with progression of breast cancer. Patients and Methods: The study included 80 women, it comprised of 45 breast cancer patients, 12 patients with benign breast lesions and 23 apparently healthy controls. ELISA method has been used for estimation the level of IL-2 and IL-4 in serum of three studied groups.Results: This study showed elevation of IL-4 level in the sera of breast cancer patients with significant differences between breast cancer and controls (p<0.001), this elevation was correlated with progression of the tumor. In addition, the elevation of serum level of IL-4 was found to be inversely related to ER and PR expression (P=<0.05). On the other hand, there was a significant decrease in the median of serum level of IL-2 in patients as compared with control groups. Conclusions: There was association between elevated serum level of IL-4 and breast cancer and this elevation was correlated with advanced stage of disease. In addition, there was no association between the statistical significant decrease of IL-2 serum level and the advanced stage of breast cancer. Keywords: Breast cancer, Cytokines, ER

Abstract

(1). autoimmune inflammation in RA, cytokineimbalance plays a great part in RA pathogenesis Rheumatoid arthritis is a chronic systemic inflammatory disease affects many tissues and organs, butprincipally attacks flexible (synovial) joints. Methotrexate is the most commonly used disease-modifyingantirheumatic drug for the treatment of rheumatoid arthritis. OBJECTIVE:The aim of this study was to evaluate the effect of methotrexate on serum levels of IL-1α and IL-8 inrheumatoid arthritis. SUBJECTS AND METHODS: Blood samples were collected from 50 patients with rheumatoid arthritis (25 patients without treatment and 25 patients are received methotraxate) and from 30 healthy age and sex matched individuals served ascontrols. Serum IL-1α and IL-8 were measured by means of enzyme-linked immune-sorbent assay.RESULTS: The present results showed that serum levels of IL-1α and IL-8 were significantly higher in RA patientsthan in healthy controls (P<0.01), furthermore, level of IL-1α was significantly decrease in patientstreated with methotraxate as compared to those patients who have received no treatment (P<0. 01). On theother hand serum level of IL-8 did not showed any significant differences between patients treated withmethotraxate and those patients without treatment (P>0. 05).CONCLUSION: These finding demonstrate that methotrexate turns out to be a good inhibitor for IL-α production. Abstract

Background: Psoriasis is an inflammatory skin disorder characterized by activation of lymphocytes, systemic and local over expression of pro-inflammatory cytokines. Objectives:- The study was designed to investigate the level of cytokines in patients with psoriasis and study the correlation of the cytokines with disease severity.Patients and Methods: Blood samples were collected from seventy patients with psoriasis for measurement of (IFNγ, IL-17 and IL-4) by ELISA technique.Results: High levels of IFNγ and IL-17 were observed in patients with psoriasis. It was found that the mean levels of IFNγ and IL-17 were significantly correlated to disease severity. Moreover, the mean level of IL-4 in psoriatic patient was found to decrease significantly according to graded severity of the disease. Statistical analysis was done by using SPSS (statistical package for social sciences) in which T-test and ANOVA test were used and chi- square (X2) test for categorical data.Conclusion: The study concluded that the level of IFNγ and IL-17 may play a key role as an indicator in the immunopathogenesis of psoriasis and markers of disease activity.Recommendation:-Using another cytokines and study effect of these cytokines with disease severity.

الخلفية :- الصدفية هو مرض جلدي التهابي يتميز بتفعيل الخلايا اللمفية وزيادة انتاج الحركيات الخلوية على المستوى الموقعي والجهازي للجسم.الاهداف:- صممت هذه الدراسة لفحص مستوى ا الحركيات الخلوية لدى المرضى المصابين بداء الصدفية ودراسة الارتباط بين هذه الحركيات الخلوية ومدى شدة المرض. المرضى وطرق العمل:- اخذت عينات دم من سبعين مريض لقياس مستوى الحركيات الخلوية ( IFNγ, IL-17و IL-4) بتقنية الايلايزا. النتائج:- ينت الدراسة ارتفاع مستوى ( IFNγوIL-17) لدى مرضى الصدفية, وتبين ان هناك فرق معنوي في معدل مستويات (IFNγ, IL-17 ) عند المصابين مقارنة مع مستوياتهما عند مجاميع السيطرة وهناك علاقة ملحوظة بين ارتفاع مستوى هذه الحركيات الخلوية وشدة المرض. كذلك وجد ان مستوى (IL-4) لدى المرضى المصابين بالصدفية يقل معنويا بشكل ملحوظ بالنسبة لشدة المرض. تم استخدام نظام SPSS لتطبيق اختبار T و اختبار الانوفا واختبار الفرق المعنويالاستنتاج:- يمكن الاستنتاج في هذه الدراسة ان مستوى الحركيات الخلوية (IFNγ and IL-17) ربما لها دور كدليل في الامراضية المناعية لداء الصدفية وتعتبر كمعلمات لدرجة فعالية المرض. التوصيات:- استخدام حركيات خلوية اخرى و دلراسة تاثيرها على شدة داء الصدفية

