Effect of Nigella Sativa (Black seed) on the serum lipids of healthy individuals

Abstract

Black seed (Nigella Sativa) is a common dietary element and has bean traditionally used as a treatment for a variety of health complains over centuries. Dyslipidaemia is an important and common medical problem.Objective: To assess the effect of N. sativa on the serum lipids of normal individuals.Design: Prospective study.Setting: Domain of the College of Medicine and IbnSina Teaching Hospital.Patients and methods: 10 healthy volunteers (8 males and 2 females) received 750 mg of powdered grains of Nigella sativa enclosed in a capsule twice daily for 28 days. Their fasting lipid profile, including triglycerides, total, LDL and HDL cholesterol were measured, before and after treatment.Results: Serum triglycerides, total and LDL cholesterol decreased significantly after treatment with Nigella sativa, P. value <0.0001. While HDL cholesterol increased significantly, P. value <0.0001.Conclusion: N. sativa exerts a favorable effect on lipid profile of healthy individual.

Abstract

Back ground: Today’s world is increasingly seeking ways to replace the synthetic drugs with the therapeutic power ofnatural products to decrease the percentage of many side effect which result from conventional treatment; one ofthese products was Nigella sativa (NgS) which was used so extensively that it became known as the seed of blessing“Habbatul Barakah” due to its powerful healing qualities for many ailments. The aim of this study was to evaluate thetherapeutic effect of Nigella sativa (powder and oil) on the healing process of extracted teeth sockets.Materials and Methods: The sample of our study consist of Forty eight rabbits to extract there upper two centralincisors under general anesthesia. The left side filled once with Nigella sativa powder and once with Nigella sativa oilmaterial, and the right side left for normal healing as a control group. The two sockets were sutured. The results werestudied radiographically and histologically after 1,2,4,6 weeks postoperatively. The radiographic examination wasperformed by using parallel technique in a digital radiographic examination and histological examination wasperformed under light microscope for the section stained with heamatoxiline and eosin.Results: Radiographically we found that NgS powder showed more radiopacity with complete disappearance oflamina dura in 6 weeks duration compared with NgS oil and control groups, while histologically we found that theNigella sativa (NgS) groups (powder and oil) illustrate an early apposition of osteoid tissue in 1st week duration withnumerous osteoblast and osteocyte in comparision to control group.In six weeks duration well developed bone filledall the portions of the socket in treated socket with NS powder with obvious complete epithelization of socket surfaceConclusion: Nigella sativa (powder or oil) seems to be bioactive materials that enhance differentiation andproliferation of progenitor cells to specialized bone formative cells, with no signs of inflammation

Abstract

Background and purpose of study: Paracetamol when used in very high dose ,can cause heavy damage in liver in man and animals. Nigella sativa has been reported to have antioxidant and hepatoprotective properties. In this study, the effective oral administration of the crude oil of Nigella sativa 0.3 ml ,has been investigated in mice following intra peritoneal of low toxic dose of paracetamol (300 mg/kg B. W.) and measured of selective parameters indicative of liver function (serum GOT, GPT and total protein). Material and Method :20 adult male albino mice Balb/ C they were randomly divided into 4 groups .The first group injected intra peritoneal with low toxic dose of paracetamol 300mg/ kg body weight then administration orally with 0.3 ml of normal saline (0.9 %w/v).The second group was the control injection intra peritoneal with 0.3 ml of normal saline then administration orally with 0.3 ml of normal saline (0.9 %w/v) .Third group administration orally with 0.3 ml of crude oil of Nigella sativa then injected intra peritoneal with 0.3 ml of normal saline (0.9 %w/v) .Fourth group injected intra peritoneal with low toxic dose of paracetamol 300mg/ kg body weight then administration orally with 0.3 ml of 0.3 ml of crude oil of Nigella sativa.After 24 hr all animals were sacrificed after weighing then take the blood samples by heart puncture for biochemical test and liver were removed out for weighing.Results and statistical analysis: Showed a significant increase (p< 0.05) in the level of serum enzymes in the first group(p) GOT (110.1±0.61 * I U/L) and GPT(80.2 ±26.3* IU/L) and also the fourth group(P&N) GOT (101.60±4.34*I U/L) and GPT(72.8±23.88* IU/L) and significant increase in total protein (7.322±2.89* gm/dl) in third group(N) and fourth group(P&N) (7.334±0.77* gm/dl) at p<0.05 as compared with control.Also there is a significant decrease (p>o.o1) in body weight (28.15 ±7.084**) and liver weight(1.7837±0.99*) at (p>o.o5).

