Abstract

Brucellosis in sheep and goats is a zoonotic infection (excluding Brucella ovis) with important effects on both public health and animal health. The disease is widespread in many areas of the world, particularly in some Mediterranean and Middle Eastern countries. Aim of the study: the present study aims to find out the prevalence of brucellosis in sheep in one of the areas of An Najaf El-Ashraf city, and study of the immune response by measuring the levels of and interleukin-10 (IL-10). Methodology: Current study conducted in the Najaf province (Al-Barakyia) during the period between September 2013- March 2014, fulfilled by using Rose Bengal and Enzyme Immunosorbent Assay (ELISA) tests for estimation of brucella antibodies. A total number of 48 serum samples were collected from sheep and stored at -20°C. Results: the frequency of positive sera by Rose Bengal test (RBT) was 20% and by ELISA technique was 25%. The level of interleukin 10 (IL-10) was estimated in all surveyed animals, 12 sera samples were showed an elevated IL-10 levels, all were positive with ELISA serological test. Conclusion: the detection of significant concentration of IL-10 among the seropositive cases may be indicated an active infection of brucella with type 2 cytokine production.Recommendations: the evaluation of immune response of vaccinated sheep with brucella REV1 vaccine, depending on the estimation of IFN-γ and IL-10 ratio that will reflect the cytokines induced during vaccination and consequently, the effective or not immune response.

Abstract

BACKGROUND: Acute myeloid leukemia (AML) is a common acute leukemia in adult. Recent studieshave shown that cytokine systems influence leukemic cell biology and clinical investigations. Amongvarious cytokines, interleukin‑6 and interleukin‑10 (IL‑6 and IL‑10) participate an essential functionin progression of the disease.OBJECTIVE: The aim of this study is to quantify of IL‑6 and IL‑10 levels in AML patients who arenewly diagnosed and evaluate lipid profile to assess the relationship between lipid profile level andbody mass index (BMI) in myeloid leukemic patients.MATERIALS AND METHODS: Samples were collected from 45 patients with AML from AL‑YarmoukTeaching Hospital in addition to another 45 healthy individuals were served as a control group duringa period from October 2015 to October 2016. Patients ages ranged from 40 to 60 years. IL‑6 andIL‑10 were measured in all patients before any treatments and compared with control group.RESULTS: There was an increase in age, white blood cell, and hemoglobin in AML patients ascompared to control, but it was not significant. There was a significant increase in neutrophils andlymphocyte count, (P = 0.001). Furthermore, there was an increase in BMI, fasting blood sugar,and lipid profile except high‑density lipoprotein cholesterol for AML patients as compared to control,but it was not significant. There was a significant increase in serum IL‑6 and IL‑10 for AML patientscompared to controls (P = 0.001).CONCLUSIONS: The present study refers that AML patients were associated with high concentrationof IL‑6 and IL‑10 in comparing to the control group.

Abstract

This study investigated whether interleukin- 10 (IL-10) gene promoter region polymorphisms are associated with susceptibility or clinical presentation of type 1 diabetes. The frequency of -1082G/A and -819C/T, polymorphisms was analyzed in 60 Egyptian patients with type 1 diabetes and in 60 healthy control subjects in a case-controlled study. However, the -819C/T not showed significance between patient and control (P=0.208) and 1082G/A were associated with T1DM disease (P<0.0001). These results suggest that the IL-10 gene promoter polymorphisms are associated with type 1 diabetes in Egyptian population.

تم اختيار عينة مكونة من 120 شخصا" ومقسمة إلى مجموعتين (60 من المرضى المصابين بمرض السكري من النوع الاول و 60 من الأشخاص الأصحاء كمجموعة ضابطة) تم التحري عن التباين الجيني في جين الانترلوكين (10) في المواقع التالية 1082G/A and -819C/T وارتباطه بمرض السكري من النوع الاول وأظهرت النتائج بانه لايوجد هنالك ارتباط بين الموقع متعدد الاشكال 819C/T ومرض السكري من النوع الاول (P=0.208) حسب الدلالات الاحصائية بين مجموعة المرضى والمجموعة الضابطه بينما اظهرت النتائج انه يوجد ارتباط وثيق بين الموقع متعدد الاشكال 1082G/A ومرض السكري من النوع الاول بين مجموعة المرضى والمجموعه الضابطه حسب الدلالات الاحصائية (P<0.0001), وهذه النتائج تشير الى وجود ارتباط وثيق بين الموقع متعدد الاشكال G/A 1082لجين انترلوكين (10) ومرض السكري من النوع الاول في المجتمع المصري.

