Abstract

Background: Tinea versicolor is a common dermatological problem ,world wide distribution ,caused by a dimorphic fungus called Malassezia furfur ,it live normally on skin as a commensal .Many factors play a role in the etiology of TV among these could be the disturbed immune systemwhich may be related to the ability of TV alter the immune system by a process calledImmunomodulation leading to subsequent infection.Immuno-inflammatory activity mediated by different cytokines could have a role in the etiologyof TV.Objective: To evaluate the serum level of the inflammatory cytokines ,IL-4andIL-8,in patients withTV as compared to immunocompramized patients and healthy groups.Patients and methods: This study enrolled 50 total patients, 15 of them were as patients controlgroup who were immunocompramized with evident skin lesion ,to be compared serologically with 15patients who had TV and normal immunity, the control group composed of20 healthy volunteers.Using ELISA test technique the following tests were done :detection of IL-4 and IL-8 in the sera ofall groups.Results and discussion: The statistical differences in the rate of detectable IL-4 level between TVand healthy control p< 0.001,while the difference was a highly significant between the healthycontrol and immunocompramized patients with TV,P<0.00338.The difference between twodiseased groups was(p< 0.219)which is non significant , when p<0. 05 was considered significant.IL-8 the results of our study showed a significant difference between the healthy control and thepatients with tinea versicolor, p-value was( 0.05).In this study there was a nice findings by which the difference between the healthy control and theimmunocompramized patients was highly significant as ,p<0.005.Conclusion :The genus Malassezia is an immunological paradox. In some circumstances, it actsas an adjuvant, activates the complement cascade, and elicits both cellular and humoral immuneresponses in healthy individuals, among which IL8 and IL-4 were notably increased . Abstract

ABSTRACT:BACKGROUND:More than half of all humans are colonized in their stomachs by Helicobacter pylori . That carriage was nearly universal among adults in developing countries suggests that in earlier times most humans carried these organisms.H. pylori was an important human pathogen that causes gastritis and is strongly associated with gastric ulcers, gastric adenocarcinomas, and mucosa-associated lymphoid tissue lymphomas. In response to H. pylori, interleukin-8 (IL-8) was secreted from host cells to attract components of the innate and adaptive immune systems to the site of infection.CagA+ cells induce higher levels of the proinflammatory IL-8 cytokine. That such effects are specific for CagA island genes had been shown by our study.OBJECTIVE:To determine the role of incidence of H.pylori CagA positive strain in induce higher level of the proinflammatory cytokine IL-8.METHODS:One hundred and fifty(150) Patients attended the Endoscopic Unit at"Gastroenterology and Hepatology Teaching Hospital/ Baghdad Medical City"were included in this study with ages range from 18 years to 65 years . The source of specimens to undergo oesophageal gastroduodenoscopy (OGD) collected from April 2009 to end of March 2010 were eligible for this study.Two types biopsy of samples for histopathology to detect H.pylori and detection of CagA gene by In Situ hybridization(ISH) and blood samples for estimation of serum IL-8 level by ELISA.RESULTS:There was a significant increase in IL-8 serum levels that P<0.05 due to the significant increase of Cag A in H.pylori positive strains.CONCLUSION:Our study revealed that increased in CagA expression in H.pylori positive strains and induced higher levels of the proinflammatory cytokines

Abstract

ABSTRACT: BACKGROUND: Multiple sclerosis (MS) is an autoimmune disease caused by a complex disorder that cluster many common diseases in children and adults, leading to chronic inflammatory disease of the central nervous system with morphological hallmarks of inflammation, demylination, axonal loss and gliosis .Factors responsible for MS and its progression are still not fully understood, but it has been suggested that both environmental determinants and genetic susceptibility are involved.OBJECTIVE :The aim of this study is to evaluate the role of inflammatory markers ;hs-CRP and IL-8 in pathogenesis of MS and to see the correlation between them.PATIENTS AND METHODS: Twenty one MS Patients (7male,14 female) attending the MS Clinic at Baghdad teaching hospital, their ages range between( 20-32) years (15) age- and sex- matched healthy control participated in the study. Investigations included serum measurements of high sensitive C-reactive protein (hs- CRP) and interleukin-8 in patients and control groups.RESULTS: The result of the present study showed significant increase in both CRP(p<0.001 ) and IL-8 (p< 0.001 ) in the serum of MS patients compared to control.CONCLUSION: There is evidence that neuroinflammatory process due to elevation of CRP and IL-8 has an important role in MS .

