Abstract

ABSTRACT:BACKGROUND:Recurrent aphthous ulcer (RAU) is a common oral disease. It is treated by a variety of agents. Lactic acid has been used in the treatment of many skin diseases.OBJECTIVE:To assess the therapeutic, prophylactic effectiveness and safety of 5% lactic acid mouthwash in management of patients with RAU.PATIENTS AND METHODS:This single blind placebo controlled therapeutic study where 40 patients with early onset RAU were recruited in Department of Dermatology -Baghdad Teaching Hospital from April 2005-April 2006. Patients ages ranged 20-60 (29.6 ±9.6) years and were divided randomly into 2 groups (20 patients for each one):Group A used 5% lactic acid mouthwash,5 ml twice daily. Group B was instructed to use distilled water as placebo in a similar way to Group A. Short term assessment of each patient of both groups was done to evaluate the therapeutic effect of drug by using oral clinical manifestation index (OCMI), mean size of largest diameter of ulcers. A long term assessment was done for each patient to evaluate the prophylactic effect of drug by using OCMI before, after 1, 2& 3 months of therapy.RESULTS:The mean of OCMI in group A started to decline directly after 4 days of therapy and to its lower level after 8 days, and it was statistically significant (P value <0.05).The change in mean of OCMI of group B after 4 and 8 days of therapy was also statistically significant (P value <0.05).The response rate after 4 days of therapy in Group A was 33.3% and in Group B was 6.44%, while the response rate after 8 days of therapy in Group A was 69.16% and in Group B was 44.98%. The difference in response rates after 4 and 8 days between groups A and B was statistically significant (P value <0.05).The mean size of ulcers in Group A started to decline directly after 4 days of therapy and to its lower level after 8 days of therapy. It was statistically significant (P value <0.05). The change in mean size of ulcers of group B after 4 and 8 days of therapy was also statistically significant (P value <0.05). The response rate after 4 days of therapy in group A was 44.92% and in Group B was 15.08%, while the response rate after 8 days of therapy in group A was 85.68% and in group B was 55.36%. The difference in response rates after 4 & 8 days between Groups A and B was statistically significant (P value <0.05). The therapeutic action of lactic acid mouth wash was statistically significant more effective than placebo. Lactic acid mouth wash showed statistically significant prophylactic effect in comparison to placebo after 1,2&3 months from starting therapy using oral clinical manifestation index, (P value <0.05).CONCLUSION:Lactic acid 5% mouthwash is a new, safe effective therapeutic and prophylactic remedy in management of RAU.

Abstract

Lactic acid is an important carboxylic acid used in different aspects of life. In thisstudy a pure isolate of homofermentative lactic acid bacteria Enterococcus faecium was isolated , and characterized by morphological examinations with physiological andbiochemical tests.The present results have revealed the ability of the isolate to produce L- lactic acid in high concentration and low concentration of D- lactic acid in contrast with control. Lactic acid has been extracted and partially purified by Amberlite IRA400 resin column, the obtained extracted parts have been tested by carboxylic acid test in addition to High Performance Liquid Chromatography (HPLC) analysis which has showed that the concentration of L-lactic acid was 0.659g/l, while D-lactic acid was 0.174g/l in crude parts with two peaks appeared, one of area 322854 for L while no visible peak appeared for D because of small area 72214 in the figure, whereas the second peak of area 203238 may returned to the interference of D and L-lactic acid . Purified parts have reached to a purity of 72.3% for L-lactic and 38.2% for D-lactic acid also with one sharp peak for L with area of 43639 and no visible peak for D-lactic due to small area of 16713.