Abstract

ABSTRACT:BACKGROUND: Ischemic heart disease is the main cause of death throughout the world. Cytokines are key regulatory glycoproteins . They are intimately associated with atherogenesis and modulate plaque morphology and stabilization. OBJECTIVE: To study the role of pro-inflammatory Interleukin-8 , and anti-inflammatory interleukin-10 in patients with Acute coronary syndromes (ACS)(acute myocardial infarction and unstable angina).METHODS: This study included (55) patients with acute coronary syndrome, (30) patients with acute Myocardial infarction ( 24 males and 6 female) and 25 patients with unstable Angina (11 males and 14 females). This project has been carried out on patients at the coronary care unit (CCU) of Baghdad teaching hospital/ Medical City/ Baghdad, during the period from April to June 2010. A twenty five, apparently healthy individuals were taken as a healthy control group. Venous blood sample was taken from each subject. Levels of interleukin-8 and Interleukin-10 were estimated by using enzyme linked immunosorbant assay . {ELISA-kit for detection of IL-8 (BioSource Europe S.A. /catalogue number; KAP1301)} and {ELISA-kit for detection of IL-10 (BioSource Europe S.A./ catalogue number; KAP1321)} RESULTS: Plasma level of pro-inflammatory cytokine (interleukin-8) showed a significant elevation in patients with acute myocardial infarction (Mean ± SD =95.26 pg/ml ± 102.89 ;P value=0.004) and unstable angina (Mean ± SD =146.43 pg/ml ± 154.45 ;P value=0.019) in comparison to control group (Mean ± SD =47.75 pg/ml ± 36.67). Serum level of anti-inflammatory cytokine (interleukin-10) showed a significant decrease in unstable angina patients (Mean ± SD =16.95 pg/ml ± 11.92 ;P value=0.012) in comparison to control group (Mean ± SD =30.29 pg/ml ± 21.17),however, it showed no significant differences among acute myocardial infarction patients (Mean ± SD =27.73 pg/ml ± 23.16 ;P value=0.500) in comparison to control group. The ratio of pro-/anti-inflammatory cytokines (interleukin-8 / interleukin-10) was significantly elevated in acute coronary syndrome; acute myocardial infarction and unstable angina patients (P=0.001, P=0.019, P=0.001 respectively) .CONCLUSION: Interleukin-8 is an important pro-inflammatory cytokine in coronary artery disease which may contribute to the plaque destabilization in acute coronary syndrome especially unstable angina, and low level of anti-inflammatory interleukin-10 which is considers as a risk factor of development of acute events in unstable angina

Abstract

Psoriasis is a multifactorial, chronic, inflammatory skin disease of unknown etiology, associated with cytokines and adipokines serum levels changes that may influence disease pathogenesis.To determine the effect of acitretin on serum IL-10, IL-23, adiponectin, Zn, Mg and copper levels in psoriasis patients.A case control study that include 50 psoriasis patients, conducted in Tikrit Teaching Hospital. They received acitretin orally for 4 weeks duration course. IL-10, IL-23, adiponectin, Zn, Cu and Mg in serum for all patients determined at the time of enrollement in the study and after 4 weeks of treatment with the drug. Acitretin reduced the mean PASI in 20 psoriatic patients from 20.97 before receiving treatment to 1.22 after receiving treatment (P=0.000). In addition, the mean serum levels for IL-10 reduced significantly (P=0.000) from (6.01±4.17) pg/ml before treatment to (1.85±0.54) pg/ml after treatment. Furthermore, Acitretin reduced the mean serum level for IL-23 from (72.08±34.14) pg/ml pre-treatment to (66.1±20.3) pg/ml, post-treatment (P=0.423). The mean serum levels of adiponectin in therapeutic group was lower (6.97±3.06) μg/ml than that of baseline value (8.9±3.96) μg/ml, but this result was statistically not significant (p=0.114). The mean serum levels of Zinc and copper in therapeutic group was lower than baseline value, but the difference was statistically not significant (p>0.05).

