Cytokines profiling as prognostic markers in newly diagnosed acute myeloid leukemia

Abstract

BACKGROUND: Acute myeloid leukemia (AML) is a common acute leukemia in adult. Recent studieshave shown that cytokine systems influence leukemic cell biology and clinical investigations. Amongvarious cytokines, interleukin‑6 and interleukin‑10 (IL‑6 and IL‑10) participate an essential functionin progression of the disease.OBJECTIVE: The aim of this study is to quantify of IL‑6 and IL‑10 levels in AML patients who arenewly diagnosed and evaluate lipid profile to assess the relationship between lipid profile level andbody mass index (BMI) in myeloid leukemic patients.MATERIALS AND METHODS: Samples were collected from 45 patients with AML from AL‑YarmoukTeaching Hospital in addition to another 45 healthy individuals were served as a control group duringa period from October 2015 to October 2016. Patients ages ranged from 40 to 60 years. IL‑6 andIL‑10 were measured in all patients before any treatments and compared with control group.RESULTS: There was an increase in age, white blood cell, and hemoglobin in AML patients ascompared to control, but it was not significant. There was a significant increase in neutrophils andlymphocyte count, (P = 0.001). Furthermore, there was an increase in BMI, fasting blood sugar,and lipid profile except high‑density lipoprotein cholesterol for AML patients as compared to control,but it was not significant. There was a significant increase in serum IL‑6 and IL‑10 for AML patientscompared to controls (P = 0.001).CONCLUSIONS: The present study refers that AML patients were associated with high concentrationof IL‑6 and IL‑10 in comparing to the control group.

Abstract

ABSTRACT:BACKGROUND:Fibromyalgia syndrome (FMS) is a common rheumatologic syndrome with multiple manifestations and associated with many diseases, it characterized by chronic wide spread muscular pain and tenderness. Further, circulatory levels of pro-inflammatory cytokines like IL-8 and others may be altered in FMS patients, possibly associated with their symptoms.The objective of the study is to measure IL-8 concentration and to evaluate its role in the pathogenesis of primary FMS.OBJECTIVE:Fifty patients with primary FMS were included in the study (37 females and 13 males) the age range of (17-65) years (Mean ± SD) (40.13 ± 12.0) years, and thirty healthy individuals volunteers (21 females and 9 males), whose age and sex matching with FMS patients, age ranging (18 - 63) years, (Mean ± SD) (36.1 ±10.0) years.METHODS:IL-8 concentration was measured in sera of patients and controls by ELISA kit. Anthropometric measurements like body mass index (BMI) and waist circumference (WCr) were taken ,besides other features like sleep disturbance , emotional distress , and fatigue were reported.RESULTS:IL-8 concentration was higher in FMS patients than controls(40.24±22.0) pg/ml vs. (19.16±7.8) pg/ml. This elevation was highly significant statistically (p=0.000).Other measurements in patients group like BMI, WCr were(28.39±5.0) Kg/m2 and (100.34±13.21) cm respectively .These values were highly significant when compared to their control group (p=0.002)and (p=0.008) respectively .Clinical features like sleep disturbance, emotional distress ,and fatigue showed highly significant difference between the two groups . No significant differences were reported with respect to age and sex.CONCLUSION:The result of the current study suggest that interleukin-8, (IL-8) might have a role in the pathogenesis of FMS.

