Kinetic Study of the Hydrolysis of synthesized Ibuprofen Ester and its Biological Activity

Abstract

It is known that the oral administration of ibuprofen caused an irritation of stomach as a side effect due to its carboxylic moiety. Ibuprofen ester was synthesized by linking the carboxylic moiety of ibuprofen and the hydroxylic group of paracetamol to reduce its side effect. Study the kinetic hydrolysis of prepared ester was examined at different values of physiological pH (1.0, 5.8, 6.4 and 7.4) at 37 ± 0.1 of 1 hour period. Measurements of absorbance were carried out by UV-Visible spectrophotometer to follow the stability of ester, it showed Pseudo first order hydrolysis. The pH- apparent rate profiles of ester was exhibited a good stability at pH 1.0 and pH 5.8. Pharmacological activity in vivo of prepared ester was evaluated in relation to analgesic and anti-inflammatory activity using the acetic acid method and the hind paw oedema inhibition, respectively. Acetyl salicylic acid (aspirin) was used as a reference drug for the above tests. The synthesized ester showed higher analgesic and anti-inflammatory action than aspirin.

من المعروف ان تناول الايبوبروفين عن طريق الفم يعمل على تهيج غشاء المعدة بسبب احتوائه على جزء حامضي في تركيبه ( الجزء الكاربوكسيلي ) و لتقليل مثل هذا التأثير , تم تحضير استر الايبوبروفين وذلك بربط الجزء الحامضي للايبوبروفين مع مجموعة الهيدروكسيل للفينول المعوض في الباراسيتامول . لقد تم قياس الاستقرارية من خلال اختبار حركية التحلل المائي للاستر المحضر , في اوساط حامضية مختلفة (عند الدالات الحامضية 1.0,5.8,6.4,7.4) عند درجة حرارة 37± 0.1°م و لمدة ساعة. تمت هذه الدراسة بقياس الامتصاصية بجهاز مطيافية الاشعة فوق البنفسجية . وقد تم قياس الفعالية الدوائية ( داخل الخلية ) لاستر الايبوبروفين , كمسكنات باستخدام طريقة حامض الخليك و كمضادات للالتهابات العضوية باستخدام طريقة قياس الورم في الرجل الخلفي للحيوان المختبري . واستخدم الاسبرين كمصدر عند مقارنة الفعالية الدوائية في كلتا الحالتين. ولقد اظهر استر الايبوبروفين فعالية بايولوجية ذات تاءثير فعال اقوى من الاسبرين فى كلا الاختبارين.

Abstract

AbstractNew fifteen polymers were prepared by modification of polyvinyl alcohol; these polymers were prepared by two steps. The first step was included reaction 1 mole of poly vinyl alcohol with 1mole of adipoyl chloride to produce ester-acid chloride polymer .Second step was included reaction the produced polymer in step one with different amines to obtained new ester-amide polymers .Four of them were reacted with acid chloride (acetyl chloride and benzoyl chloride) in DMF to produces ester imide polymers in third step. All these polymers have different physical properties which may be used in different applications. The prepared polymers were characterized by FTIR, 1HNMR spectroscopy, and by measuring their melting points, softing points, solubilities.

الخلاصةفي هذا البحث تم تحضير15 بوليمر جديد من تحوير بوليمر الكحول الفاينيلي، حضرت هذه البوليمرات بخطوتين،الخطوة الاولى تضمنت تحضير بوليمر استر –كلوريد حامض ابتداء من بولي فانيل الكحول بتفاعل 1مول منه مع 1 مول من كلوريد الاديبويل، الخطوة الثانية تضمنت تفاعل البوليمر الناتج من الخطوة الاولى مع امينات (ارماتية واليفاتية) مختلفة ليعطي بوليمرات جديدة حاوية على مجاميع استر امايد متدلية، اربعة من هذه البوليمرات تم تحويلها بخطوه ثالثة الى بوليمرات استر- ايمايد جديدة بتفاعلها مع كلوريد الحامض (كلوريد الاستيل وكلوريد البنزويل).في مذيب الDMF .هذه البوليمرات المحضرة ذات مواصفات فيزيائية جديدة مما قد يسمح لها باستخدامها في تطبيقات مختلفة. تم تشخيص هذه البوليمرات باستخدام اطيافFTIR ,NMR وقياس درجات الانصهار ودرجات التلين والذوبانية.

