A Study of Anticancer Activity for Partial Purified Urease Isolated from Lagonychium farctum Plant

Abstract

This study included two experiments, the first of which included the investigation of the urease enzyme in the plant extract of the Lagonychium farctum, which included the extracts of the seeds, leaves and stems the plant. The results, showed that the highest enzymatic and quality efficacy of urease was in seed extract of the Lagoonchium farctum. The optimal conditions for enzyme extraction were determined using different solution, pH and temperature. The Potassium Phosphate Solution at 0.02 μl pH 8 was the homogenous enzyme extract, with an enzymatic concentration of 3.195 U/ml. The optimum reaction time for enzyme extraction was 30 min, giving an enzyme activity of 3.274 U / ml. The optimum reaction time for enzyme extraction was 30 minutes where it gave enzymatic efficacy of 3.274 U/ml. The purification was by several steps including ammonium sulphate deposition with different precipitation ratios. The optimum saturation was 50 %, followed by ionic exchange on the DEAE-cellulose column and gel filtration on the Sephadex G-200 column. The purification time was 29.6 and the enzyme yield was 75 %. The second experiment included study of the effectiveness of the enzyme Urease as an Anticancer activity. Results showed that the urease enzyme had an inhibitory effect on the breast cancer line MCF7; it has been shown to be effective in the direction of breast cancer when the concentration of the partially purified enzyme is increased while in contrast to the natural cell line WRL68.

Abstract

This study aimed to investigate the effect of alkaloids extracted from Equisetum arvense on the proliferation of lymphocytes, breast cancer cell line (MCF7) and mice embryo fibroblast normal cell line (MEF) by exposing them for different concentrations for a period of 24 hours. The results showed that in the absence of phytohaemagglutinin (PHA), the alkaloid extract did not stimulate the division of human lymphocytes at all concentrations used. In contrast in the presence of PHA, the alkaloid extract decreased the division of human lymphocytes at all concentrations used. This decrease was significant compared with the control group, which gave a division ratio of 4.20%. The effect of alkaloid extract on the growth of MCF7 cancer cell line demonstrated that the lowest inhibition rate 20.07% was at the concentration 15.1 µgml, and the higher inhibition rate was 64.1% at the concentration 500 µgml. While in the MEF normal cell line, the higher inhibition rate was 11.3% at the concentration 500 µgml. In conclusion, this study showed that the alkaloid extract of E. arvense plant affects the growth of MCF7 cancer cell line and have a lower effect on the growth of lymphocytes and MEF normal cell line at high concentrations, making this plant promising candidate for the treatment of cancer but this needs more studies to prove that.

هدفت الدراسة إلى اختبار تأثير القلويدات المستخلصة من نبات ذنب الخيل Equisetum arvense على الخلايا اللمفاوية للانسان وخط خلايا سرطان الثدي البشري MCF7والخط الخلوي الطبيعي للخلايا الليفية في جنين الفأر MEF باستخدام سبعة تراكيز ولمدة تعريض 24 ساعة. اوضحت نتائج دراسة تأثير المستخلص القلويدي على انقسام الخلايا اللمفاوية في دم الانسان بدون المادة المشطرة PHA عدم وجود أي تحفيز لأنقسام الخلايا اللمفاوية ولجميع التراكيز المستخدمة بينما اظهرت نتائج دراسة تأثير المستخلص القلويدي على انقسام الخلايا اللمفاوية في دم الانسان بوجود المادة المشطرة PHA ان المستخلص القلويدي لنبات ذنب الخيل E. arvense قد سبب انخفاض في نسبة انقسام الخلايا اللمفاوية في التراكيز المستخدمة جميعها وكان هذا الانخفاض معنويا p ≤ 0.05)) مقارنة مع مجموعة السيطرة التي اعطت نسبة انقسام بلغت 4.20%. بينما اظهرت نتائج تأثير المستخلص القلويدي على نمو الخط الخلوي السرطاني MCF7 أن اقل نسبة تثبيط كانت %20.07 بتركيز 15.1 مكغم / مل واعلى نسبة تثبيط 64.1 % بتركيز 500 مكغم / مل. اما عند الخط الخلوي الطبيعي فكانت اعلى نسبة تثبيط 11.3% بتركيز 500 مكغم/مل. الاستنتاج: أوضحت هذه الدراسة بأن للمستخلص القلويدي لنبات ذنب الخيل تاثير عالي على خط خلايا سرطان الثدي MCF7 مقارنة مع الخلايا اللمفاوية للأنسان والخط الخلوي الطبيعي لجنين الفأر MEF في التراكيز العالية مما يجعل لهذا النبات دورا واعدا في علاج السرطان.

Abstract

Statins are effective to lower the cholesterol level and protect against cardiovascular disease. Recent studies showed that statins have pleiotropic effect and can be used to treat many types of diseases like neurodegenerative disorders, stroke, and cancer. Statins inhibit cell proliferation by suppression of mevalonate pathway leading to desregu-lation of cell signal transduction of some membrane receptor protein which is important for gene transcription. This study was done to evaluate the anti-proliferative activity of simvastatin and atorvastatin on MCF7 cell line alone and in combination with anathracycline chemotherapy drug (doxo-rubicin) using MTT assay. The results showed that simvastatin and atorvastatin had significant anti-proliferative effect on MCF7 cell line in dose-dependent manner with an IC50 10.10 μM and 12.3 μM respect-tively. Moreover, the combination of atorvastatin and simvastatin with (1 μM) doxorubicin had higher cytotoxic effect with an IC50 = 0.07 μM and 0.05μM respectively than doxoru-bicin alone IC50 = 1.9 μM.In conclusion, simvastatin and atorvastatin had anti-proliferative effect on MCF7 cell line and displayed significant synergism with doxorubicin which will help in enhancing efficacy of doxorubicin and decrease the adverse effect.

الستاتين وهي عقاقير تستعمل لخفض مستوى الكولسترول والحمايه ضد امراض القلب والاوعيه الدمويه . وقد اظهرت الدراسات السريريه وقبل السريريه ان الستاتينات لها تاثير عديد المظاهر ويمكن استخدامها لعلاج العديد من انواع الامراض مثل الاضطرابات العصبيه , السكته الدماغيه , والاتهابات البكتريه والسرطان.تمنع الستاتينات تكاثر الخلايا عن طريق قمع مسار الميفالونات مما يؤدي الى التاثير على تنظيم نقل الاشارات الخلويه لبعض البروتينات وهو امر مهم لنسخ الجين.وقد اجريت هذه الدراسه لتقييم النشاط المضاد للتكاثر لكل من السيمفاستاتين والاتورفاستاتين على خط خلايا
