Abstract

Background Osteoporosis has an impact on bone healing process, platelets rich plasma (PRP) ameliorated thedeleterious effect of osteoporosis on bone healing process. Autologous (PRP) could be used in many clinical fields oforal and maxillofacial bone, implant reconstructive surgery and periodontology. This study was carried out toevaluate histologically the regeneration capacity of autologous (PRP) on defect in the mandible bone ofosteoporotic rabbits.Materials and Methods Forty-eight female rabbits were used in this study, divided into four groups (12 rabbits for eachgroup), each rabbit was received intraboney defect in the mandible. Two groups were save as control groups oneof them left for normal healing (group A), while other group were receiving PRP treatment (group B). The remainingtwo groups were given 10 mg/B.W hydrocortisone i.m daily for 8 weeks to induce OP-like condition which save asexperimental groups. One of them left for normal healing (group C), while other were receiving PRP treatment(group D).After 2, 4, and 6 weeks postoperatively (4 rabbits from each groups), blood sample was taken from eachanimal for serum alkaline phosphatase, calcium, and phosphorous analysis. Then the animals were sacrificed andthe decalcified sections of the bone were studied histomorphologically. These histometric analyses includingcounting of bone cells: osteoblast, osteocyte, and osteoclast. Bone trabecular, separation, width, and number;cortical width, blood vessels number, and bone marrow space and volume assed.Results Histological examinations showed that with the use of autologous platelets rich plasma in an osteoporoticand normal rabbit, an obvious enhancement of new bone formation and neovascularization significantly more thanthat of groups without (PRP) application. The results of osteoporotic group treated with (PRP) nearly reached thelevels of normal group without PRP in all the three periods postoperatively.Biochemical serum analysis revealed anincrease in serum alkaline phosphatase and calcium concentrations in osteoporotic animals than control one, whileserum phosphorous level increased in control animals than osteoporotic ones.Conclusions This study illustrated that the (PRP) has an osteopromotive activity that accelerated bone-healingprocess in madibular bone defect in an osteoporotic rabbits as well as in normal rabbits

Abstract

Objective: the study was carried out in an attempt to find out the effects of 0.05 ml of autologous ofplatelet rich plasma on traumatic skin injury.
Materials & methods: sixteen male rabbits were used & divided into two group according to killing after trauma in to group A after 1 day & B. after 4 days each group subdivided in to control & experimental. The skin wound induced traumatically by blade 1cm length & 2mm depth in the skin of rabbit's back, the experimental wound filled with 0.05ml of autologous PRP, while the control wound left for normal healing. The wound was dressed for one day.
Result: platelet rich plasma (PRP) showed marked angiogenesis, granulation tissue formation, histological examination of biopsies revealed difference in the rate of healing process between the control & experimental groups platelet rich plasma (PRP) showed significant enhancement of healing.
Conclusion: platelet rich plasma (PRP) accelerate the soft tissue healing

Abstract

Background: Diabetes Mellitus has an impact on bone healing process, Platelet – rich plasma PRP ameliorated thedeleterious effect of diabetes on bone healing process so the aims of this study were to assess histologically theeffects of PRP on bone healing in intrabony defect of maxillary bone in alloxan – induced diabetic and nondiabeicrabbits, and to correlate the effect of time of bone healing in two experimental groups with that of controls.Materials and methods: Fifty rabbits were used, thirty – five rabbits rendered diabetic by the use of alloxan (five ofthem dead for unknown reasons), the remaining fifteen rabbits left normal. An intrabony defect was created in themaxilla of each rabbit, the defect of control group left for normal healing, while diabetic rabbits divided2groups Firstdiabetic group, the defect filled with autologous PRP as experimental group -Second diabetic groups, defect leftempty. The specimens were collected at 1, 3, 6 weeks postoperatively. Histological examination was performedunder light microscope for sections stained with heamatoxylline and eosine to assess histomorphometric analysisincluding, counting of inflammatory cells, fibroblasts, and bone cells infiltration. The regeneration levels weredetected by measuring the thickness of new bone trabeculae by a special graduated lens, at the end of each threeperiods.Results: Histological examinations showed that with the use of autologous Platelet – rich plasma in a bone of diabeticrabbit, an obvious enhancement of new bone forming and neovascularzation significantly more than that ofdiabetic group without PRP application, the results of diabetic group treated with PRP reached the levels of normalgroup in all the three periods postoperatively.Conclusions: this study illustrated that PRP has an osteopromotive activity that accelerated bone healing process inmaxillary bone of diabetic rabbits.

