STUDY THE EFFECT OF USING PROBIOTIC (VETLACTOFLORUM) ON SOME OF BIOCHEMICAL AND IMMUNOLOGICAL PARAMETERS OF BROILER CHICKENS
دراسة تاثير استخدام المعزز الحيوي (VETLACTOFLORUM) على بعض المؤشرات الحيوية والمناعية في دجاج اللحم

Abstract

In the present study its used liquid preparation of probiotics "Vetlactoflorum-M" (dissolved in diluted milk) and "Vetlactoflorum-C" (dissolved in whey), and were studied its effect on some parameters such as total protein, globulin, albumin also serum bactericidal and lysozyme activity.One hundred fifty broilers chicken breed "Ross-308" used in one day old. They were divided into three groups of 50 chicks (control" un-supplemented with probiotic" , first treatment group received “Vetlactoflorum-C” and second treatment group received “Vetlactoflorum-M”). Blood samples were taken in 7, 14, 21, 28, 35 and 42 day-old. Laboratorial analyses were conducted in Vitebsk state academy of veterinary medicine in Belarus. Level of total protein in the 2nd group to the end of the study was higher than in control group to 8.1% (P <0.05) in the third group by 3.96% (P <0.01). During observation period in chicken’s serum of supplemented probiotic groups, albumin concentration was higher relative to the control group. Similar results were observed in level of globulins. In day 42 of broilers chicken, globulin content in both treatment groups compared with the control group as higher by 2.97% (P<0.01) and 5.19% (P <0.05), respectively. The level of serum bactericidal activity in 42 day of 2nd group was higher than the control at 23.62% (P <0.01), and in the third group by 16.21% (P> 0, 05). The index of serum lysozymal activity also surpassed control group in chickens in group 2 by 11.96% (P> 0.05), and in the third group at 10.69% (P> 0.05).

في الدراسة الحالية استخدم مستحضر سائل من "Vetlactoflorum-M" الذائب في الحليب، و " Vetlactoflorum-C" الذائبة في شرش الحليب، و درست تأثيره على بعض المقاييس مثل البروتين الكلي والكلوبيولين والالبومين و كذلك النشاط القاتل للبكتريا والحال في مصل الدم. اخذت 150 فروج لحم من سلالة "روس-308" في عمر يوم واحد ، وقسمت إلى ثلاث مجموعات كل مجموعة تتكون من 50 فروجة لحم (مجموعة السيطرة لم تعطى المعزز الحيوي) ومجموعة المعالجة الاولى التي اعطيت المعزز الحيوي "Vetlactoflorum-C" ومجموعة المعالجة الثانية التي اعطيت المعزز الحيوي "Vetlactoflorum-M ". تم أخذ عينات الدم في عمر 7 و 14 و 21 و 28 و 35 و 42 يوما. أجريت الدراسة في مختبرات اكاديمية فيتبسك الحكومية للطب البيطري في روسيا البيضاء. كان مستوى البروتين الكلي في المجموعة الثانية حتى نهاية الدراسة أعلى من مجموعة السيطرة بحوالي 8.1٪ (P <0.05) في مجموعة التجربة الثالثة 3.96٪ (P <0.01) . خلال فترة المراقبة لوحظ في مصل دجاج اللحم للمجموعتين اللتين اعطيت المعزز الحيوي ان تركيز الألبومين أعلى نسبة بالمقارنة مع مجموعة السيطرة. ولوحظت نتائج مماثلة في كمية الجلوبيولين. في عمر 42 يوم كان مستوى الكلوبيولين في كلا مجموعتي المعالجة (الثانية والثالثة) بالمقارنة مع مجموعة السيطرة كان مرتفعا بنسبة 2.97٪ (P <0.01) و5.19٪ (P <0.05) على التوالي. وكان مستوى نشاط القاتل للجراثيم في مصل الدم في 42 يوم من مجموعة التجربة كان أعلى من مجموعة السيطرة بحوالي 23،62٪ (P <0.01)، وفي المجموعة الثالثة بحوالي 16.21٪(P <0.05). تجاوزت مؤشر النشاط الحال في المصل أيضا مجموعة السيطرة في افراخ الدجاج في المجموعة الثانية من 11.96٪ (P> 0.05)، وفي المجموعة الثالثة في 10.69٪ (P> 0.05).

