Effects of silymarin on coronary artery bypass grafting

Abstract

Background: Coronary artery bypass grafting (CABG) is associated with a systemic inflammatory response. Systemic inflammatory response with secretion of cytokines & activation of leukocytes & endothelial cells is induced by CABG surgery; proinflammatory cytokines such as TNF_α, IL_1β are released in to the circulation in response to CABG surgery & have been suggested to play an important role in the pathogenesis of myocardial dysfunction in ischaemia reperfusion injury. Silymarin is a polyphenolic flavonoid derived from milk thistle (Silybum marianum) that has antioxidant, anti-inflammatory, cytoprotective effects, silymarin inhibit TNF_α & IL_1β.This study was designed to elucidate the anti-inflammatory effect of silymarin in CABG patients.Patients & methods: Thirty patients scheduled for CABG surgery in a prospective, randomized study. Twenty patients received silymarin 140 mg cap. Three times daily, given 3 days before surgery, then compared to the control group. Blood sample was drawn before & after surgery for full investigation; WBCs, differential counts; Neutrophils, Monocytes, Lymphocytes, Hb, PCV, ESR, total cholesterol, LDL, HDL, Triglycerides, SGOT, SGPT. B.urea, S.creatinin.Results: There was a decrease in post operative values of WBCs, Neutrophil, monocyte, lymphocyte, ESR, total cholesterol, SGOT, SGPT, B.urea, S.creatinine in patients treated with silymarin, compared to control. There was an increase in postoperative values of HDL in treated patients compared to control.Conclusion: Silymarin treatment is associated with an effect on the measured parameters. The data suggest that this drug may have a direct anti-inflammatory effect which explains its clinical benefit.

The Efficacy of Topically Applied Silymarin in the Treatment of Herpes Labialis Ulcers

Abstract

Herpes labialis is an infection caused by the herpes simplex virus, characterized by an eruption of small and usually painful blisters on the skin of the lips, mouth, gums, or the skin around the mouth. Although there is no successful treatment available, the local use of compounds with effective anti-inflammatory and cytoprotective effects may be of value in this respect. This project was designed to evaluate clinically the local use of silymarin, a group of flavonoids with powerful antioxidant, anti-inflammatory and cytoprotective activity, in the treatment of herpes simplex ulcer. Fifty three patients with herpes labialis ulcers (HLU) were enrolled in this randomized, single blinded, placebo controlled clinical study, and they were allocated into 4 groups, treated with 1%, 3% and 5% silymarin paste and placebo formula respectively. Patient's responses to treatment were followed by clinical evaluation of healing time, size of the ulcers and pain sensation, in addition to evaluating biochemical and immunological markers of the oxidative stress and inflammatory response. HLU patients showed dose dependent improvement in the healing time, pain score and size of ulcer as a result of treatment with various concentrations (1%, 3% and 5%) of silymarin paste, associated with improvement in the oxidative stress state and immunological parameters. In conclusion, silymarin can be used locally as paste formula for the treatment of HLU, an effect which may be attributed to its antioxidant, anti-inflammatory and cytoprotective properties.

Abstract

Background: Silymarin is a polyphenolic flavonoid derived from milk thistle (Silybum marianum) that has anti-inflammatory, cytoprotective, anticarcinogenic and antioxidant effects. It has been used medicinally to treat liver disorders including acute and chronic viral hepatitis, toxin/drug induced hepatitis, and alcoholic liver disease. Objective: To evaluate the antinociceptive effect of silymarin in experimental animal model of pain.Methods: The efficacy and dose response effect of silymarin (125, 250, and 500mg/kg) were assessed against control using tail flick test in mice as a model of nociceptive pain. In this model, all doses of silymarin were given intraperitoneally 15 min before immersion of tail in hot water 50°C, and Tail Flick Latency was measured before, and after (15, 30, 60 and 120 min) administration of silymarin.Result: Silymarin in 250 and 500mg/kg significantly increase Tail Flick Latency after 15, 30, 60 and 120 min in a dose dependent manner that the maximum effect seen after 120 min compared to baseline value. Conclusion: Silymarin as a herbal drug produce a significant antinociceptive effect in experimental animal model of pain, and beside its better standardization, quality control, and safety profile, in addition to its availability and relative low cost, represent a good alternative choice for management of pain.

