Abstract

Background: Oral Lichen Planus (OLP) is a chronic inflammatory mucosal disease, presenting in various clinical formsWHO had regarded OLP as a precancerous conditions in 1978 because of its potential with cancer. Both antigenspecificand nonspecific mechanisms involved in the pathogenesis of OLP. Oral Squamous Cell Carcinoma (OSCC) isthe most common malignant neoplasm of the oral cavity representing more than 94% of oral cancer. It occurs indifferent sites and has many etiological factors. Cyclin Dl is a proto-oncogene which consider as the key protein inthe regulation of cell proliferation and its overexpression led to the occurrence and progression of malignanttumors.NF-KB p65 is a member ofNF-kB family of transcription factors that widely used by eukaryotic cells as aregulator of genes that control cell proliferation and cell survival, also plays a major role in inflammation. The aims ofthis study were to evaluate the immunohistochemical expression of Cyclin D1 & NF-KB p65 in OLP & OSCC & tocorrelate the expression of the studied markers with the clinicopathological findings and with each other.Materials and Methods: Fifty (50) formalin – fixed, paraffin – embedded blocks of both Oral Lichen Planus (25 cases) &Oral Squamous Cell Carcinoma (25 cases) were collected pro- and retrospectively were included in this study.Hematoxylin & Eosin stain was performed for each block for reassessment of histopathological examination. Animmunohistochemical staining was performed using anti Cyclin D1 and anti NF-KB p65 monoclonal antibodies.Results: Of twenty five OLPstudied cases , positive Cyclin D1 & NF-KB p65 expression was found in (84%) and (88%)ofthe cases respectively. For OSCC ,out of 25 studied cases ,positive Cyclin D1 & NF-KB p65 expression was observe in(88%) and (96%) of cases respectively. Statistically significant correlation between Cyclin D1immuno- reactivity andclinical presentation of OLP was found. Statistically significant correlation of Cyclin D1 immunoreactivity with tumorgrade andNF-kBp65 immunoreactivity with tumor stage in OSCC cases was found. Statistically a highly significantcorrelation between the expression of two studied markers in OLP and OSCC was found.Conclusion: A highly significant correlation was seen regarding the expression of both markers with each other,suggesting their cooperative role in the pathogenesis of OLP and OSCC

Abstract

Background: Odontogenic cysts include a group of osseodestructive lesions that frequently affect the jaws. Thosecysts could derive from odontogenic epithelium and occur in the tooth-bearing regions of the jaws. The aims of thisstudy were to evaluate the immunohistochemical expression of Cyclin D1 in Keratocystic Odontogenic Tumor,Dentigerous cyst and Radicular cyst in epithelium and connective tissue capsule.Materials and Methods: In this study, thirty formalin fixed paraffin embedded tissue blocks of Odontogenic cysts andTumor, consist of 14 Keratocystic Odontogenic Tumor, 8 dentigerous cysts and 8 radicular cysts were analyzedimmunohistochemically for the presence of Cyclin D1 proteins.Results: Strong to moderate expression of Cyclin D1 in epithelium was found in Keratocystic Odontogenic Tumorcases with positive cases percentage was (85.7%). Statistical significant differences (P<0.001) observed whencomparing the three lesions. Immunoreactivity of Cyclin D1 in stroma of Keratocystic Odontogenic Tumor was higherthan in dentigerous cysts and radicular cysts cases. However, the difference was not statistically significant (P<0.067).Conclusion: The results of this study propose that high expression rate of Cyclin D1 might be one of the reasons foraggressive behavior of Keratocystic Odontogenic Tumor and high recurrence rate

Abstract

Background: Cyclin D1 proto-oncogene is an important regulator of (G1 to S) phase progression in many differentcell types. The Aims of this study were to evaluate the immunohistochemical expression of Cyclin D1 inmucoepidermoid and adenoid cystic carcinoma of the salivary glands and to correlate the immunoexpression ofthis protein with the clinicopathological findings.Materials and methodsRetrospectively, twelve of archival formalin fixed paraffin embedded tissue samples of salivary Mucoepidermoid andfourteen blocks of adenoid cystic carcinomas obtained from the archives of the department of oral pathology /college of dentistry / Baghdad university, Al-Shaheed Ghazi hospital, were included in this study. Five micrometersections obtained and immunostained using monoclonal antibody against Cyclin D1. The immunoexpression wasdetected by the presence of brown stain in the nucleus of tumor cell. The proportion of cells that expressed the stainwas correlated with the clinicopathological data of the patients.Results: Cyclin D1 expression was found positive in all cases of MEC and AdCC localized in tumor cells. Nonsignificantstatistical relation (p=0.588) was detected regarding Cyclin D1 expression in both types of tumor.Significant relation was found with stage of AdCC (p=0.04) and non-significant concerning other clinicopathologicalparameters in both tumor types.Conclusion: Weak expression of Cyclin D1 in MEC and AdCC might be explained by Cyclin D1 does not represent anexclusive factor consequently; other factors might be involved in the proliferation, progression and metastasis of bothtumor types.