Abstract

Abstract

AbstractBackground: There are many modalities used in treatment of AA. However, there is no universally accepted therapy. Lactic acid has been tried in diversity of skin diseases such as warts, xerosis, wrinkles, chemical peeling and repigmentation of vitiligo. Objective: The present work has been arranged to evaluate the efficacy and tolerability of topical 15% lactic acid solution in the treatment of patchy alopecia areata.Patients & Methods: A study was designed as a single blind therapeutic trial. Twenty nine patients suffering from 59 patches of alopecia areata were included in the study. Their ages ranged from 1.5-61 years with a mean ±SD of 23.88 ± 14.3 years. Patches were divided into three groups. First group (22 patches) were treated with topical 15% lactic acid solution twice weekly. The second group (17 patches) were treated with topical 15% lactic acid solution thrice weekly. The third group (20 patches) were randomly selected from patients with multiple patches; they were treated with tap water for comparison. Patients with alopecia totalis, alopecia universalis and treated cases were excluded from the study. Patients were regularly evaluated for hair regrowth and side effects every 2 weeks for 3 months. Results: In the first group, the final hair regrowth assessment revealed hair regrowth of 68% of the treated patches .In the second group, 82.4% of the treated patches showed complete hair regrowth. In the control group, the final hair regrowth assessment showed 30% of the patches with hair growth. After four weeks of therapy, erythema with mild scaling was noticed in 90% of treated patches of the first group and in 100% of patches in the second groups. These features disappeared completely at the end of treatment.Conclusion: Topical 15% lactic acid solution is an effective, safe and tolerable method of treating patchy alopecia areata with minimal side effects and cost benefit therapy. Thrice weekly application of topical 15% lactic acid solution achieved better rate of hair regrowth in a shorter period than twice weekly application.

الملخص:المقدمة: هنالك طرق علاجية متعددة لعلاج مرضى الحاصة البقعية. و لكن لا يوجد علاج موحد لهذا المرض. محلول حامض اللابنيك استخدم في امراض جلدية متعددة مثل الثئاليل, جفاف الجلد, التجاعيد الجلدية, القشط الكيميائي و البهق.الغرض من الدراسة: لقياس فاعلية و تقبل محلول حامض اللبنيك 15% الموضعي لعلاج المرضى المصابين بداء الحاصة البقعية.المرضى و طرق العمل: هذه الدراسة تمثل دراسة علاجية مفردة. تسعة و عشرون مريضاَ مصاباَ بداء الحاصة البقعية (داء الثعلب) تم ادخالهم في هذه الدراسة، 22 ذكرا و 7 اناث, تراوحت أعمارهم بين1.5-61 سنة و بمعدل 23.88 سنة ±14.28 سنة. كانوا من المرضى المصابين بالمرض لفترة لا تتجاوز الثلاثة اشهر ممن لم يسبق لهم العلاج. طريقة العلاج: تضمنت وضع محلول حامض اللابنيك 15% الموضعي على المناطق المصابة بداء الحاصة البقعية مرتين اسبوعيا للمجموعة الاولى و ثلاث مرات للمجموعة الثانية. المرضى المصابين باكثر من بقعة مرضية اخذت واحدة كمجموعة ثالثة وضعت الماء المجرد كمجموعة مقارنة. بلغ عدد البقع الداخلة في البحث تسعة و خمسين بقعة مصابة بداء الثعلب.النتائج: أظهرت ان (82%) من البقع التي عولجت بحامض اللابنيك ثلاث مرات اسبوعيا و (68%) من البقع التي عولجت بحامض اللابنيك مرتين اسبوعيا تحسناَ واضحاَ في مرضهم بينما (30%) فقط من البقع في المجموعة المقارنة اظهرت نموا للشعر. الاعراض الجانبية التي لوحظت اثناء فترة الدراسة كانت قليلة وبسيطة مثل الحكة و الاحمرارالجلدي لكن هذه الاعراض اختفت بصورة تلقائية بدون الحاجة الى ايقاف العلاج.الاستنتاج: أمكن لنا أن نستنتج من هذه الدراسة ان استخدام محلول حامض اللابنيك 15% الموضعي هوعلاج فعال لمعالجة داء الحاصة البقعية ويمكن استخدامه كبديل للعلاجات الاخرى المستخدمة في علاج هذا المرض وهي طريقة سهلة، بسيطة، وذات تكلفة منخفضة ويمكن للمريض استخدامها في البيت دون الحاجة للقدوم الى المستشفى لمرات عديدة .