Abstract

Thirty serum samples were collected from patients with chronic hepatitis B who were admitted to Hepatology and Gastroenterology Teaching Hospital in Baghdad during period from beginning of February / 2008 to the end of February / 2009, also serum samples were collected from 30 healthy HBs Ag carrier was discovered accidentally through attending blood bank for donation of blood and a total of 30 apparently healthy blood donors were included in the study served as a control group. The concentrations of some cytokines Pg/ml (IFN-γ, GM-CSF, IL-1α, IL-6, IL-8 and IL- 2 receptor) have been estimated in the sera of the studied groups and the results showed that mean levels of gamma-interferon, interleukin-1 alpha, interleukin-8 and interleukins-6 in sera of carriers group were (109.6, 251.6, 232.9 and 166.8 respectively) more than the level of these cytokines in sera of chronic HBV patients (75.1, 94.7, 159.6 and 110.9 respectively) and healthy control (48.4, 10.1, 11.3 and 15.4 respectively), while the mean level of GM-CSF was elevated in sera of CHB patients (110.8) than both of healthy control and carrier group (17.3 and 41.2 respectively).Finally the interleukin-2 receptor was represented with similar level in sera of chronic patients and carrier group ( 165.6 and 167.9 respectively) with non significant difference, whereas all other statistical comparisons among the three main groups showed highly significant difference.

جمع 30 نموذج من الدم من مرضى إلتهاب الكبد الفايروسي المزمن نوع (ب) من المراجعين للمستشفى التعليمي لأمراض الجهاز الهضمي والكبد في بغداد فضلاً عن 30 نموذج من الأصحاء الحاملين للمستضد السطحي لفايروس التهاب الكبد نمط ( ب) من المراجعين لمصرف الدم المركزي في بغداد و30 نموذج لأشخاص أصحاء ظاهرياً لغرض استعمالها كمجموعة سيطرة ، للمدة من بداية شباط 2008 ولغاية نهاية شباط 2009.تم قياس تركيز بعض السايتوكينات في مصل المجاميع المدروسة إذ أظهرت مستوى السايتوكينات الآتية gamma-interferon, interleukin-1 alpha, interleukin-8, interleukins-6 إرتفاعاً في المرضى الحاملين (109.6, 251.6, 232.9,166.8) بالمقارنة مع المزمنين((75.1, 94.7, 159.6, 110.9 ومجموعة السيطرة(48.4, 10.1, 11.3, 15.4), بينما كان مستوى السايتوكين GM-CSF مرتفعاً في المرضى المزمنين(110.8) بالمقارنة مع الحاملين والأصحاء(17.3,41.2) ، من ناحية أخرى كان مستوى السايتوكين interleukin-2 receptor متشابهاً في المرضى المزمنين والحاملين( 165.6, 167.9) مع عدم وجود فرق معنوي, بينما أظهرت جميع المقارنات اﻹحصائية الأخرى وجود فروق معنوية بين المجاميع الثلاثة.

Abstract

Acute cholecystits is an acute inflammation of the gallbladder. A Total of 52 Cholecystectomy patients from a Al- Sadar Teaching hospital and 52 healthy volunteers were enrolled . Blood specimen collected by sterile container from both . The serum levels of pro-inflammatory cytokines (IL-1, TNF-α & IL-6) were significantly increased in gallstone patients sera comparing to healthy controls at (p<0.05) .While anti-inflammatory cytokine IL-4 concentrations in gallstone patients sera showed significant reduced comparing to healthy controls at (p<0.05). And regulator cytokine IL-17 concentrations in gallstone patients sera showed not significant reduced comparing to healthy controls at (p<0.05), . Finally IgG concentrations in gallstone patients sera showed significant elevation comparing to healthy controls at (p<0.05).The elevation of pro-inflammatory cytokines including (IL-1 , IL-6 & TNF-α) indicated the presence of inflammatory response associated with gallstone. The reduction of anti-inflammatory cytokine (IL-4) and regulator cytokine (IL-17) indicated the presence of immune predisposition in patients for gallstone.

يعد التهاب المرارة من الالتهابات الحادة ..جمعت عينات دم من 52 مريض يخضع للعملية (استأصال المرارة) في مستشفى الصدر التعليمي و52 اصحاء متطوعين لغرض السيطرة. جمعت عينات الدم في قناني معقمه. كان مستوى المدورات اللمفاوية البادئة للالتهاب والمتمثله (IL-1, TNF-α & IL-6)مرتفعة وبفارق معنوي عن السيطرة عند مستوى الاهمية (p<0.05) بينما المضادة للالتهاب والمتمثله بال IL-4منخفضة وبفارق معنوي عن امصال السيطرة أما ال IL-7 و الذي يعد من المدورات اللمفاوية المنظمة منخفظا ولكن ليس بفارق معنوي ( P< 0.05). كما ارتفع مستوى IgG في امصال المرضى مقارنة بالسيطره.