Abstract

AbstractBackground: Adipose tissue is the source of a wide variety of molecules involved in the regulation of energy output and carbohydrate metabolism. Among these, cytokines, such as tumor necrosis factor (TNF)-α and interleukin (IL)-6, appear to play a role in modulating insulin sensitivity in peripheral tissues and have been associated with the development of insulin resistance in adults. Aim of study: The aim of the study was to ascertain the relationship among the degree of adiposity, circulating fasting insulin levels, with plasma IL-6 in an obese child and adolescent population with or without glucose intolerance.Subjects and Materials: Eighty obese children and adolescents were studied. Plasma interleukin (IL)-6 were measured for those obese with normal oral glucose tolerance Test (OGTT), and obese with glucose intolerance. These patients were compared with eighty healthy children matched for age and sex.Results: Plasma fasting insulin concentration and HOMA (Homeostatic model assessment) were significantly higher in glucose intolerance obese patients [(28.2±14.9µIU/ml),(5.7±4.8)] compared with their results in normotolerance obese patients [(20.9±8.3µIU/ml),(4.5±2.7)] and control subjects[(14.8±6.3µIU/ml),(3.2±1.9)] (p 0.005 ,0.001) respectively, Plasma levels of (IL-6) in glucose intolerance obese patients were significantly higher (2.9±1.3 ng/ml) compared with normotolerance obese patients (2.0±1.1 ng/ml) and control subjects (1.5±0.9 ng/ml)(p 0.005).Interleukin (IL-6) was positively correlated with BMI, (r=0.37; p 0.001), waist and hip circumferences(r=0.35; r=0.33) respectively, (p 0.001), insulin levels (r=0.30; p 0.001), TG and VLDL-C (r=0.27; p 0.001)Conclusion: Plasma IL-6 appears to be involved in glucose metabolism, insulin resistance, and dyslipidemia in obese children and adolescents with glucose intolerance.

الخلاصةالخلفية العلمية: الأنسجة الدهنية هي مصدر لطائفة واسعة من الجزيئات المشتركة في تنظيم إنتاج الطاقة والتمثيل الغذائي للكاربوهيدرات ,من بين هذه الجزيئات الحركيات الخلوية(السايتوكينات) مثل عامل نخر الورم (TNF-) والانترلوكين-6(IL-6) ويبدو انها تلعب دورا في تحوير حساسية الانسولين في الأنسجة المحيطية و كذلك ارتبطت مع تطور مقاومة الانسولين في البالغين الهدف من الدراسة: ان الهدف من الدراسة هو التحقق من العلاقة ما بين درجة السمنة و مستويات الانسولين الصيام الدوار مع نسبة الانترلوكين (IL-6) في بلازما الأطفال البدناء والمراهقين المصابين وغير المصابين بعدم تحمل الكلوكوز.المواد والطرق: تم دراسة 80 من الأطفال و المراهقين البدينين , حيث تم قياس نسبة الانترلوكين-6 (IL-6) في بلازما هؤلاء البدناء ذو التحمل الطبيعي للكلوكوز الفموي والبد ناء المصابين بعدم تحمل الكلوكوز. قورنت هذه النتائج مع نتائج ثمانين من الأطفال الأصحاء من نفس الفئة العمرية والجنس.النتائج : أظهرت النتائج وجود زيادة نوعية في نسبة تركيز الأنسولين و HOMA عند البد ناء المصابين بعدم تحمل الكلوكوز ](28,2± 14,9مايكرو وحدة قياسية/مليلتر) (5,7 ± 4,8 )[ عند مقارنتها عند البد ناء ذو التحمل الطبيعي للكلوكوز الفموي ](20,9 ± 8,3 مايكرو وحدة قياسية/مليلتر) (4,5±2,7) [ والأشخاص الأصحاء ](14,8 ±6,3مايكرووحدة قياسية/مليلتر) (3,2 ± 1,9[(l ] 0,005 p< و0,001 [p< على التوالي. كذلك اظهرت النتائج وجود زيادة نوعية في نسبة الانترلوكين-6 في بلازما البد ناء المصابين بعدم تحمل الكلوكوز](2,9 ±1,3 نانوغرام/مليلتر) [عند مقارنة نسبها في البد ناء ذوي التحمل الطبيعي للكلوكوز الفموي](2,0 ± 1,1 نانوغرام/مليلتر)[ والأشخاص الأصحاء(1,5 ±0,9 نانوغرام/مليلتر), (0,005p<), كذلك أظهرت الدراسة وجود علاقة موجبة بين نسبة الانترلوكين-6 في بلازما المرضى المصابين بعدم تحمل الكلوكوز مع دالة كتلة الجسم (0,37 r= 0,001 P<), محيط الخصر و الورك (0,33و0,35 r=)على التوالي (0,001p<), نسبة الانسولين (0,30 r=)(0,001 p<) والدهون الثلاثية والدهون ذات الكثافة الخفيفة جدا ( 0,27 =r )( 0,001 p<)الخلاصة : يبدو أن مستوى الانترلوكين-6 في بلازما الدم تشارك في استقلاب الكلوكوز,مقاومة الانسولين,واضطراب الدهون لدى الاطفال واليافعين البدناء المصابين بعدم تحمل الكلوكوز.

