Effects of Metformin on Sperm Parameters in Mice: as a model for human being

Abstract

Background:Metformin is anti-hyperglycemic effect due to decrease of hepatic glucose production. Another effect, leading decrease insulin resistance, regarded a safe drug, treatment of choice for overweight.Objective:To investigate the effects of 3 concentration of MF administered orally to male mice on sperm parameters as model for human being.Materials and Methods:In current study, 80 male mice were taken with age ranging between 1.5-2 month and weights ranging between 25-30 grams males classified into four groups of a control group (G1) and treated group (G2, G3 and G4) doses of MF were employed, 0.2, 0.4, 0.6 mg/mice daily to groups, given orally for 6 weeks and then the animals were scarified.ResultsResults have revealed a significant difference (P < 0.05) in sperm concentration for 3 groups compared to G1 group. Progressive motility (%) appeared with significant difference (P< 0.05) for G3 group as compared to other treated and G1 group. Furthermore, the result illustrated a significant decreased (P< 0.05) in the abnormal sperm morphology for G3 group compared with treated and control. However, sperm agglutination (%) confirmed that the G3 group exhibited highly statistical significant decrease (p≤0.001) compared to G1, G2 and G4.ConclusionsFrom results of the present study, it was concluded that the administration of MF doses reduce sperm concentration .while, 0.4mg dose improve certain sperm parameters.

Abstract

Background:Vinblastine Sulphate (VBL) is administered with other anticancer drugs for treatment of metastatic testicular carcinoma.Objective:The aim of this study was to investigate the effects of different concentrations of vinblastine on the outcome of sperm parameters and histological changes of the seminiferous tubules in mice.Methods:Thirty fivehealthymaturemalemice(78-) weeks old and weight of (2325-) g were involved in the present study. They were divided into six treated groups (0.092, 0.14, 0.18, 0.23, 0.28 and 0.4 mg/kg) with control group (5 mature males mice/group) according to periods of orally administration of each week throughout 6 weeks. Males were scarifiedattheendofadministrationperiodforassessmentofsperm function parameters. The testes were obtained and prepared for histological sections. Diameter of seminiferous tubules and thickness of layers of proliferated spermatogenic cells were assessed for all groups of male mice.Results:A significantdecrease(P<0.05)wasobservedinthespermconcentrationaftertreatmentwiththedoses 4, 5 and 6 when compared with control group and highly significantdecrease(P<0.001)inthesperm viability percentage was appeared for all doses groups when compared with control group. On the other hand, highly significantdecrease(P<0.001)appearedinthespermmotilityaftertreatmentwith all doses when compared with control group.Diameter of seminiferous tubules and thickness of proliferated spermatogenic cells layers were decreased gradually throughout periods of orally vinblastine administration. Highly significantdecrease(P<0.001) in the diameter of seminiferous tubules and thickness of proliferated spermatogenic cells layers was appeared following the treatment with high doses when compared with control.Conclusion:High doses of vinblastine sulphate have negative effects on sperm parameters and histological feature testis in mice.

Abstract

Background: Male obesity is associated with physiological changes that can negatively affect male fertility. Obese men have demonstrated a 3-fold increase in the prevalence of oligozoospermia when compared with men with normal Body Mass Index (BMI). Objective: To estimate the role of aromatase enzyme in decreasing sperm parameters in obese infertile men. Subjects, Materials and Methods: Seventy obese infertile male (patients) and twenty obese fertile male (control) are enrolled in this study. Seminal fluid analysis done according to criteria of WHO (2010). Results: A significant negative correlation was found between human cytochrome enzyme and sperm concentration, progressively motile sperm, non-progressively motile sperm, immotile sperm and normal morphology.Conclusion:The present study showed clearly that male obesity negatively impacts fertilitythrough direct association between increased (BMI) and semen parameters. Theresults suggest that increased estrogen as a result of aromatization in the fattytissue, reflected through the increase in human cytochrome enzyme (aromatase),may be an important mechanism for the hypoandrogenemia and altered spermparameters