Abstract

The present study aimed to determine the antibacterial activity of lactic acid bacteria metabolites against Listeria monocytogenes which was isolated from some foods in local markets of Erbil and Koya cities; 225 food samples were examined for the presence of Listeria species particularly L. monocytogenes from October 2010 to March 2011. The studied samples included 70 samples of raw chicken meat, 55 samples of raw milk, 50 samples of cheese and 50 samples of raw red meat. According to motility test, hemolysin production, sugar fermentation test, 8 isolates of L. monocytogenes were identified, 1 (2 %) isolate from cheese, 2 (4 %) isolates from red meat and 5 (7.3 %) isolates from chicken meat. The percentages of contaminated foods for Listeria species from chicken meat, raw milk, cheese and red meat were 20, 9, 20 and 22% respectively. With respect to the biocontrolling attempt, the effect of cell free extract (CFE) of lactic acid bacteria (LBA) which is considered as safe against isolated L. monocytogene was used as biocontrolling agent. The CFE of lactic acid bacteria which isolated from local dairy products, was tested for its controlling ability by agar diffusion method, the CFE showed inhibition zone from (15-23 mm) in diameter. Also the minimum inhibitory concentration MIC of CFE was determined at three additive percentages (0.5, 1.0 and 1.5ml to 10 ml of nutrient broth), the inhibition percentage ranged from (28.75 - 48.97) at (0.5 %), (52.13-95.55) at (1 %) and (82.93 -98.15) at (1.5 %) additive percentage of CFE of LAB.

هدفت الدراسة الحالية إلى تحديد الفعالية التثبيطية للنواتج الايضية لبكتريا حامض الاكتيك ضد بكتريا Listeria monocytogen المعزولة من بعض الاغذية المحلية المتواجدة في اسواق مدينة اربيل وكويه، جمعت 225 عينة من نماذج غذائيه مختلفة بشكل عشوائي من الاسواق المحلية. تَضمّنتْ العيناتُ المدروسةُ 70 عيّنةِ من لحمِ الدجاجِ الخامِ، 55 عيّنةِ من الحليبِ الخامِ، 50 عيّنةِ من الجبنِ و50 عيّنةِ من اللحومِ الحمراءِ الخامِ. فُحِصتْ ِجميع العينات الخاضعة في الدراسة الحالية لمدى تلوثها بجرثومة اللستيريا وخصوصا (Listeria monocytogens). طبقاً لإختبارات حركة جرثومة L. monocytogens و انتاجها لانزيم الحال للدم وقدرتها على تخمير السكريات تم تشخيص 8 عزلات للجرثومة، بمعدل عزلة واحده وبنسبة 2 % من عينات الجبن، وعزلتين وبنسبة 4 % من عينات مِنْ اللحمِ الأحمرِ، وخمسة عزلات بنسبة 7.3 % من لحم الدجاج. تراوحت نسب التلوث لعينات لحمِ الدجاجِ، حليب خام، جبـن ولحم أحمر بجرثومة Listeria 20، 9، 20 و 22 % على التوالي اما بخصوص دراسة السيطرة الحيوية biocontrolling على نمو وبقاء (Listeria monocytogenes)، اذ استخدم رواشح المزرعة البكتيرية CFE لبكتيريا حامض اللبنيك LAB والتي تعتبر آمنة وطبيعية، ضدّ عزلات (Listeria monocytogenes) في السيطرة الحيوية. اذ استخلص الراشح من بكتيريا حامض اللبنيك المعزولة من بعض منتجات الالبان المحلية. تم اختبار قدرة التثبيط الحيوي لـ LAB بطريقة الانتشار بالحفر والتي تم فيها تركيز الراشح الى ضعفين. اذ أظهرت نتيجة الاختبار هذا ان الراشح المستخلص من LAB القدرة على تثبيط النمؤ البكتيري، اذ تراوح قطر منطقة التثبيط (15-23 مليمتر). حدد التركيز الادنى المثبط CFE باستخدام ثلاث نسب من الاضافات (0.5, 1.0 و1.5 %) لـ CFE، اذ اظهر الراشح نسبة تثبيط (28.75 - 48.97) عند نسبة الاضافة (0.5 %)، (95.55 - 52.13) عند نسبة الاضافة (1 %) و(98.15 - 82.93) عند نسبة الاضافة (1.5 %).