Abstract

ABSTRACT:BACKGROUND: Osteoarthritis is a degenerative joint disease, occurring primarily in older persons. Although many factors initiate this disease, progress to overt OA needs many biological substances like IL-6 and IL-8 that reduce cartilage repair ability or like reactive species that induce cartilage destruction through direct degradation of matrix components. OBJECTIVE: To shed the light on the expression of serum IL-6 and IL-8 levels, and production of serum malondialdehyde level (marker of oxidative stress), then estimation of the correlation among serum IL-6, IL-8 and MDA levels in patients with knee osteoarthritis.PATIENTS AND METHODS: This study was conducted in AL-Kadhemiya Teaching Hospital during the period from April 2011 to December 2011. The study included two groups: knee osteoarthritis( KOA) group of patients and the control group. Each group was further sub-divided into: obese & non-obese according to their BMIs. Sixty osteoarthritic patients aged 40-70 years were chosen depending on medical history, clinical examination, and radiographic observations. 10 ml of blood needed for assessment of the above makers. Thirty healthy control (age and sex-matched) were enrolled in this study. RESULTS: Mean serum IL-6 and IL-8 levels were significantly higher in KOA patients compared to control group for both (P<0.001), with the highest levels seen in the obese KOA group. Mean serum MDA level was significantly higher in patients with KOA compared with that in control (P<0.001). Significant negative correlation was observed between serum IL-6 and IL-8 levels. CONCLUSION: IL-6, IL-8 & MDA have important role in the pathogenesis of KOA. Receptors of IL-6 and IL-8 on chondrocytes could be considered as target for future therapy of any arthritic inflammation and also to consider MDA reduction in patients with knee osteoarthritis, as part of treatment of osteoarthritis.

Abstract

The result of interleukin-8 level through acute and chronic phases in pneumonia patients sera was showed high level in patients compared with healthy persons (0.194 ± 0.046, 0.095 ± 0.006 pg / ml , respectively) with a significant difference statistically (P˂0.05). The results of interleukin 8 levels in the age group ( 2 months -3 years ) higher than the age groups ( age 15-60years ) (age 60-85years ) (0.154 ± 0.04, 0.131 ± 0.02, 0.113 ± 0.2 pg / ml , respectively) with no significant difference (P˂0.05).

ظهرت نتائج قياس الانترليوكين 8 خلال الطورين الحاد والمزمن في مصل مرضى ذات الرئه كانت باعلى المستويات مقارنه بالاصحاء (0.194 ± 0.046 , 0.095± 0.006 بيكو غرام /مل على التوالي) مع فرق معنوي احصائيا (P˂0.05).كذلك في الفئه العمريه (شهرين-3سنوات ) اظهرت نتائج قياس انترليوكين 8 اعلى المستويات مقارنه مع بقيه الاعمار (15-60 سنه) (60-85 سنه) (0.154± 0.04, 0.131± 0.02, 0.113±0.2 بيكو غرام /مل على التوالي )مع بفرق غير معنوي .P˂0.05

Abstract

Infection with hepatitis HBV and HCV affects the liver and results in abroad spectrum of disease outcomes . The role of any given cytokine may be specific or overlapping to generate downstream effects that result from a maze of complex interactions . We evaluated the association between IL- 8 levels of patients with HBV , HCV infections and blood induces and some biological parameters.The study was carried on : - 30 patients chronically infected with HBV (12 males + 18 females ) , the mean age of the patients were 34.03 ± 12.78 years old -30 patients infected with HCV ( 17 males + 13 females ) with mean age of the patients 44.86 ± 17.75 years old. - 28 subjects (control) ( 20 males + 8 females ) , mean age of 28.32 ± 9.32 years old had negative HBs Ag and anti-HCV Ab for the duration of 17/9/20112 to 30/12/2012.Blood samples (6ml) were collected , 2ml for hematological measurements and 4 ml to obtain serum after centrifugation and stored at -84c0 in a deep freezer. Results show lower significantly differ between HBV patients and healthy control in RDW% , Neutrophils and Basophils count . While the mean levels of IL – 8 in sera significantly differ between HBV patients and healthy control( 17.72 ± 14.87 , 8.20 ± 3.15 pg/ml respectively In HCV patients there were lower significantly differ in red blood cells count , concentration of hemoglobin , haematocrit % , RDW% ,platelets, white blood cells count , lymphocytes and basophils . The mean levels of IL – 8 in sera and other parameter did not significantly differ between patients and control. Negative relationship between IL – 8 and lymphocytes , eosinophils in patients with HCV rather than HCV patients, also positive relationship between IL – 8 and ALT, AST enzymes.