Abstract

Objective : The study aimed to study the effect of rapid freezing and thawing on Malondialdehyde concentration and abnormal sperm chromatin percent and sperm parameters of Asthenozoospermic patients Methodology : This study was performed in the laboratory of Intra cytoplasmic Injection and freezing of sperm in the fertility center of Al-Sader Medical City / Al-najaf Alashraf City during the period from May, 2012 to April 2013 , The study included 30 of Asthenozoospermic patients, the statistical analysis of this study performed by using Student's t – test and paired t – test at the level of probability 0.05Results : the results showed that is sperm rapid freezing process of patients who were Included in the study by used sperm freezing media ( Sperm Freeze) and doing the thawing processes after one month From freezing leads to a significant decreasing (p<0.05) In the sperm concentration, sperm progressive motility percent, sperm viability percent and normal sperm morphology percent. and a significant increasing (p<0.05) In The Malondialdehyde concentration and abnormal Sperm chromatin percent and no significant difference(p<0.05) In round Cell concentration in comparison with their ranges after activation Conclusion : It can be concluded From this study that sperm rapid freezing and thawing processes had a negative effects on the semen and sperm parameters and Malondialdehyde concentration and abnormal sperm chromatin percent In the all samples of study included patients .Recommendation: Adding Anti oxidants to the sperm freezing media to decrease the negative effects of freezing and increase the sperm parameters .

الهدف : تهدف الدراسة الحالية إلى محاولة التعرف على تأثير عمليات التجميد السريع والإذابة في معايير المني و النطف وتركيز ألمالون داي ألالديهايد MDA والنسبة المئوية لكروماتين النطف غير السوي لمرضى وهن النطف Asthenozoospermicالمنهجية: أنُجزت هذه الدراسة في مختبر الحقن ألمجهري وتجميد النطف في مركز الخصوبة في مدينة الصدر الطبية / محافظة النجف ألاشرف , وامتدت الدراسة من المدة أيار 2012 إلى نيسان2013 وشملت الدراسة 30 من مرضى وهن النطف. وكان معدل العمر ( 1.23 ± 33.1 ) سنة و أنجزت التحليلات الإحصائية الخاصة بهذه الدراسة باستخدام اختبار – ت (Student's t – test) و اختبار – ت (paired t – test) على مستوى الاحتمالية 0.05 النتائج:. أظهرت النتائج أن عملية التجميد السريع لنطف المرضى المشمولين في الدراسة باستعمال وسط تجميد النطف (Sperm Freeze) , وإجراء عملية الإذابة بعد مرور شهر واحد أدت إلى انخفاض معنوي (P<0.05) في كل من تركيز النطف والنسبة المئوية للنطف ذات الحركة التقدمية و النسبة المئوية لعيوشية النطف و النسبة المئوية للنطف السوية و حدوث زيادة معنوية (P<0.05) في معدلات كل من تركيز المالون داي ألديهايد MDA و النسبة المئوية لكروماتين النطف غير السوي وعدم وجود فرقا" معنويا" (P<0.05) في تركيز الخلايا الدائرية مقارنة مع معدلاتها بعد التنشيط .الاستنتاجات :يستنتج من هذه الدراسة بأن عمليات التجميد السريع والإذابة للنطف تمتلك تأثيرات سلبية في معايير المني و النطف وتركيز ألمالون داي ألالديهايد MDA والنسبة المئوية لكروماتين النطف غير السوي في كل عينات المرضى المشمولين بالدراسة التوصيات :إضافة مضادات الأكسدة لاوساط تجميد النطف لغرض التقليل من تأثيرات التجميد وتحسين معايير النطف .

Abstract

Defective Sperm parameters was suggested to play an important role in determining fertility and was proven to be closely related to fertilization and pregnancy rates in the natural fertilization process as well as in ART.To evaluate the influence of defective sperm parameters including concentration, morphology and motility on intracytoplasmic sperm injection outcome represented by fertilization rate, cleavage rate, embryo quality and pregnancy rate.This a cohort study included 60 infertile couple attended the clinics of infertility treatment center Al-Sadder teaching hospital and underwent intracytoplasmic sperm injection. They are classified into three groups according to their spermiogram and ICSI outcome evaluated for each group and compared with each other.The main results of this study showed no significant difference in fertilization rate,cleavage rate ,embryo quality and pregnancy rate among studied groups i.e., no significant difference in ICSI outcome between normozoospermic group and those with defective spermiogram. Intracytoplasmic sperm injection outcomes represented by fertilization rate, cleavage rate embryo quality and pregnancy rate were identical with no significant differences between couples with defective sperm parameters and those with normal spermiogram.