Abstract

Lactic acid was used as difunctional spacer could reacted with glycerol gave its corresponding ester derivative with remain the hydroxyl groups of lactate units which reacted with carboxyl group of Mefenamic acid forming ester attachment, the prepared pro drug have many development, it could enhanced the drug with easier hydrolysis through ester-ester groups with extended the arm pendant substituted drug. The action of polymeric drugs depends on hydrolytic and cleavage of the drug moiety from the polymer, this give advantages of late and sustained release of drug over long time with decrease of side effects, the modification percentage test was carried out, also it was characterized by FTIR and 1H-NMR spectroscopes. Controlled drug release was deliberate by using different pH values at 37℃. Thermal stability of the drug polymer was observed, indicated the protection of drug with longer expire date. Abstract

Lactic acid discovered at 1780 and its had many application in food industries such as acidulant ,preservative and material for organic synthesis. Commercial Lactobacillus delbrueckii isolate of has been used in the production of lactic acid from sweet bovine whey supplemented with local renewable resources The study used cane sugar molasses and date waste juice (5,10,15,20,25)% (w/v) as carbon sources and yeast extract ,urea ,ammonium salphate percentage as nitrogen sources (0.2 , 0.4 , 0.6 , 0.8 ) % (w/v) . the study showed: * The commercial isolate was ferment the un supplemented whey during 48 hr . at 45c and pH 6 , its lactic acid production as 3.5 gm/100 ml whey with conversion efficiency 18.6%. * When the molasses was added (5, 10, 15, 20, 25)% (w/v) the study showed increased lactic acid amount to (4.7, 5.8, 6.9, 18.5, 33.9)gm/100 ml whey with conversion efficiency (44.7, 19.3, 22.5, 28.3, 81.5) % respectively.*On the other hand our study showed the addition of date waste juice (5,10,15,20,25) % (w/v) increased lactic acid amount as (4.8, 6.7, 10.7, 20.7, 29.5) gm /100 ml whey with conversion efficiency (43.7, 22.8, 20.8, 44.7, 52.8) % respectively.*The generally treatment (whey and 25% molasses) was selected as a fermentation medium for the production of lactic acid it is also supplemented by different nitrogen sources, the results showed:*The addition of ammonium salphate (0.2, 0.4, 0.6, 0.8) % (w/v) increased lactic acid amount to (4.8, 5.3, 5.5, 5) gm /100 ml whey with conversion efficiency (28.3, 75.73, 66.5, 71.42) % respectively. and The addition of urea as nitrogen source did not exert any change in the amount of lactic acid product. While The addition of yeast extract (0.2, 0.4, 0.6, 0.8) % (w/v) the result illustrated increased lactic acid amount was (4.9, 6.5, 21.7, 31.9) gm /100 ml whey with conversion efficiency (71.22, 66.77, 86.55, 89.65) % respectively. On the other hand molasses 25% and whey +0.8%yeast extract was selected as best medium for lactic acid production.

اكتشف حامض اللاكتيك عام 1780 وله استعمالات عديدة في الصناعات الغذائية منها مادة محمضة وحافظة ومادة تفاعل للتخليق العضوي واستخدمت عزلة تجارية من بكتريا Lactobacillus delbrueckii لانتاج حامض اللاكتيك من شرش الابقار الحلو والذي تم دعمه بمصادر متجددة محلية ، اذ استخدم مولاس قصب السكر وعصير حثالة التمر كمصدر للكاربون بنسب (5 ،10 ، 15 ،20 ،25 ) % (وزن/حجم) في حين دعم وسط التخمر بكل من خلاصة الخميرة واليوريا وكبريتات الامونيوم بنسب (0,2، 0,4 ، 0,6، 0,8 ) % (وزن/حجم) كمصدر للنتروجين حيث•تمكنت العزلة للبكتريا من تخمير الشرش غير المدعم خلال 48 ساعة عند درجة حرارة 45م ورقم هيدروجيني 6 وكانت كمية حامض اللاكتيك الناتجة 3,5 غم /100مل شرش وبكفاءة تحويل بلغت 18,6 % .•ادى اضافة المولاس بنسب (5 ، 10 ، 15 ، 15، 20 ، 25 ) % (وزن/حجم ) الى زيادة كمية الحامض المنتجة وكانت كمية حامض اللاكتيك (4.7 ، 5.8 ،6.9 ، 18.5 ،33.9 ) غم/100 مل شرش وبكفاءة تحويل بلغت ( 44.7 ، 19.3 ، 22.5 ،28.3 ، 81.5 ) % على التوالي .•كما اوضح اضافة عصير حثالة التمر بنسب (5 ، 10 ، 15 ، 20 ،25 ) % (وزن/حجم ) الى زيادة كمية الحامض وكانت (4.8 ، 6.7 ، 10.7 ،20.7 ،29.5 ) غم/100مل شرش وبكفاءة تحويل بلغت (43.7 ، 22.8 ، 20.8 ، 44.7 ، 52.8 ) % على التوالي .•انتخبت المعاملة (شرش +25 % مولاس ) كوسط تخميري لانتاج حامض اللاكتيك ودعم بمصادر نتروجينية مختلفة حيث بينت النتائج بأن :• اضافة كبريتات الامونيوم بنسب (0.2 ، 0.4 ، 0.6 ، 0.8 ) % (وزن/حجم ) الى زيادة كمية الحامض وكانت (4.8 ،5.3 ، 5.5 ،5 ) غم/100مل شرش وبكفاءة تحويل بلغت (28.3 ، 75.73 ، 66.5 ، 71.42) % على التوالي .•كما اوضح بأن اضافة اليوريا كمصدر نتروجيني الى زيادة ملحوظة في انتاج حامض اللاكتيك • كما اوضح بأن اضافة خلاصة الخميرة الى زيادة كمية الحامض وكانت ( 4.9 ، 6.5 ، 21.7 ، 31.9 ) غم/100 مل شرش وبكفاءة تحويل بلغت (71.22 ، 66.77 ، 86.55 ، 89.65 ) % على التوالي .•بين الوسط المكون من الشرش + (مولاس 25 +0.8 خلاصة خميرة ) % بانه الوسط المثالي لانتاج حامض اللاكتيك من بكتريا Lb. delbrueckii