Abstract

Background:Oligozoospermia is one of male infertility type which related with sperm lowering count in seminal fluid under limited reference. There are many factors leads to oligozoospermia like, congenital defects, endocrinal disorders, genetic abnormalities, immunological factors , life style , or in some cases unknown reasons. Immune and some non-immune cells produce immuno-chemical messengers proteins called (Interleukins) work as alerts proteins and contribute in immune body defend. These mediators found in all body fluids, also found in seminal fluid,and it’s presence of certain concentrations reflects the state of health of the body.Objective:Measure the level of interleukin-6 in fertile (normospermia) and infertile (Oligozoospermia).Materials and Methods:Microscopically and macroscopically ejaculates analysis according to WHO (2010) criteria for eighty -seven ejaculate from normospermia and oligozoospermia patients with age rang (21-47)yrs. , in duration of ( Nov- 2013 to Mar- 2014), study divide in to three groups according to the concentration ranges in to (normospermia > 15X 106 cells/ml), (Mild to Moderate Oligozoospermia 15-5 X106cells/ml) , (Severe Oligozoospermia< 5X106). Then store all samples in(-20) , IL-6 was detected through the (ELISA) specific way and kit .Results:Interleukin-6level is less in normospermia group than Oligozoospermia groups at (P<0.05), IL-6 level not correlated with sperm count, except weak correlation in severe group at (P≤ 0.05) and (t α=0.05).Conclusions:There are no correlation between IL-6 level and sperm cells concentration but the variation of interleukin-6 level among the groups indicate that some sperms parameters influence on interleukin-6 levels among the groups like sperm motility or morphology and also that the causative factors which lead to oligozoospermia also effect on IL-6 level and effect on spermatogenesis

Abstract

Toxoplasmosis is the general term for infection and disease in man and animal caused by a parasite called Toxoplasma gondii, this disease is prevalent among different categories of people, Heart disease are one of them. Because of the dormancy status of T. gondii, there is ambiguity and variation in the immunity of patients. This study aimed to investigate the immune status of heart disease patients infected with toxoplasmosis. Two hundred and fifty samples of both heart disease patients and controls had been tested by ELISA technique to detect anti-Toxoplasma Abs (IgG and IgM). The positive samples with toxoplasmosis were tested to detect the level of interleukins (IL-10, IL-12 and IL-17). The result of this study revealed the prevalence of toxoplasmosis among heart disease patient in about (43.33% IgG and 0.66% IgM). The results of interleukins appeared high level of IL-12 (82.85 pg./ml) in heart disease patients infected with toxoplasmosis when compared with control (60.97 pg./ml) with significant difference between them at p≤0.05, while there was no significant difference in IL-10 between patients and controls. The results also showed decreased level of IL-17 in patients with heart disease. The increased level of IL-12 in patient with heart disease infected with toxoplasmosis indicate to the activity of cell mediated immunity in those patients and may be lead to provoke an inflammatory status in the heart valves and muscle then worse heart disease to be sever and need complicated treatments.

