The Acute and Chronic Effects of Flavonoids on The Absorption and Tissue Distribution of Selenium in Rats

Abstract

The present study was designed to evaluate the effects of acute- and chronic use of the flavonoids silibinin, epigallocatechin gallate (EGCG), quercetin and rutin on the absorption and tissues distribution of selenium (Se) after single oral dose of Na-selenite. In the first part, thirty rats were allocated into 5 groups: 1st group treated with olive oil and served as control; the other 4 groups were treated with either silibinin (100mg/kg), EGCG (25mg/kg), quercetin (50mg/kg) or rutin (500mg/kg), administered orally as oily solutions for 30 days. Then, all groups received orally single doses of Na-selenite (0.5mg/kg) 2 hrs after administration of the last doses of the flavonoids and the vehicle. In the second part, similar protocol was followed as in the first part, except for the duration of flavonoids treatment, where only single doses were administered. The animals were sacrificed 3 hrs after Se administration. Blood samples, brains, kidneys and livers were obtained for evaluation of Se content using atomic absorption spectrometry. Chronic use of flavonoids increased serum and tissues Se significantly compared to control. While acute use did not change tissues Se levels, but significantly decreasing serum Se in all flavonoids treated groups, except for EGCG-treated group. In conclusion, chronic use of flavonoids increases serum and tissues levels of Se, while single doses approach reveal a significant decrease in serum levels without affecting tissues distribution; highly significant positive correlation between serum and kidney Se were also reported.

Abstract

This experiment was carried out at Animal Farm, Department of Animal Production, College of Agriculture, University of Tikrit for the period between 17/2/2012 and 17/2/2013. 42 Awassi lambs obtained from the ewe bred at the college farm were used in the study .Growth and development measurements were recorded for the lambs between birth and yearling months).The lambs were assigned to 7 age groups : birth , 2 ,4 ,6 ,8 ,10 and 12 months with an average live body weights : 4.09,12.72,23.00,33.60,44.00,55.21 and 63.37 kg for the age groups ,respectively . The results obtained revealed a significant increase(p<0.05) of the daily weight gain with the advancement of age with the exception of the first period(birth-2 months ).The highest daily weight gain(182 gm /day) was recorded for the period (8-10 months).However, a decrease of the daily weight gain (158 gm/day ) was recorded at the last period (10-12 months) .The highest dressing percentage(54.21%) as measured on the basis of chilled carcass weight (chilled carcass weight /empty body weight ) was found for the lambs slaughtered the age of 12 month. Eye muscle area and fat thickness were increased significantly(p<0.05)with age advancement .The highest values of both traits were found for lambs slaughtered 12 months (17.36 cm2 and 3.80 mm, respectively).Physically dissected tissues (bone, muscle and fat) appeared to be significantly affected (p<0.05) with the exception of bone tissue. Lean meat % decreased from 63.22% at birth to 45.50% at the age of 12 months. Alternatively, fat % increased from 13.64% at birth to 40.10% at the same age.

أجريت هذه التجربة في الحقل الحيواني التابع لكلية الزراعة / جامعة تكريت للفترة من 17/2/2012 ولغاية 17/2/2013 استخدم في التجربة 42 حملا عواسيا تم الحصول عليها من مواليد النعاج الموجودة في الحقل الحيواني للكلية واجريت عليها القياسات الخاصة بالنمو والتطور منذ الولادة ولغاية عمر سنة ( 12 شهر) حيث تم تقسيمها الى 7 سبعة مجاميع عمرية ( 0 ، 2 ، 4 ، 6 ، 8 ، 10 ، 12 شهرا ) وبواقع 6 ستة حملان في كل مجموعة, بلغ معدل أوزانها ( 4.09 ، 12.72 ، 23.00 ، 33.60 ، 44.00 ، 55.21 ، 63.37 كغم) لكل مجموعة على التوالي . وقد اظهرت نتائج الدراسة ارتفاعا معنويا (p>0.05) في معدل الزيادة الوزنية اليومية نتيجة تقدم الحيوان بالعمر عدا بداية التجربة (من الولادة ولغاية عمر 2 شهر)اذ بلغت اعلى زيادة في الفترة من 8-10 شهر (182غم/يوم) ، في حين انخفضت من عمر 10-12 شهر لتصبح (158غم/يوم) كما ارتفعت نسبة التصافي المحسوبة على اساس وزن الذبيحة البارد / وزن الجسم الفارغ خلال مراحل النمو اذ بلغت اعلى نسبة عند عمر 12 شهر (54.21%) ، وازدادت كل من مساحة العضلة العينية وسمك طبقة الدهن معنويا (p>0.05) بتقدم العمر اذ بلغت اعلى قيمة لهما عند عمر 12 شهر ( 17.36سم2) (3.87 ملم) على التوالي. اما فيما يخص توزيع الانسجة الرئيسة المفصولة فيزيائيا فقد تاثرت معنويا (p>0.05) عدا العظام حيث انخفضت نسبة اللحم الشرح(lean ) من ( 63.22%) عند الولادة لتصبح (45.50%) عندة عمر 12 شهرا في حين ارتفعت نسبة الدهن من (13.64%) عند الولادة لتصبح (40.10% ) عند عمر 12 شهرا.