Abstract

Bacteriocins are ribosomal synthesized antimicrobial peptides produced by one bacterium that are active against other bacteria, either in the same species (narrow spectrum), or across genera (broad spectrum).Thirty isolates of Lactic acid bacteria obtained from fermented milk and Yoghurt from various locations in Alnajaf were analyzed by PCR to rapid screening of pediocin, plantaricin and enterocin genes that present on the bacterial chromosome or on plasmidsMultiplex PCR method was used to detect the presence of bacteriocin gene by using specific primers to amplify a fragment from bacteriocin structural gene. Eighteen strains produced one of the PCR fragments with specific primers. Highest frequency of occurrence 11(61.1%) of isolates produced fragment of 428 bp indicate to presence of plantaricin gene, 5 (27.8%) produced fragment of 332 bp indicate to presence of pediocin gene whereas 2(11.1%) produced fragments of 412 bp indicate to presence of enterocin gene. The agar well diffusion methods was used to detect the selected strains with antibacterial activities against P. aeruginosa . The supernatant of the thirty LAB screened for bacteriocin production, eighteen (60%) were gave inhibition zones (12-20mm) onto the indicator pathogenic strain. Among LAB isolates, strain M5 plantaricin producer the most effective antibacterial compounds against P. aeruginosa. (20 mm as diameter of inhibition). The LAB isolates were: Lactobacillus plantarum, Pediococcus and Enterococcus. L. plantarum had the highest frequency of occurrence 11(61.1%), while, Pediococcus and Enterococcus had 5 (27.8%) and 2(11.1%) respectively.

البكتريوسين هي ببتيدات رايبوسومية الصنع لها فعالية ضد الاحياء المجهرية تنتج من قبل بكتريا ولها تاثير فعال ضد بكتريا اخرى من نفس النوع (فعالية محددة ) او ضد اجناس اخرى (فعالية واسعة) .تم الحصول على ثلاثين عزلة من بكتريا حامض اللاكتيك من اللبن المتخمر والمعلب من مناطق متعددة في محافظة النجف .اجريت غربلة سريعة لمعرفة قابلية العزلات على انتاج البكتريوسين pediocin و plantaricinو enterocin من خلال الكشف عن جينات هذه البكتريوسنات باستعمال تقنية تفاعل البلمرة المتسلسل مع بادئات خاصة بها .اظهرت النتائج ان ثمانية عشر فقط من العزلات المدروسة اظهرت احدى الحزم الخاصة .اذ كانت 11 (61.1%) عزلة اظهرت حزمة بطول 428 زوج قاعدي والتي تشير الى وجود جين ال plantaricin وهي النسبة الاعلى .و 5(27.8 %) منها اظهرت حزم بطول 332 زوج قاعدي والتي تشير الى وجود جين ال pediocin . فيما كان 2 (11.1 %) منها اظهرت حزمة بطول 412 زوج قاعدي مما تشير الى وجود جين ال enterocin .كما استعملت طريقة الانتشار بحفر الاكار لتاكيد فعالية البكتريوسينات المنتجة من بكتريا حامض اللاكتيك ضد بكتريا ممرضة Pseudomonas aeruginosa

