Interleukine-6 Level in Saliva of Patients with Chronic Periodontitis: A Case-Control Study

Abstract

Background:Periodontal diseases are infectious diseases in which periodontalpathogens trigger chronicinflammatory and immune responses. Interleukine-6 is a multifunctional cytokine playing a central role ininflammation and tissue injury.The aim of the study IS to determine the level of Interleukin-6(IL-6) in saliva of patientswith chronic periodontitis compared to healthy subjects.Materials and Methods:The total subjects of the present study is 60, divided into 3 groups; 20 patients with chronicperiodontitis with pocket depth(PD ≥4 mm)(group I), 20 patients with pocket depth(PD <4 mm) with clinicalattachment loss (group II), and 20 healthy controls with pocket probing depth (PPD ≤ 3 mm) without clinicalattachment loss (group III). Un-stimulated salivary sample was taken from each subject and was investigated for thepresence of Interleukine-6by using Enzyme‑linked Immunosorbent Assay (ELISA) technology.Results:Mean IL-6 levels in saliva in patients with chronic periodontitis (98.40 ± 18.44 ng/L)was significantly higher thanin controls (11.67 ± 3.32; p=0.001). Also a significant difference in IL-6 levels in saliva was observed between the PPD ≥4 mm and PPD < 4 mm groups and between PPD ≥ 4 mm and control groups, as well as statistically significantdifferences were observed between PPD < 4 mm and control groups (P < 0.001).Conclusion:The Interleukine-6 level in saliva can be considered as one of inflammatory biomarker indicators ofseverity of periodontitis

Abstract

Objectives: The aim of this study is to estimate the roles of interleukine-6 in breast cancer Iraqi women and to assess the correlation between the serum levels of IL-6 with the progression of disease stage. Method: Fifty patients with proved diagnosis of breast cancer by clinical examinations presented to the Al alwyah hospital for gynecological diseases and obstetrics /Breast cancer early detection clinic) in Baghdad. All were females and didn’t have received any chemo/or radiological treatment .also 20 sample of apparently healthy women were involved as a control. The blood samples (5ml) were drown from all of the studied cases in order to be used for measuring their serum level of (IL-6) by using Enzyme –Linked Immunosorbent Assay (ELISA) .Result: Serum (IL-6) level were increased with high significant difference in Breast cancer patients and showed significant correlation in all disease stages as compared to control group with the highest level in stage I patients group.Conclusion: serum level of (IL-6) is highly correlated with breast cancer and might have detecting and diagnostic usefulness .

يعد سرطان الثدي من أكثر أنواع السرطانات التي تصيب النساء في العالم ، وقد تناولت الكثير من الدراسات والبحوث العوامل المسببة لهذا المرض من أجل التوصل للتشخيص المبكر والعلاج الناجح له. تناولت هذه الدراسة بحث العلاقة بين هذا السرطان وتركيز الانترليوكين السادس كما تناولت العلاقة بين تركيز هذا الانترليوكين مع أعمار المريضات و مراحل تقدم المرض . أجريت هذه الدراسة على 50 مريضة شُخصن بسرطان الثدي واللاتي دخلن الى مستشفى العلوية التعليمي (جميعهن من الأناث)، بالأضافة الى مجموع 20 امرأة (سيطرة) وتضم 20 امرأة سليمة مع مراعاة فائتهن العمرية ، في الفترة مابين كانون الثاني 2011 الى أب 2011. تم سحب عينة (5 مللتر) من الدم الوريدي من 70 من الحالات الداخلة في الدراسة (المريضات والسليمات) لغرض استخدامه في الدراسة المناعية والتي يشمل قياس الانترليوكين السادس بواسطة تقنية ELISA . بينت النتائج ارتفاع في مستوى الانترليوكين السادس حيث كانت العلاقة عالية المعنوية مقارنة بمجموعة السيطرة .كما تبين وجود علاقة معنوية بين مستوى الانترليوكين السادس في جميع مراحل المرض مقارنة بمجموعة السيطرة مع تسجيل اعلى مستوى في المراحل الاولية . بينما كانت العلاقة غير معنوية بين مستوى هذا الانترليوكين و اعمار المرضى.

The Salivary Inflammatory Biomarkers (Interleukin-6, C - reactive protein) in Relation with Caries-Experience among a Group of 12 Year Old Obese Boys

Abstract

Background: Obesity and dental caries are multifactorial diseases related to poor eating habits and show a closerelationship with the sociodemographic characteristics of individuals presenting these diseases. This research aimedto investigate the severity of dental caries among group of obese boys aged 12 year in relation to salivary interleukin-6 (IL-6) and C-reactive protein (CRP) of unstimulated whole saliva in comparison with normal weighted boys of thesame age.Materials and Methods: The study group included 40 obese boys, with an age of 12 year. The control group included40 normal weighted boys of the same age. The total sample involved for nutritional status assessment using BodyMass Index specific for age and gender according to CDC growth chart (2000). The diagnosis and recording ofdental caries conducted by using (D1-4MFS and d1-4mfs) index according to the criteria of Muhlemann (1976). Thecollection of unstimulated whole saliva was performed under standardized condition. Salivary samples werechemically analyzed for measuring IL-6 and CRP.Results: The caries experience among study group was lower than that among control group for both dentitions, withsignificant differences for D2, ds and dmfs and highly significant difference for d4. Salivary inflammatory biomarkers(IL-6, hs-CRP) were slightly higher among study group compared with control group with no significant differencebetween them. Salivary IL-6 and hs-CRP were negatively correlated with dental caries of both dentitions amongstudy group with significant correlation between IL-6 and D2, while they were correlated positively with dental cariesof both dentitions among control group with highly significant correlation between IL-6 and D3 and significantcorrelation between IL-6 and d4.Conclusion: Obesity and dental caries are associated with increased levels of salivary interleukine-6 and C-reactiveprotein, this making both obesity and dental caries as a state of inflammation that exacerbating immune responsesin the body

Abstract

Summary:Background: Cytokines produced by inflammatory cells play a pivotal role in synovial inflammation and joint destruction in rheumatoid arthritis.Patients and Methods: The cytokine serum levels were measured by EASIA (Enzyme amplified sensitivity immunoassay) in sera from 50 RA patients, and 40 healthy donors. Cytokine levels were compared in different RA subpopulations (positive or negative rheumatoid factor (RF), long term or recent onset disease, high or low disease activity). In addition, the possible association with other demographic and clinical parameters (gender, age, etc) was also analyzed. Results: It was demonstrated that IL-2, IL-6 and IFN-δ levels were elevated in serum samples of RA patients as compared with apparently healthy controls. Maximum elevation of TNF-α was recorded in a few number of patient's sera. There were non significant differences between control and RA patient groups in serum TNF-α level. Conclusions: Assessing the serum IL-2, IL-6, IFN-δ and TNF-α levels may be helpful in the confirmation of the RA activity. Due to the chronic course of this disease, other inflammatory markers must be identified in order to provide early therapeutic strategies to these patients.Key words: Rheumatoid Arthritis, Interleukine-2, Interleukine-6, Tumor Necrosis Factor- α, Interferon- δ.