Abstract

AN experiment conducted at the department of veterinary public health, college of veterinary medicine, Mosul-Iraq, during September to October 2009 to study the effect of Mycofix ® plus (MY) on broiler health performance during aflatoxicosis Eighty, one- day- old commercial male broiler chicks (Ross 308), were distributed to 4 dietary treatment groups with 2 replicates of 10 chicks each. Birds were reared for 35 days. All birds were fed on diet with or without aflatoxin (AF), and with or without (MY) in feed. The treatment groups were as follow: G1 (0 part per million (ppm) AF without MY); g2 (2.5 ppm AF without MY ); g3 (0 ppm AF & 0.25% MY ); G4 (2.5 ppm AF & 0.25 % MY ). Body weights and feed intake were recorded weekly. At 35 of bird’s age, five birds were randomly selected from each group for estimation of WBCs, RBCs, HB and ESR; glucose, total protein, cholesterol, and triglycerides; serum levels of AST,ALT and ALP enzymes; antibody titers and CV% against Newcastle disease (ND) vaccine; relative weights of bursa of Fabricious , thymus, spleen, proventriculus, gizzard, kidney, liver, heart and pancreas, in addition to recording liver lesions. Results show that MY was effective in ameliorating the negative effect of AF on growth performance (body weight gain ,feed consumption and feed conversion ratio); on relative weights of affected internal body organs and the score lesion of liver; on blood picture (WCS, HB and glucose); serum enzymes (AST, ALT and ALP); antibody titer and CV% against ND vaccine in comparison to AF contaminated feed and control groups.

الخلاصةأجريت هذه الدراسة في قسم الصحة العامة البيطرية ، كلية الطب البيطري /جامعة الموصل / العراق ، خلال المدة من ايلول إلى تشرين الاول 2009 لمعرفة تأثير المايكوفكس(Mycofix® plus (MY على الأداء الصحي والانتاجي لفروج اللحم أثناء تعرضه لسم الافلا ، اذ تم توزيع ذكورفروج اللحم (روس 308) بعمر يوم واحد إلى 4 مجاميع وبواقع مكررين (10 افراخ لكل منهما). تم تربية الافراخ لمدة 35 يومًا وغذيت مع أو بدون سم الافلا (AF) او المايكوفكس (MY). وزعت مجاميع الافراخ على النحو التالي: المجموعة الاولى :صفر جزء في المليون سم الافلا (AF) وخالية من المايكوفكس ؛ المجموعة الثانية : 2.5 جزء في المليون سم الافلا (AF) وخالية من المايكوفكس ؛ المجموعة الثالثة : 0.25 % مايكوفكس MY)) وخالية من سم الافلا ؛ المجموعة الرابعة 2.5 جزء في المليون سم الافلا (AF) و 0.25 % مايكوفكس . تم تسجيل وزن الجسم واستهلاك العليقة أسبوعيًا. وفي 35 يوم من عمر الطيور تم اختيار خمس طيور عشوائياً من كل مكرر للمجاميع الاربعة وذلك لتقدير الاعداد الكلية لخلايا الدم الحمراء ، وخضاب الدم ( (HB وسرعة ترسيب كريات الدم الحمراء (ESR) ، ومستوى الجلوكوز و البروتين الكلي و الكوليسترول و والدهون الثلاثية و ومستويات الانزيمات AST ، ALT و ALP. و قياس معيار الأجسام المضادة ومعامل اختلافها ضد لقاح مرض نيوكاسل في مصل الدم . كما واخذت الأوزان النسبية لجراب فابريشيا ، الغدة الصعترية ، الطحال ، المعدة الغدية ، القانصة ، الكلى ، الكبد ، القلب والبنكرياس اضافة إلى تسجيل آفات الكبد. اشارت النتائج إلى أن المايكوفكس (MY) كان فعالا في تقليل التأثير السلبي لسم الافلا على أداء النمولافراخ فروج اللحم (زيادة وزن الجسم واستهلاك العليقة ونسبة تحويلها) وعلى الأوزان النسبية لأعضاء الجسم الداخلية المتأثرة و آفات الكبد وكذلك على صورة الدم (WBCs ، HB والجلوكوز) وانزيمات المصل (AST، ALT و ALP) ومعيار الأجسام المضادة ومعامل اختلافها ضد لقاح مرض نيوكاسل مقارنة مع المجاميع المستهلكة لسم الافلا .