يمكن ان تسبب الجرعة السامة من عقار الباراسيتامول ضررا في كبد الانسان او الحيوان , لذا استخدمت الحبة السوداء التي لها صفات مضادة للاكسدة وحماية الكبد. لقد استخدمت في هذه الدراسة الزيت الخام للحبة السوداء التجاري عن طريق الفم وبجرعة 0.3 مل وتم حقن الحيوانات بالجرعة السامة الواطئة 300 ملغم / كغم من وزن الجسم من عقار الباراسيتول داخل البريتون . وقيست مستويات انزيمات الكبد في المصل GOT ,GPT والبروتين الكلي . قسمت 20 من ذكور الفئران البيض من طراز بالب / سي عشوائيا الى اربع مجاميع المجموعة الاولى حقنت داخل البريتون بالجرعة السامة الواطئة من عقار البراسيتول 300ملغم / كغم من وزن الجسم ثم جرعت ب 0.3 مل من محلول الملح الوظيفي (0.9 %غم / لتر )عن طريق الفم . المجموعة الثانية اعتبرت مجموعة سيطرة حيث حقنت داخل البريتون ب 0.3 مل من محلول ملحي ثم جرعت فمويا ب 0.3 مل من محلول الملح الوظيفي (0.9 %غم / لتر )عن طريق الفم .جرعت المجموعة الثالثة عن طريق الفم ب 0.3 مل من زيت الحبة السوداء وحقنت داخل البريتون ب 0.3 مل من محلول الملح الوظيفي (0.9 %غم / لتر ). المجموعة الرابعة حقنت داخل البريتون بالجرعة السامة الواطئة من عقار البراسيتول 300ملغم / كغم من وزن الجسم ثم جرعت عن طريق الفم ب 0.3 مل من زيت الحبة السوداء .بعد 24 ساعة من التجربة تم التضحية بجميع الحيوانات بعد وزنها وجمعت عينات الدم عن طريق ثقب القلب لاجراء الاختبارات البايوكيميائية واخذ الكبد للوزن.اظهر التحليل الإحصائي زيادة معنوية (p< 0.05) في مستويات انزيمات المصل في المجموعة الاولى (p) GOT (110.1±0.61 * I U/L) و GPT(80.2 ±26.3* IU/L) وكذلك المجموعة الرابعة (P&N) GOT (101.60±4.34*I U/L) و GPT(72.8±23.88* IU/L)وزيادة معنوية في قيمة البروتين الكلي (7.322±2.89* gm/dl) في المجموعة الثالثة(N) و المجموعة الرابعة(P&N) (7.334±0.77* gm/dl) عند p<0.05مقارنة مع مجموعة السيطرة . كذلك هنالك انخفاض معنوي (p>o.o1) في وزن الجسم (28.15 ±7.084**) ووزن الكبد (1.7837±0.99*) عند (p>o.o5.

included the problem that is there is weakness in some of the physical force players when applying the required activity, Another problem is that some athletes use stimulants to get better results illegally, and this results in serious health harms, The goals beyond the research is to identify the impact of eating different doses of Nigella Sativa accompanied by training on some the composition blood of Physical Force Players,And included the theoretical material. Subjects related directly to the research matter, and the title of the thesis, are discussed as well as the most important previous similar studies dealt with the methodology of the research and the field procedures the researcher user an experimental methodology the population was Diyala province clubs for the sport of physical force while the sample is (9) athletes players The sample was given training program concurrently with the food program by giving the sample of Nigella Sativa in three different daily doses after training (1gram for the first group, 1.5 gm. for the second and 2gm. For the third group) , Consisted Chapter Four of displaying and analyzing and discussing the findings of researcher for pre and post-tests through physical and some the composition blood test and the researcher concluded that the effectiveness of Nigella Sativa through the results in(white blood cells and red blood cells) ,The recommendation is the third dose (2g Nigella Sativa seeds) is to be adopted due to its high effectiveness in high tension activities according to the results of the three experimental groups.

تكمن مشكلة البحث بوجود ضعف لدى بعض لاعبي القوة البدنية في إنجاز الفعالية المطلوبة مما أدى إلى لجوء البعض منهم لتناول المنشطات كوسيلة للحصول على نتائج متقدمة وبطريقة الكسب غير المشروع ، وهدف البحث إلى التعرف على تأثير تناول جرع مختلفة من الحبة السوداء على بعض مؤشرات الدم للاعبي القوة البدنية،كما احتوى على المواد النظرية وأهم المواضيع التي تخدم البحث بصورة مباشرة وأهم الدراسات المشابهة. وأشتمل البحث على منهجية البحث وإجراءاته الميدانية إذ أستخدم الباحث المنهج التجريبي ، أما مجتمع البحث فقد تمثل في أندية محافظة ديالى لرياضة القوة البدنية وبلغت عينة البحث (9) لاعبين، وتم إجراء القياسات القبلية لمؤشرات الدم وأجري العمل مع العينة بالمنهج التدريبي والمنهج الغذائي الخاص بالحبة السوداء بشكل يومي بعد التدريب وبثلاث جرع مختلفة على المجاميع الثلاثة (المجموعة الأولى 1غم، المجموعة الثانية 1.5غم ،المجموعة الثالثة 2غم).وأحتوى البحث على عرض وتحليل ومناقشة النتائج التي توصل إليها الباحث للقياسات القبلية والبعدية لبعض مؤشرات الدم، وإستنتج الباحث بوجود تأثير إيجابي للحبة السوداء في بعض مؤشرات الدم،ويوصي الباحث باعتماد الجرعة الغذائية ذات الوزن(2غم) من الحبة السوداء لفاعليتها في الفعاليات ذات الشدد العالية والتي أظهرتها النتائج بين المجاميع الثلاثة.