Abstract

Background: Atopic dermatitis is a chronic inflammatory disease that causes eczema, rashes, and itching. It is most common in younger people & adolescence. Recurrent relapses are a characteristic feature of atopic eczema. Anti-inflammatory therapy of exacerbations is aimed to control effectively disease activity and permit a return to basic dermatological therapy as soon as possible. Oxidative stress & inflammatory responses are thought to be responsible for the pathogenesis of atopic dermatitis. Silymarin is a polyphenolic flavonoid derived from the seed of milk thistle (Silybum marianum (L.) Gaertner) that has anti-inflammatory, antioxidant and cytoprotective effect. The aim of this study was to evaluate the anti-inflammatory effect of topical preparation of silymarin in atopic dermatitis.Patients & method: Forty-three patients with atopic dermatitis (26 female, 14 male, 3 - 34 years old) were participated in the single blind, placebo controlled, 8-week trial. Of these, 10 patients were treated with 0.05 %, 10 patients were treated with 0.1 %, 10 patients were treated with 0.2 % topical preparation of silymarin, twice daily. The other 10 patients were received placebo twice daily.Results: There was a statistically significant improvement. A statically significant was seen in mean total body surface area involvement over time. Mean total body surface area was 25.675% at baseline & decreased to 1% at 2-4 week.Silymarin ointment was proven to act rapidly, to be highly effective, excellent improvements in pruritis, erythema, & skin irritation were noted in the patients who received 0.1 % & 0.2 % silymarin ointment topical treatment in comparison with the control group.Conclusion: Topical treatment of silymarin is more effective, safe & may represent a breakthrough drug in the manegment of atopic dermatitis.

Abstract

This study aimed to investigate the Ameliorative effect of ethanolic extracted of silymarin from Silybum marianum seeds and to compare it’s with the commercial silymarin extract(legalon) against oral dosing of Nickel chloride effects on sperms concentration, motility ,viability, sperm abnormality and male fertility and gonadal hormones .Twenty adult male rabbits aged 5 to 6 month and weighted 1.250-1.500 kg divided into four equal groups,1stgroup served as control groupreceived 1 ml of normal saline (NaCl 0.9%),2ndgroup received (1mg/100g B.W )NiCl2 orally, 3rdgroup received same dose of NiCl2 plus (0.1mg/100g B.W)silymarin extract, the fourth group received same dose of NiCl2 plus (0.1mg/100g B.W) legalon for 35 days, Results showed negative effects of NiCl2 which causedsignificant(p≤0.05) decrease in sperm concentration ,viability, motility and fertility while sperm abnormality was significantly increase, also NiCl2 caused significant (p≤0.05) decrease in serum progesterone, estradiol and testosterone. while silymarin extract and legalon adverse the negative effects of NiCl2 and causing ameliorative effects on all the studied parameters .

Abstract

The present study aimed to investigate the role of silymarin as an antioxidant and/or it activity in induction of theendogenous antioxidants in intact adult male rats. Seventy males were randomly devided into control and silymarin treatedgroups (35 each), and were drenched with drinking water and silymarin suspension (200 mg/ kg b.w) daily for 40 days. Eachgroup was allocated to 5 equal subgroups; sacrificed before treatment (0 day), and after 10, 20, 30, and 40 days of treatment.At the end of each period, males were anaesthesized, dissected and blood samples were obtained for assessment of MDA,SOD, CAT and GSH concentrations. liver samples (1 g) have been removed and homogenized for assessment of liversubcellular MDA, SOD, CAT and GSH concentrations. At the end of each periods, serum and liver subcellular MADconcentrations showed no significant changes between groups, whereas SOD, CAT, and GSH concentrations significantlyincreased at 10, 20, 30, and 40 day periods in silymarin treated males compared with control. It can be concluded thatsilymarin antioxidant activity is of pharmacological value not only as an antioxidant by itself but also as an inducer ofendogenous enzymatic and non-enzymatic antioxidants even in normal intact male.