Abstract

Rheumatoid arthritis (RA) is a chronic inflammatory disease in which interleukin (IL)-10 gene plays an important role. There are, however, controversial reports that IL-10 promoter polymorphism may be an independent marker of susceptibility and severity of RA. The aim of the present study was to examine the IL-10 promoter polymorphism among RA patients. The aim of this study was to investigate the association between IL-10 G-1082A promoter polymorphism of interleukin-10 gene in some Iraqi rheumatoid arthritis patients using ARMS-PCR technique.We examined 70 RA patients (60 females and 10 males; aged: 20 – 74 years, mean: 51.7 years) were recruited from a private clinic in different region of Salahaddin governorate and 50 healthy individuals (40 females and 10 males; age: matching with patients) who were the staff of Tikrit University and hospital workers. The venous blood samples (5 ml) were collected from all the participants at the time of clinical examination, aliquoted in plain and EDTA vacutainers for the determination of erythrocyte sedimentation rate, C-reactive protein, presence of RFs. Genomic DNA was isolated from the whole-blood samples of all the patients and control subjects. The IL-10 -1082 G/A polymorphism genotyping Amplification Refractory Mutation System- Polymerase Chain Reaction (ARMS-PCR) method was used.ESR rate and RF positivity Levels of CRP were found as high as ≥ 20, 86.3% among patients, while all the healthy individuals were negative for this test except for a few number.The genotype GA for the −1082 G/A polymorphism of the IL-10 gene promoter was the most frequently observed genotype in both patients and controls, however, the distribution of the GG and AA genotype in patients and control was different. While genotype AA has a higher frequency than GG in patients (27.04vs.23.04), genotype GG was more frequently observed than AA in controls (46.08vs. 0.08).

يعد مرض التهاب المفاصل الرثوي من الأمراض الالتهابية المزمنة التي يلعب فيها جين الانترلوكين 10 (IL-10) دورا هاما كذلك أشارت بعض التقارير إلى أن التباين الوراثي للبروموتر الخاص بهذا الجين من الممكن اعتماده مؤشراً إلى حساسية وخطورة مرض التهاب المفاصل الرثوي.وتهدف هذه الدراسة الى امكانية تحديد العلاقة بين التباين الوراثي في الموقع (1082 GA) للبروموتر التابع للجين IL-10 ومرض التهاب المفاصل الرثوي في العراق بالاعتماد على تقنية ARMS-PCR.أجريت الدراسة على عينة تتكون من 70 مريضا إناثا وذكورا وبمعدل عمري 51.7 سنة من مناطق مختلفة لمحافظة صلاح الدين، و 50 شخصا إناثا وذكورا بوصفهم مجموعة سيطرة لها معدل الفئة العمرية ذاتها، لتحديد التباين الوراثي في الموقع 1082 GA للبروموتر التابع للجين IL-10 بالاعتماد على تقنية ARMS-PCR.وقد بينت نتائج الدراسة ارتفاعا ملحوظا في مستويات الـ ESR وRF و CRP ≥20 , 86.3 %. كذلك بينت الدراسة بان النمط الوراثي لتبين GA في الموقع 1082 التابع للبروموتر الخاص بالجين IL-10 gene اكثر تكرارا من بقية الانماط الوراثية لدى المرضى ومجموعة السيطرة وان تواجد الانماط الوراثية AA,GG كان مختلفاً لدى المرضى ومجموعة السيطرة اذ تبين بان النمط الوراثي AA كان تكراره لدى المرضى اعلى من الأصحاء (27.04vs.23.04) في حين النمط الوراثي GG كان تكراره لدى الأصحاء أعلى مما هو في المرضى (46.08 vs. 0.08).

Abstract

BACKGROUND: Myelofibrosis (MF) is largely documented by an abnormal cytokine expression, which in turn couldcontribute to bone marrow fibrosis, angiogenesis, and constitutional symptoms. To gain additional pathogeneticinsight regarding cytokine phenotype correlations in MF, this study estimated the level of interleukin‑10 (IL‑10)abnormality and Janus kinase (JAK2 V617F) mutation in primary MF.OBJECTIVE: The objective of this study was to assess serum IL‑10 level and it’s relation with the presence ofJAK2 V617F mutation in patients with MF.MATERIALS AND METHODS: JAK2 V617F mutation detection was performed in 32 patients with MF usingamplification refractory mutation screening‑polymerase chain reaction. IL‑10 level was estimated in 36 patientswith MF using enzyme‑linked immunosorbent assay technique compared to 27 healthy controls who wereenrolled for comparison.RESULTS: The study showed higher significant (P ≤ 0.002) increase of IL‑10 in patients with MF with cutoffvalue ≥8.9510 pg/ml and area under the curve value of 0.749 (P < 0.001) and also in patients with serum levelof IL‑10 more than 13.6 pg/ml characterized with significant lower white blood cell count, nonsignificant differencewith lower hemoglobin level, normal platelet count, and smaller size splenomegaly. About 59% of the studiedprimary MF (PMF) patients had JAK2 positive, 63% of them were male. There was no significant correlationbetween IL‑10 and JAK2.CONCLUSION: JAK2 mutation and IL‑10 as anti‑inflammatory cytokines may play a role in the pathogenesis andhematological presentation of patients with PMF. High IL‑10 level may predict good prognosis in patients with PMF.