Abstract

This study is designed for isolation and identification of some saponin compounds from nigella sativa(seeds), and to determine the inhibitory effect of these compounds were investigated on the growth of some bacteria, which include: Staphylococcus aureus, Bacillus subtilis, Salmonella typhi, Klebsiella pneumoniae, Proteus vulgaris and Pseudomonas aeruginosa and used the antibiotics (Gentamicin, Tetracycline) as control samples. The saponin compounds were further analyzed by IR and identificated by thin layer chromatography (TLC plate). The saponin compounds showed significant inhibiting effect on the all bacteria which was used in this study compared to commercial antibiotics as a control.

تم في هذه الدراسة فصل وتشخيص بعض مركبات الصابونينات من ثمار نبات حبة السوداء Nigella sativa وتحديد التاثير التثبيطي لهذه المركبات في نمو عدد من الجراثيم: Staphylococcus aureus و Bacillus subtilis و Pseudomonas aeruginosa و Klebsiella pneumonia و Proteus vulgaris و Salmonella typhi . واستخدم المضاديين Gentamicin و Tetracycline .وتم تشخيص مركبات الصابونينات المفصولة باستخدام طيف الاشعة تحت الحمراء (IR) كما استخدمت تقنية الطبقة الرقيقة في عملية التشخيص Thin layer chromatography (TLC) . اذ اظهرت مركبات الصابونينات فعالية تثبيطية عالية على جميع انواع الجراثيم المستخدمة قيد الدراسة مقارنة بعينات السيطرة.

Abstract

Aims: To find out the effect of topical Nigella Sativa oil on healing of chemically induced oral ulcer. Materials and Methods: Twelve rabbits were used and divided into control and treated groups, the ulcers were induced by injection of 0.3 ml 1%formalin in rabbit cheek mucosa and were treated by daily application of black cumin (N.S) twice on the ulcer site for 3days then the animals were sacri-ficed 5days later. Results: Nigella Sativa (N.S) showed marked anti inflammatory activity on a chemi-cally induced oral ulcer in experimental animals ,histological examination of biopsies from ulcers after three days of treatment revealed difference in the rate of epithelization and healing process of ulcers between the two groups, N.S treatment showed significant enhancement of healing of the ulcers. Con-clusions: Nigella Sativa oil has an obvious effect on the rate of healing process of oral ulcer.

Abstract

ABSTRACT:BACKGROUND:Acne vulgaris is the most common chronic inflammatory disease of the skin. Despite the vast array of treatment modalities available for acne, there is considerable dissatisfaction in acne treatment among patients and doctors. Cost limitation, adverse effect or lack of efficacy limit the use of current therapy. Herbs and their extracts have been used for treatment of skin disease for centuries. One of the most popular herbs in our society is Nigella sativa; this plant had diverse clinically useful activity.OBJECTIVE:To use nigella sativa oil as natural plant remedy in the treatment of acne vulgaris.METHODS:This study was conducted in the Department of Dermatology and Venereology in Baghdad Teaching Hospital, Iraq during the period between October 2005 and April 2006. In this single-blind randomly controlled therapeutic study, ninety three patients were enrolled, their ages ranged from 13-23 years. There were 51` females and 42 males. The patients were divided into 2 groups; group A (47 patient) instructed to use nigella sativa oil lotion twice daily for 2 months, while group (46 patient) used a control solution. Acne vulgaris was graded as mild and moderate acne by counting the number of papules and pustules. Statistical analysis was carried out by computer using t-test, f-test, and chi-square when needed.RESULTS:Eighty-one patients completed the study. In group A (43 patients), we noted a significantly reduced mean lesion count of papules and pustules after 2 months of therapy. The response of patients to treatment was good in 58%, moderate in 35% and no response in 7%. The satisfaction of patients with treatment was full in 67%, partial in 28%, and no satisfaction in 5%. In group B (38 patients), the lesions showed no significant reduction after 2 months and the response of patients to treatment was good in 8%, moderate in 34%, and no response in 58%. The satisfaction of patients with treatment in this group was full in 8%, partial in 24%, and no satisfaction in 68%. The mean difference in outcome after 8 weeks between the 2 study groups was statistically significant.CONCLUSION:The nigella sativa oil lotion has proved its efficacy as a topical therapy for acne vulgaris. This is a new natural plant extract, which lacks any side effects