المقدمة:- داء السكري لھ تاثیر ضار على عملیة التئام العظم ،المصل الغني بالصفیحات الدمویة یقلل من ھذا التاثیر الضار ،ویعمل على تسریع عملیة التئام العظم في الارانب المصابةبداء السكري.ھذه الدراسة اجریت لتقییم التطبیق السریري والسلوك النسیجي للمصل الغني بالصفیحات الدمویة والمأخوذ من نفس دم الحیوان وتأثیره على تسریع عملیة التئام العظم.الدراسات الحدیثة تتجھ نحو استخدام المصل الغني بالصفیحات الدمویة الذاتي في مختلف المجالات الطبیة مثل جراحة الوجھ والفكین ،والجراحات التعویضیة،وجراحة اللثة.طریقة العمل والمواد المستخدمة:- في ھذه الدراسة تم استخدام خمسین أرنبا، خمسة عشر ارنبا لم یتم حقنھم بایة مادة وتم حقن خمسة وثلاثین أرنب بعقار الالوكسان لتحویلھم الىأرانب مصابة بداء السكري (خمسة ارانب ماتوا بعد الحقن لاسباب غیر معروفة)، تم تقسیم الارانب الخمسة والاربعین الى ثلاثة مجموعات : الاولى :طبیعیة،تم حفر فتحة في الفكالعلوي ولم تعامل بأیة مادة. الثانیة:أرانب مصابة بداء السكري ،قسمت الى مجموعتین :- في الاولى تم حفر فتحة في الفك العلوي وملئت بمصل غني بالصفیحات الدمویة من نفس دمالحیوان.أما الثانیة فقد تم حفر فتحة في الفك العلوي ولم تعامل بأیة مادة. أخذت العینات للفترات 6,3,1 أسابیع بعد اجراء العملیة. أجریت النتائج النسیجیة بأستخدام المجھر الضوئي حیثصبغت الشرائح بصبغة الھیماتوكسیلین والایوسین لتسھیل اجراء القیاسات النسیجیة لبیان ترسب الخلایا الالتھابیة ،الارومات اللیفیة والخلایا العظمیة فضلا عن قیاس سمك الشرائحالعظمیة المكونة جدیدا في نھایة كل فترةالنتائج: نتائج الفحص النسیجي أوضحت زیادة معنویة في نسبة تكون العظم وزیادة ثخن الصفیحات العظمیة في العظم الجدید لمجموعة الحیوانات المصابة بداء السكري والتي عوملفیھا عظم الفك بالمصل الغني بالصفیحات الدمویة مقارنة بتلك التي في مجموعة الحیوانات المصابة بداء السكري ولم یعامل فیھا العظم بھذا المصل.الاستنتاجات: استخدام المصل الغني بالصفیحات الدمویة أدى الى زیادة أسرع وأفضل في تكوین عظم الفك وزیادة عدد الخلایا المكونة للعظم بنسبة واضحة وأكثر من المجموعةالمصابة بداء السكري والتي لم تعامل بھذا المصل.

Abstract

The purpose of this study is to evaluate the effects of OSTEON, PRP and the combination of both of them on the healing of experimentally induced bone defects in rabbits. Materials and Meth-ods: Fifteen domestic rabbits were used in this study; four bone defects were made in the tibia of each rabbit. The defects were filled with OSTEON, PRP, OSTEON / PRP and the other left unfilled as con-trol. Specimens were collected at one, two and four weeks after surgery. Radiographical and histologi-cal examinations were used to evaluate healing depending of the amount of new bone formed. Results: It was found that the OSTEON / PRP combination led to a significant healing compared to other groups at first week only. On the other hand PRP alone showed better result than OSTEON alone in the three periods. Conclusion: It was concluded that PRP plasma exert a beneficial effect on healing pro-cess when added to bone substitute material OSTEON.

Abstract

Abstract The role of platelet-rich plasma in the pain relief and treatment of many orthopedic problems had gained lot of studies & practice. Yet, it hadn’t been practiced in our locality. Thus, the study of its role in the treatment of certain enthsiopathies (plantar faciitis, achillis tendinitis and lateral epicondylitis) had been planned for. A total of 63 cases of the three diseases were chosen for a case control study. They were divided into two groups: the case group who had been treated with local injection of autologous platelets rich plasma (prepared by the Trima accel cell separating machine) and the control group who were treated by local steroid injections. Pre and three post-treatment follow up of cases were done to assess the pain perception level using the simple visual analog scale (VAS). Results had shown a statistically significant reduction in pain among cases compared to control. These results were comparable to many studies elsewhere in the world. This had led us to conclude the advice to encourage this type of therapy on a large scale of patients in the future with more detailed further studies about.