Abstract

This study included (50) samples of fasting healthy people in Ramadan males and females with ages between (80-19) years and (50) samples of healthy people who are not infected with any apparent disease without fasting males and females were considered a control group of the same categories Age above. In this study, blood samples were collected for healthy people in the case of fasting and control group after fasting period for fasting group (10-15) days and then follow the same people after (20) days and then the serum was separated, where the serum divided into two parts .The biochemical parameters in part one glucose (Glu), total cholesterol (TC), high-density lipoprotein cholesterol (HDL), low-density lipoprotein cholesterol (LDL), very low-density lipoprotein cholesterol (VLDL), triglyceride(TG) and phospholipids was measured by using kits obtained from regional and international suppliers. The measurement of percentage of fatty acids in the three parts (CE, TG, PL) was performed by Capillary Gas Chromatography (CGC). The results showed that there was a significant increase in the percentage and level of saturated fatty acids (SFA), a significant decrease in monounsaturated fatty acids (MUFA) and a significant decrease in polyunsaturated fatty acids (PUFA), in the cholesterol ester (CE) part of serum lipids for the fasting group compared with the control group.Also, the results of this study showed that there was a significant increase in (SFA), a significant decrease in (MUFA), and a significant increase in (PUFA) in triglyceride (TG) part in serum of the fasting group compared to the control group.The results showed that there was a significant increase in (SFA), a significant decrease in (MUFA) and a significant decrease in (PUFA) in phospholipids (PL) part in the serum for the fasting group compared with the control group.

Abstract

In this paper, several specimens from unsaturated poly ester resin with different filler materials such as (Fe2O3, CaCO3, CaO, Cement, Ca(OH)2( have beenprepared. Three weight percent have been used as (10%,20%, and 30% wt)from eachfiller materials. Casting samples are grinded by 800 grit using SiC grinding paper andthen washed by distilled water. Dry sliding wear has been done using pin on desktest in different period of wear time as (5,10,15,20,25,30 min). it has been shown thataddition of cement with percent (10 % wt) get lower wear rate for unsaturatedpolyester resin.

Abstract

New hydrazide compounds (A2) and (A9) were prepared from their corresponding esters (A1, A8) .These esters were also prepared from their precursors 5-ethoxy carbonyl-(4-methoxyphenyl)-6-methyl-1,2,3,4-tetrahydropyrimidine-2-thione (A1) via multicomponent reaction type and from hippuric acid respectively. The hydrazide compounds were then allowed to react with some aldehydes forming the corresponding hydrazones (A3-7) and (A10-14). The synthesized compounds were characterized by IR, 1H-NMR, 13C-NMR, and Mass spectroscopies and well discussed. .

تــم تحضير هــيدرازيدات جديدة وتـمثل المــركبات (A2 و A9) من استراتها المقابلــة (A1 و A8) وهذه الاسترات تـم تـحضيرها من مـوادها الأولـية5 – ايثوكسي كــاربونـيل (4 – ميثوكسي فنيل ) – 6 – مثيل –4,3,2,1 –رباعي هايدرو بيريمدين –2 – ثايون ( (A1عن طريق تفاعل متعدد المكونات وكذلك من حامض الهبيوريك على التوالي . هذه الهيدرازيدات تم مفاعلتها مع بعض الألدهايدات مكونة الهايدرازونات المقابلة (7– A3) و (14–A10 ). تم تشخيص المركبات المحضرة بالطرق الطيفية المتمثلة بـ IR,1H-NMR,13C-NMR و طيف الكتلة وتمت مناقشتها.

Abstract

Cetane improver is an additive that improves the cetane number of diesel fuel. Now the world focus on using cetane improvers which are friend to environmental that biodegradable and reduce exhaust of gases such as carbon monoxide and oxide of nitrogen. Cetane improver was prepared by reacting methyl ethyl ester of long chain fatty acids with nitric acid in the presence of acetic anhydride in nitration process, while the ester can be produced from castor oil through transesterification.In this work the optimum conditions of nitration reaction which includes mole ratio of nitric acid, temperature and time of reaction were studied according to the results of percent of yield and absorbance peak area of OH group of starting materials.It was found that 10 is best mole ratio of nitric acid to esters, 10-20˚c temperature for 3hours time.The nitrated products were analyzed as cetane number enhancers adding it to diesel fuel at 0.1 and 0.05% concentration and it was found that the cetane number of diesel fuel was increased at 25% percentage. Abstract