Abstract

ABSTRACT:BACKGROUND: Hepatic ischemia-reperfusion (I/R) injury may occur in a variety of clinical settings and this remains a significant problem. Oxygen free radicals, produced on reperfusion have been shown to play a major role in hepatic I/R injury. Various therapeutic effects have been described for Nigella sativa. Additionally, it has been presented that Nigella sativa (orientalis) oil has protective effect against ischemia reperfusion injury to various organs. Therefore, it seems possible that the administration of Nigella sativa (orientalis) oil might protect the liver against the ischemia reperfusion injury . OBJECTIVE: To determine whether Nigella sativa (orientalis) oil prevents hepatic ischemia-reperfusion injury to the liver. METHODS: Thirty-six rats were divided into three groups as control (Group 1), I/R group (Group 2), and Nigella sativa oil (NS) treatment group (Group 3). All rats underwent hepatic ischemia for 60 min followed by 60 min period of reperfusion. Rats were intraperitoneally infused with only 0.9% saline solution in group 2. Rats in group 3 received NS oil (0.2 mL/kg) intraperitoneally, before ischemia and before reperfusion. Blood samples were harvested from the rats, and then the rats were sacrificed. Serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) levels were determined. RESULTS: The levels of liver enzymes in group 3 were significantly lower than those in the group 2. CONCLUSION: Our results suggest that Nigella sativa oil treatment protects the rat liver against to hepatic ischemia-reperfusion injury.

Abstract

Background: The aim of this in vivo study was to evaluate the additive beneficial role gained from dip coating ofhydroxyapatite (HA) coated cp Ti implants in Nigella Sativa (Black Seed) oil extract and its effect on the mechanicaland histological properties of bone-implant interface.Materials and Methods: 48 cp Ti screws (3mm in diameter, 8mm in length) were implanted in tibia bones of 12 NewZeeland rabbits. 24 screws were electrophoretically coated with HA within 4 minutes (control group) and 24 wereelectrophoretically coated with HA for 4 minutes and dip coated in black seed oil extract (BSO) for 5 seconds(experimental group). The samples were distributed equally over two healing intervals 6 and 12 weeks, in everyhealing interval each tibia bone(right and left) of 6 rabbits received two samples, control and experimental.Results: After each healing interval two tests were performed: Mechanical (Removal Torque) and Histological tests.Mechanical test showed that the mean torque values for HA&BSO coated implants were significantly higher thanthose for implants coated with HA only. Also the histological analysis revealed improved quality of bone response toimplants coated with HA&BSO.Conclusions: HA and BSO extract produced a couple of synthetic and natural biomaterials act in harmony andcomplementary to each other in enhancing healthy and strong bony integration of cp Ti implants

Abstract

Background: The black seed or Nigella sativa has been used for centuries to promote health and fight disease. Thisplant has a great focus for research due to its antibacterial, antifungal, anti-tumor, and hypotensive effects. Thisstudy was conducted to assess the effect of the black seed oil extract on sensitivity of mutans streptococci and theadherence to tooth surface in comparison to chlorhexidine gluconate in vitro.Materials and methods: Four different concentrations of black seed oil extract (1%, 5%, 10%, and 20%) were preparedusing ethanol as a solvent for the evaluation of the antimicrobial activity of the black seed oil extract against mutansstreptococci isolated from saliva of volunteers and compared with 0.2% chlorhexidine gluconate using agar diffusiontest, followed by determination of the Minimum Bactericidal Concentration (MBC) of the black seed oil extract.Three concentrations (1%, 5%, and 10%) were used in the adherence study whereby a stainless steel wires werethreaded from one end in the roots of previously cleaned, polished and sterilized first premolars, which were thenimmersed in 10 ml of the agent for 2 minutes, followed by washing with sterilized deionized water. The teeth werethen immersed in 10ml Brain Heart Infusion Broth and inoculated with 2% of bacterial isolates and incubatedaerobically at 370C for seven days. A positive score was given to the microbial growth on wire, teeth and bottleindicating a non-effective treatment and vice versa.Results The study showed inhibition zones for black seed oil extract which were found to be increased as theconcentration of the extract increased. The MBC of the black seed oil extract against mutans streptococci was 10%.The results also showed that the oil extract was effective in inhibiting the adherence of mutans streptococci to toothsurface at a concentration of 10%.Conclusion: The black seed oil extract has a bactericidal effect against mutans streptococci at a concentration of10%, and can inhibit the adherence of these microorganisms to tooth surface