Abstract

Background: The preparation of Platelet-rich plasma (PRP) is minimally invasive way, simple, low cost to obtainnatural autologous growth factors and is now being widely used in different fields of medicine for its ability toincrease the regeneration potential of tissue. The aim of this study was to investigate the effect of local application ofautologous PRP gel on acceleration rate of osseointegration period by clinical assessment accomplished bydetermining the changes in implant stability during 3 months healing period using resonance frequency analysis(RFA).Materials and methods: A total of 28 dental implants were inserted in edentulous maxillae or mandibles of 13 patientsusing a split mouth design, i.e. each patient was received at least two dental implants at the same session, oneimplant was implanted in association with PRP which was placed locally in one site, to serve as PRP group, and theother implant was placed without PRP, to serve as a control group. Both groups were followed with repeated implantstability measurement by means of resonance frequency analysis at different time intervals (at the time of implantplacement, 8th week, and 12th week postoperatively).Results: There was no obvious statistically significant difference in mean ISQ between PRP and control groups (P >0.05) at baseline, 8 weeks, and 12 weeks postoperatively.Conclusions: Within the limitations of the present study, no appreciable clinical effect was observed to acceleratethe rate of osseointegration of sandblasted acid-etched endosseous dental implants when using topical applicationof autologous PRP gel into the prepared drill holes

Abstract

Background: Painful elbow joint over the lateral epicondyle especially with resisted wrist extension are common signs of lateral epicondyle tendinopathy, also called tennis elbow. Objective: To evaluate the clinical outcome of local platelet rich plasma (PRP) injection in patients with chronic tennis elbow compared with a steroid (Depomedrol 40 mg) injection.Methods: A total of 88 patients with chronic tennis elbow were treated at Al-Kindy Teaching Hospital and private clinics. All patients had chronic pain for about 24 weeks or more and had failed first line treatment. The patients dividing into two groups, Group A injected with PRP (n = 44), and group B injected with depomedrol 40 mg (n = 44). A good clinical result was demarcated as 25% or more progress on the visual analog scale for pain. All patients followed for 6 months in both group for clinical successful result.Results: At three months (n = 44), in group A reported a perfection of 58.2% in their pain scores while 49.3% in the group B (N = 44). At 6 months follow up, the group A informed a perfection of 74.3% in their pain scores while 58.4 % in the group B. The local elbow tenderness recording at three months was 37.4% in the group A, while in the group B was 48.4%. At six months, 16.1% versus 30.2% recounted major elbow tenderness (P = .009) in groups (A and B) respectively.The clinical improvement rates at three months revealed no changes between both groups while it showed more significant clinically changes in group A ( 87.1%) than in group B (70.1 %) with P value = 0.008 after six months follow up.Conclusion: No important changes were found at 3 months in both groups, but at 6months, clinical significant perfections in patients treated with PRP group (group A)

Abstract

Background: Anterior disc displacement with reduction (ADDWR) is the most common form of the internal derangement (ID) of temporomandibular joint (TMJ). It is a painful progressive dysfunction and clinically characterized by reciprocal clicking due to shift in the disc anteriorly in relation to the condyle and fossa during mandible elevation. Minimally invasive therapy such as intra-articular injection of platelet-rich plasma (PRP) has been used. PRP is a natural autologous product with a high platelet concentration obtained by centrifugation process to enhance tissue healing through several growth factors (GFs), which are released after endogenous activation. The aim of this study is to assess this technique which is increasingly used today as a safe, simple modality.Materials and Methods: sixty patients with ADDWR were participated (Females 41, males 19; age 20-39 years). Patients were divided into two groups; PRP was used as study group and conservative therapy was used as control group. 1 ml of PRP was obtained by a single step centrifugation. Pain, joint sounds and maximum mouth opening were clinically evaluated before and two months after therapy.Results: There was statistically significant improvement in pain score, clicking and maximum mouth opening in patients treated with PRP. No complications were recorded immediately and two months after therapy. Conclusion: This study support the effectiveness of PRP injection in treatment of ADDWR as a safe modality in reducing pain, clicking and increase mouth opening