ھدفت الدراسة الحالية الى التحري عن دور السليمارين بوصفه مضادا للأكسدة و/أو فعاليته في تحفيز انتاج مضادات الأكسدة الداخلية في ذكور الجرذان السليمة. تم توزيع ٧٠ جرذا ذكرا ناضجا على مجموعتي سيطرة ومعاملة بالسليمارين ( ٣٥ جرذا لكل منھا) وجرعت ماء الشرب ومعلق السليمارين ( ٢٠٠ ملغم/كغم من وزن الجسم)، على التوالي، يوميا ولمدة ٤٠ يوما. قسمت كل مجموعة الى ٥ مجموعات ثانوية متساوية العدد، تمت التضحية بھا قبل المعاملة (اليوم الصفر) وبعد ١٠ و ٢٠ و ٣٠ و ٤٠ يوما. في نھاية كل مدة، تم في مصل الدم، وعينات من أكبادھا لغرض قياس نفس المعايير في GSH و CAT و SOD و MDA أخذ عينات دم لغرض قياس تركيز في مصل الدم والسوائل الخلوية عدم وجود فروقات معنوية بين MDA السوائل الخلوية للأكباد. في نھاية كل مدة، أظھرت تراكيز في مصل الدم والوائل الخلوية مرتفعة معنويا في مجموعة المعاملة GSH و CAT و SOD المجموعتين، بينما كانت تراكيز كل من بالمقارنة مع السيطرة ابتداءً من اليوم العاشر من المعاملة واستمرت في الارتفاع مع تقدم مدة المعاملة. يستنتج أن فعالية السليمارين المضادة للأكسدة ذات قيمة دوائية ليس بوصفھا مضادة للأكسدة فحسب بل أنھا تحفز انتاج مضادات الأكسدة الداخلية حتى في الحيوانات السليمة.

Abstract

This study was conducted to investigate if silymarin can prevent the adverse effect of oxidative stress induced by carbon tetrachloride(CCl4) on some characters of male rat reproductive system. Twenty male albino rats were divided into 4 groups, 5 for each included, control group, CCl4 group, silymarin group and silymarin plus CCl4 group. Groups with silymarin received dose (150mg/kg) and group with CCl4 received (3ml/kg) at the end of experiment. The period of the study was 35 days. CCl4 can induce male reproductive toxicity through damage testis structure, weights, reduction in epididymis, sperm count, morphology, viability and motility. Silymarin plays its antioxidant role with scavenging free radical resulted by CCl4 and showed significant increasing in body and testis weight, normal sperms counting and vital sperms in compared to CCl4 group

انجزت هذه الدراسة لمعرفة كفاءة السلمارين في منع التاثير الضار للاجهاد التاكسدي الذي يحثه رباعي كلوريد الكربون CCl4 على بعض خصائص الجهاز التكاثري لذكور الجرذان البيض. عشرون ذكراً من الجرذان البيض قسمت الى اربع مجموعات، خمس حيوانات لكل منها، تضمنت المجموعات المختبرية، مجموعة السيطرة، المجموعة المعاملة بـCCl4 3m/Kg))، مجموعة السلمارين (150mg/Kg) ومجموعة السلمارين مع CCl4. كانت مدة الدراسة 35 يوما. اظهر CCl4 تاثيرا سميا على الجهاز التكاثري من خلال تضرر نسيج الخصية وانخفاض وزن البربخ ،عد النطف،الشكل الطبيعي للنطف، حيوية وحركة النطف. اظهر السلمارين فعاليته المضادة للاكسدة لازالة الجذور الحرة الناتجة عن تاثير CCl4 ولوحظ ارتفاعا معنويا في وزن الجسم والخصى، النطف الطبيعية، عد النطف وحيوية النطف بالمقارنة مع المجموعة المعاملة بـCCl4