إن إصابة الكبد بأحد فيروسات الكبد C , B يتسبب في التهاب الكبد، وإن استمرارية الإصابة الفيروسية للخلايا الكبدية تسبب تطور المرض إلى الحالة المزمنة والتي تعد مشكلة مرضية خطيرة قد تقود إلى تشمع الكبد Cirrhosisفسرطان الخلايا الكبديةHepatocellular Carcinoma ( HCC ) . وكرد فعل فأن الخلايا المصابة تقوم بتصنيع وإفراز الحركيات الخلوية cytokines عالية التخصص وذات تأثيرات تفاعلية معقدة ، منها IL- 8 الذي يرتفع مستواه في الدوران بزيادة شدة الالتهاب ، وبهذا فإنه يعد علامة دالة على تطور المرض . جاءت هذه الدراسة للتعرف على مستوى IL – 8 في الدوران ، ومعرفة التغيرات الحاصلة في صورة الدم وبعض القيم الكيموحيوية لدى الحاملين لفيروسي التهاب الكبد Bأو Cمن مرضى الغسيل الدموي في مستشفى بعقوبة التعليمي، ومن الاشخاص المسجلين لدى دائرة صحة ديالى / قسم وقاية الأمراض الوبائية والانتقالية . جمعت عينات من الدم الوريدي من 30شخص ) 12ذكور + 18 إناث ) حامل لفيروس التهاب الكبد الفيروسي Bو 30شخص ( 17 ذكور + 13 إناث ) حامل لفيروس التهاب الكبد C قورنت مع 28شخص 20 )ذكور + 8إناثNegative HBs Ag & anti – HCV Ab ( وعدت مجموعة سيطرة للمدة من 2012/9/17 ولغاية 2012/12/30 . أظهرت النتائج وجود انخفاض معنوي في أعداد خلايا ا لدم الحمراء ، حجم الخلايا المرصوصة ، تركيز خضاب الدم ، سعة انتشار الحمر، أعداد الصفائح الدموية ، أعداد خلايا الدم البيضاء ، أعداد الخلايا اللمفاوية ، إضافة إلى أعداد الخلايا القاعدية لدى حاملي فيروس HCV مقارنة بالسيطرة . أما مرضى التهاب الكبد الفيروسي HBV فقد أظهرت فروق معنوية في سعة انتشار الحمر ، خلايا العدلات وأعداد الخلايا القاعدية مقارنة بالسيطرة. وأظهر مستوى IL-8 ارتفاع معنوي لدى مرضى التهاب الكبد الفيروسي HBV دون HCV، فيما لم تختلف معنويا مستويات إنزيمات AST, ALT على الرغم من اجتيازها القيم الطبيعية وهذا يسري كذلك علىALKP ,TBIL ,DBIL . وجود علاقة ارتباط سالبة بين مستوى IL-8 والخلايا اللمفاوية والحمضية لدى مرضى التهاب الكبد الفيروسي HCV دون HBV ، مع وجود علاقة ارتباط قوية موجبة بين IL- 8 وإنزيمات الكبد ALT , AST .

Significance of Interleukin-8 in Cystitis and Schistosomal Chronic Cystitis

Abstract

One of the important mediators of early inflammatory response in cystitis and an agent for attraction of neutrophils to the inflammation site is IL-8, in addition to its role in regulation angiogenesis. This study was carried out to assess the level of IL-8 biomarker in urine of cystitis patients (schistosomal chronic cystitis and non schistosomal cystitis). Eighty eight Iraqi individual were divided to 50 cystitis patients and 38 healthy control, of whom urine sample and bladder biopsy were collected. Diagnosis of cystitis patients carried out by cystoscopy followed by histopathological examination and was classified to schistosomal chronic cystitis and non-schistosomal cystitis. Urine IL-8 level of studied groups was measured by ELISA test.According to the result of ELISA technique, the mean urine concentration (pg/ml) of IL-8 increased significantly in cystitis patients 224.86 pg/ml than healthy controls 51.70 pg/ml. In regard to the age of cystitis patients urine level of this marker was significantly different (P<0.05), but there was not any significant increase in IL-8 urine level according to sex, while IL-8 was with significant increase in Schistosomal chronic cystitis than non-Schistosomal cystitis. Lastly urine IL-8 showed specificity in diagnosis of cystitis 91.39% and more sensitivity 96.34%. Result of this study showed significant increase in urinary IL-8 concentration in cystitis patients, with significant correlation between IL-8 level with Schistsoma infection and age