Abstract

This study was carried out to evaluate the anti-inflammatory effect of losartan in 18 female rats which were divided into 3 groups. Respiratory disease were experimentally induced by the intraperitoneal injection and spray inhalation of ovalbumin (OVA) in first and second group while third group left as negative control group. First group were treated with losartan orally at dose rate of 5 mg/kg body weight, on the other hand second group considered as positive control group. ELISA test were used to estimate the concentration of TNF-α, IL-4 in BALF and total IgE in serum samples. Total WBC, neutrophil, eosinophil and lymphocyte were counted in bronchoaleveolar lavage (BAL). First and second rat groups show signs of pulmonary disease. Losartan treated group showed significant (p<0.05) decrease in concentration of TNF-α (152.483 pg/ml), IL-4 (39.733 pg/ml) in BALF and total serum IgE (56.006 pg/ml) in comparison with positive control group. A significant decrease (p<0.05) were also detected for total WBC (101.33 x103), neutrophil (8.83 x103), eosinophil (15.50 x103 ) and lymphocytes (15 x103 ) in BALF of losartan treated rat group in compare to positive control group.

Abstract

Background:Osteoarthritis (OA) is a degenerative joint disease caused by gradual loss of cartilage. Telmisartan is an angiotensin II receptor antagonist, and act as a partial agonist on the nuclear peroxisome proliferator-activated receptor-gamma (PPAR-γ), that has been reported to exert anti-inflammatory effects.Objective:To evaluate the potential anti-inflammatory effect of telmisartan in patients with knee OA.Methods:Forty-two patients with painful knee OA were allocated into 2 groups, group (1): patients treated with naproxen tablets (500 mg/12 hr), telmisartan tablets (40 mg/day) and omeprazole (20 mg/day) for 3 months, while group (2): patients treated with naproxen tablets (500 mg/12 hr) and omeprazole (20 mg/day) for 3 months. The serum levels of IL-1β, high-sensitivity C-reactive protein (hs-CRP), TNF-α and erythrocyte sedimentation rate (ESR) were measured before and after 3 months of treatment.Results:Telmisartan when used in combination with naproxen resulted in significant decrease in serum levels of IL-1β and hs-CRP, higher than that produced by naproxen when used alone. The mean TNF-α level and ESR was decreased non-significantly in both study groups.Conclusion:Administration of telmisartan as an adjuvant therapy to naproxen in knee OA patients produced a significant decrease in the serum levels of IL-1β and hs-CRP, though no clear effect on TNF-α and ESR was noticed after 3 months treatment. Accordingly, many promising preventive strategies emerged from the available results since telmisartan relatively reduce the inflammatory burden in OA patients. Abstract

Toxoplasma gondii is an opportunistic pathogen in which the reactivation of a latent infection can cause death in congenitally infected fetuses, newborns, and immunocompromised patients. This study aimed to determine the seropositivity of toxoplasmosis infection and the possible association with Interleukin-12 (IL-12) and Interleukin-23 (IL-23) cytokines in breast cancer patients. In this study, 190 women were enrolled. All serum samples were tested for T. gondii immunoglobulins (IgG and IgM) antibodies and (IL-12, IL-23) levels using ELISA technique. The result of this study showed that breast cancer patients recorded the highest percentage of toxoplasmosis infection. There were no positivity rates for anti- Toxoplasma IgM in breast cancer patients while the positivity percentage for anti- Toxoplasma IgM among the control group was (7.00%). Furthermore, the seroprevalence of anti- Toxoplasma IgG was the highest in the age group (31- 40) years in patients with breast cancer while the highest mean titer of the IL-12 is restricted to ages (21-30) years in the control groups who are seropositive to anti- Toxoplasma IgG. Although, in patients with breast cancer who are seropositive to anti- Toxoplasma IgG, the highest mean titer of the IL-23 was in ages (21-30) years. Since most immunosuppressive patients are exposed to various possible risk factors including Toxoplasma primary infection or reactivation, so it is important to diagnose and treat toxoplasmosis in breast cancer patients to reduce the consequences of the infection.

Abstract

ABSTRACT: BACKGROUND: Type 1 diabetes is characterized by a complete or near-complete insulin deficiency caused by animmune-mediated selective destruction of the insulin-producing β-cells in the Islets of Langerhans. Inflammatory mechanisms play a key role in the pathogenesis of type 1 diabetes.Many findings suggest that the Islet autoantibody status in type 1 diabetes is linked to diseaseactivity.OBJECTIVE: To investigate the hypothesis that the systemic immunoregulatory balance, as defined by levels ofcirculating cytokines, is associated with Islet autoantibody status. METHODS: Cytokines (IL-2, IL-4, IL-5,IL-10, TNF-β and INF-γ) and Islet autoantibodies (ICA, GADA, IA-2)were measured in 56 patients with insulin dependent diabetes mellitus (IDDM) and 20 healthycontrol patients. RESULTS:The three proinflammatory cytokines measured [interleukin-2 (IL-2) , interferon gamma (IFN-γ)and tumor necrosis factor-β (TNF-β)], both TNF-β (50.0 ±5.9) (63.4± 5.4) and INF-γ (13.8 ± 10.9)(13.7 ± 5.5) showed a significant increase (P <0.05) with Islet autoantibody positivity, while theother three cytokines,(IL-4,IL-5 and IL-10), only IL-4 showed a positive increase (54.4 ± 1.4) withIslet autoantibody positivity although it is non- significant association.