EFFICACY EVALUATION OF INACTIVATED LOCAL VACCINE AGAINST NEWCASTLE DISEASE IN BROILERS

Abstract

Locally prepared vaccine, precipitated with alum and sodium hydroxide (IAPSHV) in embryonated chicken eggs. Different programs were carried out to evaluate its efficacy. Two hundred and fifty broiler chicks were divided equally into five groups. Different vaccination programs were used a follow. G1: vaccinated with live attenuated vaccine (B1) by intraocular route at 1, 10, 17 and 27 days of age. G2: vaccinated with live attenuated vaccine (B1) by intraocular route at 1 day of age and vaccinated also with (IAPSHV) at 1 day of age. G4: Vaccinated with (IAPSHV) only, by subcutaneous route at 1 day of age. G5: Control group left with out any vaccine. Indirect (ELISA) and Haemagglutination inhibition tests (HI) were used for evaluation of antibodies titer before and after vaccination, challenge test also used with virulent strain of Newcastle disease virus. Results showed that G3 gave the highest titer by both ELISA and (HI) test and low mortality in challenge test. It was concluded that (IAPSHV) at 7 day of age with attenuated live vaccine at the same age gave the best result than other groups. The applied program, and (IAPSHA) vaccine is recommended.

حضر محلياً لقاح مبطل مرسب بالشب وهيدروكسيد الصوديوم (IAPSHV ) في بيض مجنن, أتبعت برامج مختلفة للتلقيح لغرض تقيم كفاءته. قسم 250 فرخة دجاج لحم إلى خمسة مجاميع متساوية وأتبعت البرامج المختلفة الآتية: م1: لقحت بلقاح حي مضعف (B1 ) بطريقة التلقيح بالعين في الأعمار1و10و17و27 يوم. م2: لقحت بلقاح حي مضعف (B1 ) بطريقة التلقيح بالعين بعمر يوم واحد فضلاً عن لقاحIAPSHV تحت الجلد بعمر يوم واحد. م3: لقحت بلقاح حي مضعف (B1 ) بطريقة التلقيح بالعين بعمر 7 أيام ولقاح IAPSHV بعمر 7 أيام كذلك. م4: لقحت بلقاح IAPSHV تحت الجلد فقط بعمر يوم واحد. م5: مجموعة سيطرة بدون أي تلقيح. أستخدم فحص ايلايزا غير المباشر وفحص اثباط التلازن الدموي لتقييم معيار الأضداد قبل وبعد التلقيح كما نفذ فحص التحدي بعتر ضارية لفايروس نيوكاسل. أظهرت النتائج بأن م3 أعطت أعلى معيار للأضداد في اختباري ايلايزا واختبار اثباط التلازن الدموي وأقل هلاكات في اختبار التحدي. يستنتج بأن التلقيح بلقاح IAPSHV بعمر سبعة أيام مع لقاح حي مضعف بنفس العمر أعطى أفضل النتائج من بقية برامج المجاميع وهو البرنامج واللقاح المرشح.