داء المقوسات هو مصطلح عام للأصابة والمرض في الإنسان والحيوان يسببه طفيلي يعرف بالتوكسوبلازما، هذا المرض منتشر بين فئات مختلفة من البشر، ومن ضمنهم مرضى القلب . بسبب الحالة الساكنة (الكامنة) للطفيلي هنالك اختلاف وغموض في الحالة المناعية للمرضى المصابين بالطفيلي . هدفت هذه الدراسة إلى التحقيق في الحالة المناعية لمرضى القلب المصابين بداء المقوسات , وقد تم اختبار مائتين وخمسون عينه من كل من مرضى القلب والسيطرة(الأصحاء) بأستعمال تقنية اللأليزا (تقنية الانزيم المرتبط) للكشف عن الأجسام المضادة لطفيلي التوكسوبلازما , كذلك تم اختبار العينات الموجبة للتوكسوبلازما لتحديد مستويات الأنترلوكينات (10-IL, 12-IL ,17-IL) . أضهرت نتائج هذه الدراسة انتشارطفيلي التوكسوبلازما بين مرضى القلب بنسبة 43.33% للجسم المضاد ( IgG) و0.66 % بالنسبة للجسم المضاد( IgM) . أما نتائج اختبار الأنترلوكينات أضهرت أرتفاع مستوى الأنترلوكين 12- IL (lm ̸ pg 82.85 ) في مرضى القلب المصابين بطفيلي التوكسوبلازما عند مقارنتها مع عينات السيطرة الأنترلوكين 12 -IL ( ml ̸ pg 60.97 ) حيث اضهرت فرق معنوي بينهم عند0.05≥p , بينما لا توجد اختلافات واضحة في مستوى الأنترلوكين 10-IL بين مرضى القلب وعينات السيطرة , كذلك أضهرت النتائج أنخفاض في مستوى الأنترلوكين 17 في مرضى القلب. زيادة مستوى الأنترلوكين 12 في مرضى القلب المصابين بداء المقوسات يشير الى نشاط المناعة الخلوية في مرضى القلب ، ويمكن أن يؤدي إلى إثارة حالة التهاب في صمامات القلب والعضلات ثم أمرض القلب الأكثر شدة والحاجة للمعالجة المعقدة.

8.INTERLEUKIN-4 SINGLE NUCLEOTIDE POLYMORPHISM C-590T POLYMORPHISMS IN RELATION TO ASTHMA

Abstract

Background:Single nucleotide polymorphisms in the promoter regions of genes encoding for some interleukins may associate with occurrence of asthma.Objective:To investigate the association of single nucleotide polymorphisms of interleukin-4 (IL-4) (C-590T) and asthma.Methods:Forty-five patients with asthma and 40 apparently healthy subjects (represent the control group) were enrolled in this study. Blood samples were collected from both patients and controls. DNA was extracted from blood samples and gene fragments corresponding to IL-4 C-590T were amplified with specific primers using conventional PCR technique. Results:The heterozygote genotypes of IL-4 C-590T (CT) showed significant association with asthma (OR = 3.922, 95% CI= 1.153-13.339, P = 0.028). Detection of interleukin-6 and interleukin-8 in serum from women with recurrent spontaneous abortion

Abstract

To investigate the role of IL-6 and IL-8 in the immune-regulatory mechanisms involved in the recurrent spontaneous abortion of the first trimester of pregnancy. Serum level of IL-6 and IL-8 were determined in 25 women of age (20-35) years who had a spontaneous abortion of unknown aetiology during the first trimester of pregnancy .They were compared with the corresponding levels of 20 pregnant and non-pregnant women as control groups .cytokine levels were measured by (ELISA) technique .The women with spontaneous abortion had highly significant (P < 0.01) increased serum level of IL-8 and highly significant (P < 0.01 ) decreased level of IL-6 compared to those with normal pregnant and non-pregnant women. The results of this study may be related to the underlying aetiopathogenetic mechanisms , however , There is sufficient evidence for their use as predictive markers of pregnancy outcome.