Abstract

This study was aimed to investigate the effect of sperm parameters on the response to in vitro sperm activation (ISA) and to compare sperm parameters according to the age and type of infertility.One hundred twenty-eight subjects employed in this study, with age range (18-49) years and duration of infertility (1-28) years. From each subjects semen sample was obtained and seminal fluid analysis (SFA) was done. Simple medium for assisted reproductive techniques; (SMART) was used for in vitro sperm activation by direct swim-up technique and centrifugation Swim-Up technique. Sperm concentration, sperm motility (%), sperm grade activity (%), normal sperm morphology (%) and sperm agglutination (%) were assessed. Crude data were statistically analyzed. From the results of present study, it was appeared that most sperm parameters were significantly (P<0.05) enhanced post-activation in vitro as compared to pre-activation using SMART medium by direct swim-up technique and centrifugation Swim-Up technique. Sperm concentration for post-activation was significantly reduced (P<0.05) compared to pre-activation groups and the percentages of progressive sperm activity with grade A, total progressive sperm motility (grades A and B) and normal sperm morphology were significantly increased (P<0.05). Based on the results of this study, it can be concluded that sperm motility and concentration were considered most important parameters that have the importance of activation and sperm morphology one of the sperm parameters that have role of the influence on the sperm qualities after activation.

صممت هذه الدراسة لتقييم مدى تأثير متغيرات النطف على الاستجابة للتنشيط خارج الجسم ولمقارنة متغيرات النطف مع العمر ونوع العقم. شارك في هذه الدراسة مئة وثمانية وعشرون شخص، وكان متوسط أعمارهم (18-49) وفترة العقم (1-28) سنة. أخذت عينة السائل المنوي من كل مريض واجري لها فحص السائل المنوي. استعمل الوسط الزرعي البسيط للتقنيات المساعدة على الإنجاب لتنشيط النطف خارج الجسم باستخدام تقنية سباحة النطف للأعلى مباشرة وتقنية الطرد المركزي. تم تقيم تركيز النطف والنسبة المئوية للنطف المتحركة والنسبة المئوية لنشاط النطف والنسبة المئوية لشكل النطف الطبيعي والنسبة المئوية لتكتل النطف.كما تم تحليل النتائج الأولية إحصائيا. أظهرت نتائج الدراسة الحالية تحسن معنوي (P<0.05) في اغلب متغيرات النطف بعد التنشيط خارج الجسم عند المقارنة مع قبل التنشيط باستخدام وسط سمارت وتقنية سباحة النطف للأعلى مباشرة وتقنية الطرد المركزي. تركيز النطف بعد التنشيط انخفض بشكل معنوي( P<0.05) عند المقارنة مع قبل التنشيط والنسب المئوية لكل من الحركة التقدمية (A) للنطف والحركة التقدمية الكلية((A+B للنطف وشكل النطف الطبيعي أظهرت زيادة معنوية(P<0.0).لذلك نستنتج اعتمادا على معطيات هذه الدراسة بأن حركة النطف وتركيزها من أهم الصفات التي لها أهمية بالتنشيط خارج الجسم وشكل وشكل النطف الطبيعي واحد من الصفات التي لها دور بالتأثير على صفات النطف بعد التنشيط خارج الجسم.

Abstract

The toxic effect of drugs and environmental chemicals on the human reproductive system has become a major health concern. Lead is recognized as the most dangerous of the toxic metals and a major threat to the biological systems of both human and animals. In this study we detect the effect of two types of antioxidants: flaxseed and zinc, on the toxicity of low dose of lead acetate on reproductive organs of male mice, this study was conduct.Two groups of mature male mice,exposed to low dose of lead, were administered either zinc sulphate (0. 5mg/kg body weight/day) or flaxseed oil (2.5% of total daily food intake), for 6 weeks. The results showed that lead caused a significant reduction in the total body weight, testicular weights & adverse effects on sperm parameters , while the concomitant administration of antioxidants caused elevation in these weights with significant improvement in sperm quality as compared to the only lead treated group. It is concluded that lead, like all other heavy metals, could induce oxidative stress and the concomitant administration of antioxidants (zinc or flaxseed) can ameliorate the toxic effect of leadon reproduction .