Abstract

Taldalafil is a long acting phosphodiesterase-5 inhibitor widely used in thetreatment of erectile dysfunction. The abuse of tadalafil, which is a worldwideproblem, is associated with many side effects. Sylimarin considered as a herbal drug,its active constituents are silybin, isosilybin, silydianin and silychristin. Acontroversy exist with regard to the effects of tadalafil and silymarin on gonadalfunction in animal species. The present study aimed to investigate the effects oftadalafil and/or sylimarin on gonadal function adult male rates. Twenty four male ratsare used in the study, they divided into 4 groups (n=6) and gavaged daily for 1 monthas follows; 1-control(0.5ml distilled water), 2-tadalafil (10mg tadalafil/kg ), 3-silymarin (100 mg silymarin/ kg) and 4-tadalafil + silymarin ( 10 mg tadalafil and 100silymarin mg/ kg). Tadalafil significantly increases serum testosterone and decreasesLH and FSH, it adversely affect the process of spermatogenesis (sperm count andpercentage of motile, dead, and abnormal sperms). While, silymarin significantlyincreases serum testosterone, LH and FSH and improve spermatogenesis. Serumtestosterone remains significantly higher in tadalafil+silymarin group compared withcontrol group, while FSH and LH and parameters of spermatogenesis becameinsignificantly different from those in control group.

التادالافیل ھو مانع طویل الامد لانزیم فوسفودیستریس- ٥یستخدم على نطاق واسع في علاجضعف الانتصاب. سوء استعمال التادالافیل،الذي ھو مشكلة في جمیع أنحاء العالم،یرتبط مع العدید من الآثارالجانبیة. سیلیمارین یعتبر من الادویةالعشبیة، ومكوناتھ النشطة ھي (silybin, isosilybin, silydianin and.silychristin)وھنالك جدل فیما یتعلق بآثار التادالافیل والسیلیمارین على وظیفة الغدد التناسلیة في أجناسالحیوانات. الھدف من ھذه الدراسة ھو التحقیق من آثار تادالافیل و/ أو سیلیمارین على وظیفة الغدد التناسلیة فيذكور الجرذان البالغة . أستخدم أربعة وعشرون ذكراً من الفئران في الدراسة، وقسمو إلى ٤مجموعات )٦جرذان في كل مجموعة( وجرعت یومیا لمدة شھر واحد على النحو التالي: -١مجموعة السیطرة )٠٫٥مل ماءمقطر( -٢مجموعة التادالافیل )١٠ملغم تادالافیل/ كجم(، -٣مجموعة السیلیمارین ) ١٠٠ملغ سیلیمارین/ كجم(و -٤مجموعة التادالافیل + السیلیمارین )١٠ملغ تادالافیل/ كجم و ١٠٠ملغ سیلیمارین/ كجم(. التادالافیل أدىإلى زیادة معنویة في ھرمون التستوستیرون ونقصان في FSHو ،LHوانھ اث َر سلباً على عملیة تكونالحیوانات المنویة )عدد الحیوانات المنویة والنسبة المئویة للحیوانات المنویة المتحركة، المیتة والغیر طبیعیة(.بینما السیلیمارین أدى إلى زیادة معنویة في ھرمون التستوستیرون ، FSHو LHوحسن عملیة تكونالحیوانات المنویة. أما في مجموعة تادالافیل + سیلیمارین فأن ھرمون تستوستیرون لایزال أعلى معنویاً مقارنةمع مجموعة السیطرة، في حین أن FSHو LHودلالات تكون الحیوانات المنویة أصبحت غیر مختلفة معنونیاًمقارنتاً مع تلك الموجودة في مجموعة السیطرة