The Role of Interleukin-10 in Women with Metastatic Invasive Ductal Carcinoma

Abstract

Background: Breast cancer is the malignant tumor that forms from the uncontrolled growth of abnormal breast cells. It usually affects tissues involved in milk production (Ductal and lobular tissues). It is the most common malignancy in women and it remains one of the greatest health threats facing women around the world as we enter the 21st century.Objectives: To estimate the role of IL-10 in the progression of invasive ductal carcinoma.Patients and Method: Seventy three metastatic invasive ductal carcinoma Iraqi women were admitted to Nuclear Medicine Hospital in Baghdad and 15 samples of apparently healthy women were involved as a control. The blood samples (2 mL) were drown from all studied cases in order to be used for measuring their serum level of IL-10 by using Enzyme-linked Immune Sorbent Assay technique.Result: The results have shown that the high significant relation was that of elevated IL-10 levels, which increased in 98.6% of patients.Conclusion: The result indicates that the highl significant elevation of serum IL-10 concentration are strongly asso¬ciated with metastatic invasive ductal carcinoma, this may be helpful in adding further vision in the diagnosis of women with this type of cancer.

يعد سرطان الثدي من اكثر انواع السرطانات التي تصيب النساء في العالم ، وقد تناولت الكثير من الدراسات والبحوث العوامل المسببة لهذا المرض من أجل التوصل للتشخيص المبكر والعلاج الناجح له. تناولت هذه الدراسة حصراً النوع الخبيث والأكثر انتشاراً لسرطان الثدي وهو " سرطان القنوات المنتشر" حيث تركزت على بحث العلاقة بين هذا السرطان وتركيز الانترليوكين العاشر ، كما تناولت العلاقة بين تركيز هذا الانترليوكين وكل من اعمار المريضات وحجم الورم وعدد الغدد اللمفاوية المصابة لدى مريضات سرطان الثدي العراقيات. اجريت هذه الدراسة على 73 مريضة ادخلن لمستشفى الطب والإشعاع الذري في بغداد ، بالإضافة الى 15 امرأة سليمة (سيطرة)، حيث تم سحب عينة دم (2 مل) من جميع الحالات (المرضى والسليمات) لغرض استخدامها في قياس تركيز الانترليوكين باستخدام تقنية (ELISA) . بينت النتائج إن للانترليوكين العاشر قد ارتفع في 98.6% من المريضات (عالي المعنوية), لكن مع الزيادة في العمر او حجم الورم او عدد الغدد اللمفاوية المصابة لدى المريضات فان تركيز هذا الانترليوكين لم يتأثر( غير معنوي).

Abstract

Background: Hepatitis G virus and GB virus C (GBV-C) RNA viruses that were independently identified in 1995, and were subsequently found to be two isolates of the same virus. Blood transfusion is the main risk factor for HGV transmission Immune-mediated mechanisms are believed to play an important pathogenic role in hepatitis G virus infection. Interleukin-1 (IL-1) plays an important role in the inflammatory process. implying that IL-1 may play a role in viral clearance and suggesting that IL-1 has direct antiviral activity, so that IL-1 induces novel antiviral pathways within a cell. Interleukin 10 ( IL-10 )is secreted by T helper-2 type cells (Th2) which may down regulate cell-mediated immune effecter mechanisms important in the host defense against intracellular pathogens.Objectives : This prospective study aimed to estimate interleukins 1 and 10 levels in acute hepatitis (G) in Iraqi patients with leukemiaMethods: This cross sectional study was carried out at the national center of hematology , almustansiriya University and department of microbiology in Baghdad medical college in Baghdad, Iraq from January till August 2012. The study involved 33 patients (19 males & 14 females) diagnosed with hepatitis G infection as having a positive IgM antibodies by ELISA technique .Serum IL-1 and IL-10 levels were measured in 33 patients with hepatitis G virus infection and 40 leukemia patients without HGV infections as a control group by ELISA technique .Results: Twenty three patients (70%) with hepatitis G infection had elevated serum IL-1 alpha levels (the normal detection level is below 5 pg/ml), the patient serum IL 1 alpha range 2-37 pg/ml, with a mean 27.813 and SD 32.765 ). There was also a high significant correlation between serum IL-1 alpha level and hepatitis G infection (p value= ± 0.004) when compared with the control group by the t –test Also serum IL-10 level was below the normal detection limit (5-20 pg/ml) in 20 (60%) patients .The 33 patients had serum IL-10 levels (range 0.6 - 9 pg/ml, with a mean 2.213 & SD± 4.745 ) . There was a significant correlation between serum IL-10 level & hepatitis G infection when compared with the control group (p<0.0386 by t- test),so the viral infection may suppress the T helper 2 cells by the gamma interferon . This elevation in IL1 alpha which is an inflammatory cytokine produced by antigen presenting cells , may suggest that IL-1 is an important interleukin to eradicate hepatitis G virus infection.Conclusions: This study showed that serum Th2 cytokines are suppressed (IL10 ) while serum inflammatory cytokine (IL 1 alpha ) is elevated in a proportion of patients with hepatitis G virus infection. However, the elevated IL 1 alpha cytokine levels may represent a systemic response and not as a result of increased local production within the liver.Keyword : Hepatitis G virus , interleukin 1, interleukin 10 .