الإنترلوكين- 8 هو أحد العوامل المهمة في الاستجابة الإلتهابية المبكرة من التهاب المثانة فضلاً عن دوره في جذب خلايا العدلة الى مواقع الإلتهاب ودوره في تنظيم التكون الوعائي. الهدف من هذه الدراسة هو تقييم مستوى المؤشر البايولوجي الانترلوكين- 8 في بول مرضى التهاب المثانة البلهارزي المزمن وغير البلهارزي.تم تقسيم ثمانية وثمانين فرد عراقي الى قسمين، الأول مكون من (50) مريضاً بالتهاب المثانة و(38) شخصاً من الأصحاء. تم جمع عينات بول وخزع من المثانة وتم تشخيص مرضى التهاب المثانة من خلال ناظورالمثانة وفحص الأنسجة ومن ثم تصنيف المرض الى إلتهاب المثانة البلهارزي المزمن والتهاب المثانة غيرالبلهارزي، وقد تم قياس الانترلوكين- 8 في بول المجموعات المدروسة من خلال تقنية الاليزا.وفقاً لنتيجة تقنية الاليزا ظهر زيادة إحصائية في تركيز الانترلوكين-8 في بول مرضى التهاب المثانة عن الاصحاء. أما فيما يتعلق بعمر مرضى التهاب المثانة فقد كان هناك إختلافاً إحصائياً بمستوى الانترلوكين- 8 بين الأعمار المختلفة، ولكن لم يكن هناك أي زيادة في تركيز الانترلوكين في البول حسب الجنس. كذلك لوحظ وجود اختلافات معنوية في تركيز الانترلوكين- 8 في مرضى التهاب المثانة البلهارزي المزمن والتهاب المثانة غير البلهارزي. وأخيراً أظهرالانترلوكين-8 دقة 91.39٪ وحساسية أكثر96.34٪ لتشخيص المرض.أظهرت نتائج هذه الدراسة وجود زيادة في تركيز الانترلوكين-8 في بول مرضى التهاب المثانة فضلاً عن وجود علاقة ذات دلالة إحصائية بين مستوى الانترلوكين- 8 في الادرار والاصابة بالبلهارزيا والعمر

Abstract

Cytokines may be implicated in the patho-physiological mechanism of preterm and term labor. Many studies indicates cytokines as predictors of preterm delivery and explain partially mechanism of preterm uterine contraction. The objective of this study was to find out whether preterm labor is associated with raised maternal serum concentration of intreleukine-6 and interleukine-8 levels.Fifty-pregnant women in preterm labor (group A), 25 term pregnant women in labor (group B), and 25 women preterm not in labor (group C(. The serum cytokines levels were measured by enzyme linked immunosorbant assay (ELISA(. The result showed that IL6 and IL8 significantly higher during preterm labor and term labor in compares with preterm group did not in labor. No statistical difference was found in cytokines levels between women in preterm labor with rupture membranesand those with intact membranes. There was a significant higher serum concentration of IL6 in late preterm labor (34weeks-36+6days) compared with early preterm labor (24weeks-33weeks+6days). In conclusion; our data presented that higher serum cytokines levels (IL-6 &IL-8) in patient with preterm labor and term labor than preterm not in labor. The increase of IL-6 &IL-8 level during labor can be related with the possible role of these cytokines in the immunological mechanism of labor beginning

The Prevalence of Human Papilloma Virus among Breast Cancer Women in Relation with IL-8 Level in Kirkuk Province