لغرض التحري عن دور الحركيات الخلوية (8,6) في آليات التنظيم المناعي المتضمنة لدى النساء ذوات الاجهاضات العفوية المتكررة في الاشهر الاولى من الحمل . تم التحري عن مستوى تلك الحركيات الخلوية (6، 8) في مصول خمس وعشرون امرأة بأعمار (20- 35)سنة يعانين من اجهاضات عفوية غير معروفة السبب في الاشهر الاولى للحمل وتم مقارنة ذلك مع عشرون امرأة كمجاميع سيطرة (عشرة نساء حوامل وعشرة نساء غير حوامل) . تم قياس مستوى الحركيات باستخدام تقنية الامتزاز المناعي المرتبط بالانطيم (ELISA) . أظهرت نتائج الدراسة وجود ارتفاع معنوي عالي في مستوى السايتوكين الخلوي(8) وانخفاض معنوي عالي في مستوى السايتوكين الخلوي-6 (P<0.01) مقارنة بالنساء الحوامل والنساء غير الحوامل . ان نتائج تلك الدراسة ذات علاقة بالاليات المرضية لدى النساء ذوات الاجهاضات العفوية ويمكن استخدام تلك الحركيات الخلوية كمؤشرات تخمينية لنتائج الحمل .

Abstract

Recurrent Spontaneous Abortion (RSA) is the most painful experience for couples expecting a child. This study aimed to determine the relevance of IL-2 and IL-6 in recurrent spontaneous abortion (RSA). Serum samples were collected from 90 women attend Al Kadhmiya teaching hospital in Baghdad. 60 women (first group) had recurrent abortion the women were negative for rubella virus, herpes simplex virus and toxplasma gondii. And they were negative from bacterial infection eg. Niesseria gonorrhea and Chlamydia trachomatis. The histopathological tests for fetus abnormalities were negative in this group, and 30 women (second group) with successful pregnancy (normal delivery). All samples were analyzed for IL-2 and IL-6 by commercially available Enzyme-Linked Immunosorbent Assay (ELISA) kits. The data showed highly significant increase in the serum level of IL-2 in group 1 compared with group 2 (P<0.001). However, IL-6 showed highly significant increase level in group 2 compared with group 1. In addition, there was no significant correlation between these two markers in studied groups. The data of this study strengthen the possibility that high level of IL-2 and low level of IL-6 may explain the role of type-1 cytokines in the pathogenicity of recurrent spontaneous abortion.

يعد الإسقاط الذاتي المتكرر من التجارب الأليمة في حياة الزوجين الراغبين في إنجاب الأطفال. أجريت هذه الدراسة للتحري عن دور الانترلوكين 2 و6 في إحداث الإسقاط الذاتي المتكرر حيث جمعت تسعون عينة مصل من مجموعتين من نساء ادخلن إلى مستشفى الكاظمية التعليمي المجموعة الأولى ستون عينة كانت من نساء عانوا من الإجهاض الذاتي المتكرر واللاتي سبق واثبت خلوهن من الإصابة بفيروس الحصبة الألمانية (الحميراء) Rubella virus, وفيروس الحلأ التناسلي Herpes simplex virus وداء المقوسات gondii Toxoplasma وكذلك عدم إصابتهن ببكتريا المسببة السيلان, Niesseria gonorrhea والكلاميديا Chlamydia trachomatis وكذلك أثبتت الدراسة النسيجية من عدم وجود تشوهات في أجنة هذه المجموعة. المجموعة الثانية كانت من ثلاثون عينة كانت من نساء وضعن حملهن بصورة طبيعية. جميع العينات فحصت بطريقة الاليزا (ELIZA) للتحري عن الانترلوكين 2 و 6 , أظهرت النتائج فروق معنوية عالية في مستويات الانترلوكين 2 في مصول المجموعة الأولى مقارنة مع المجموعة الثانية (p< 0.001) , وكذلك كانت معدلات الانترلوكين 6 في مصول المجموعة الثانية مرتفعة مقارنة مع المجموعة الأولى. بالإضافة إلى ذلك لم تظهر النتائج ارتباط معنوي بين هذين الواصمين في المجاميع قيد الدراسة . نتائج هذه الدراسة دعمت احتمالية دور(السايتوكاين 1) في أمراضية الإسقاط التلقائي المتكرر في حالات ارتفاع الانترلوكين 2 وانخفاض الانترلوكين 6.