Abstract

Background and objective: Many drugs and chemical substances may induce sexual dysfunction and have a negative impact on male fertility. Lipid-lowering agents like statins and fibrates are usually recommended for cardiovascular diseases prevention. However, the combination of atorvastatin and fenofibrate is the subject of questions concerning their effect on androgenic activities in this study. This study was designed to evaluate the efficacy and toxicity of atorvastatin, fenofibrate, and their combination on sperm parameters, sexual hormones, and histopathology of the testis. Methods: Twenty-four male rats were divided into four groups, each of six. The first group served as a control. The second, third, and fourth groups received atorvastatin, fenofibrate, and their combination, respectively. After 28 days, blood samples were collected for hormones and other parameters evaluations. Testis and epididymis were taken for histopathology studies and sperm parameters assessments.Results: Fenofibrate showed a significant reduction in sperm viability and serum levels of luteinizing and estradiol hormones. The combination of both drugs significantly reduced the sperm count and viability in normal male rats. Both atorvastatin and its combination mildly changed the histological structures of the testis, reduced number of spermatozoa, and arrested spermatogenesis.Conclusion: The findings from this prospective study suggested that there was no harmful effect of atorvastatin usage on conventional sperm parameters and sexual hormones. Fenofibrate usage was not adversely associated with sperm parameters other than sperm viability. The combination of atorvastatin and fenofibrate significantly reduced the sperm count and viability. No deleterious effects of the drugs were seen on the testosterone, which was the essential hormone for all stages of spermatogenesis, sperm production, maturation, and transport.

Abstract

Background:Alt men have the natural desire to have their own biological children. Cryopreservation of male gametes is an important aspect of human fertility preservation. With the advancement in assisted reproductive technology, indication for sperm cryopreservation is expanding.Objectives:To evaluate the effect of cryopreservation by using Sperm Freeze TM kit on some sperm parameters for infertile patients.Materials and methods:Ninety five semen samples were taken from male patients. The seminal fluid analysis was performed according to the standard criteria of WHO (1999). The patients were divided into four groups depending on their sperm concentration and motility: Normozoospermic, oligozoospermic, asthenozoospermic and oligoasthenozoospermic patients. Cryopreservation in liquid nitrogen was done by using Sperm Freeze TM kit for the samples and thawing was performed after three months.Results:In general, the post-thawing study revealed a considerable highly significant (P <0.001) decrease in the sperm parameters which include the means of sperm concentration. sperm motility, progressive motility and normal sperm morphology as compared to that of pre-cryopreservation. Whereas the percentage of immotile sperm was highly significant (p<0.05) increase as compared to pre-cryopreservation in all groups except for oligozoospermic patients in which there was a significant (p<0.05) increase.Conclusions:After semen cryopreservation, all parameters were significantly decreased.

Abstract

The study had been performed at college of science/Babylon university, the study included 60 male rabbits. The study aimed to determine the effect of extract of Rashid seeds Lepidium sativum in the sperm parameters of adult male rabbits. The In Vivo study include the estimation of the medium effect dose (MED50) for terpen extraction by using Dose-Response Curve. The mean of MED50 was 53.3 mg /kg body weight of terpen extraction. The effect dose of terpene revealed insignificant increase (P in sperm parameters, which included the rate of sperm concentration in the testis and epididymis tail, and the average number of sperm in one gram of testicular and sperm motility percent, grade activity, and sperm viability percent of epipdidymis caudal, and non-significant decreased (P> 0.05) in the abnormal sperm morphology percent in the epipdidymus caudal.

أجريت هذه الدراسة في كلية العلوم, جامعة بابل, وقد شملت اختيار 30 ذكرا" أرنبا". واستهدفت الدراسة معرفة تأثير مستخلص بذور الرشاد Lepidium sativum في معايير نطف ذكور الأرانب البيض, إذ شملت الدراسة داخل الجسم الحي تقدير الجرعة المؤثرة للنصف لمستخلص التربين, باستخدام منحني الجرعة-الاستجابة, وقد بلغ المعدل الحسابي للتركيز المؤثر 53.3 ملغم/كغم من وزن الجسم لمستخلص التربين. أظهر التركيز المؤثر لمستخلص التربيني زيادة غير معنوية (P ) في معايير النطف, والتي شملت معدل تركيز النطف في الخصية وذيل البربخ, ومعدل عدد النطف في غرام واحد من الخصية والنسبة المئوية للنطف المتحركة, ودرجة نشاط النطف والنسبة المئوية لحيوية النطف لذيل البربخ, وأنخفاض غير معنوي (P ) في النسبة المئوية للنطف اللاسوية في ذيل البربخ.