AN INVESTIGATION ON LIGHT COLOR AND STOCKING DENSITY ON SOME PRODUCTIVE PERFORMANCE OF BROILERS

Abstract

This study was designated to investigate the effect of light color and stocking density on some productive performance of broilers. A total of 675 Ross 308 one-dayold broiler chicks were used in this study. The birds were exposed to white light (WL) as a control , red light (RL), blue light (BL), green light (GL), and Blue – Green mix light (BGL) by a light-emitting diode system (LED) applied for 24 hours daily in separated rooms. The birds were randomly divided and housed into 9 wooden sealed pens of 1m2in three replicates for each density 12, 15 and 18 birds/m2in the room. The results showed that the broilers reared under BGL significantly increased (P< 0.05) body weight (BW) and weight gain (WG) at 1st, 3rdand 5thweek. Obviously, stocking density had no significant effect on BW and WG at 1stweek but significantly increased under 12 birds/m2at 3rdand 5thweek. Feed intake (FI) recorded a significant increase (P< 0.05) in broilers reared under WL at 1stweek, but no statistical difference was found at 3rdweek whereas, FI significantly increased (P< 0.05) in broilers reared under BL at 5thweek. The stocking density affected FI values which were significantly differed (P< 0.05) at 1st, 3rdand 5thweek under 12 birds/m2compared with other densities. The results of the study revealed a significant increase (P< 0.05) of Feed conversion ratio (FCR) in broilers reared under BL at 1stweek and RL at 3rdweek but there was no significant difference between groups at 5thweek. The results indicated a significant difference of FCR value in 18 birds/m2at 1st week but there were no significant differences at 3rdand 5thweek at different densities. In conclusion, the results of this study indicated that chickens reared under five different color lighting schedules with three bird densities showed that broilers reared under mixing blue – green light under 12 birds/m2has a significant positive effects on production performance compared with other light programs and bird densities

Abstract

The present study was carried out in order to demonstrate the effect of dietary supplementation of garlic and neomycin in reducing the experimental infection of broilers with Salmonella typhimurium &the improvement of productive, physiological parameters all over the duration 42 days.200 broiler chicks(Hubbard flex) were divided into four groups: T1 (negative control):chicks fed on diet without any addition without Salmonella challenge,T2:chicks fed on diet mixed with 5% garlic powder (at 3 day age) plus Salmonella challenge,T3:chicks fed on diet with neomycin 0.5% (at 3 day age) plus Salmonella challenge,T4 (positive control):chicks fed on diet free of any addition ,but challenged with Salmonella. The results showed that the addition of garlic powder to the diet of broilers caused significant reducing S.typhimurium infection resembling to that of neomycin addition both produced significant decreasing(P≤0.05) in the number of excreted Salmonella in the feces and the mortality rate in the treatments T2 and T3 in comparison with T4.The productive parameters showed significant increasing(P≤0.05)in body weight and weight gain inT1,T2,T3 when compared withT4 in spite of no significant differences in total food consumption and feed conversion ratio among the four treatment, on the other hand the blood parameters showed significant increasing(P≤0.05) in RBC count and PCV in T1,T2,T3 in comparison with T4, while the percentage of hemoglobin showed non-significant increasing in T1,T2,T3when compared with T4,and there are significant increasing in WBC count in T2,T3,T4 when compared with T1, also there are significant decreasing in the three treatments when compared with T4 in L/H ratio, while there was no significant difference in differential WBC between all the treatments, As conclusion, the results of our study had demonstrated the antibacterial activity of garlic and neomycin against the experimental infection with Salmonella via enhancement of resistance and immunity besides the improvement of the productive and blood parameters.