Abstract

The study aimed at evaluating the frequency of HPV in breast cancer women in relation with serum IL-8 level. The study that carried out in Kirkuk city from 15th of January 2017 to 15th of June 2017, a total of 150 breast cancer women who admitted to Kirkuk oncology cancer and 96 healthy control women whose ages were between 30-75 years. Patients and control were investigated for detection of human papilloma virus (HPV) 16 E7 protein and interleukin-8 (IL-8) by ELISA technique. The rate of breast cancer women with HPV 16 E7 (30.67%) was higher than that in control women (3.125%) with highly significant relation. The highest rate of breast cancer women was within the age group 40-49 years with no significant relation between breast cancer and age. The study showed that the highest means of IL-8 level (36.92 pg/ml) were found in breast cancer women without HPV, 29.09 pg/ml in HPV positive and 6.45 pg/ml in the control group with highly significant relation. There was a highly significant relation of WBCs, lymphocytes and neutrophils count and Hb level among the study groups concerning HPV. WBCs count and hemoglobin level were higher in the control group than in breast cancer women. The mean absolute lymphocytes count was higher in breast cancer women with HPV infection than those without HPV and the control group (4.13, 1.80 and 1.89 respectively), while the least mean of neutrophils count was reported in breast cancer women with HPV infection. Conclusions: It was concluded that there was a highly significant relation between HPV and EBV infection with breast cancer.

تهدف الدراسة الى تقييم الانتشار المصلي لفيروس الورم الحليمي البشري في النساء المصابت بسرطان الثدي مع علاقته به مستوى IL-8. أجريت الدراسة الحالية في مدينة كركوك (مركز كركوك للأمراض السرطانية) في الفترة الممتدة من 15 كانون الثاني 2017ولغاية 15 حزيران 2017 وشملت الدراسة 150 امرأة مصابة بسرطان الثدي و96 امرأة غير مصابة كمجموعة سيطرة في الدراسة حيث تراوحتاعمار المجموعتين من 30 الى 75 سنة ، وتم اجراء فحوصات الـ ELISA للكشف عن فايروسالورم الحليمي البشري في المجموعتين فضلا عن الكشف عن مستوى IL-8.اظهرت النتائج ان نسبة إصابة مريضات سرطان الثدي بفيروس البشري الحليمي (30,67%) والتي كانت اعلى مما هو الحال في مجموعة السيطرة (3,125%). كانت أعلى نسبة للمريضات المصابات بسرطان الثدي ضمن الفترة العمرية 40-49 سنة ولم يكن هنالك أي تأثير مع سرطان الثدي. قد أظهرت الدراسة ان اعلى مستوى لـ IL-8 ( 36,92 بيكوغرام/مليلتر) وجد في مريضات سرطان الثدي مقارنة بالمريضات المصابات بفايروس الورم الحليمي البشري (29,09 ، 28.74 و18,74 بيكوغرام/مليلتر على التوالي) ومجموعة السيطرة (6,45) مع وجود علاقة إحصائية بهذا الصدد. وان هنالك علاقة إحصائية قوية بين تعداد خلايا الدم البيضاء والخلايا العدِلة والخلايا اللمفية ومستوى الهيموغلوبين في مجاميع البحث بما يخص الإصابات بفايروس الورم الحليمي. كان تعداد خلايا الدم البيضاء ومستوى الهيموغلوبين اعلى في مجموعة السيطرة مقارنة بمريضات سرطان الثدي بينما كان تعداد الخلايا اللمفية عاليا في مريضات سرطان الثدي اللواتي يحملن الإصابة بفايروس الورم الحليمي البشري مقارنة بغير المصابات ومجموعة السيطرة ( 4,13 ، 1,80 و1,89 على التوالي). كان اقل تعداداً للخلايا العدلة قد سُجّل في مريضات سرطان الثدي المصابات بفايروس الورم الحليمي البشري. من هذا يُستنتج ان هنالك علاقة قوية بين سرطان الثدي والاصابة بفايروس الورم الحليمي البشري ومستوى IL-8.

Abstract

Background: Cytokines are the messengers of the immune system.They are mostly secreted by macrophages and lymphocytes and their production is induced in response to injury or infections.Objective: Biopsy speciemens from the middle body of the stomach were obtained from 18 patients with gastric carcinoma, 32 patients with atrophic gastritis, 50 patients with chronic gastritis and 20 healthy subjects and IL-8 and IL-1B levels were detrmined.Methods: Sandwich-type enzyme-linked immunosorbent assay kit was used to determine the IL-8 and IL-1B levels.Results: IL-8 and IL-1B levels were significantly higher in gastric carcinoma patients than patients with atrophic gastritis and both significantly higher than healthy subjects. Conclusions: IL-8 and IL-1B levels were recommended to be used in differentiation between gastric carcinoma and atrophic gastritis.