Abstract

Background: Hepatitis C infection is often asymptomatic but chronic infection with hepatitis C virus is associated with an increased risk for the development of fibrosis, cirrhosis, and hepatocellular carcinoma. Objective: To assess the expression of cyclin D1 in liver tissues of patients with chronic hepatitis C virus and related with different parameters such as gender, age, stage of fibrosis and histological active index.Patients and Methods: Twenty one formalin fixed, paraffin-embedded liver tissue blocks were included (16 males and 5 females), age range (15-60) years. They were collected from the archives of histopathology laboratories of Hepatology and Gastroenterology Teaching Hospital in Baghdad, Iraq, the present study done in department of Microbiology – College of Medicine – University of Diyala during the period from 1st August 2014 till 1st March 2015. All the samples are related to the period between 2009 to 2012. Histopathological sections were made for these liver biopsies and stained by hematoxylin and eosin stain for definitive diagnosis. The molecular detection of cyclin D1 in those tissue blocks were performed by using immunohistochemistry.Results: Distribution of 21 patients with chronic active hepatitis diagnosed by 3rd generation enzyme linked immunosorbant assay revealed that 16 were males are higher (76.19%) than females 5(23.8%). The mean age was 38 years, most infection occurs within age group 31-45 years. The majority of the liver cases were diagnosed to have histological active index was 5/18 total number was 10 cases. Among liver tissues, 5 out of 21 (23.8%) showed 1/6 and 2/6 stage of fibrosis. Expression of cycline D1 demonstrated that 16 cases was positive while 5 was negative. Statistical analysis revealed significant differences between expression of cycline D1 and age, grade, HAI, but not significant with stage of fibrosis. Conclusion: Cyclin D1 play important role in development and progression of hepatitis C virus, these findings support the concept that the cell cycle regulation may play a role in initiation and development of hepatocellular carcinoma; need to be confirmed by further large scale studies.

خلفية الدراسة: يعد التهاب الكبد الفايروسي النمط ج أحد الأمراض المعدية التي تؤثر في المقام الأول على الكبد، الناجمة عن فيروس التهاب الكبد الوبائي. العدوى غالبا ما تكون غير متناظرة. ولكن العدوى المزمنة بفيروس التهاب الكبد الفايروسي النمط ج ترتبط بزيادة خطر التعرض لتليف الكبد وتطورة الى سرطان الكبد.الهدف من الدراسة: لتقييم التعبير Cyclin D1 في أنسجة الكبد من المرضى الذين يعانون من التهاب الكبد فايروسي النمط ج المزمن وعلاقته مع عوامل مختلفة مثل الجنس والعمر ومراحل التليف ومؤشر نشط النسيجي.المرضى وطرائق العمل: ادراجت واحد وعشرون عينه مثبته بالفورمالين ومطمورة بشمع البرافين من المرضى المصابين بالتهاب الكبد الفايروسي النمط ج المزمن بعد تشخيصهم باستخدام الجيل الثالث من اختبار ارتباط الخميرة للامتزاز المناعي ، 16عينه كانت من ذكور و 5 عينات كانت من الإناث، الفئة العمرية تراوحت مابين (15-60) سنة. تم جمعها من أرشيف مختبر الانسجة المرضية في مستشفى الكبد والجهاز الهضمي في بغداد، العراق، العمل الحالي تم انجازة في فرع الاحياء المجهرية – كلية الطب – جامعة ديالى خلال الفترة من 1 أغسطس 2014 حتى 1 مارس 2015. جميع العينات تعود للفترة ما بين عام 2009 إلى عام 2012. حضرت المقاطع النسيجية من الخزع وصبغت باستخدام صبغتي الهيماتوكسلين والايوسين لتشخيص النهائي كما تم إجراء الكشف الجزيئي عن التعبير Cyclin D1 في جميع العينات النسيجية باستخدام الطريقة الكيميائية المناعية النسيجية.النتائج: كان توزيع المرضى الذين يعانون من التهاب الكبد المزمن باستخدام الجيل الثالث من اختبار ارتباط الخميرة للامتزاز المناعي, ان 16 من العينات كانت للذكور (76.19٪) بينما كانت 5 (23.8٪) تعود للاناث. كان متوسط العمر 38 عاما، تحدث معظم العدوى داخل الفئة العمرية 31-45 سنة. تم تشخيص الغالبية العظمى من حالات الكبد لديهم مؤشر نشط النسيجي وكان 5/18 عدد 10 حالات. بين أنسجة الكبد، 5 من أصل 21 (23.8٪) وأظهرت الدراسة ان 1/6 و 2/6 مرحلة التليف. اظهرت الدراسة ان ان 16 حالة كانت ايجابية التعبير cycline D1 في حين كانت 5 سالبة. وكشف التحليل الإحصائي بوجود فروق ذات دلالة إحصائية بين التعبير عن D1 cycline والعمر، الجنس،النشط النسيجي ، ولكن لم تكن كبيرة مع مرحلة التليف.الاستنتاجات : التعبير Cyclin D1 يلعب دورا هاما في تطور وتقدم الاصابة ب فيروس التهاب الكبد النمط ج وهذه النتائج تدعم فكرة أن تنظيم دورة الخلية قد تلعب دورا في بدء وتطوير سرطان الكبد. تحتاج إلى تأكيد من إجراء المزيد من الدراسات على نطاق واسع.