Abstract

This experiment was designed to evaluatethe thyroid gland activity (hyperthyroidism and/or hypothyroidism) on some blood parameters in broilers. Thirty, oneday old broiler chicks were divided randomly into three equal groups. First group (G1) was control, second group (G2) Thyroxine was added to the drinking water (1 mg/L; 4 wks) serves as hyperthyroidism chicks and third group (G3) carbimazole was added to the drinking water (0.1 % ; 4wks) serves as hypothyroidism chicks. The blood samples were collected from overnight fasting birds at 15 and 30 day of experiment for the determination of serum glucose, total cholesterol (TC), total protein, albumin, and insulin concentration. At the end of experiment birds in all groups administered glucose 2g/kg/B.W. to evaluated glucose tolerance test, after 30, 60 and 90 mintfrom the glucose load, a blood sample was collected to determine serum glucose and insulin concentration. The results revealed that a significant increase in serum glucose and cholesterol concentration was observed in G3 as compared to the G1 and G2groupsat day 15 and 30 of experiment,at that timea significant decreasedwas showed in G2 group at day 30 of experiment as compared with G1 and G3 groups. A sharp descending of insulin line was showed after 90 minute in G1 group as compared with G2 and G3 groups in glucose tolerance test. In conclusions, that the effect of thyroxine and carbimazole administration to induced hyperthyroidismand hyporthyroidismrespectively caused hypoglycemia and hypolipidemic effects,and the modulations in body components according to thyroid gland condition are at least partly interposed by modulation in the control of glucose via insulin hormones in broilers.

Using species-specific PCR technique to detect Toxoplasma gondii in broiler chickens

Abstract

Two groups of broiler chickens were used in this study. One was reared under typical conditions at the animal house of Veterinary Medicine College/Mosul University- Iraq, while the other group was reared under common commercial farm conditions. Fifty and 80 birds from the two respective groups were sacrificed at 49 days of age for detecting Toxoplasma gondii by using Species-specific PCR technique. Results of Latex agglutination test indicated, principally, that 29.3% and 49.2% of the serum samples were positive for the birds of both groups, respectively. Titer figures ranged between 1:20 to 1:320 where the highest value was 1:160 (39.3%) and the lowest was 1:20 (5.8%). Confirmation of 38 and 64 serum samples, using Latex agglutination test was performed by PCR technique, from the two respective groups of chickens. Of those, 8 samples from the college birds and 35 from the commercial farm birds were confirmed positive by giving band of 133 bp, according to specific primers designated on gene B1.Based on these results, pursuing the PCR technique is considered, so far, a most sensitive method for Toxoplasma gondii detection. Also, positive PCR results are counted on as an early marker for reactivation and useful means in monitoring therapies.

استخدمت في هذه الدراسة مجموعتين من فروج اللحم بعمر 49 يوم للكشف عن المقوسة الكوندية باستخدام تقنية تفاعل البلمرة المتسلسل، تم ذبح 50 دجاجة كانت مرباة تحت ظروف تربية مثالية في بيت الحيوانات التابع لكلية الطب البيطري في جامعة الموصل بينما ذبحت 80 دجاجة كمجموعة ثانية مرباة في حقول التربية التجارية. أظهرت النتائج أولا أن 29,3% و 49,2% كانت موجبة لاختبار تلازن اللاتكس في المجموعتين على التوالي. معايير الأضداد تراوحت بين 20:1 و 320:1 وكانت أعلى قيمة (39,3%) عند المعيار 160:1 وأقل قيمة (5,8%) عند المعيار 20:1. تم تأكيد النتائج الموجبة (38 و 64) لكلا المجموعتين من الدجاج في اختبار تلازن اللاتكس بواسطة تقنية تفاعل البلمرة المتسلسل، كانت ثمانية عينات من فروج الكلية و 35 عينة من دجاج الحقول موجبة بتفاعل البلمرة المتسلسل من خلال ظهور حزمة بوزن جزيئي 133 زوج قاعدي اعتمادا على البادئ الخاص المصمم على جين ب 1. اعتمادا على النتائج المستحصلة من تقنية تفاعل البلمرة المتسلسل لهذا تعتبر هذه الطريقة حساسة في الكشف عن المقوسة الكوندية.