Abstract

Summary:Background: There is a general acceptance which illustrated that auto antibodies act as a central immunological disturbance in most of the auto immune diseases, among these auto immune disease lies the SLEPatients and Methods: Thirty five patients with SLE were compared to twenty age and sex matched, control subjects and studied for the presence of auto antibodies, plus IL-4 and IL-6 using Elisa method and immune fluorescent method (for ANA only)Results: Data showed that IL-6 detectable levels were statistically significant in patients with positive anti ds-DNA, but not significant statistically in ANA positive patients although it was detected in 24 (70.6%) of positive ANA patients, while there was no statistically significant correlation between IL-4 detectable level and autoantibodies production.Conclusion: There is apparently a positive correlation between IL-6 and anti ds DNA production in lupus patients.Key words: Autoantibodies, Interleukin-4, Interleukin-6, systemic lupus erythematosus

Interleukin-8 and Interleukin-10 in Patients with Ischemic Heart Disease

Abstract

ABSTRACT:BACKGROUND: Ischemic heart disease is the main cause of death throughout the world. Cytokines are key regulatory glycoproteins . They are intimately associated with atherogenesis and modulate plaque morphology and stabilization. OBJECTIVE: To study the role of pro-inflammatory Interleukin-8 , and anti-inflammatory interleukin-10 in patients with Acute coronary syndromes (ACS)(acute myocardial infarction and unstable angina).METHODS: This study included (55) patients with acute coronary syndrome, (30) patients with acute Myocardial infarction ( 24 males and 6 female) and 25 patients with unstable Angina (11 males and 14 females). This project has been carried out on patients at the coronary care unit (CCU) of Baghdad teaching hospital/ Medical City/ Baghdad, during the period from April to June 2010. A twenty five, apparently healthy individuals were taken as a healthy control group. Venous blood sample was taken from each subject. Levels of interleukin-8 and Interleukin-10 were estimated by using enzyme linked immunosorbant assay . {ELISA-kit for detection of IL-8 (BioSource Europe S.A. /catalogue number; KAP1301)} and {ELISA-kit for detection of IL-10 (BioSource Europe S.A./ catalogue number; KAP1321)} RESULTS: Plasma level of pro-inflammatory cytokine (interleukin-8) showed a significant elevation in patients with acute myocardial infarction (Mean ± SD =95.26 pg/ml ± 102.89 ;P value=0.004) and unstable angina (Mean ± SD =146.43 pg/ml ± 154.45 ;P value=0.019) in comparison to control group (Mean ± SD =47.75 pg/ml ± 36.67). Serum level of anti-inflammatory cytokine (interleukin-10) showed a significant decrease in unstable angina patients (Mean ± SD =16.95 pg/ml ± 11.92 ;P value=0.012) in comparison to control group (Mean ± SD =30.29 pg/ml ± 21.17),however, it showed no significant differences among acute myocardial infarction patients (Mean ± SD =27.73 pg/ml ± 23.16 ;P value=0.500) in comparison to control group. The ratio of pro-/anti-inflammatory cytokines (interleukin-8 / interleukin-10) was significantly elevated in acute coronary syndrome; acute myocardial infarction and unstable angina patients (P=0.001, P=0.019, P=0.001 respectively) .CONCLUSION: Interleukin-8 is an important pro-inflammatory cytokine in coronary artery disease which may contribute to the plaque destabilization in acute coronary syndrome especially unstable angina, and low level of anti-inflammatory interleukin-10 which is considers as a risk factor of development of acute events in unstable angina