: التهاب الكبد الفايروسي نوع (G) هو فايروس نوع RNA اكتشف عام 1995.ينتقل بواسطة الدم عادة والتفاعلات المناعية تلعب دور مهم في التهاب الكبد الفايروسي مع هذا الفايروس . مثلا الانترلوكين الاول يلعب دور مهم في التهاب الكبد الفايروسي ويلعب دور مهم في ازالة الفايروس من الجسم وممكن ان يعتبر بأن له دور مباشر ضد الفيروس داخل الخليه اما الانترلوكين العاشر الذي يفرز من الخلايا اللمفاويه المساعده النوع الثاني(Th2) وهو يلعب دور مباشر لإحباط المناعة المتخصصة بواسطة الخلايا التي تساعد على مكافحة الفايروس الهـــــدف : هدف هذه الدراسة هو لقياس الانترلوكين الاول والعاشر لدى المرضى المصابين بالتهاب الكبد الفايروسي نوع(G) طريقة العمــــــل : هذه الدراسة تم اجراؤها في المركز الوطني لإمراض الدم التابع للجامعة المستنصريه وفرع الاحياء المجهريه في جامعة بغداد كلية الطب من كانون الثاني الى اب في عام 2012 وشملت هذه الدراسة 33 مريض (19 ذكر و14 انثى) مصابون بمرض التهاب الكبد الفايروسي نوع G وذلك بتشخيص المستضاد IgM بواسطة ELISAتم قياس الانتلوكين الاول والعاشر في دم المرضى وعند 40 مريضا باللوكيميا ولكنهم غير مصابين بالتهاب الكبد الفايروسي (العينه ألمقارنه) بواسطة ELISA النتائــــــج : وجد ان 23 مريضا من اصل ثلاثة و ثلاثون مريضا مصابا بالتهاب الكبد الفايروسي نوع(G) (70%)عندهم ارتفاع في الانترلوكين الاول في الدم فوق الحد الطبيعي الذي هو اقل من (5Pg/ml) وكان الانترلوكين الاول لدى المرضى يتراوح بين(2-37pg/ml) وكان المعدل هو 27.813pg/ml وكان هناك علاقة احصائيه0.004p< بين الانترلوكين الاول عدى المرضى المصابين بالتهاب الكبد الفايروسي نوع(G) وعند مقارنتهم مع مجموعه المقارنه بواسطة فحصt- test بينما كان الانترلوكين العاشر اقل من الحد الطبيعي الذي هواقل من (5pg/ml) عند 20 مريضا (60%).وكان الانترلوكين العاشر لدى المرضى يتراوح بين (0.6-9pg/ml) وبمعدل2.213pg/ml وكان هناك علاقة احصائيه .0386p<0. بين معدل الانترلوكين العاشر لدى المرضى عند مقارنتهم مع مجموعه المقارنه مما يدل على ان الفايروس ربما يهبط عمل (Th2) بواسطة الانترفيرون نوع كاما وهذا الارتفاع بالانترلوكين الاول الذي هو من نوع الذي يسبب الالتهابات ربما مهم في عملية مكافحة الفايروسالخلاصــــــــة : هذه الدراسه ربما تدل على ان الانترلوكين العاشر يهبط اقل من الطبيعي بواسطة كبح عمل (Th2) وان الانترلوكين الاول يزداد الذي هو من النوع المسبب للالتهابات وهذا الارتفاع في الانترلوكين الاول هو ارتفاع مركزي وليس فقط في الكبد المفاتيح : الانترلوكين الاول ,الانترلوكين العاشر ,فيروس الكبد نوعG