Abstract

This study was conducted to evaluate the effect of Protexin® and levamisole as an immune modulator. A total of 125 unsexed (Ross 308) broiler chicks with an average initial body weight of 43g to all chicks at the age of one day were randomly divided into five equally treated groups (25 chicks per pen ).Group 1 was considered as control group fed with commercial ration without any additives, group 2 was fed with commercial ration with the addition (daily) of 1g protexin® /liter of drinking water 1- 10 days, 1g protexin /2 liters of drinking water 10-20 days and 1g protexin® /4 liters of drinking water 20 day- end of the study, group 3 was fed with commercial ration with the addition (each 3 days a week) of 1g protexin® /liter of drinking water 1- 10 days, 1g protexin® /2 liters of drinking water 10-20 days and 1g protexin® /4 liters of drinking water 20- end of the study, group 4 was fed with commercial ration with the addition of levamisole 10 µg/kg B.w. daily, group 5 was fed with commercial ration with the addition of levamisole 10 µg/kg b.w. 3 days a week. The experiment lasted for five weeks (35 days). Chicks were vaccinated against New castle disease (ND) on age ( 10,20,30) days , by drinking water method, also vaccinated against Infectious Bursal Disease (IBD) on age (14) days by drinking water method. At days 1, 12, 22 and 35 blood samples were collected to estimate the immune response by ELISA test for ND and at 35 day for IBD. The titer of antibody (Ab) increased significantly (P ≤0.05) against NDV and IBD in G2 (Protexin® daily) as compared with G1(control group) at the age of 22 and 35days.

Abstract

Seventy five broiler chicks were used to determine the immunization of in ovo inoculation or intramusclular (I/M) injection of sonicated oocyts (SO) antigen against E. tenella manifestation. The chicks were divided in to 3 equal groups compromising of 25 chicks each. The 1st group inoculated in ovo with SO of E. tenella and repeated by I/M injection at 21 days old. The 2nd group injected I/M with SO at day old and repeated at 21 days old, The birds of the 3rd group remains as control. All birds were challenge by 50,000 sporulated oocysts of E. tenella at day 28. No significant (P > 0.05) group difference was detected between the immunized and non-immunized groups for mean, WBC count, H/L ratio, lesion score, mortality and oocysts shedding until E. tenella challenge at day 28. After challenge test the in ovo and I/M immunized groups showed a great protective immunity against E. tenella infection documented by significantly (P < 0.05) reduced mortality, lesion score and decreased fecal oocyst shedding, compared with non-immunized group. It is concluded that in ovo and I/M immuinzation of sonicated oocyst stimulates a remarkable protection in broilers following E. tenella infection comparing with non-immunized groups. There was no significance difference between the two procedures of immunization.

Abstract

The aim of this study is to evaluate the possible protective effect of preparation which synthesized in Vitebsk State Academy of Veterinary Medicine adsorbent ((Toxicom)) 5gkg diet against the toxic effects of mixed mycotoxins in growing broiler chickens .Total 75 chicks, one week age, were divided into 5 treated groups, 15 birds for each. The first group (G1) fed a contaminated ration with mycotoxin and supplemented with (Toxicom) 5g/kg of diet and vaccinated with Infectious Bursal Disease (IBD) vaccine at 15 and 22 days of age. The second group (G2) was fed a ration contaminated with mycotoxin and vaccinated with IBD vaccine at 15 and 22 days of age and not supplemented with (Toxicom). The third group (G3) was fed an intact broiler ration and vaccinated with IBD vaccine at 15 and 22 days of age. The fourth group (G4) was only fed a contaminated ration with mycotoxins and not vaccinated. The fifth group (G5) was fed intact broiler ration and not vaccinated as a control group. The diet was naturally contaminated with many mycotoxins and analyzed by ELISA and the level of mycotoxins were as follows : Aflatoxin B1 0.001 mg/kg, Dezoxivalenol 1.24 mg/kg, Zearalenon 0.068 mg/kg, Ochratoxin 0.005 mg/kg, T2 toxin 0.09 mg/kg, Fuminisen B1 0.2 mg/kg. It was concluded that this preparation which synthesized in Vitebsk State Academy of Veterinary Medicine is protect chicken blood parameters in comparison with the other groups.