Background: acrylic resin denture base consider a common denture base material for its acceptable cost, aesthetic and easy processing but still has disadvantages including easy of fracture and low impact strength.Material and method: The experimental group was prepared by addition of 15% phosphoric acid 2-hydroxyethyl methacrylate ester (PA2HEME) with polymethyl methacrylate monomer; the experimental groups was compared with the control one. The specimens were prepared according to ADA specification No. 12 with dimension 65 mm x 10 mm x2.5 mm (length x width x thickness respectively). The prepared specimens were tested by three-point flexural strength utilizing Instron Universal Testing Machine (WDW, Layree Technology Co.), Shore D hardness tester used to measure hardness test. Statistical analysis used student T- test, mean and standard deviation.Results: The result of PA2HEME group showed high significant reduction comparing to the control group for both transverse strength and hardness test.Conclusion: Mixing 15% of PA2HEME with 85% methyl methacrylate (MMA) can reduce the mechanical properties of new modified polymethyl methacrylate (PMMA) acrylic resin

Abstract

Biodiesel is gaining considerable attention as a renewable sourceof energy, as an alternative to petroleum fuel and can be used in enginewithout modification in this paper, the profile of preparation of fatty acidmethyl ethyl ester from castor oil via a base-catalyzed transesterification wasinvestigated.. The variables chosen for the study were reaction time, oil to(methanol and ethanol) mixture ratio, and reaction temperature at constantconcentration of KOH(catalyst) .The effects of these variables on viscosity andspecific gravity were studied.

Abstract

sensitive and environmentally benign spectrometric method was developed for quantifying Meprobamate (MEP). The analyzed MEP was derivatized into a colored complex and determined spectrometrically. The colorimetric analytical parameters were optimized and validated. Low limit of detection (LOD) was achieved down to 1.88×10-6 mol/l while the limit of quantification (LOQ) was extended over the range of 1.97×10-6 - 1.35×10-3 mol/l. The high precision has been denoted by the 1.54% value of the coefficient of variation. The recovery was 96.07%, while the RSD (n=3) was 1.05 - 1.19%. The apparent molar absorptivity (Ɛ) obtained within 1154.7 - 1691.9 L.mol-1.cm-1. The outcomes have signified the high sensitivity of the technique. Furthermore, the avoidance of any traditional sample preparation steps prior to the analysis makes the method an excellent environmental approach. There were no analytical interferences with the common composites. The method has been successfully verified for the quantification of MEP content in bulk pharmaceutical matrix and can be applied to determine other environmentally toxic carbamates

Abstract

Ten Amide-ester copolymers were synthesized, whereby the sequence distribution and the used dicarboxylic acids were varied. The electric conductivity of the copolymers are determined in the pure and doped states. The results showed that two factors are important in determining the conductivity levels: the flexibility of the polymer chains, and the efficiency of conjugation along the chains. Thus acidic units that give rise to coiled chains structure, and hence flexibile segments gives high conductivity compared with acid units that give rise to extended structure and rigid segments like terephthalic acid. The introduction of saturated units that interrupts the chains conjugation decrease the electrical conductivity of the polymers.The temperature dependence of the electrical conductivity (σ) of the investigated copolymers was studied in the temperature range (303-373K). A plot of ln (σ) versus (1/T) reveals that two regions with an inflection point at the glass transition temperature Tg of the polymers. The Tg of the investigated polymers determined by this method was found to be influenced by the nature of the repeating unit of the polymers and their sequence distribution.

حضرت عشرة من بوليمرات الأستر أمين المشتركة والمختلفة في توزيع السلسلة والحامض الكاربوكسيلي الثنائي المستخدم. تم قياس التوصيلية الكهربائية للبوليمرات المشتركة في حالتها النقية والمشوبة. أظهرت النتائج ان العاملين المهمين اللذان يؤثران على التوصيلية الكهربائية هما مرونة السلسلة البوليمرية وكفاءة التناوب على طول السلسلة. أما المجموعة الحامضية التي تزيد من الشكل الملتوي للجزيئة فإنها تزيد من مرونة السلسلة وتؤدي الى زيادة التوصيلية مقارنة مع المجاميع الحامضية الأخرى التي تزيد من الشكل الممتد للسلسة وبوجود مقاطع صلدة ضمن السلسلة مثل حامض التريفثاليك. قطع التناوب في السلسلة بإدخال مجاميع مشبعة يقلل من التوصيلية. درس تأثير الحرارة على التوصيلية الكهربائية (σ) للبوليمرات قيد الدراسة ضمن مدى حراري (303-373 ) كلفن. رسم العلاقة بين ا للوغريتم الطبيعي (σ) ومقلوب درجة الحرارة أظهر منطقتين تفصل بينهما نقاط واضحة عند درجة الانتقال الزجاجي Tg للبوليمرات. إن درجة الانتقال الزجاجي للبوليمرات المعينة بهذه الطريقة تتأثر بعاملين: طبيعة الوحدات المتكررة الداخلة في تركيب البوليمر وتوزيعها في السلسلة.