Abstract

Several approaches have been taken in the prevention of dental caries; the main approach is to render enamel surface more resistance to attack of acidogenic bacteria. In this study one hundred dental plague samples with 20-30 mg of plagues were collected. These patient's samples were collected during March to May 2014 from Baghdad dental clinics. Culturing of dental plagues was inoculated on different selective, enrichment and diagnostic media to be obtain pure culture of Strep. mutans. Streptococcus mutans is considered the main cause of destruction of dental tissues, with the incidence 13%. The inhibitory effect of Lactobacillus gasseri against Streptococcus mutans isolates was studied by well-diffusion method on solid media. The results showed that the Lactobacillus gasseri has a highly effects of inhibition zone on the Strep. mutans growth (92.30%) and the inhibition diameters zone were more than 12 mm. Also this effect compared with other antibacterial drugs, which were showed that the isolates had different responses of resistance to Ampicillin (AM) 5(38.4%), Azithromcin (AZM) and Lincomycin (LN) 2(15.4%) for each, Gentamycin (CN) 4(30.7%), so that the Cefotaxim (CTX) antibacterial recorded lowest resistance 1(7.6). Hence, this study reveals the potential of Lactobacillus gasseri as an alternative and complementary treatment medicine for dental caries by inhibiting the growth of Streptococcus mutans. Also these results may provide a basis for alternative therapies to prevent or treatment of dental caries.

Abstract

The first section covered the introduction of the research and the researcher discussed the importance of lactic acid in the effectiveness of running 800 meters as well as interest in physical qualities of the game. The researcher used the descriptive method on a subject of (5) Iraqi team runners of the applicants representing the in the event ran 800 meters and was presented in this section the mechanism of work and how the variables were chosen and the method of conducting the main experiment, The researcher concluded with a set of conclusions and recommendations presented in the fifth section. The most important conclusion is that endurance exercises according to the Cosmin test followed by the 800 meter walkers need to reconsider the improvement of operations chemical in muscle cells to increase the ability of the racers to produce and carry lactic acid and continuously take into account the principle of privacy in endurance exercises, and the most important recommendations must be adopted sequential methodology when evaluating the endurance exercises for those running 800 meters.

شمل الباب الاول على مقدمة البحث وتطرق فيها الباحث الى اهمية حامض اللاكتيك في فعالية ركض 800 متر فضلا عن الاهتمام بالصفات البدنية الخاصة باللعبة اما الباب الثاني شمل مجموعة من المواضيع التي تخص فهموم حامض اللاكتيك واهميتة في ركض المسافات المتوسطة وشمل الباب الثالث منهجية البحث إذ اعتمد الباحث المنهج الوصفي وكانت عينة البحث قد اختيرت بالطريقة العمدية البالغة (5) عدائين من المتقدمين يمثلون منتخب العراق في فعالية ركض 800 متر وتم في هذا الباب عرض الية العمل وكيف تم اختيار المتغيرات وطريقة اجراء التجربة الرئيسة اما الباب الرابع تم فيه عرض النتائج ومناقشتها في ضوء ما حصل عليه الباحث من درجات وبعدها توصل الباحث الى مجموعة من الاستنتاجات والتوصيات تم عرضها في الباب الخامس وكانت من اهم الاستنتاجات هو أن تدريبات التحمل على وفق أختبار كوسمين التي يتبعها مدربو راكضي مسافة (800) متر تحتاج إلى أعادة النظر في تحسين العمليات الكيميائية داخل الخلايا العضلية لزيادة قدرة الراكضين على انتاج وتحمل حامض اللاكتيك وبصورة مستمرة تراعي مبدأ الخصوصية في تدريبات التحمل، ومن اهم التوصيات لابد من إعتماد المنهجية التتبعية عند تقييم تدريبات التحمل لكلٍ من راكضي فعاليتي (800) متر