Abstract

Introduction: Temporomandibular joint internal derangements are progressivepainful conditions that associated with limitation of mouth opening that caused bydisruption within the internal aspects of the TMJ.The aim of this study was to evaluate the management of internal derangementof TMJ regarding pain intensity in temporomandibular joints and muscles ofmastication and mouth opening.Materials and methods: This study was conducted in the Department of oral andmaxillofacial surgery-college of dentistry Tikrit University. Twenty-one patients,14 women and 7men with age range between 18-33 years old, were diagnosedwith internal derangement of TMJ that were treated by intraarticular PRP.Results: show significant reduction of pain experienced by the patient after 12weeks(P- < 0.0001). And show significant improvement of maximal interincisalmouth opening (P-<0.000).These finding demonstrating clearly that a significantimprovement in both pain , mouth opening and clicking in the TMJ was achievedafter the management.Conclusion: PRP injection to TMJ space for treatment of internal derangement aresafe, effective, non-invasive method that help to reduce pain, joint noise andincrease mouth opening, thus improving the jaw function.

Abstract

This study aimed to evaluate the benefits of using platelet– rich plasma (PRP) on wound healing. The study performed on ten adult male dogs, aged 2-3 years and their weight were 18-25 kg. Under the effect of deep sedation and local subcutaneous anesthesia at the site of wound, four of (2x2) cm of full-thickness square cutaneous wounds were induced, two on each side of the lateral thoracic region of each animal. A pair of left wounds was treated by injection with 5 mL of autonomous PRP (treatment group), 2 mm lateral to the wound edges and in the wound center., immediately after wounding while, right wounds, were injected with a single dose of 5ml of sterile saline (control group) by using the same procedure as in treatment group. The clinical and histopathological evaluation of each group for five weeks, histological biopsies were taken in the days (3, 7, 14, 21, 28 and 35) of the experiment. The clinical evaluation of PRP treated wounds showed that the wound healing process (% contraction, % re-epithelization and % total wound healing) were significantly more (P<0.05) than the control wounds during the five weeks study. The histopathological results revealed that PRP treated wounds have enhanced cellularity, increased vascularity, with increased amount of granulation tissue accompanied by increase numbers of skin appendages suggesting enhanced cutaneous regeneration, than those in untreated wounds. Depending on the clinical and histopathological findings, this study confirms that local implantation of PRP leads to acceleration and improvement cutaneous wound healing.

هدف الدراسة الى تقييم فوائد استخدام الصفائح الدموية الغنية بالبلازما على التئام الجروح. أجريت هذه الدراسة على 10 كلاب محلية وكان اغلبها من الذكور, عمرها من 2-3 سنوات وكانت تزن من 18-25 كغ. تحت تأثير التهدئة العميقة والتخدير الارتشاحي الموضعي, استحداث أربعة من الجروح الجلدية مربعة الشكل بأبعاد 2×2 سم في كل حيوان, اثنان على كل جانب من جوانب الوجه الوحشي لمنطقة الصدر. تم علاج زوج من الجروح اليسرى عن طريق الحقن بجرعة 5 مل من PRP الذاتية (مجموعة العلاج) ، 2ملم وحشيا لحافات الجرح وفي مركز الجرح, مباشرة بعد استحداث الجرح, بينما مجموعة السيطرة فقد شملت على الجروح اليمنى والتي حقنت بجرعة 5 مل من المحلول الملحي, بنفس طريقة الحقن التي اتبعت في مجموعة المعالجة. تم تقييم ومقارنة شفاء الجروح سريريا )عيانيا) ونسجيا في مجموعتي السيطرة والمعالجة مدة خمسة اسابيع حيث تم اخذ الخزع النسجية في الايام (3, 7, 14, 21, 28, 35) من التجربة. أظهر التقييم السريري للجروح المعالجة بالـ PRP أن عملية التئام الجروح (نسبة التقلص ٪, اعادة بناء الظهارة ٪, والشفاء الكلي للجروح ٪) كانت أكثر بكثير (P <0.05) من جروح السيطرة خلال دراسة الخمسة أسابيع. كشفت النتائج النسيجية أن الجروح المعالجة بالـ PRP عززت الخلوية، زيادة الأوعية الدموية، مع زيادة كمية النسيج الحبيبي مصحوبة بزيادة أعداد الملحقات الجلدية مما يوحي بتجديد جلدي محسّن، أكثر من الجروح غير المعالجة. اعتمادا على النتائج السريرية والتشريح المرضية، تؤكد هذه الدراسة أن الغرس الموضعي للـ PRP يؤدي إلى تسريع وتحسين شفاء الجروح الجلدية.
